간이비가림시설에서의 포도 내한성 발현 및 생육 특성 (Characteristics of Cold Hardiness and Growth of Grapevines Grown under Rain Shelter Type Cultivation System in the Vineyard)

  • 본 연구에서는 노지재배 포도의 내한성을 증대시킬 수 있는 방법을 모색하고자 간이비가림시설 설치에 따른 식물체의 반응을 조사하였다. 간이비가림시설에서 월동한 식물체의 탄수화물 함량은 노지 처리구에 비해 높았으며, 'Muscat Bailey A'의 줄기보다 '거봉'과 '캠벨얼리' 품종에서 탄수화물의 함량이 높게 나타났다. 노지에 비해 간이비가림시설에서 발아율과 신초의 생장도 양호하였으나 방충망, 차광막을 처리한 구에서는 유의차가 나타나지 않았다. 포도 잿빛곰팡이병의 발생은 간이비가림시설에서 발생이 적었다. Stilbene 화합물 함량은 노지보다 간이비가림 처리구에서 함량이 약간 더 높게 나타났으며 가장 많이 함유된 stilbene은 배당체 형태인 t-piceid였으며, resveratrol, piceatannol, c-piceid에 비해 상당히 고함량을 나타내었다. Chalcone isomerase, manganese superoxide dismutase, proline rich protein 2, temperature induced lipocalin의 transcripts는 시간이 지날수록 유전자의 발현량이 더 많이 축적되었고, 대체적으로 간이비가림시설의 삽수에서 저온 스트레스에 더 강하게 발현되었다. 처리구별 온도차이는 없었으나 노지에 비해 간이비가림시설에서 풍속이 크게 감소하였으며, 간이비가림시설 내에서 차광막을 설치한 경우 풍속의 감소효과가 현저하게 나타났다. 겨울철의 간이비가림시설과 방충망을 설치함으로써 극저온에 의한 포도나무의 저온피해를 줄일 수 있을 것이다.

Screening Differential Expressions of Defense-related Responses in Cold-treated 'Kyoho' and 'Campbell Early' Grapevines

  • Low temperature is one of the major environmental factors that affect productivity including reduced growth and budding of vines, and changes of metabolic processes in grape (Vitis spp.). To screen the specific expression of abiotic stress-related genes against cold treatment in 'Kyoho' and 'Campbell Early' grapevines, expression of various defense-related genes was investigated by RT-PCR and real-time PCR. Among the 67 genes analyzed by RT-PCR and real-time PCR, 17 and 16 types of cDNA were up-regulated, while 5 and 6 types were down-regulated in cold-treated 'Kyoho' and 'Campbell Early' grapevines, respectively. Genes encoding carotene (Cart3564 and Cart4472), chalcone isomerase (CHI), cytochrome P450 (CYP), flavonol synthase (FLS), endo-${\beta}$-glucanase precursor (Glu), glutathione peroxidase (GPX), glutathione-S-transferase (GST), leucine-rich repeats (LRR), manganese superoxide dismutase (Mn-SOD), phenylalanine ammonia lyase (PAL), polygalacturonase-inhibiting protein (PGIP), proline rich protein 2 (PRP2), small heat shock protein (sHSP), temperature induced lipocalin (TIL), and thaumatin-like protein (TLP) were up-regulated, while those encoding CBF like transcription factor (CBF1), chitinase-like protein (CLP), cold induced protein (CIP), glycerol-3-phosphate acyltransferase (GPAT), and mitogen-activated protein kinase (MAPK) were down-regulated by low temperature treatment in both in 'Kyoho' and 'Campbell Early'.

Interaction of Hepatitis C Virus Core Protein with Janus Kinase Is Required for Efficient Production of Infectious Viruses

  • Chronic hepatitis C virus (HCV) infection is responsible for the development of liver cirrhosis and hepatocellular carcinoma. HCV core protein plays not only a structural role in the virion morphogenesis by encapsidating a virus RNA genome but also a non-structural role in HCV-induced pathogenesis by blocking innate immunity. Especially, it has been shown to regulate JAK-STAT signaling pathway through its direct interaction with Janus kinase (JAK) via its proline-rich JAK-binding motif ($^{79}{\underline{P}}GY{\underline{P}}WP^{84}$). However, little is known about the physiological significance of this HCV core-JAK association in the context of the virus life cycle. In order to gain an insight, a mutant HCV genome (J6/JFH1-79A82A) was constructed to express the mutant core with a defective JAK-binding motif ($^{79}{\underline{A}}GY{\underline{A}}WP^{84}$) using an HCV genotype 2a infectious clone (J6/JFH1). When this mutant HCV genome was introduced into hepatocarcinoma cells, it was found to be severely impaired in its ability to produce infectious viruses in spite of its robust RNA genome replication. Taken together, all these results suggest an essential requirement of HCV core-JAK protein interaction for efficient production of infectious viruses and the potential of using core-JAK blockers as a new anti-HCV therapy.

한국인 성씨에 따른 이하선 탕개 내 Pr,Db,Pa의 유전자빈도에 관한 연구 (A Study of Gene Frequency of Proline-rich Protein, Double-band Protein and Pa Protien in Patotic Saliva according to Family Name of Korean People)

  • After dividing 372 Korean people of 47 different family names into 307 people of 28 indigenous family name groups and 65 people of 19 immigrated family name groups and investigating Pr. Db, Pa gene frequency of each family name groups based on phenotype of parotid saliva character the author have got following conclusions. 1. The gene frequencies of indigenous family name groups were Pr1=0.686, Pr2=0.314, Pr gene frequencies of immigrated family name groups were Pr1=0.7, Pr2=0.3. 2. The gene frequencies of indigenous family name groups were Db==0.021, Db-=0.979, Pr gene frequencies of immigrated family name groups were Db+=0.023, Db-=0.977. 3. The gene frequencies of indigenous family name groups were Pa+=0.248, Pa-=0.752, Pr gene frequencies of immigrated family name groups were Pa+=0.206, Pa-=0.794. 4. The Pr gene frequencies of immigrated family name groups were in the middle of those of Chinese people and indigenous people groups. 5. There was no significant difference of Db gene frequencies between indigenous and immigrated family name groups. 6. Pa gene frequencies of immigrated family name groups were similar to those of Chinese people.

Amino Acid Profiles of Tropical Legumes, Cooper (Glycine wightii), Tinaroo (Neonotonia wightii) and Siratro (Macroptilium atropurpureum), at Pre-blooming and Blooming Stages

  • An experiment was conducted to investigate amino acid composition of three tropical legumes (Cooper (Glycine wightii), Tinaroo (Neonotonia wightii) and Siratro (Macroptilium atropurpureum)) at two different stages (pre-blooming and blooming stages). Chemical composition and totally 16 amino acids of these plants were analysed for comparison of their composition among species at different growing stages and characterizing the amino acid pattern of these legumes. Crude protein content of the plants ranged from 16% to 27% on a dry matter basis. The total amount of 16 amino acids analyzed in this experiment was highest at 89.7 mg/16 g N in Cooper at pre-blooming and lowest at 80.9 mg/16 g N in Glycine at blooming stage. Total amount of amino acids in each legume species tended to slightly decrease with their maturity but no statistical difference was found. The percentage of aspartic acid, glutamic acid and proline in the total amount of amino acids was dominant at 9% to 13%, and that of methionine was less than 1.6%. In this experiment it was concluded that three tropical legumes were rich in crude protein content and characterized by 16 different amino acids with lower sulfur-containing amino acid as methionine.

Physicochemical Profiles of Chub Mackerel Scomber japonicus Bones as a Food Resource

  • The present study was conducted to characterize physicochemical properties of chub mackerel bones (CMB) to evaluate its potential as a food resource. The proximate composition of CMB showed 40.4% moisture, 13.8% crude fat, 15.2% crude protein, and 28.7% ash. The major minerals in CMB were calcium (26.27 g/100 g) and phosphorus (15.88 g/100 g). The amino acids were rich in glycine, proline, glutamic acid, and alanine. The contents of total and neutral lipids, glycolipid and phospholipid were shown to be 16.05%, 95%, 2.32%, and 3.15%, respectively. The major fatty acids were C22:6, C16:0, C18:1, C20:5, C18:0, C17:0, C14:0, C20:1 in order. The fatty acid contents of total and neutral lipid were in a range of 39.25% and 44.54% for saturated and 33.61% and 34.05% for polyunsaturated, respectively. The breaking strength and hardness of intact CMB were 10.01 and $50.03kgf/cm^2$, whereas those of CMB heated for 45 min at $121^{\circ}C$ were 0.40 and $1.94kgf/cm^2$, respectively.

유제품의 치매 및 인지기능 저하 예방 효과 (Preventive Effects of Dairy Products on Dementia and Cognitive Decline)

  • The prevention of cognitive decline and dementia is an increasingly important global public health priority due to an increase in the percentage of the elderly population. Dementia, a severe cognitive disorder, not only negatively impacts the patients' quality of life but also creates a substantial burden for caregivers. This review introduced recent advances regarding the protective effects of dairy product intake against dementia and cognitive decline. Recent epidemiological studies have suggested that specific components of dairy products including bioactive peptides, colostrinin, proline-rich polypeptides, α-lactalbumin, vitamin B12, calcium, and probiotics might promote healthy brain function during aging. Additionally, oleamide and dehydroergosterol in Camembert cheese have been suggested as agents capable of reducing microglial inflammatory responses and neurotoxicity. The intake of neuroprotective and anti-inflammatory compounds in meals is safe and easy, hence nutritional approaches, including dairy product consumption, serve as a promising intervention for the prevention of neurodegenerative disorders.

당뇨병 환자의 이하선 타액내 단백질의 다형현상에 대한 연구 (A Study on the Polymorphisms in Parotid Salivary Proteins of the Patients with Diabetes Millitus)

  • The purpose of this study was to evaluate the polymorphosms in parotid salivary proteins of the patients with diabetes mellitus. Saliva from the parotid glands was collected from 94 healthy Korean adults who were live in Kwang-ju and from 33 diabetes mellitus patients who had more than 140mg/dl of fastingblood sugar for one week. Diabetes mellitus patient group was subdivided to insulin dependent diatetes mellitus (IDDM) and non-insulin dependent diabetes mellitus (NIDDM). In the saliva collected from the parotid glands, parotid acidic protein(Pa), proline-rich protein(Pr) and double band protein(Db) were analyzed to evaluate the distribution of phenotype using alkaline slab polyacrylamide gel electrophoresis. The results were as follows : 1. The parotid acidic protein (Pa) was found more frequently in the diabetes mellitus patient group than in the control group, but the difference was not statistically significant. 2. The Pr(1-2) type was found more frequently in the control group, but the Pr(1-1) and Pr(2-2) type were found more freqnently in the diabetes mellitus patient group and the difference of phenotypic distribution was statistically significant between the two groups. (p<0.05) 3. The parotid acidic protein(Pa) and Pr(1-2) type were found more frequently in the noninsulin dependent diabetes mellitus (NIDDM) patients than in the insulin dependent diabetes mellitus patients, though the difference was not statistically significant.

Transcriptional profiles of Rhizobium vitis-inoculated and salicylic acid-treated 'Tamnara' grapevines based on microarray analysis

  • The transcriptional profiles of 'Tamnara' grapevine (Vitis labruscana L.) to Rhizobium vitis were determined using 12,000 gene oligonucleotide microarray chips constructed with 6,776 unigenes based on the EST sequencing. Among them, 95 clones were up-regulated more than three times and 90 were down-regulated more than 5-times in the R. vitis-inoculated grapevines relative to the control vines. Treatment of salicylic acid showed that 337 clones were upregulated and 52 clones were down regulated in grapevines. Microarray analysis, reverse transcription-polymer chain reaction, and slot blot hybridization analysis revealed that 5, 14, and 64 clones were up-regulated and 10, 12, and 61 clones were down-regulated in wounded, salicylic acid-treated, and R. vitis-inoculated 'Tamnara' grapevine leaves, respectively. The expression patterns of ${\beta}$-1,3-glucanase, proline-rich protein, and lipoxygenase genes of 'Tamnara' moderately resistant to R. vitis were similar to those of resistant 'Concord' and 'Delaware' grapevines. However, chalcone synthase genes in 'Tamnara' grapevines showed similar expression patterns to susceptible grapevines 'Neomuscat' and 'Rizamat'. Further expression studies with various clones for each gene should be conducted to elucidate their roles in resistant responses against pathogens or other stimuli in grapevines. These results could provide better resources for understanding the mechanism of defense responses against crown gall disease and clues for identifying new genes that may play a role in defense against R. vitis in grapevines.

Development and evaluation of semi-nested PCR for detection of the variable lipoprotein haemagglutinin (vlhA) gene of Mycoplasma Synoviae in chicken

  • This study aimed to develop a semi-nested polymerase chain reaction assay for the direct detection of Mycoplasma synoviae (M. synoviae) from clinical samples using three newly designed oligonucleotide primers specific to the variable lipoprotein haemagglutinin (vlhA) gene and differentiate M. synoviae field strains based on a nucleotide deletion or the insertion of the proline-rich repeat (PRR) region of the vlhA gene. The developed semi-nested polymerase chain reaction (PCR) assay revealed positive results in 12 out of 100 clinical samples collected from chickens showing lameness and joint swelling. Six positive samples were selected randomly for sequencing, and sequence analysis revealed 96.3-100% nucleotide identities compared to the reference sequences. Phylogenetic analysis showed that sequences of the strains in this study were closely related to WVU1853 (Spain), CK.MS.UDL.PK.2014.2 (Pakistan), and F10-2AS (USA) strains, but they were distinct from the M. synoviae-H vaccine strain sequence. M. synoviae obtained from these samples were identified as types A and C with a length of 38 and 32 amino acids, respectively. These results indicated that the specific and sensitive semi-nested PCR could be a useful diagnostic tool for the direct identification of clinical samples, and the sequence analysis of the partial vlhA gene can be useful for typing M. Synoviae.
