Silk Filament Progression with Backcross Breeding Generations in Tropical Tasar Silkworm, Antheraea mylitta D

  • Silk filament progression study applying backcross breeding with recipient parent Jata and donor parent Daba ecoraces of tropical tasar silkworm, Antheraea mylitta Drury conducted during 2006 to 2008, revealed introgression of filament denier (10.2 d) superior to both parents at BC4 level. Also, the silk waste (0.35%) and filament breaks (2.6 nos) were reduced compared to both parents, while the filament length (973 m) and non break filament (296 m) improved over donor and could attain closer to recipient parent. The best performance of Jata $\times$ Daba at F1 level, with highest silk filament length (1646 m) of 12.5 d denier, denotes heterosis impact on silk trait with parental heterogenousity, an advantage to exploit silk filament yield. The progression of quality in terms of finer filament denier, less silk waste and least number of filament breaks over both parents and improvement in filament length and non break filament over donor parent except for a marginal shortage against recipient at BC4 level indicates the prospects of Jata and Daba ecoraces as source of breeding material for qualitative improvement of tasar silk filament. The study suggests that the commercially important finer denier of tasar silk filament can be attained with minor reduction in silk yield of wild Jata ecorace by adopting repeated backcrossing for four generations with semi domesticated Daba ecorace.

MicroRNA-766-3p Inhibits Tumour Progression by Targeting Wnt3a in Hepatocellular Carcinoma

  • Recent studies have indicated that microRNAs (miRNAs) play an important role in hepatocellular carcinoma (HCC) progression. In this study, we showed that miR-766-3p was decreased in approximately 72% of HCC tissues and cell lines, and its low expression level was significantly correlated with tumour size, TNM stage, metastasis, and poor prognosis in HCC. Ectopic miR-766-3p expression inhibited HCC cell proliferation, colony formation, migration and invasion. In addition, we showed that miR-766-3p repressed Wnt3a expression. A luciferase reporter assay revealed that Wnt3a was a direct target of miR-766-3p, and an inverse correlation between miR-766-3p and Wnt3a expression was observed. Moreover, Wnt3a up-regulation reversed the effects of miR766-3p on HCC progression. In addition, our study showed that miR-766-3p up-regulation decreased the nuclear ${\beta}-catenin$ level and expression of Wnt targets (TCF1 and Survivin) and reduced the level of MAP protein regulator of cytokinesis 1 (PRC1). However, these effects of miR-766-3p were reversed by Wnt3a up-regulation. In addition, PRC1 upregulation increased the nuclear ${\beta}-catenin$ level and protein expression of TCF1 and Survivin. iCRT3, which disrupts the ${\beta}-catenin-TCF4$ interaction, repressed the TCF1, Survivin and PRC1 protein levels. Taken together, our results suggest that miR-766-3p down-regulation promotes HCC cell progression, probably by targeting the Wnt3a/PRC1 pathway, and miR-766-3p may serve as a potential therapeutic target in HCC.

모델링을 통한 과학영재 학생들의 메타모델링 지식 발달 단계 분석 (Analysis of Progression Levels for Meta-modeling Knowledge of Science Gifted Students through Modeling)

  • 이 연구에서는 모델링을 통해 드러난 학생의 과학 메타모델링 지식 발달 단계를 탐구하는데 목적이 있다. 이를 위해 5명의 영재학생들에게 양초연소와 관련된 모델링을 요구하였으며, 모델링의 모든 과정은 관찰되어지고 이후 개별 면담이 진행되었다. 연구결과, 2명의 학생이 1단계로 분류되었으며, 3명의 학생은 2단계로 분류되었다. 1단계 학생들은 모델의 생성, 모델을 이용한 예측 활동이 전혀 수행되지 않았으며, 그들에게는 관찰이 가장 의미가 있는 활동이었으며 이러한 관찰을 통해 모델을 생성하였다. 반면, 2단계의 학생들은 모델의 생성, 모델을 이용한 예측, 실험 수행 및 관찰, 예측과 자료의 비교를 모두 수행하였다. 하지만 모델의 생성과 이를 이용한 예측에서 상대적으로 강한 수행을 보였다. 그들은 실험에서 얻는 자료를 절대적인 모델을 확인하는 용도로만 인식하였다. 3, 4단계에 해당하는 학생은 관찰되지 않았다. 이러한 연구결과는 현재 영재학생들이 모델을 객관적 실체로 인식하는 단계에 머물러 있으며, 이들을 진정한 과학자로 양성하기 위해서는 영재학생들이 모델의 주관성을 인식할 수 있도록 돕는 교육이 필요함을 보여준다.

Gomisin G Suppresses the Growth of Colon Cancer Cells by Attenuation of AKT Phosphorylation and Arrest of Cell Cycle Progression

  • Colorectal cancer is one of the leading causes of cancer related death due to a poor prognosis. In this study, we investigated the effect of Gomisin G on colon cancer growth and examined the underlying mechanism of action. We found that Gomisin G significantly suppressed the viability and colony formation of LoVo cells. Gomisin G reduced the phosphorylation level of AKT implying that Gomisin G suppressed the PI3K-AKT signaling pathway. Gomisin G also induced apoptosis shown by Annexin V staining and an increased level of cleaved poly-ADP ribose polymerase (PARP) and Caspase-3 proteins. Furthermore, Gomisin G remarkably triggered the accumulation of cells at the sub-G1 phase which represents apoptotic cells. In addition, the level of cyclin D1 and phosphorylated retinoblastoma tumor suppressor protein (Rb) was also reduced by the treatment with Gomisin G thus curtailing cell cycle progression. These findings show the suppressive effect of Gomisin G by inhibiting proliferation and inducing apoptosis in LoVo cells. Taken together, these results suggest Gomisin G could be developed as a potential therapeutic compound against colon cancer.

보행 시 Wedged 인솔과 보행진행 각이 슬관절 외측 밀림(Lateral Thrust)에 미치는 영향 (The Influence of Wedged Insole and Foot Progression Angle on Lateral Thrust of Knee During Walking)

  • The purpose of this study was to identify the influence of wedged insole and foot progression angle (FPG) on lateral thrust of knee in healthy subjects. Fifteen healthy male subjects were recruited from Suncheon First College, in Suncheon. The subjects randomly walked at the comfortable velocity under five conditions: bare footed, medio-lateral $10^{\circ}$ wedged insoles, toe-in and toe-out gait. The lateral thrust was measured by a accelerometer with telemeter during walking. Data was collected while each subject walked for about 10 gait cycle on a flat, level walkway at their normal speed. The middle three gait cycle were used for averaged peak value of lateral acceleration. The three averaged peak value of lateral acceleration were collected under each condition at heel strike. The results showed that averaged peak value of lateral acceleration increased significantly in medial wedged insole and toe-in gait and decreased significantly in lateral wedged insole and toe-out gait as compared with bare footed (p<.05). These results suggest that wedged insole as well as walking strategy, such as foot progression angle, may prevent progression of degenerative knee osteoarthritis.

Iron-Saturated Lactoferrin Stimulates Cell Cycle Progression through PI3K/Akt Pathway

  • Iron binding lactoferrin (Lf) is involved in the control of cell cycle progression. However, the molecular basis underlying the effects of Lf on cell cycle control, as well as its target genes, remains incompletely understood. In this study, we have demonstrated that a relatively low level of ironsaturated Lf, Lf($Fe^{3+}$), can stimulate S phase cell cycle entry, and requires Akt activation in MCF-7 cells. Lf($Fe^{3+}$) immediately induced Akt phosphorylation at Ser473, which subsequently induced the phosphorylation of two G1-checkpoint Cdk inhibitors, $p21^{Cip/WAF1}$ and $p27^{kip1}$. The Lf($Fe^{3+}$)-induced phosphorylation of Cdk inhibitors impaired their nuclear import behavior, thereby inducing cell cycle progression. However, the treatment of cells with a PI3K inhibitor, LY294002, almost completely blocked Lf($Fe^{3+}$)-stimulated cell cycle progression. LY294002 treatment abrogated Lf($Fe^{3+}$)-induced Akt activation, and prevented the cytoplasmic localization of $p27^{kip1}$. Higher levels of $p21^{Cip/WAF1}$ were also detected in the cytoplasmic sub-cellular compartment as a measure of cellular response to Lf($Fe^{3+}$). Consequently, the degree of phosphorylation of retinoblastoma protein was enhanced in response to Lf($Fe^{3+}$). Therefore, we conclude that Lf($Fe^{3+}$), as a potential antagonist of Cdk inhibitors, can facilitate the functions of E2F during progression to S phase via the Akt signaling pathway.

Long-term assessment of periodontal disease progression after surgical or non-surgical treatment: a systematic review

  • The primary aim of this systematic review was to assess the evidence on periodontal disease progression after treatment in patients receiving supportive periodontal therapy (SPT) and to identify predictors of clinical attachment level (CAL) loss. A protocol was developed to answer the following focused question: In adult patients treated for periodontitis, what is the disease progression in terms of CAL loss after surgical or non-surgical treatment? Randomized controlled clinical trials, prospective cohort studies, and longitudinal observational human studies with a minimum of 5 years of follow-up after surgical or non-surgical treatment that reported CAL and probing depth changes were selected. Seventeen publications reporting data from 14 investigations were included. Data from 964 patients with a follow-up range of 5-15 years was evaluated. When the CAL at the latest follow-up was compared to the CAL after active periodontal therapy, 10 of the included studies reported an overall mean CAL loss of ${\leq}0.5mm$, 3 studies reported a mean CAL loss of 0.5-1 mm, and 4 studies reported a mean CAL loss of >1 mm. Based on 7 publications, the percentage of sites showing a CAL loss of ${\geq}2mm$ varied from 3% to 20%, and a high percentage of sites with CAL loss was associated with poor oral hygiene, smoking, and poor compliance with SPT. The outcomes after periodontal therapy remained stable over time. Disease progression occurred in a reduced number of sites and patients, mostly associated with poor oral hygiene, poor compliance with SPT, and smoking.

Serum CEA Level Change and Its Significance Before and after Gefitinib Therapy on Patients with Advanced Non-small Cell Lung Cancer

  • Objective: The aim of this study was to explore change and significance of serum carcino-embryonic antigen (CEA) before and after gefitinib therapy in patients with advanced non-small-cell lung cancer (NSCLC). Methods: Forty patients with advanced NSCLCs in III~IV stages were selected as study objects given gefitinib therapy combined with routine local radiotherapy until tumor progression or intolerable toxicity. After treatment, all patients were divided into control and non-control groups according to the results of evaluation based on RECIST 1.1 (Response Evaluation Criteria in Solid Tumors in 2009). Peripheral fasting blood from all patients was collected in the early morning and serum CEA was assessed by electro-chemiluminescence immunoassay (ECLIA) before and after treatment. Before treatment, patients were divided into high CEA group (CEA level > 50 ng/mL) and low CEA group (CEA level ${\leq}$ 50 ng/mL). Adverse reactions were noted and progression-free survival (PFS) in both groups was recorded after long-term follow-up that ended in December, 2012. Results: There was no difference between control and non-control groups in CEA level before treatment (P>0.05), whereas serum CEA decreased more markedly lower in the control group after treatment (P<0.01). All patients were divided into high CEA group (26) and low CEA group (14) according to serum CEA level. There was no statistically significant difference between two groups in adverse reactions (P>0.05) but the rate in former group was lower. Additionally, survival rates at 9 and 12 months in high CEA group were clearly higher than in the low CEA group (P<0.01). Conclusions: Serum CEA level can serve as a biochemical index to evaluate the prognosis with gefitinib treatment for NSCLC.

Serum Level of Mast Cell Tryptase in Patients with Oral Squamous Cell Carcinoma: Lack of Correlation with Clinicopathologic Factors

  • Background: Mast cells can influence tumor progression via different pathways and increased mast cell density has been demonstrated in oral squamous cell carcinoma (OSCC). It has been shown that the serum tryptase level is elevated with some malignant tumours and may thus be a useful parameter. However, there are no data available about OSCC. The main aim of this study was the evaluation of mast cell tryptase (MCT) level in OSCC patient serum. Materials and Methods: In this cross-sectional, analytic study, the circulating levels of MCT were assessed in sera of 55 OSCC patients and 34 healthy individuals with ELISA technique. Results: The serum MCT level in OSCC patients was 12-14 ng/ml, which was not significantly higher than the healthy control group. While the serum level of MCT was higher with larger tumours, there was no apparent correlation with clinico-pathological features such as patient age, gender, tumor location, stage, nodal status, distant metastasis, histological grade and smoking. Conclusions: Our findings showed that despite the results obtained from studies of other malignant tumors, serum level of MCT in OSCC patients could not be a credited as a reliable indicator of the presence or progression of tumours.

학습 발달과정 탐색을 통한 계절의 변화 교육과정 및 교수 계열 제안 (A Proposal of Curriculum and Teaching Sequence for Seasonal Change by Exploring a Learning Progression)

  • 이 연구에서는 학습 발달과정을 탐색을 통해 계절의 변화에 대한 교육과정과 교수 계열을 제안하고자 하였다. 학습 발달과정을 알아보기 위해 Wilson (2005)이 제안한 '구인 모델링 방식'의 4가지 단계(구인특화, 평가 문항 개발, 평가 결과 기술, 측정 모델)를 적용하였다. 구인특화 단계에서는 계절의 변화의 하부 구인으로 '계절에 따른 그림자의 길이', '계절에 따른 별자리의 위치', '남반구와 북반구의 계절', '계절에 따른 태양의 일주운동', '계절의 원인과 현상'을 설정하였으며, 선행 연구 결과를 바탕으로 각 하부 구인의 하위 정착점인 수준 1에서 상위 정착점인 수준 4까지를 나타낸 구인구성도를 작성하였다. 평가 문항 개발 단계에서는 구인구성도를 토대로 C-E (choose and explain) 형태의 문항 3개, CR (constructed response) 형태의 문항 2개로 구성된 총 5개의 평가 문항을 개발하였으며, 초, 중, 고등학교 학생 383명을 대상으로 적용하였다. 평가 결과 기술 단계에서는 평가 문항에 대한 학생 응답 결과를 구인구성도를 토대로 범주화하는 과정을 거쳤으며, 이 범주들을 학생 능력에 따라 4수준으로 분류하고 1-4점의 점수를 부여하였다. 측정 모델 단계에서는 Rasch 모델의 부분점수 모형을 적용하여 학생들의 응답 결과를 통해 작성한 학습 발달의 경로가 선행 연구를 기반으로 작성한 구인구성도와 일치하는지 비교하였다. 이를 통해 구인구성도를 수정함으로써 최종적으로 계절의 변화에 대한 가설적인 학습 발달과정을 작성하였다. 최종적으로, 연구 결과를 바탕으로 계절 변화에 대한 교육 과정 개정 방향과 효과적인 교수 계열을 제안하였다.
