Alcohol Production from Whey in Batch and Continuous Culture of Kluyveromyces fragilis.

  • In order to develop the whey beverage, we examined the optimum conditions for alcohol fermentation by Kluyveromyces tragilis ATCC 46537. The optimum conditions for alcohol production by K. fragilis ATCC 46537 were as follows; pH 4.5, $30^{\cir}C$, with a supplement of 50 g/l of lactose. To develop a continuous production of alcohol from whey, we compared batch fermentation with continuous iermentation in conjunction with UF system. Batch fermentation produced 11.0 g/l of alcohol, whereas pseudocontinuous and continuous fermentation with UF system produced 8.5 g/l of alcohol. To increase the alcohol production, we added 50 g/l of lactose to both fermentations. Batch fermentation with lactose supplement produced 15.7 g/l of alcohol and continuous fermentation with lactose supplement in conjunction with UF system produced 15.0 g/l of alcohol. These results clearly demonstrate that the UF system can be used to increase the alcohol production from whey, supplemented with exogenous lactose.

Treatment of Wastewater from Purified Terephtalic Acid (PTA) Production in a Two-stage Anaerobic Expanded Granular Sludge Bed System

  • The wastewater treatment with a two-phase expanded granular sludge bed (EGSB) system for anaerobic degradation of acetate, benzoate, terephtalate and p-toluate from purified terephtalic acid (PTA) production was studied. The feasibility and effectiveness of the system was evaluated in terms of organic oxidation by chemical oxygen demand (COD), gas production, bacterial adaptability and stability in the granular sludge. Average removal efficiencies 93.5% and 72.7% were achieved in the EGSB reactors under volumetric loading rates of $1.0-15kg-COD/m^3/day$ and terephtalate and p-toluate of 351-526 mg/L, respectively. Gas production reached total methane production rate of 0.30 L/g-COD under these conditions in the sequential EGSB reactor system. Higher strength influent COD concentration above 4.8 g-COD/L related to field conditions was fed to observe the disturbance of the EGSB reactors.

  • The effects of agitation and aeration for exopolysaccharide(EPS) production through batch cultivation of an Enterobacter sp. isolated from the composter were investigated. During the EPS fermentation under conditions of constant agitation speed from 200 to 900 rpm and constant aeration rate of 0.5-2.5 vvm, the low yields of EPS(4.8-5.2g/L) was observed as the viscosity increase of culture broth. With the stepwise increases in agitation speed and aeration rate, the EPS production and the viscosity of EPS were increased 1.3~1.4 times and 2.3~3.6 times higher than those of the fixed conditions, respectively. Therefore, these stepwise increases were considered as the key operating parameters for enhancing EPS production. The max. EPS(6.8g/L) and viscosity(14,000cp) were obtained when the agitation speed was increased from 300 to 900 rpm for 54hrs at 1.5 vvm.

  • A strain of Saccharomyces cerevisiae BY-366 was found to produce a strong sucrose-hydrolyzing enzyme Using this strain, the optimal culture conditions for the production of invertase were investigated. The results are as follows : 1. For enzyme production, optimal temperature, initial pH and critical concentrations of sucrose and raffinose were 3$0^{\circ}C$, 5.0 and 3.0%, respectively. 2. Enzyme production was reached maximum by organic nitrogen source, 0.3% yeast extract plus 0.5% bactopeptone. 3. It was appeared the presence of 0.1 M Mn2+ and Fe2+ ion was essential factors, on the other hand, 0.1 M Ag+ and Hg2+ ion almost block in yeast growth and enzyme production. 4. Invertase productivity was reached maximum within 3 days on stationary culture with medium-composed of sucrose 3%, bactopeptone 0.5%, yeast extract 0.3%, KEHPO. 0,1%, MgSO4.7H2O 0.05%.

Improvement of Amidase Production by a Newly Isolated Delftia tsuruhatensis ZJB-05174 Through Optimization of Culture Medium

  • The R-amidase production by a newly isolated strain of Delftia tsuruhatensis ZJB-05174 was optimized in this paper. Effects of factors such as carbon sources, nitrogen sources, and inducers on amidase production were investigated. The medium composition was optimized using central composite designs and response surface analysis. The optimal medium components for enhanced amidase production were found to be as follows: glucose, 8.23 g/l; yeast extract, 11.59 g/l; 2,2-(R,S)-dimethylcyclopropane carboxamide, 1.76 g/l; NaCl, 1 g/l; ${KH_2}{PO_4}$ 1 g/l; and ${K_2}{HPO_4}$ 1 g/l. A maximum enzyme production of 528.21 U/l was obtained under the optimized conditions, which was 4.7 times higher than that obtained under initial conditions.

  • The effect of AcNPV infection conditions such as serum concentration, pH, CaCl2, lysosomotropic agent, cell density at infection, agitation, aeration and nutritional supplementattion on recombinant protein production in Spodoptera frugiperda 21 cells was investigated using tissue culture flask, bottle and spinner flask. It was shown that serum, CaCl2, pH and cell density at infection affected recombinant production. The lysosomotropic agent did not significantly influence recombinant protein production.

Pseudoperonospora humuli의 실험실상의 난포자 형성 (Laboratory Production of Oospores in Pseudoperonospora humuli)

  • In pseudoperonospora humuli, the cause of hop downy mildew, environmental and host factors affecting laboratory production of oospore were examined. After 7 days incubation of leaf disk inoculated with sporangia on water, additional incubations were carried out under different conditions of temperature and moisture. Oospore production was also compared between very susceptible (Nugget) and resistant (Fuggle) hop cultivars. Oospores were not produced at 18$^{\circ}C$ regardless of other incubation conditions. Leaf disks failed to produce oospore when incubated on water for up to 18 days at 8$^{\circ}C$. No oospores formed on infection sites without necrosis. However, abundant oospores were produced at necrotized infection sites when inoculated leaf disk incubated on dry filter paper for 5 days at 8$^{\circ}C$. Both susceptible and resistant hop cultivars produced abundant oospores. In the measurement of optimal temperature for oospore production, oospores were produced at 6 to 12$^{\circ}C$ Most abundant oospores were produced at 8$^{\circ}C$. We suggest that proper combination of low temperature, dryness and necrosis may be a critical environmental factors for oospore production of P. humuli.

Optimization of Culturing Conditions for Improved Production of Bioactive Metabolites by Pseudonocardia sp. VUK-10

  • The purpose of the present study was to investigate the influence of cultural and environmental parameters affecting the growth and bioactive metabolite production of the rare strain VUK-10 of actinomycete Pseudonocardia, which exhibits a broad spectrum of in vitro antimicrobial activity against bacteria and fungi. Production of bioactive metabolites by the strain was high the in modified yeast extract-malt extract-dextrose (ISP-2) broth, as compared to other tested media. Glucose (1%) and tryptone (0.25%) were found to be the most suitable carbon and nitrogen sources, respectively, for optimum production of growth and bioactive metabolites. Maximum production of bioactive metabolites was found in the culture medium with initial pH 7 incubated with the strain for four days at $30^{\circ}C$, under shaking conditions. This is the first report on the optimization of bioactive metabolites by Pseudonocardia sp. VUK-10.

Bacillus polymyxa YL38-3의 세포외 cytosine deaminase 생성의 최적 배양 조건 (Optimum culture conditions for production of extracellular cytosine deaminase by bacellus polymyxa YL 38-3)

  • The strain YL 38-3, which was capable of producing extracellular cytosine deaminase, was isolated and taxonomically examined. The isolated strain was identified to be Bacillus polymyxa YL 38-3. The optimal conditions for the enzyme production from Bacillus polymyxa YL 38-3 were investigated. The enzyme production was reached maximum level in the medium containing 0.5% glucose, 0.2% beef extract, 0.5% NaCl and 0.1% $KH_{2}PO_{4}$ (pH 6.0). And the enzyme showed the highest activity when the strain YL 38-3 was cultivated at $35^{\circ}C$ for 24 gours under the initial pH 6.0. By the additions of peptone the extracellular enzyme production was inhibited, meanwhile the intracellular enzyme production was highly stimulated. It was, therefore, deduced that peptone was related to the secretion mechanism of the enzyme from this bacterial cell.

Concurrent Production of Methanol and Dimethyl Ether from Carbon Dioxide Hydrogenation : Investgation of Reaction Conditions

  • The concurrent production of methanol and dimethyl ether from carbon dioxide hydrogenation has been studied under various reaction conditions. First, the methanol synthesis was compared with the concurrent production method. For the methanol synthesis, the ternary mixed oxide catalyst (CuO/ZnO/Al2O3) was used and for the coproduction of methanol and dimethyl ether, silica-alumina was mixed with the methanol synthesis catalyst to be a hybrid catalyst. The results show that the co-production provides much higher per-pass yield than methanol synthesis even at very short contact time. The effects of temperature, contact time, pressure and catalyst hybrid ratio on the product yields and selectivities were also determined in the co-production.
