Prostaglandin $F_{2a}$에 의한 유우난소질환의 치료효과 (Therapeutic Effect of Prostaglandin $F_{2a}$ on Cows with Ovarian Disease)

  • A total of 600 Holstein cows in Chonnam province were examined to make a diagnosis on the ovarian diseases. By clinical signs and rectal examinations, 120 cows were founded to have ovarian diseases such as 40 cows with suboestrus, 40 cows with persistent corpus luteum, 40 cows with luteal cysts. The Prostaglandin $F_{2{\alpha}}$ (PG $F_{2{\alpha}}$) was administered intramuscularly and intradermic vaginally ; also intraovarian injections and intrauterine infusions was made. Comparisons were made on the effect of PG $F_{2{\alpha}}$ on the ovarian diseases. The results obtained were summarized as follows. 1. Induction of normal estrum after PG $F_{2{\alpha}}$ treatment was 75.0~82.5%(mean 78.3%). The conception rate was 83.3~84.8% (mean 84.0%) and average services per conception was 1.6~l.8(mean 1.7). Days from treatment to estrum was 3.9~4.3(mean 4.1) days. 2. Average conception rate was different according to the routes of administration: intraovary 91.7%, intra-uterus 88.0%, intramuscle 80.9% and intradermic vagina 75.0%, respectively.

Extracellular Prostaglandin $E_2$ Upregulation Effect of the Methanol Extract of Artemisia argyi

  • Since 15-hydroxyprostaglandin dehydrogenase (15-PGDH) is the key metabolic enzyme of prostaglandin $E_2$ ($PGE_2$), inhibition of 15-PGDH is supposed to facilitate various physiological functions by increasing $PGE_2$. Methanol extract of Artemisia argyi (AAME) inhibited 15-PGDH ($IC_{50}$: $13.13{\mu}g/mL$) with relatively low cytotoxicity ($IC_{50}$: $415.00{\mu}g/mL$) and elevated extracellular $PGE_2$ levels in HaCaT cells. Real-time PCR analysis showed that AAME decreased significantly mRNA expression of PG transporter (PGT) in HaCaT cells. These results indicate that AAME could be applicable to functional materials as a 15-PGDH inhibitor and PGT expression inhibitor for the upregulation of extracellular $PGE_2$ level.

과배란유도방법에 따른 난포액내의 Prostaglandin $E_2$$F_{2{\alpha}}$ 농도의 변동에 관한 연구 (Prostaglandin $E_2$ and $F_{2{\alpha}}$ Concentrations in Follicular Fluid after Different Ovarian Hyperstimulation Methods)

  • Follicular fluid(FF) prostaglandin $E_2$(PG$E_2$) and $PGF_{2{\alpha}}$ levels were compared in 3 groups of spontaneously ovulatory women undergoing ovulation induction with clomiphene citrate (CC) alone or with human menopausal gonadotropin(hMG) (14 patients), hMG(9 patients), or pure FSH/hMG(11 patients) for the purpose of in vitro fertilization. FF volume aspirated did not differ significantly according to the maturity of the oocyte. According to hyperstimulation regimens, the volume of FF from which preovulatory occytes were obtained was significantly less in the hMG-treated group than in the other groups. In follicles of preovulatory oocytes, FF PG$E_2$ values were significantly lower in the FSH treated group than in the Cc.treated or hMG-treated group, and FF $PGF_{2{\alpha}}$ values were significantly higher in the hMG-treated group than in the CC-treated or FSH-treated group. In follicles of immature or atretic oocytes, there was no significant difference in FF PG$E_2$ and PG$F_{2{\alpha}}$ concentrations of the similar morphology of the oocyte according to hyperstimulation regimens. In all cycles, FF PG$E_2$ and PG$F_{2{\alpha}}$ values of preovulatory oocytes were not significantly different from those of immature oocytes, but those of atretic oocytes were relatively lower than those of fertilizable oocytes and it was statistically signifincant in PG$E_2$ values of CC-treated group. In all treatment groups, FF PG$E_2$ and PG$F_{2{\alpha}}$ levels did not show and close relationship with the success of fertilization in vitro and of pregnancy after embryo transfer. Above results suggested that FF PG$E_2$ and PG$F_{2{\alpha}}$ be involved in oocyte maturation and ovulation, but their relationship with the success of in vitro fertilization was not found.

Monitoring the Sonographic Ovarian Dynamics and Pregnancy Rate in Cyclic Murrah Buffalo Cows Synchronized with Prostaglandin F2α

  • The objective of this research work was to know ovarian dynamics and pregnancy rate of cyclic Murrah buffalo cows with induced estrus by administration of prostaglandin F2α (PGF2α) and timed artificial insemination (TAI) with frozen thawed semen. A total of 31 female buffaloes were selected for the study. The buffalos having matured CL observed by ultrasonography were given one intra muscular injection of cloprostenol 500 ㎍ and TAI was performed using frozen thawed semen of Indian Murrah buffalo bull. Results showed that 90.32% (significantly, at p < 0.05) cows explore the sign of heat after injection of PG and 67.85% (significantly, at p < 0.05) cows were become pregnant out of 28 inseminated (TAI) cows. In the 28 inseminated (TAI) cows, average number of smaller and larger size of follicles were non-significantly (p > 0.05) higher at day 3 post PG injection, but the medium size of follicles was nonsignificantly (p > 0.05) higher at PG injection. At day 3 post PG injection the diameter of follicles was significantly (p < 0.05) higher, but the diameter of CL was significantly (p < 0.01) lower compared at PG injection. At PG injection the diameter of largest follicle was non-significantly differences (p > 0.05) in between pregnant and non-pregnant cows. But at day 3 post PG injection it was significantly (p < 0.01) higher in pregnant cows compared to non-pregnant cows. The number of small, medium, and large follicles at PG injection and at day 3 post PG injection were non-significantly (p > 0.05) difference in between pregnant and non-pregnant buffalo cows. Finally, it is concluded that the CL was effectively regresses and induced the sign of heat in buffalo cows and after AI the cows were become pregnant with significant rate. The study will help to the veterinarian and researcher to know the efficacy of PG injection and AI for reproductive efficiency in buffalo cows.

한국 재래 산양에 있어서 Prostaglandin $F_{2\alpha}(PGF_{2\alpha})$의 투여가 호르몬 함량 및 난포의 발육에 미치는 영향 (The Effects of Prostaglandin $F_{2\alpha}(PGF_{2\alpha})$ Treatment on Hormone Concentrations and Follicular Development in Early Postpartum Korean Native Goats)

  • 본 실험은 한국재래산양에 있어서 분만 후 초기에 prostaglandin $F_{2{\alpha}}$(PG $F_{2{alpha}$)의 투여가 호르몬 농도 및 난포발육에 미치는 효과를 검토하기 위하여 수행되었던 바 그 결과는 다음과 같다. 1. 분만한 한국재래산양에 혈장중 PG $F_{2{\alpha}}$의 농도는 PG $F_{2{\alpha}}$처리 후 2일에 PG $F_{2{\alpha}}$처리군 (GR-1 및 GR-2)에서 각각 6.35$\pm$0.5 및 0.22$\pm$0.2pg/$m\ell$로서 저하한 후 점차 증가하여 4일 후 21.18$\pm$1.6 및 4.21$\pm$0.4 pg/$m\ell$로서 절정에 도달하였다. 2. 혈장중 PG $F_{2{\alpha}}$의 농도는 PG $F_{2{\alpha}}$ 처리 전 9.08$\pm$1.2 및 5.44$\pm$1.8 pg/$m\ell$였으며 처리후 농도는 다소 저하하였으나 처리 군간에 유의차는 없었다. 3. 혈장중 estradiol-l7$\beta$( $E_2$)농도는 PG $F_{2{\alpha}}$ 처리 후 2일까지 점차 감소한 후 3일에 증가하여 4일에 최대치에 도달하였다. 4. 혈장중 luteinizing hormone (나T)의 농도는 PG $F_{2{\alpha}}$ 처리 후 약간의 증가가 있었으나 유의한 차이는 없었다. 5. 혈장중 prolactin (PRL)의 농도는 PG $F_{2{\alpha}}$처리후 감소되었으나 저농도의 수준으로 남아 있었다. 6. PG $F_{2{\alpha}}$의 처리 후 소형 및 중형의 난포의 수는 거의 유사하였으나 4mm 이상 대형 난포의 수는 숫적로 증가하는 경향을 보였다. 이상의 결과로 보아 PG $F_{2{\alpha}}$ 및 estradiol-17$\beta$는 난포의 직경과 관계가 있다고 암시되며 PG $F_{2{\alpha}}$는 난포발육을 촉진하였다고 생각된다.

피부 각질세포에 대한 비파엽 에탄올 추출물의 PGE2 조절 작용과 상처치료 효과 (Prostaglandin E2 Up-regulation and Wound Healing Effect of the Ethanol Extract of Eriobotryae Folium in Human Keratinocyte)

  • Prostaglandin (PG) $E_2$ is an important mediator of skin wound healing without excessive scarring and gastric ulcer healing. However, $PGE_2$ has a short lifetime in vivo because it is metabolized rapidly by 15-hydroxyprostaglandin dehydrogenase (15-PGDH). Ethanol extract of Eriobotryae folium (EFEE) elevated intracellular and extracellular $PGE_2$ levels in HaCaT cells and inhibited 15-PGDH ($ED_{50}$ : $168.4{\mu}g/mL$) with relatively low cytotoxicity ($IC_{50}$ : $250.0{\mu}g/mL$). Real-time PCR analysis showed that mRNA expression of cyclooxygenase (COX)-1 and COX-2 enzymes were increased and prostaglandin transporter (PGT) was decreased in HaCaT cells by EFEE. Moreover, wound healing effect of EFEE ($168.4{\mu}g/mL$) was comparable to that of TGF-${\beta}1$ (300 pg/mL) as a positive control. These results demonstrate that EFEE may be valuable therapeutic materials for the treatment of $PGE_2$ level dependent diseases.

Effects of Kinematic Taping Therapy on Dysmenorrhea

  • The present study aimed to investigate the effects of kinematic taping therapy on menstrual distress, pain, and $prostaglandinF2{\alpha}$. The experimental subject was a 24 years old woman with the pain of over 5 points on the dysmenorrhea measurement test and no unusual reactions on the taping test. The menstrual distress was measured by the Menstrual Distress Questionnaire (MDQ), and the dysmenorrhea was measured by the Visual Analogue Scale (VAS) before and after the intervention. The prostaglandin $F2{\alpha}(PGF2{\alpha})$ was measured on the first day of menstruation before the intervention and 24 hours after the taping therapy by the blood test. The intervention of kinematic taping was conducted by the instructor of the International Kinematic Taping Academy, and the taping was applied to the lower abdomen and the waist. According to the difference of menstrual distress before and after the intervention, the menstrual distress turned out to be decreased to 96 points after the intervention from the 115 points before the intervention. The dysmenorrhea also reduced 3 points on the VAS from 7 points to 4 points before and after the intervention. The $PGF2{\alpha}$ decreased from $26pg/m{\ell}$ to $20pg/m{\ell}$ before and after the intervention. Besides, the $PGF2{\alpha}$ decreased from $26pg/m{\ell}$ to $20pg/m{\ell}$ before and after the intervention. Results suggest that kinematic taping therapy could be useful to reduce the menstrual distress, pain, and $PGF2{\alpha}$.

Development of Highly Sensitive Analytical Method for Evaluation of Evening Primrose Oil's Enhancing Effect in Prostaglandin E1(OP 1206) Biosynthesis

  • This study aimed to develop and validate highly sensitive determination method of a prostaglandin ($PGE_1$, OP 1206) in human plasma by LC-MS/MS using column switching. Plasma stored at $-30^{\circ}C$ and treated with methanol effectively inhibited interferences synthesized post-sampling. Samples were added with internal standard and were separated by reversed-phase HPLC with a cycle time of 30min. The method was selective for OP 1206 and the regression models, based on internal standard, were linear across the concentration range 0.5-50 pg/mL with the limit of quantification of 0.5 pg/mL (limit of quantitation, LOQ) for OP 1206. The calibration curve of OP 1206 standards spiked in five individual plasma samples was linear ($r^2=0.9999$). Accuracy and precision at the concentrations of 0.5, 1.5, 5.0 and 40 pg/mL, and at the lower LOQ of 0.5 pg/mL were excellent at 20%. OP120 < 6 was stable in plasma samples for at least 24 hours at room temperature, 24 hours frozen at $-70^{\circ}C$, 24 hours in an auto sampler at $6^{\circ}C$, and for two freeze/unfreezing cycles. The validated determination method successfully quantified the concentrations of OP 1206 in plasma samples from simulated administrating a single $5{\mu}g$ OP 1206 formulation. Thus, this novel LC-MS/MS technique for drug separation, detection and quantitation is expected to become the standard highly-sensitive detection method in bioanalysis and to be applied to many low dose pharmaceutical products.

초치료 및 난치 폐결핵 환자의 투베르쿨린 피부검사와 혈장 Prostaglandin $E_2$ (Tuberculin Skin Test and Plasma Prostaglandin $E_2$ In Patients of New and Intractable Pulmonary Tuberculosis)

  • 연구배경: 결핵균의 제거에는 세포성 면역이 필요하며, Prostaglandin(PG) 특히 PG $E_2$가 세포성 면역억제에 관여 한다고 알려져있다. Indomethacin은 체내에 증가된 PG $E_2$를 억제시킨다고 한다. 이에 항결핵 화학요법으로 치유가 불기능한 난치 폐결핵 환자들에게 Indomethacin을 면역조절제로서의 적용 기초로 삼고자 투베르쿨린 피부 검사와 혈장 PG $E_2$치를 측정 하였다. 방법: 객담 항산성 도말검사에 양성인 폐결핵 환자 48명을 대상으로 하여 결핵 치료 과거력(초치료와 난치), 흉부 정면 방사선사진(경증과 중증)과 투베르쿨린 피부검사(음성과 양성)로 6군으로 분류하였다. 이 6군 중 1군(2명; 경증, 투베르쿨린 피부검사 음성반응)을 제외하고 방사선 면역방법으로 혈장 PG $E_2$치를 측정하였다. 결과: 1) 초치료 환자의 혈장 PG $E_2$치는 경증과 중증, 투베르쿨린 피부검사의 음성과 양성에 따른 유의한 차이는 없었다. 2) 난치 환자의 혈장 PG $E_2$치는 투베르쿨린 피부검사의 음성과 양성에 따른 유의한 차이는 없었다. 3) 초지료 환자군과 난치 환자군의 혈장 PG $E_2$치를 비교하였을때 난치 환자군에서 통계학적으로 유의하게 낮았다. 4) 투베르쿨린 피부검사의 음성과 양성에 따른 혈장 PG $E_2$치의 유이한 차이는 없었다. 5) 남자와 여자에 따론 혈장 PG $E_2$치의 유의한 차이는 없었다. 6) 10대, 20대, 30대, 40대에 따른 혈장 PG $E_2$치의 유의한 차이는 없었다. 7) 투베르쿨린 피부 검사 경결의 크기와 혈장 PG $E_2$치 간의 상관관계는 없었다. 결론: 이상의 결과로서 난치 환자들의 혈장 PG $E_2$치가 저지들의 예상과는 달리 초치료 환자보다 높지 않았고 병소의 크기, 투베르쿨린 피부검사 경결의 크기와 혈장 PG $E_2$치간 관련이 없었기에 난치 환자들에게 Indomethacin을 면역조절제로 적용하는것에 대한 더 광범위한 연구가 필요할 것으로 사료된다.

  • Prostaglandins (PGs) and Leukotrienes (LTs) have been implicated in the genesis of pulpal and periapical inflammation. In this study, the relationships among $PGE_2$, 6-keto-PG $F_1{\alpha}$ (a stable metabolite of $PGI_2$) and $LTB_4$ concentrations in inflamed pulp and periapical lesions were discussed. Pulp tissue were obtained in routine endodontic treatment and periapical lesions in periapical surgery after clinical diagnoses were made. These specimens were divided into four groups as normal pulp group (Control group), acute pulpitis group, chronic pulpitis group, and periapical lesion group. Pulp tissue and periapical lesions were stored in liquid nitrogen. The concentration of $PGE_2$, $PGI_2$ and $LTB_4$ were measured with ELISA. The data were analyzed by one-way ANOVA. Significantly higher levels of $PGE_2$, 6-keto-PG $F_1{\alpha}$ a and $LTB_4$ were found in acute pulpitis group than chronic pulpitis group and periapical lesion group(p<0.05). Periapical lesion group showed significantly higher mean concentrations of $PGE_2$ and $LTB_4$ than chronic pulpitis group. In control and chronic pulpitis group, significant higher levels of $PGI_2$ than $PGE_2$ and $LTB_4$ were found. These results suggested that the high levels of $PGE_2$ and $LTB_4$ in periapical lesions may be due to rich endothelium., fibroblast and lymphocyte known as the main producers of $PGE_2$ and $LTB_4$. $PGI_2$ may be thought to one of the most abundant PGs in normal pulp tissue.
