Nitric Oxide Production Inhibitory and Anti-Oxidative Activities of Phenolic Compounds from the Barks of Ulmus davidiana

  • The Barks of Ulmus davidiana (Ulmaceae) have been used for the treatment of insecticide, anti-boil and anti-fungi in Korean traditional medicine. Four phenolic compounds were isolated from 80% Acetone extracts. The structures of these compounds were elucidated as (+)-catechin, (+)-catechin 7-O-${\beta}$-D-glucopyranoside, (+)-catechin 7-O-${\beta}$-D-xylopyranoside and procyanidin B-1. These phenolic compounds showed significant nitrogen monoxide(NO) production inhibitory activity in IFN-${\gamma}$, LPS stimulated RAW 264.7 cell and also showed significant antioxidative activity on DPPH radical. Phenol성 물질이 첨가된 Corn Oil-in-Water Emulsion의 산화에 미치는 Surfactant Micelle의 영향 (Effect of Surfactant Micelle on Lipid Oxidation in Corn Oil-in-Water Emulsion with Phenol Compounds)

  • 5% corn oil과 17 mM Brji 700으로 제조한 oil-in-water emulsion(O/W)에서 과량의 잉여 surfactant가 phenol이 첨가된 O/W의 산화에 미치는 영향을 알아보기 위하여 Brji 700을 $0{\sim}2%$의 농도로 O/W에 첨가한 경우, 첨가된 surfactant의 양이 증가할수록 droplet의 particle size는 커지는 경향을 나타내었다. 또한 continuous phase의 surfactant 농도도 높아지며 phenol 함량도 상대적으로 높아지는 것을 알 수 있었다. Phenol류가 100 ppm의 농도로 첨가된 O/W에 과량의 surfactant$(0{\sim}2%)$를 첨가하고 30일간 저장 기간별 hydroperoxide의 생성량을 측정한 결과, 대조구에 비해 낮은 hydroperoxide 함량을 보여 surfactant를 첨가하지 않은 실험구와 유사한 경향을 보였으나, hydroperoxide 함량은 surfactant를 추가하지 않은 실험구 보다 더 낮은 값을 나타내었다. 또한 phenol 종류 및 surfactant 농도별 hydroperoxide 생성억제 효과와 headspace aldehyde 생성 억제 효과는 BHT>procyanidin B3-3-O-gallate>(+)-gallocatechin>(+)-catechin 및 2%>1%>0% surfactant 첨가의 순서로 나타났다. 이러한 결과는 phenol성 물질과 과량의 surfactant micelle에 의해 emulsion으로부터 continuous phase로 hydroperoxide의 physical location 변화가 발생하는 것을 나타낸다.

원적외선 처리가 포도씨의 기능성분에 미치는 영향 (The Effect of Far-Infrared Irradiation on Functional Components of Grape Seed)

  • 원적외선 조사가 포도씨 기능성분의 변화에 미치는 영향을 알아보고자 가열출력(0, 900, 1800 W) 및 가열시간(0, 20, 40 min)을 달리하여 원적외선 조사 후 마이크로파를 이용하여 포도씨 추출물을 제조하고 기능성분으로서 가용성 고형분, 기계적 색도, 카테킨, 총 페놀, 총 플라보노이드 함량 및 전자공여능과 아질산염 소거능을 각각 측정하였다. 가용성 고형분 함량 및 백색도 및 적색도는 유의적인 차이가 없는 것으로 나타났다. Cathechin류는 가열시간에 따라 catechin, procyanidin $B_2$, epicatechin 함량의 변화가 있음을 확인하였으며, 총 catechin 함량은 총 카테킨 함량은 900 W 및 20분 처리 시 가장 높게 나타났다. 총 페놀성화합물 및 플라보노이드 함량 또한 가열출력 900W 및 가열시간 20분 일 때 가장 높게 나타났다. 그에 따라 전자공여능도 가열출력 900 W 및 가열시간 20분 일 때 가장 높은 것으로 나타났다. 이상의 결과는 원적외선 처리에 의해 총 페놀성 화합물 함량, 총 플라보노이드 함량이 증가됨을 볼 수 있었으며, 특히 cathechin류의 변화가 있음을 확인할 수 있었다. 또한 이로 인해 항산화능도 증가됨을 알 수 있었다. 또한 포도씨의 원적외선 가열처리 최적조건을 설정하고자 가열출력 및 가열시간을 달리하여 중심합성실험계획으로 반응표면분석한 결과 가열출력 621.82~818.18 W, 가열시간 16.3~19.83 min의 범위가 최적 가열조건범위로 설정되었다.

루이보스차(Asphalthus linearis)의 추출방법에 따른 페놀릭류 함량 변화연구 (Determination of Phenolic Contents in Rooibos (Asphalthus linearis) Tea Depending on the Steeping Temperature and Time)

  • 루이보스티에서 5종의 phenolic acid (gallic acid, chlorogenic acid, caffeic acid, p-coumaric acid, trans-ferulic acid)와 9종의 flavonoid (procyanidin b1, aspalathin, rutin, vitexin, hyperoside, isoquercitrin, luteolin, quercetin, chrysoeriol)를 UPLC-MSMS를 이용하여 동시 분석하였다. 14종 페놀릭류를 동시 분석하기 위하여 기기조건과 유효성을 검증하였고 확립된 분석방법을 이용하여 시중에 유통중인 루이보스티 30건을 채취하여 페놀릭류를 분석하였다. 루이보스티 1 g 혹은 1티백에 뜨거운 물 100 mL을 가하여 3분, 6분, 30분이 경과 후 그리고 차가운 물 ($25-30^{\circ}C$)에 30분 우려낸 루이보스티의 페놀릭류 함량을 구하였다. 루이보스티에서 전체 실험대상 페놀릭류 중 rutin과 aspalathin이 가장 많이 추출되어 나왔으며 각각 물질의 함량은 제품별로 달랐다. 페놀릭류 성분의 추출효율은 14종 페놀릭류의 총합 기준으로 뜨거운 물 30분 > 6분 > 3분 > 차가운 물 30분 순으로 높았다.

Chemical Characterization of Neutral Extracts Prepared by Treating Pinus radiata Bark with Sodium Bicarbonate

  • A neutral extract (NE), that is soluble in cold water and has excellent antioxidant activity, from Pinus radiata pine bark was prepared by sodium bicarbonate treatment, and its chemical characteristics were investigated. NE was prepared by treating P. radiata bark with 0.8% NaHCO3 aqueous solution with a 5 : 1 liquor-to-bark ratio at boiling temperature for 1 h, resulting in 44% yield and final pH of 6.66. The yield of NE was 11% higher than that of the hot water extract (HWE) due to the increase in the solubility of polyphenols, the main component in the bark, by NaHCO3 treatment. NE was characterized through FT-IR, NMR, and MALDI TOF MS analyses. The results indicated that NE is mostly composed of proanthocyanidins (PAs) consisting of procyanidin (PC) units. The acetylated neutral extract (Ac-NE) had weight average molecular weight (${\bar{M}}w$) of 5,300 Da. The Ac-NE had wide molecular weight distribution and its polydispersity (${\bar{M}}w/{\bar{M}}n$) was 6 times higher than that of pure PA. The antioxidant activity of NE was determined by 2-diphenyl-1-picrylhydrazyl (DPPH) free radical scavenging assay and showed that NE had comparable antioxidant activity with pure PA.

Changes in Functional Constituents of Grape (Vilis vinifera) Seed by Different Heat Pretreatments

  • Changes in functional constituents of grape (Vitis vinifera) seeds prepared by three different heat pretreatments were determined and compared with those of non-treated grape seed. The recovery of grape seed oils was generally increased by roasting, steaming and microwave processes, although the recovery of specific constituents varied among three heat pretreatments. The recovery of MeOH extracts of the seeds increased following the roasting process, whereas that of MeOH extracts decreased gradually with steaming and microwave treatments. Levels of four catechins in grape seeds: (+)-catechin, procyanidin B$_2$, (-)-epicatechin, and (-)-epicatechin gallate, were decreased with increased roasting and steaming time, but were unaffected by microwave treatment. During the three different heat pretreatments, levels and compositions of fatty acid did not change, whereas those of phytosterol compositions decreased greatly. These results suggest that a mild heat pretreatment, controlled for temperature and time, is needed to prevent a considerable loss in the level of valuable functional components in grape seed.

Antioxidant and Anti-inflammatory Activity of Stem Bark Extracts from Ulmus davidiana var. japonica

  • Ulmus davidiana var. japonica is a deciduous tree used in traditional medicine. In this study, we examined the antioxidant and anti-inflammatory activity of extracts and compounds isolated from U. davidiana var. japonica stem barks for development of cosmetic phyto-materials. Phytochemical examination of the stem bark led to the isolation and characterization of three known compounds, (+)-catechin (1), (+)-catechin-7-O-${\beta}$-D-apiofuranoside (2), and procyanidin B3 ((+)-catechin-($4{\alpha}{\to}8$)-(+)-catechin) (3). Their bioactivities including antioxidant (DPPH ree radical scavenging assay) activity, anti-inflammatory (nitric oxide nhibition assay) were evaluated. Most of the crude extracts and isolates indicated significant antioxidant potential compared with BHT and ${\alpha}$-tocopherol as controls. Futhermore, all compounds showed higher inhibitory activities for NO production in Raw 264.7 cells than the L-NMMA using the positive control. Eespecially, (+)-catechin (1) and (+)-catechin-7-O-${\beta}$-D-apiofuranoside (2) which could inhibit more than 90% of the NO production at a concentration of 100 ${\mu}g/m{\ell}$, implying excellent anti-inflammatory activity.

Fruit Characteristics and Variation of Phenolic Compounds in the Fruit of Hawthorn (Crataegus pinnatifida Bunge) Selected from Korea and Chinese Cultivars

  • In order to select superior tree from Korea, five major phenolic compounds including (-)-epicatechin(EC), chlorogenic acid (ChA), hyperoside (HP), isoquercitrin (IQ), and procyanidin B2 (PC-$B_2$) in hawthorn fruit were evaluated. We also compared these hawthorn fruits of five clones with Chinese hawthorn cultivars. HPLC with a diode-array detector was used to determine the contents of the individual compounds. Hawthorn fruits of five clones (selected from different area of Korea), and four Chinese hawthorn cultivars grown in the Korea Forest Research Institute (Suwon) were utilized. With their high functional components, Jungsun is the clone including the highest contents of EC (11.26 mg/g) and PC-$B_2$ (24.46 mg/g). The clone of Chuncheon 15 had highest HP (0.53 mg/g) and IQ (0.41 mg/g). From the results, the clone of Jungsun and Chuncheon 15 can be evaluated to be selected breeding material for cultivar development.

Smilax riparia 잎의 항산화 성분 (The Anti-oxidative Compounds of Smilax riparia Leaves)

  • Rhizoma of Smilax china has been used as anti-inflammatory and analgesic, antiedemic agent in Korean folk medicine. In order to investigate the efficacy of anti-oxidative activity, the activity-guided fractionation and the isolation were performed. Each fractions ($H_2O$ fraction, 20%, 40%, 60%, 100% MeOH fractions and CHCl$_3$ fraction) was examined antioxidant activity by 1,1-diphenyl-2-picrylhydrazyl (DPPH) free radical scavenging potential. It was revealed that 40%, 20% MeOH fractions and $H_2O$ fractions have significant anti-oxidative activity. From 40% and 20% MeOH fractions two flavonoid glycosides and one procyanidin were isolated and elucidated apigenin-7-Ο-$\alpha$-L-rhamnopyranosyl(1\longrightarrow2)-$\beta$-D-glucopyranoside, apigenin-7-Ο-$\alpha$-L-rhamnopyranosyl(1\longrightarrow6)-$\beta$-D-glucopyranoside and catechin(4$\alpha$\longrightarrow6)epicatechin through their physicochemical data and IR, FAB-MS, $^{13}$ C-NMR, and $^1$H-NMR analysis with authentics, respectively. The isolated compounds were examined by DPPH method. Apigenin-7-Ο-$\alpha$-L-rhamnopyranosyl(1\longrightarrow2)-$\beta$-D-glucopyranoside and catechin (4$\alpha$\longrightarrow6) epicatechin showed powerful radical scavenging activities on DPPH radical among three compounds.

Effects of Fruit By-product Extracts Supplementation on Growth Performance and Nutrient Digestibility in Growing Pigs

  • A total of 96 pigs [(Landrace ${\times}$ Yorkshire) ${\times}$ Duroc] with an initial BW of $27.94{\pm}0.92kg$ were used in a 6-week experiment to determine the effects of dietary supplementation with fruit by-product extracts on growth performance and nutrient digestibility in growing pigs. Pigs were randomly allotted to four treatments : 1) CON (basal diet), 2) PRO (CON + 0.5% procyanidin), 3) HES (CON + 0.5% hesperidin), 4) TAN (CON + 0.5% tannin). There were six replications per treatment with four pigs per pen. Supplementation had no effect (p>0.05) on average daily gain, average daily feed intake, and G/F. The apparent total tract digestibility (ATTD) of dry matter and nitrogen was increased (p<0.05) in the HES treatment relative to the PRO treatment. Pigs fed the HES and TAN diets had greater (p<0.05) ATTD of energy than pigs fed PRO diet. The ATTD of ash was greatest (p<0.05) in HES treatments. In addition, the ATTD of calcium was greater (p<0.05) in HES treatments than in CON and PRO treatments. Overall, the results of this study indicated that dietary supplementation with 0.5% fruit by-products did not affect growth performance, but inclusion of 0.5% hesperidin increased nutrient digestibility in growing pigs.
