Facile Synthesis of 2-(p-Methylallylaminophenyl)propionic Acid (Alminoprofen) (2-(p-메틸알릴아미노페닐)프로판산의 합성)

  • A new method for the synthesis of alminoprofen, which is a non-steroidal antiinflammatory agent, was described. Ethyl 2-phenyl-propionate(4) was prepared by Friedel-Crafts reaction of benzene with ethyl ${\alpha}-chloro-{\alpha}(methylthio)acetate(1)$, followed by methylation and desulfurization of the resultant ethyl 2-(methylthio)phenylacetate(2). Ethyl 2-(p-aminophenyl)propionate(6) was obtained by nitration of (4) and successive reduction of ethyl 2-(p-nitrophenyl)propionate(5). Alminoprofen was synthesized by reaction of (6) with methallyl chloride, followed by hydrolysis of the resultant ethyl 2-(p-methylallylaminophenyl)propionate (7).

Waste Activated Sludge for Start-up Seed of Thermophilic Anaerobic Digestion (고온 혐기성 소화공정의 start-up seed로서의 호기성 폐 활성슬러지 이용가능성 연구)

  • Since there are very limited numbers of thermophilic anaerobic digesters being operated, it is often difficult to start up a new one using sludge from an existing reactor as a seed. However, for obvious reasons it seems few attempts have been made to compare the start-up performance of thermophilic anaerobic digestion using different sources of seed sludges. The purpose of this study was to evaluate the start-up performance of anaerobic digestion using aerobic Waste Activated Sludge (WAS) from a plant and mesophilic Anaerobic Digested Sludge (ADS) as the seed source at both mesophilic ($35^{\circ}C$) and thermophilic ($55^{\circ}C$) temperatures. In this study, two experiments were conducted. First, thermophilic anaerobic reactors were seeded with WAS (VSS = 4,400 mg/L) and ADS (VSS = 14,500 mg/L) to investigate start-up performance with a feed of acetate as well as propionate. The results show that WAS started to produce $CH_4$ soon after acetate feeding without a lag time, while ADS had a lag time of 10 days. When the feed was changed to propionate, WAS removed propionate down to below the detection limit of 10 mg/L, while ADS removed little propionate and produced little $CH_4$. Second, in order to further compare the methanogenic activity of WAS and ADS, both mesophilic and thermophilic reactors were operated. WAS acclimated to anaerobic conditions shortly and after acclimating it produced more $CH_4$ than ADS. WAS at mesophilic temperature biodegraded acetate at the same rate as for thermophilic. However WAS at mesophilic temperature biodegraded propionate at a much faster rate than at thermophilic. WAS as the seed source of anaerobic digestion resulted in much better performance than ADS at both mesophilic and thermophilic temperatures for both acetate and propionate metabolism.

Selction of Useful Chemicals Reducing Soybean -Sprout Rot (콩나물 부패경감에 유용한 약제 선발)

  • This study was carried out to select useful chemicals to control pathogens inciting soybean-sprout rot. Pathogen strains were isolated from decaying soybean seedlings. The isolated strains were identified as Pseudomonas spp. (Pse. strains) through microbiological test, however, no rot-inciting fungus was isolated. Eight food additives were tested with different concentrations in controlling pathogens and harmfulness. Five chemicals(over 5% cone.) including sodium hypochlorite apparently inhibited the growth of Pse. strains on media plate, however, sodium hypochlorite was discarded due to its severe germination inhibition. Propionic acid and acetic acid inhibited the growth of Pse. strains more effectively than calcium propionate and sodium propionate relatively. As no harmful effect on seed germination and no growth retard of soybean sprouts were observed by those chemicals with lower concentration(0.5∼1.0%), these chemicals were considered to be applicable to sprout rot control judged by the effectiveness and permissible concentration as food additives.

  • Flourination of Ethyl, ${\alpha},{\beta$}-dichloro-${\beta$}-phenyl propionate, Ethyl ${\alpha},{\beta$}-dibromo ${\beta$}-phenyl propionate, Ethyl ${\alpha},{\beta$}-dichloro ${\beta$}-(p-chloro-phenyl) propionate, and dibromostyrene by potassium fluoride were investigated in presence of dimethyl formamide, diethylene glycol, and diethylene glycol monomethyl-ether. The reactivity of these organic halogen esters and hydrocarbons towards potassium fluoride was checked further by means of radioactive fluorine-18. tracer. Generally, the reaction gave monofluoride together with dehalogenated olefin. The formation of olefine was increased when the reaction was done at high reaction temperature in presence of diethylene glycol, whereas the lower reaction temperature in presence of diethyleneglycol monomethyl ether favored the formation of mono fluoride in a good yield. The procedures and methods of the identification of monofluorides were described and the feasibility of this reaction of fluorine containing ester including the F-18 labelled compounds was discussed.

  • Backgroung : The efficacy of oral corticosteroids in the treatment of chronic asthma is undisputed, but their long-term use is associated with adverse side-effects, including supression of the hypothalamic-pituitary adrenal axis function, osteoporosis, weight gain, hypertension and impaired glucose tolerance. The introduction of inhaled corticosteroids in the early 1970's represented a significant therapeutic advance in the management of asthma, since these compounds combined high topical potency with low systemic activity. Fluticasone propionate is a new topically active synthetic glucocorticosteroid that combinds a high degree of efficacy with negligible systemic bioavailability. This study was perfomed to determine the effect of inhaled fluticasone propionate on the adreocortical supression in patients with bronchial asthma or chronic obstructive pulmonary disease. Method : The adrenocortical function was assessed by measurement of plasma cortisol concentration at 8 o'clock in morning and free cortisol in 24 hour urine collection at interval. Absolutely, no steroid was taken during pretreatment period of 10days. There after each subject inhaled fluticasone aerosol, in daily doses of 500 or 1000micrograms for 12days. The dose was delivered by metered dose inhaler(MDI). Results : The serum cortisol and 24hour urinary free cortisol were not decreased during the treatment period in patients with inhaled fluticasone propionate in daily doses of 500 micrograms. In contrast, serum cortisol was significantly decreased on 9th and 12th day(p less than 0.05). And, 24hour urinary free cortisol was also significantly decreased on 3rd and 12th day of treatement period(p less than 0.05) in patients with inhaled fluticasone in daily doses of 1000 micrograms. Conclusion : These results suggested that endogenous cortisol secretion was not supressed after short-term inhalation of fluticasone in daily dose of 500 micrograms, but in daily dose of 1000 micrograms, the endogenous cortisol secretion was supressed.

The Effect of Substrates and Nitrate on Biological Phosphorus Release (생물학적 인 방출시 유기물 및 질산염에 대한 영향)

  • In this study, effects of substrates and nitrate on biological phosphorus release in EBPR(enhanced biological phosphorus removal) process were examined using batch test apparatus at anaerobic conditions. The sludge used in this experiments was taken from SBR(sequencing batch reactor) treating swine wastewater at aeration period. Phosphorus release rates obtained with substrates of FSW(fermented swine wastewater), acetate, propionate, domestic wastewater and methanol were 6.19, 5.99, 1.52, 1.2 and $1.03mgP/gVSS{\cdot}hr$, respectively. Those observed with acetate and FSW were 4~5 times greater than those with propionate, methanol and domestic wastewater. Therefore phosphorus release rates were significantly affected by type of substrate added at anaerobic condition. Phosphorus release was greatly affected by concentration of nitrate in anoxic condition. Comparing to acetate, propionate and FSW, phosphorus release was observed after almost completely depletion of nitrate concentration with methanol and domestic wastewater added as substrate. In the cases supplied with acetate, propionate and FSW, phosphorus release rates were less influenced by a nitrate concentration than those with methanol and domestic wastewater.

Behaviors of Pollutants and Microorganisms in an Anaerobic Digestion of Propionate Containing High Ammonia Nitrogen Level (고농도 암모니아성 질소를 함유한 프로피온산의 혐기성 분해시 오염물질 및 미생물 거동)

  • Behaviors of simple organic compound and granular sludge in an upflow anaerobic sludge blanket (UASB) reactor treating propionate at high ammonia nitrogen levels were investigated for 12 months. The UASB reactor achieved about 80% removal of chemical oxygen demand (COD) at ammonia nitrogen concentration up to 6000 mg-N/L. At higher concentration of ammonia nitrogen, the propionate in the effluent increased whereas the acetate was very low. At ammonia nitrogen concentration of 8000 mg-N/L, the volatile suspended solids (VSS) increased sharply due probably to the decrease of the content of extracellular polymer (ECP) although methane production was very low. The specific methanogenic activity (SMA) using formate, acetate, and propionate as substrate to granules decreased as ammonia nitrogen concentration increased. The ammonia nitrogen concentration $I^{50}$, causing 50% inhibition of SMA were 2666, 4778 and 5572 mg-N/L, respectively. The kinetic coefficients of ammonia inhibition using formate, acetate, and propionate as substrate were 3.279, 0.999 and 0.609, respectively. The SMA using formate was severely affected by ammonia nitrogen than those using acetate and propionate. This result indicated that the hydrogenotrophic methanogens was most affected by ammonia nitrogen. Granules were mainly composed of microcolonies of methanothrix-like bacteria resembling bamboo-shape, and several other microcolonies including propionate degrader with juxtapositioned syntrophic associations between the hydrogen-producing acetogens and hydrogen-consuming methanogens.

Clinical Characteristics of Patients after Aryloxyphenoxy Propionate Herbicide Ingestion (Aryloxyphenoxy propionate 계열 제초제 중독환자의 임상 양상)

  • Purpose: No studies have been conducted to investigate the acute toxicity of aryloxyphenoxypropionate herbicides in humans following ingestion. Therefore, this study was conducted to investigate the clinical characteristics of aryloxyphenoxypropionate herbicide poisoning and provide guidance for physicians treating patients who have ingested these types of herbicides. Methods: A retrospective observational case series was conducted using ten patients with history of aryloxyphenoxy propionate herbicide. Data were collected for clinical manifestation, management and final outcome. Results: The most common symptoms were gastrointestinal irritation and an altered mental state (Glasgow Coma Scale<15). An elevated lactate level was a common laboratory abnormality, and prolonged QTc interval was commonly observed. These clinical features normalized within one day of supportive treatment. Conclusion: The acute toxicity of aryloxyphenoxypropionate herbicides in humans is manageable with supportive treatment. However, physicians should take into account depressed consciousness, the possibility of arrhythmia, and an elevated lactate level when planning their treatment strategy.

Effect of Inhaled Fluticasone Propionate on Hypothalamic-Pituitary-Adrenal Axis in Bronchial Asthma Patient (기관지 천식환자에서 Fluticasone Propionate 흡입제가 Hypothalamic-Pituitary-Adrenal Axis에 미치는 영향)

  • Background : Inhaled glucocorticoids are the medical treatment of choice in asthma patients. Fluticasone propionate is one of the most effective inhaled corticosteroids and has been reported to have minimal effect on the hypothalamic-pituitary-adrenal axis at the recommended dose. However, reports of long-term trials characterizing their systemic safety with chronic use are rare. This study was designed to evaluate the long-term safety of inhaled fluticasone propionate to the hypothalamic-pituitary-adrenal axis. Method : This study was conducted on 21 patients to evaluate the adrenal response to rapid ACTH stimulation test after 6 months of treatment with fluticasone propionate from $200\;{\mu}g$ to $750\;{\mu}g$ daily. The serum cortisol levels was measured to assess its effect on the hypothalamic-pituitary-adrenal axis just prior to the injection, at 30 minutes and 60 minutes after an intramuscular injection of synthetic ACTH. Result : The mean dose of inhaled fluticasone propionate was $355\;{\mu}g$ per day(SD=$174\;{\mu}g$, range=$200\;{\mu}g$ to $750\;{\mu}g$). The mean serum cortisol levels of the patients was $11.0\;{\mu}g/d{\ell}$(SD=$6.4\;{\mu}g/d{\ell}$) prior to the injection, $20.0\;{\mu}g/d{\ell}$ (SD=$7.7\;{\mu}g/d{\ell}$) after 30 minutes, and $23.0\;{\mu}g/d{\ell}$(SD=$6.3\;{\mu}g/d{\ell}$) after 60 minutes. Sixteen patients of the 21 patients had a normal response(> $18\;{\mu}g/d{\ell}$), and 5 out of the 21 patients had serum cortisol levels below the normal range after the rapid ACTH stimulation test. Conclusion: Adrenal suppression occurred in 5 out of 21 patients with 6 months treatment with inhaled fluticasone propionate.
