Molecular Genetics of Emericella nidulans Sexual Development

  • Many aspergilli that belongs to ascomycetes have sexuality. In a homothallic or self-fertile fungus, a number of fruiting bodies or cleistothecia are formed in a thallus grown from a single haploid conidia or ascospores. Genome-sequencing project revealed that two mating genes (MAT) encoding the regulatory proteins that are necessary for controlling partner recognition in heterothallic fungi were conserved in most aspergilli. The MAT gene products in some self-fertile species were not required for recognition of mating partner at pheromone-signaling stage but required at later stages of sexual development. Various environmental factors such as nutritional status, culture conditions and several stresses, influence the decision or progression of sexual reproduction. A large number of genes are expected to be involved in sexual development of Emericella nidulans (anamorph: Aspergillus nidulans), a genetic and biological model organism in aspergilli. The sexual development process can be grouped into several development stages, including the decision of sexual reproductive cycle, mating process, growth of fruiting body, karyogamy followed by meiosis, and sporulation process. Complicated regulatory networks, such as signal transduction pathways and gene expression controls, may work in each stage and stage-to-stage linkages. In this review, the components joining in the regulatory pathways of sexual development, although they constitute only a small part of the whole regulatory networks, are briefly mentioned. Some of them control sexual development positively and some do negatively. Regarding the difficulties for studying sexual differentiation compare to asexual one, recent progresses in molecular genetics of E. nidulans enlarge the boundaries of understanding sexual development in the non-fertile species as well as in fertile fungi.

Novel biological strategies to enhance the radiation therapeutic ratio

  • Successful anticancer strategies require a differential response between tumor and normal tissue (i.e., a therapeutic ratio). In fact, improving the effectiveness of a cancer therapeutic is of no clinical value in the absence of a significant increase in the differential response between tumor and normal tissue. Although radiation dose escalation with the use of intensity modulated radiation therapy has permitted the maximum tolerable dose for most locally advanced cancers, improvements in tumor control without damaging normal adjacent tissues are needed. As a means of increasing the therapeutic ratio, several new approaches are under development. Drugs targeting signal transduction pathways in cancer progression and more recently, immunotherapeutics targeting specific immune cell subsets have entered the clinic with promising early results. Radiobiological research is underway to address pressing questions as to the dose per fraction, irradiated tumor volume and time sequence of the drug administration. To exploit these exciting novel strategies, a better understanding is needed of the cellular and molecular pathways responsible for both cancer and normal tissue and organ response, including the role of radiation-induced accelerated senescence. This review will highlight the current understanding of promising biologically targeted therapies to enhance the radiation therapeutic ratio.

Toll-Like Receptor-Mediated Free Radical Generation in Clonorchis sinensis Excretory-Secretory Product-Treated Cholangiocarcinoma Cells

  • Clonorchiasis, caused by direct contact with Clonorchis sinensis worms and their excretory-secretory products (ESPs), is associated with chronic inflammation, malignant changes in bile ducts, and even cholangiocarcinogenesis. Our previous report revealed that intracellular free radicals enzymatically generated by C. sinensis ESPs cause NF-${\kappa}B$-mediated inflammation in human cholangiocarcinoma cells (HuCCT1). Therefore, the present study was conducted to examine the role of upstream Toll-like receptors (TLRs) on the initial host innate immune responses to infection. We found that treatment of HuCCT1 cells with native ESPs induced changes in TLR mRNA levels in a time-dependent manner, concomitant with the generation of free radicals. ESP-mediated free radical generation was markedly attenuated by preincubation of the cells with TLR1-4-neutralizing antibodies, indicating that at least TLR1 through 4 participate in stimulation of the host innate immune responses. These findings indicate that free radicals triggered by ESPs are critically involved in TLR signal transduction. Continuous signaling by this pathway may function in initiating C. sinensis infection-associated inflammation cascades, a detrimental event leading to progression to more severe hepatobiliary diseases.

Estimation of LOCA Break Size Using Cascaded Fuzzy Neural Networks

  • Operators of nuclear power plants may not be equipped with sufficient information during a loss-of-coolant accident (LOCA), which can be fatal, or they may not have sufficient time to analyze the information they do have, even if this information is adequate. It is not easy to predict the progression of LOCAs in nuclear power plants. Therefore, accurate information on the LOCA break position and size should be provided to efficiently manage the accident. In this paper, the LOCA break size is predicted using a cascaded fuzzy neural network (CFNN) model. The input data of the CFNN model are the time-integrated values of each measurement signal for an initial short-time interval after a reactor scram. The training of the CFNN model is accomplished by a hybrid method combined with a genetic algorithm and a least squares method. As a result, LOCA break size is estimated exactly by the proposed CFNN model.

Efficacy of Caffeine in Promoting Hair Growth by Enhancing Intracellular Activity of Hair Follicles

  • Caffeine is widely used in cosmetics and hair care products. Although its efficacy in stimulating hair growth has been confirmed in recent studies, its mechanism of action remains unelucidated. The present study aimed to determine the effects of caffeine on hair growth, with a focus on intracellular hair follicle activity. Experiments included in vitro and ex vivo tests, and a clinical study. Caffeine enhanced the cellular activity and potassium channel opening. It also promoted human hair follicle elongation. Immunohistochemical staining showed that the Ki-67 signal was significantly higher in cells treated with caffeine. These efficacies of caffeine were comprehensively demonstrated in clinical results, wherein caffeine-containing shampoo improved hair density after 24 weeks of testing. Collectively, the results of this study demonstrated that caffeine promoted hair growth and inhibited the progression of hair loss by enhancing intracellular activity of hair follicles.

  • Litorilituus sediminis is a Gram-negative, aerobic, novel bacterium under the family of Colwelliaceae, has a stunning hypothetical protein containing domain called von Hippel-Lindau that has significant tumor suppressor activity. Therefore, this study was designed to elucidate the structure and function of the biologically important hypothetical protein EMK97_00595 (QBG34344.1) using several bioinformatics tools. The functional annotation exposed that the hypothetical protein is an extracellular secretory soluble signal peptide and contains the von Hippel-Lindau (VHL; VHL beta) domain that has a significant role in tumor suppression. This domain is conserved throughout evolution, as its homologs are available in various types of the organism like mammals, insects, and nematode. The gene product of VHL has a critical regulatory activity in the ubiquitous oxygen-sensing pathway. This domain has a significant role in inhibiting cell proliferation, angiogenesis progression, kidney cancer, breast cancer, and colon cancer. At last, the current study depicts that the annotated hypothetical protein is linked with tumor suppressor activity which might be of great interest to future research in the higher organism.

키토산 기반 나노방출제어시스템의 세포주기진행 유전자 발현 증진 효과 및 기전 (Effect and mechanism of chitosan-based nano-controlled release system on the promotion of cell cycle progression gene expression)

  • 목적: 이전 연구에서 치은섬유아세포 혹은 성견 구개 연조직에 trichloroacetic acid (TCA)를 적용하는 것이 세포주기진행 유전자 발현의 변화를 일으키는 것으로 밝혀졌다. 이에 따라 본 연구에서는, hydrophobically modified glycol chitosan (HGC)기반의 나노방출제어시스템을 이용한 TCA 및 상피세포성장인자(EGF)의 순차적 방출시스템에서 이 효과를 검증하기 위하여 다양한 세포주기진행 유전자들의 발현을 규명하였다. 재료 및 방법: TCA와 EGF를 담지하는 HGC기반 나노방출제어시스템을 제작하였다. 실험군은 대조군(CON); TCA-담지형 나노방출제어시스템 투여군(EXP1); TCA- 및 EGF-담지형 나노방출제어시스템 투여군(EXP2)으로 정의되었다. 24시간 및 48시간 배양 시 37개 세포주기 유전자들의 발현을 분석하였다. 영향인자로서의 유전자 및 상관관계에 대해서도 분석하였다. 결과: Cyclins (CCNDs), cell division cycles (CDCs), cyclin-dependent kinases (CDKs), E2F transcription factors (E2Fs), extracellular signal-regulated kinases (ERKs)와 같은 다수의 유전자들과 기타 다른 세포주기 유전자들의 발현이 EXP1과 EXP2에서 상향조절되었다. E2F4, E2F5, GADD45G와 같은 세포주기차단 유전자들의 발현도 상향조절되었으나, 또다른 세포주기차단 유전자인 SMAD4의 발현은 하향조절되었다. 다중회귀분석에서 CCNA2, CDK4 그리고 ANAPC4가 ERK 유전자 발현에 가장 영향력 있는 유전자로 선정되었다. 결론: HGC기반 순차적 나노방출제어시스템을 이용한 TCA 및 EGF의 적용은 다양한 세포주기진행 유전자들의 발현을 상향조절함이 밝혀졌고, 이를 토대로 한 구강연조직증대시스템 개발의 가능성이 확보되었다.

입술 움직임 영상 선호를 이용한 음성 구간 검출 (Speech Activity Detection using Lip Movement Image Signals)

  • 본 논문에서는 음성인식을 위한 음성구간 검출과정에서 유입될 수 있는 동적인 음향에너지 이외에 화자의 입술움직임 영상신호까지 확인함으로써 외부 음향잡음이 음성인식 대상으로 오인식되는 것을 방지하기 위한 한 가지 방법이 제시된다. 우선, 연속적인 영상이 PC용 영상카메라를 통하여 획득되고 그 입술움직임 여부가 식별된다. 다음으로, 입술움직임 영상신호 데이터는 공유메모리에 저장되어 음성인식 프로세서와 공유한다. 한편, 음성인식의 전처리 단계인 음성구간 검출과정에서 공유메모리에 저장되어진 데이터를 확인함으로써 화자의 발성에 의한 음향에너지인지의 여부가 입증된다. 최종적으로, 음성인식기와 영상처리기를 연동시켜 실험한 결과, 영상카메라에 대면해서 발성하면 음성인식 결과의 출력에 이르기까지 연동처리가 정상적으로 진행됨을 확인하였고, 영상카메라에 대면치 않고 발성하면 연동처리시스템이 그 음성인식 결과를 출력치 못함을 확인하였다. 또한, 오프라인하의 입술움직임 초기 특정값 및 템플릿 초기영상을 온라인하에서 추출된 입술움직임 초기특정값 및 템플릿 영상으로 대체함으로써 입술움직임 영상 추적의 변별력을 향상시켰다. 입술움직임 영상 추적과정을 시각적으로 확인하고 실시간으로 관련된 패러미터를 해석하기 위해 영상처리 테스트베드를 구축하였다, 음성과 영상처리 시스템의 연동결과 다양한 조명환경 하에서도 약 99.3%의 연동율을 나타냈다.

Early Detection of hyperemia with Magnetic Resonance Fluid Attenuation Inversion Recovery Imaging after Superficial Temporal Artery to Middle Cerebral Artery Anastomosis

  • Objective : Cerebral hyperperfusion syndrome (CHS) manifests as a collection of symptoms brought on by heightened focal cerebral blood flow (CBF), afflicting nearly 30% of patients who have undergone superficial temporal artery (STA)-middle cerebral artery (MCA) anastomosis. The aim of this study was to investigate whether the amalgamation of magnetic resonance imaging (MRI) fluid-attenuated inversion recovery (FLAIR) and apparent diffusion coefficient (ADC) imaging via MRI can discern cerebral hyperemia after STA-MCA anastomosis surgery. Methods : A retrospective study was performed of patients who underwent STA-MCA anastomosis due to Moyamoya disease or atherosclerotic steno-occlusive disease. A protocol aimed at preventing CHS was instituted, leveraging the use of MRI FLAIR. Patients underwent MRI diffusion with FLAIR imaging 24 hours after STA-MCA anastomosis. A high signal on FLAIR images signified the presence of hyperemia at the bypass site, triggering a protocol of hyperemia care. All patients underwent hemodynamic evaluations, including perfusion MRI, single-photon emission computed tomography (SPECT), and digital subtraction angiography, both before and after the surgery. If a high signal intensity is observed on MRI FLAIR within 24 hours of the surgery, a repeat MRI is performed to confirm the presence of hyperemia. Patients with confirmed hyperemia are managed according to a protocol aimed at preventing further progression. Results : Out of a total of 162 patients, 24 individuals (comprising 16 women and 8 men) exhibited hyperemia on their MRI FLAIR scans following the procedure. SPECT was conducted on 23 patients, and 11 of them yielded positive results. All 24 patients underwent perfusion MRI, but nine of them showed no significant findings. Among the patients, 10 displayed elevations in both CBF and cerebral blood volume (CBV), three only showed elevation in CBF, and two only showed elevation in CBV. Follow-up MRI FLAIR scans conducted 6 months later on these patients revealed complete normalization of the previously observed high signal intensity, with no evidence of ischemic injury. Conclusion : The study determined that the use of MRI FLAIR and ADC mapping is a competent means of early detection of hyperemia after STA-MCA anastomosis surgery. The protocol established can be adopted by other neurosurgical institutions to enhance patient outcomes and mitigate the hazard of permanent cerebral injury caused by cerebral hyperemia.

Expression of Egr3 in mouse gonads and its localization and function in oocytes

  • Objective: The early growth response (Egr) family consists of four members (Egr1, Egr2, Egr3, and Egr4) that are zinc finger transcription factors. Among them, Egr3 is involved in transcriptional regulation of target genes during muscle spindle formation and neurite outgrowth. We previously showed that the immunoreactive Egr3 is localized on oocyte spindle and accumulate near the microtubule organizing center during meiosis I in mice. Egr3 was also shown to be localized on spermatocytes. We herein investigated if Egr3 is expressed in mouse gonads and if Egr3 blockade results in any defect in oocyte maturation. Methods: Expression of Egr3 in mouse gonads was examined by reverse transcription-polymerase chain reaction. Full-length Egr3 and truncated Egr3 (${\Delta}Egr3$) complementary RNAs (cRNAs) with Xpress tag at N-terminus and DsRed2 at C-terminus, and small interfering RNA (siRNA) targeting Egr3 were microinjected into mouse oocytes at germinal vesicle stage. Localization of microinjected Egr3 was examined by confocal live imaging and immunofluorescence staining. Results: Egr3 mRNA was detected in mouse ovaries and testes from 1 to 4 week-old mice. An uncharacterized longer transcript containing 5'untranslated region was also detected in 3 and 4 week-old gonads. Microinjected Xpress-Egr3-DsRed2 or Xpress-${\Delta}Egr3$-DsRed2 localized to nuclei and chromosomes during meiotic progression. Microinjection of these cRNAs or Egr3 siRNA in oocytes did not affect meiotic maturation. Immunofluorescence staining of Egr3 in Xpress-${\Delta}Egr3$-DsRed2-injected oocytes showed a positive signal only on meiotic spindle, suggesting that this antibody does not detect endogenous or exogenous Egr3 in mouse oocytes. Conclusion: The results show that Egr3 localizes to chromosomes during meiotic progression and that certain antibodies may not faithfully represent localization of target proteins in oocytes. Egr3 seems to be dispensable during oocyte maturation in mice.
