Serological Approach for Selection of Bradyrhizobium japonicum Strain with Superior Symbiotic Effectiveness (Bradyrhizobium japonicum의 공생효과(共生效果) 우수균주(優秀菌株) 선발(選拔)을 위한 면역혈청학적(免疫血淸學的) 접근(接近))

  • Symbiotic effectiveness and significance of Bradyrhizobium japonicum strains in five serogroups which were consisted of three corresponding serotype strains, respectively, indigenous to Korean soils were evaluated in terms of utility of strain serogroup for symbiotic improvement on soybean plant. The nodulation by rhizobia of each serogroup on four soybean cultivars(Danweonkong, Kwangkyo, Pangsakong, and Eunhakong) was good in order of USDA 123 > YCK 150 > YCK 117 > YCK 141 > USDA 110 serogroup members. Shoot dry weight of soybean was relatively high with USDA 110 serogroup members as well as with YCK 141 serogroup members, whereas the effectiveness of USDA 123 serogroup members was the lowest among the serogroups examined. In particular, Pangsakong among soybeans inoculated with five-serogroup members was positively outstanding on nodulation and shoot dry weight of the plant. Overall, symbiotic parameters of serogroup members associated with soybean plant such as nodule number, nodule mass, $N_2$ase activity, and shoot dry weight showed significantly different responses at level of 1% probability among both rhizobial serogroups and soybean cultivars, respectively. The rate in symbiotic similarity of the members of each serogroup from F-test ($$P{\leq_-}0.05$$) was 100% for nodule No., 90% for $N_2$ase activity. and 80% for soybean shoot dry weight. Taken together, the results indicated that the serological grouping of B. japonicum could be strongly useful for improving the symbiotic effectiveness hetween soybean and Rhizobium.

Application of Hot Air for Shelf-Life Extension of Peach(Prunus persica L. Batsch) (열풍 처리 복숭아의 저장 중 품질 변화)

  • To improve their shelf-life, domestic peaches were treated with hot air($46^{\circ}C$) for 0~9 hour, and the spoilage rate, changes of physico-chemical and sensory properties were investigated. The control showed spoilage at day 4, and then 50% of control showed spoilage on day 6. However, 12.5% of samples treated with hot air($46^{\circ}C$) for 6 hr showed spoilage on day 4. After 6 days, the spoilage rates of peaches treated with hot air treated for 3~9 hr were 16.7, 25.0, and 25.0%, respectively. Weight change of control was higher than that of hot air-treated samples for 6 days storage. There were no differences in pH of samples among the treatments, but pH increased with storage time. Hardness of hot air-treated samples was higher than that of control in the initial stage of storage. And then hardness of control sample was decreased faster than that of the hot air-treated samples for 6 days of storage. No significant changes in lightness and redness of the samples were observed after hot air treatment. Changes in yellowness of control occurred faster than that of hot air-treated samples during storage. The sensory parameters including taste, flavor, color, texture and overall acceptance at initial period were not different among treatments. The scores for taste, flavor, texture and overall acceptance of control decreased faster than those of hot airtreated samples during storage.

Effects of Leucine on in Vivo Protein Synthesis in Skeletal Muscles of Fed and Food-Deprived Rats (Leucine이 정상 또는 굶게 된 쥐의 골격근육의 단백질 생합성에 미치는 영향)

  • In vivo effects of leucine on skeletal muscle protein synthesis in fed and I-day food deprived young rats were examined. Animals assigned to leucine group were given a single i.p. injection of 80 or 160flmoles of leucine while control group animals were saline sham injected. The rate of protein synthesis was measured by the amount of $^{14}\textrm{C} incorporated into muscle protein after a single injection of $^{14}\textrm{C}-tyrosine, IO$\mu$ Ci/l00g B.W. Examined muscles were two different types of hind limb muscles. the oxidative solues and the glycolytic EDL and plantaris. Administered leucine elevated the concentration of free leucine in soleus muscles by 4-6.8 times the normal level. A massive dose of leucine, 160 flmoles, stimulated protein synthesis in the EDL and plantaris by 24 %, 29 % respectively of straved rats. The soleus of I-day food deprived rats and both types of muscles in fed rats did not respond to the injected leucine. The synthesis rate of the EDL and plantaris was supressed to one-half of the normal while the soleus that was not stimulated by leucine maintained a relatively normal rate, 78 %, of protein synthesis after I-day of food deprivation. Thus, in vivo stimulatory effect of leucine appears to be not a general phenomenon but to be related to the degree of catabolic condition developed by stress such as food deprivation. Although anabolic effects of leucine observed in this study was limited, any applicability of this special property of leucine to human subjects for the purpose of protein sparing in skeletal muscles remains to be examined.

Preparation and biological evaluation of 99mTc tricarbonyl cysteine (테크네슘-99엠 트리카보닐 시스테인의 제조 및 생물학적 특성 평가)

  • This paper describes the development of $^{99m}Tc$ tricarbonyl cysteine as potential renal function diagnostic radiopharmaceutical and evaluation of its biological characteristics using experimental animals. l-Cysteine was labeled efficiently with $^{99m}Tc$ tricarbonyl precursor $([^{99m}Tc(CO)_3(H_2O)_3)]^{+})$ under 30 min heating at ${75^{\circ}C}$. Labeling yield and stability were analyzed by high performance liquid chromatography (HPLC). The biodistribution property of $^{99m}Tc$ tricarbonyl cysteine in mice and its dynamic imaging profiles in rabbits were carried out. To investigate the excretion mechanism of $^{99m}Tc$ tricarbonyl cysteine, tubular transport inhibition test with probenecid was adopted. $^{99m}Tc$ tricarbonyl cysteine was obtained with a high labeling yield under the moderate condition. The results of biodistribution experiments of $^{99m}Tc$ tricarbonyl cysteine in ICR mice at 3 and 90 min provided that $^{99m}Tc$ tricarbonyl cysteine was very highly accumulated in the kidney and bladder, thereby almost 99% of $^{99m}Tc$ tricarbonyl cysteine was excreted within 90 min post injection. The same results were confirmed by the whole body dynamic images for 30 minutes and static images in rabbits at given time intervals after injection. Renogram of $^{99m}Tc$ tricarbonyl cysteine in rabbits showed that its $T_{max}$ and $T_{1/2}$ of $^{99m}Tc$ tricarbonyl cysteine were $2.33{\pm}0.56$ and $4.30{\pm}0.79$ min, respectively. The $T_{max}$ of $^{99m}Tc$ tricarbonyl cysteine with probenecid pretreatment was $2.30{\pm}0.17$ min, whereas $T_{1/2}$ of that with probenecid pretreatment was $17.0{\pm}32.47$ min. $T_{1/2}$ of $^{99m}Tc$ tricarbonyl cysteine with probenecid pretreatment was significantly different, as compared to the result without probenecid (p<0.0001). The results showed that the excretion of $^{99m}Tc$ tricarbonyl cysteine was extremely affected by probenecid. Therefore, $^{99m}Tc$ tricarbonyl cysteine was rapidly excreted from the kidney principally by the tubular secretion.

An Experimental Study on the Influence of High Quality Fly ash and Water-Binder Ratio on Properties of the Ternary System Concrete (3성분계 콘크리트의 특성에 미치는 고품질 플라이애쉬의 치환율 및 물-결합재비 영향에 관한 실험적 연구)

  • Recently the press and institute recognized fly ash as it had excellent performance. Its research and applications are on the rise largely as a substitute for cement. On the contrary, it is in a situation that the regulation of high quality fly ash remains at a low level. Accordingly, this study was to establish 8000 class of fineness of fly ash and three levels of substitute like 15%, 3 0%, and 45% in order to analyze the replacement ratio and effect of water-binder ratio for fly ash that affected the properties of ternary system concrete. As a result of experiment by planning water-binder ratio for two levels like 40% and 50%, it increased the fluidity in a fresh state, and it decreased the air content. This study has found out the setting acceleration and reduction of heat of hydration. As for the strength property in a set state, this study has shown the tendency of being equal or higher in age 28 days.

  • Beans are acknowledged to be food resources, which have more abundant proteins and fats. The constituent parts of beans (i.e. aspartic aid, glycine, arginine) are effective against diabetes, and dietary fiber contained in the beans has an important property to maintain insulin sensitivity. Based on these, using streptozotocin (STZ)-induced diabetic rats, this study examined how the rat-eye soybean, which is principal products of the Imsil province, is effective to attenuate and/or prevent the development of diabetes mellitus. We divided rats into the non-diabetic and diabetic group, and diabetic group was further subdivided into six experimental groups [DC, diabetic control; DI, diabetes with insulin treatment (4∼6 IU/rat); DB, diabetes with black bean; DY, diabetes with yellow soybean; DS, diabetes with rat-eye soybean; DSS, diabetes with vinegar-fermented rat-eye soybean. All bean treatment (1.5 mg/l g body weight).]. Food efficiency ratio (FER), body weight and insulin sensitivity in diabetic rats were significantly reduced compared to those in normal control animals. These reductions were obviously attenuated by administration of a variety of beans used in this study (20∼30%), and the recovery effects were comparable to the results obtained by insulin treatment. Taken together, this study suggests that all beans used may have an essential property to improve and/or attenuate the development of diabetes mellitus in rats.

Effect of Incorporation Times of Green Barley and Hairy Vetch on Rice Yield in Paddy Soil with Liquid Pig Manure (돈분액비를 시용한 녹비보리 및 헤어리베치의 혼입시기가 벼 수량에 미치는 영향)

  • BACKGROUND: Soil incorporation of green manure crop(GMC) and liquid pig manure(LPM) is one of the methods for reduction of chemical fertilizer and the increase of crop yield. The objective of this study was to select optimal incorporation time of GMCs on growth and nutrient property in paddy soil treated LPM. METHODS AND RESULTS: The kinds of GMCs were Hordeum vulgare L.(green barley, GB) and Vicia villosa roth(hairy vetch, HV). The effects of GMCs on rice yield were investigated under different incorporation times of GMCs(LPM1: at 25 days before rice transplantation, LPM2: at 18 days before rice transplantation, LPM3: at 11 days before rice transplantation). In GB treatments, the biomass was greater in the order of $$LPM3{\geq_-}LPM2{\geq_-}LPM1$$. Contents of N, P and K ranged 1.21~1.28, 0.36~0.38 and 1.41~1.45%, respectively, regardless of incorporation times. The amounts of nutrient supply in GB treatments were higher in LPM1 than those in other treatment conditions. In GB treatments, rice yields in LPM1, LPM2 and LPM3 were 523, 525 and 526(increasing yield 3% than control) kg/10a, respectively. In HV treatments, the amounts of nutrient supply were higher in the order of $$LPM3{\geq_-}LPM2{\geq_-}LPM1$$. Rice yields were 530 kg/10a for LPM1, 531 kg/10a for LPM2, 535 (increasing yield 5% than control) kg/10a for LPM3 in HV treatments, respectively. CONCLUSION(s): The optimum incorporation time of green barley and hairy vetch was at 11 days before rice transplantation(LPM3) in paddy soil with liquid pig manure.

Analysis of the Physicochemical Properties and Antioxidative Activity of Napa Cabbage Pickle (저장기간 동안 배추 피클의 이화학적 특성 및 항산화 활성 분석)

  • The principal objective of this fundamental research was to analyze the physicochemical properties the and antioxidative activity of Napa cabbage pickle (NCP) for development of low-salt pickles. NCP-1 was a smaller than NCP at amount soy sauce (10%). The pH of NCP and NCP-1 were $4.51{\pm}0.15$, $4.85{\pm}0.08$ immediately after preparation. The pH of NCP and NCP-1 was reduced to $4.08{\pm}0.05$ and $4.31{\pm}0.12$ over time during 60 days of storage. The acidity of the NCP and NCP-1 immediately after preparation were 0.51% and 0.38% and increased to 0.67% and 0.56% after 60 days of storage. The salinity for the NCP ranged from 1.71-2.22% and NCP-1 ranged from 1.18-1.63%. The L value, which indicates the lightness, was the highest at day 0 and the lowest at 60 day. The tensile strength value of NCP was $10.9{\pm}0.05kgf/cm^2$ and NCP-1 in $11.84{\pm}0.11kgf/cm^2$ at day 0 and then significantly decreased with time in storage. The cutting force of NCP was $1004{\pm}7.12gf/cm^2$ and NCP-1 in $845{\pm}5.27gf/cm^2$ at day 0, which increased over time in storage. The overall acceptability of NCP was the highest at day 30, but the overall acceptability of NCP-1 was the highest at day 45. NCP-1 extracts at day 60 showed the highest antioxidant activity of 66.04%, whereas the NCP extract at day 0 showed the lowest antioxidant activity of 45.41%. These results showed that depending on the content of the seasoning pickle difference in the antioxidant activity. Thus, the best pickled Napa cabbage is determined by a smaller amount soy sauce in NCP-1, and the results could provide a basis for improving the availability and quality of Napa cabbage.

Changes in the Quality of Peaches (Prunus persica L. Batsch) Treated by UV-C Irradiation during Storage (Ultraviolet-C 조사에 의한 복숭아의 저장 중 품질변화)

  • To prolong the shelf-life of domestic peaches, samples were treated with UV-C (0~10.0 $kJ/m^2$) and the spoiling rate and changes in physico-chemical and sensory properties were investigated. No spoiled peaches were observed within the first four days of storage in the control and 2.5 $kJ/m^2$ UV-C treatment groups. However, spoilage was observed in these groups on day six, and 29.17% and 25.0% of the samples showed spoilage on 10 day, respectively. Moreover, samples treated with greater than 5.0 $kJ/m^2$ of UV-C showed a higher percentage (41.67% or higher) of spoilage than those of the control or 2.5 $kJ/m^2$ UV-C treatment groups on 10 day. Additionally, weight changes in the peaches were the lower in the control group and 2.5 $kJ/m^2$ UV-C treatment group than in those treated with 5.0 $kJ/m^2$ of UV-C treatment or higher during 10 days of storage. There was no difference in pH among treatments, regardless of storage time. The hardness of the samples was not changed immediately after UV-C treatment, but that of samples treated with 5.0~10.0 $kJ/m^2$ of UV-C decreased rapidly after four days, when compared to the control and 2.5 $kJ/m^2$ UV-C treatment groups. No significant changes in the lightness and redness of the samples were observed in response to UV-C treatment, however, UV-C treatment led to a slight decrease in the yellowness of the samples. The initial taste, flavor, color, texture, and overall acceptance did not differ among control and UV-C treatments. The sensory score of the samples was the highest after 2 and 4 days of storage, while it decreased thereafter. In general, samples in the control and the 2.5 $kJ/m^2$ UV-C treatment groups showed higher sensory quality than those treated with UV-C at 5.0 $kJ/m^2$ or higher.

Changes of Peach (Prunus persica L. Batsch) Quality during Storage after Treatment with Aqueous Chlorine Dioxide (이산화염소수를 처리한 복숭아의 저장 중 품질 변화)

  • To prolong the shelf-life of domestic peaches, samples were treated with 30 ppm of aqueous chlorine dioxide ($ClO_2$) for 0~15 minute, after which the spoilage rate, changes in physico-chemical and sensory properties of treated samples were investigated. The control showed spoilage at day 4, and then 31.25% of control showed spoilage at day 8. However, samples treated with aqueous $ClO_2$ had no spoilage at day 4. On day 6, only 6.25% of samples treated with aqueous $ClO_2$ for 5 min showed spoilage. After 8 days, no spoilage was observed for samples treated with aqueous $ClO_2$ for 10 and 15 min. The weight change of the control was higher than that of aqueous $ClO_2$ treated samples for 8 days of storage. There were no differences in pH of samples among the treatments, but they were increased by storage time. Although there were no initial differences in the firmness of samples among the treatments, firmness of the control sample was decreased faster than those of the aqueous $ClO_2$ treated samples for 8 days of storage. No significant changes in lightness, redness and yellowness of the samples by aqueous $ClO_2$ treatment were observed during storage. The sensory parameters including taste, flavor, color, texture and overall acceptance at the initial period were not different among treatments. However, all scores of the control were decreased faster than those of aqueous $ClO_2$ treated samples during storage.
