Adhesion Properties of Indigenous Dadih Lactic Acid Bacteria on Human Intestinal Mucosal Surface

  • Dadih is Indonesian traditional fermented buffalo milk believed by the natives to have beneficial effects on human health. This may be due to the probiotic properties possessed by the lactic acid bacteria (LAB) involved in its fermentation process. It was discovered that ten strains of dadih lactic isolates possessed some probiotic properties in vitro. In this study, the adhesion properties of dadih LAB, in comparison with documented probiotic strains, were investigated in vitro by using mucin extracted from human faeces and Caco-2 cells as the models for human intestinal mucosal surface and intestinal cells respectively. The adhesion results showed the distinction of Lactobacillus reuteri IS-27560 in adhering to both mucus layer and Caco-2 cells. The competition assay for adhesion to the mucus layer between dadih LAB and selected pathogens indicated the competence of Lactococcus lactis IS-16183 and Lactobacillus rhamnosus IS-7257 in significantly inhibiting the adhesion of Escherichia coli O157:H7. Accordingly, these two strains may be potential candidates for use as probiotic strains. Overall, the adhesion properties of all dadih LAB strains were relatively comparable to that of Lactobacillus casei Shirota and Lactobacillus rhamnosus GG, the documented probiotic strains.

Rapid Identification of Lactobacillus and Bifidobacterium in Probiotic Products Using Multiplex PCR

  • Lactic acid bacteria (LAB) are beneficial for the gastrointestinal tract and reinforce immunity in human health. Recently, many functional products using the lactic acid bacteria have been developed. Among these LAB, Lactobacillus acidophilus, Lactobacillus rhamnosus, Bifidobacterium longum, and Bifidobacterium bifidum are frequently used for probiotic products. In order to monitor these LAB in commercial probiotic products, a multiplex PCR method was developed. We designed four species-specific primer pairs for multiplex PCR from the 16S rRNA, 16S-23S rRNA intergenic spacer region, and 23S rRNA genes in Lactobacillus acidophilus, Lactobacillus rhamnosus, Bifidobacterium longum, and Bifidobacterium bifidum. Using these primer pairs, 4 different LAB were detected with high specificity in functional foods. We suggest that the multiplex PCR method developed in this study would be an efficient tool for simple, rapid, and reliable identification of LAB used as probiotic strains.

Lactobacillus sakei BK19의 어류 병원성 세균에 대한 항균활성 (Antibacterial Activity of Lactobacillus sakei BKl9 against Fish Pathogenic Bacteria)

  • 본 실험의 목적은 Gram 양성균 뿐만 아니라 Gram음성세균에 대한 넓은 항균활성 및 내산성 그리고 항생제 내성을 갖고 있는 유용한 probiotic 후보균주를 선발하여 어류의 세균성 질병 예방 흑은 치료를 함으로써 양식산업의 효율성을 증대시키고자 한다. 20균주의 유산균을 김치, 양식 넙치의 장 그리고 각종 젓갈류 중에서 분리하여 어류 병원성 세균에 대한 항균능을 MRS agar상에서 agar spotted method에 의해 조사하여 Vibrio anguillarum, Edwardsiella tarda 그리고 streptococcus sp.에 대해 항균활성 이 있는 7균주를 1차 선발 하고 이들중 항균스펙트럼 이 넓은 BK19를 최종선발하여 동정한 결과 Lactobacillus sakei BKl9로 나타났다. L. sakei BK19의 cell free supernatant를 pH 중화 및 catalase 처리한 후 어병세균에 대하여 항균활성을 확인하였다. L. sakei BK19 supernatant의 V.anguillarum 및 V.alginolyticus에 대한 항균성 기작은 주사전자현미경 관찰을 통하여 확인하였고 세포벽을 붕괴 시킴으로써 성장을 저해하고 사멸시키는 것으로 확인되었다.

  • Lactic acid producing bacteria were isolated from baby feces and characterized to be used as a probiotic with anti Helicobacter pylori functions. The selected bacteria had inhibition activity on the adherance and growth of H. pylori. These bacteria had additional beneficial characteristics for the probiotic such as antibacterial activity, antitumor activity, immunostimulation activity, resistance to antibiotic and bile salt, ability to bind to the intestinal cells, and safe for the human use.

Effect of Lactobacillus acidophilus based probiotic product supplementation on the blood profile, fecal noxious gas emission, and fecal shedding of lactic acid bacteria and coliform bacteria in healthy adult Beagle dogs

  • The aim of this study was to evaluate the effect of Lactobacillus acidophilus probiotic (LAP) product supplementation on the blood profile, fecal noxious gas emission, and fecal shedding of lactic acid bacteria and coliform bacteria in healthy adult Beagle dogs. In total, 14 Beagle dogs with an average initial body weight of 10.19 ± 0.61 kg were randomly assigned into two dietary treatments,with and without LAP supplementation, for a 28-day feeding trial. At the end of the experiment, there was no significant (p > 0.05) difference in the concentration of serum total cholesterol, high-density lipoprotein cholesterol (HDL-C), low-density lipoprotein cholesterol (LDL-C), white blood cell (WBC), red blood cell (RBC), blood lymphocyte percentage, fecal hydrogen sulfide (H2S) and total mercaptans (R.SH) emission, and fecal coliforms counts. However, the serum concentrations of the triglyceride and fecal ammonia (NH3) emission of the LAP treatment were significantly (p < 0.05) decreased in the group compared with the CON dogs. Fecal total lactic acid bacteria counts were significantly (p < 0.05) increased in the LAP treatment. In conclusion, the supplementation of LAP in Beagle dog diets could decrease the blood triglyceride level and enhance the gut Lactobacillus count which may have positive effects on dogs.

Probiotic Bacillus amyloliquefaciens B-1895 Improved Growth of Juvenile Trout

  • The aim of this study was to evaluate a new Bacillus amyloliquefaciens B-1895 probiotic as a feed additive for farmed trout. Final weight, absolute and average daily gain of fish, and average daily growth rate were higher in the group that received the probiotic than in the control group (p<0.05). Moreover, the probiotic-fed trout had more intense growth rates than the control group (higher by 15.7%; p<0.05). A decrease in feed ratio was also observed in the group that received probiotic (25% decrease; p<0.05), indicating more efficient digestion and assimilation of feed. In general, the introduction of probiotic in the feed did not adversely affect the functional status of the fish. In young trout of the control group, when assessing the general chemical composition of the organism in the muscle tissue revealed significantly (p≤0.001) higher level of moisture content by 5.1% and lower by 11.0% dry matter content. In muscle, the protein content was higher by 1.33% (p≤0.001) and fat content by 2.1% (p≤0.001) in experimental fish. Generally, Lactobacilli, Enterococcus, Vibrio, Bacillus, and coliform bacteria were found in the intestinal samples of rainbow trout. Significant reliable difference (p≤0.05) between the samples of experimental and control groups was noted in the content of Bacillus bacteria. In the control group, 5.0±0.4×103 CFU/g was detected, while in the experimental group 8.4±0.8×104 CFU/g. Overall, the data indicate that probiotic bacteria B. amyloliquefaciens B-1895 has no adverse effect on selected microorganisms in the study fish.

젓갈 프로바이오틱 생균의 내산성 및 내담즙 특성 (Some Probiotic Properties of Some Lactic Acid Bacteria and Yeasts Isolated from Jeot-gal.)

  • In order to select probiotics with a high survival rate in gut and the growth inhibition of virulent pathogens to human beings or animals, we have screened lactic acid bacteria and yeasts from Jeot-gal to assess resistance against the artificial gastric acid and bile juice. Lactic acid bacteria and yeasts isolated were incubated for 24 h in artificial bile juice after incubation for 2 h in artificial gastric acid. Especially, strain HW 161 and strain NK 181 showed the higher survival for 2 h incubation in artificial gastric acid. All of 3 strains of lactic acid bacteria and 2 strains of yeast were showed higher growth rate than the control in artificial bile. The antimicrobial activity of lactic acid bacteria and yeasts was also investigated to prove efficacy as probiotic organisms. Lactic acid bacteria were shown the inhibition of Gram positive and negative bacteria, but yeasts narrow inhibition.

The dual probiotic and antibiotic nature of Bdellovibrio bacteriovorus

  • Bdellovibrio bacteriovorus is a predatory bacterium which attacks and consumes other bacterial strains, including the well known pathogens E. coli O157 : H7, Salmonella typhimurium and Helicobacter pylori. This remarkable activity has been the focus of research for nearly five decades, with exciting practical applications to medical, agriculture and farming practices recently being published. This article reviews many of the exciting steps research into this bacterium, and similar bacteria, has taken, focusing primarily on their use as both an antibiotic to remove harmful and pathogenic bacteria and as a probiotic to help curb and control the bacterial populations within the intestinal tract. Owing to the unique and dual nature of this bacterium, this review proposes the use of "amphibiotic" to describe these bacteria and their activities.

전통식품 유래 유산균의 해조류 발효 및 Probiotic 특성 (Seaweed Fermentation and Probiotic Properties of Lactic Acid Bacteria Isolated from Korean Traditional Foods)

  • 해조류의 발효가 가능하고 probiotic 특성이 우수한 유산균을 분리 선발한 후 이들의 미역과 다시마 발효능을 검토하였다. 미역 및 다시마 발효가 가능한 균주를 김치 젓갈, 된장으로부터 331 균주를 순수 분리하여 해조류 구성 다당(alginate, cellulose) 분해능, 균의 생육, 항균 활성 등을 비교 검토한 결과 4균주(stain No. 162, 164, 192, 196)가 우수하였다. 선발 균주 모두 인공위액, 인공담즙액, NaCl에 높은 생존율을 나타내었고 이들 4균주 중 No. 192가 가장 우수하였으며 Enterococcus faecium으로 동정되었다. 미역과 다시마를 이용하여 선발 유산균을 배양한 결과 No. 192 균주가 발효특성이 가장 양호하였으며, No. 162, 164, 196 균주도 양호하였다. 선발 유산균을 이용한 미역과 다시마에서 성장이 가능하였으며 발효 후 미역과 다시마 발효물의 항산화 활성이 증진되었다.

In Vitro Probiotic Properties of Indigenous Dadih Lactic Acid Bacteria

  • The aim of this research was to identify candidate probiotic lactic bacteria among indigenous dadih lactic isolates. Dadih is an Indonesian traditional fermented milk of West Sumatra which is fermented naturally. Viability of the strain is critical in determining the capacity of lactic bacteria to induce immune stimulation as well as to colonize in the intestinal tract. Therefore, LAB are proposed to exert health promoting or probiotic effects in human, such as inhibition of pathogenic microflora, antimutagenic, and the reduction of cholesterol levels. This manuscript reports in vitro probiotic properties of indigenous dadih lactic bacteria, especially some important colonization factors in GI tract, such as lysozyme, acid and bile tolerance. Bile Salt Hydrolase (BSH) activity, spectrum of bacteriocin, and antimutagenic activity of bacterial cells were also assessed. Twenty dadih lactic isolates were screened further for their tolerance to low pH, at pH 2 and 3 as well as their bile tolerance. There were ten isolates classified as acid and bile acid tolerant, and further screened for lysozyme tolerance, BSH activity. The spectrum of bacteriocin activity of isolates was assayed using cell-free neutralized supernatants by agar spot test against variety of pathogens. Lc. lactis subsp. lactis IS-10285, IS-7386, IS-16183, IS-11857 and IS-29862, L. brevis IS-27560, IS-26958 and IS-23427, Leu.mesen.mesenteroides IS-27526, and L. casei IS-7257 each has good survival rate at low pH values and in the presence of lysozyme, and short lag time in the presence of 0.3 % oxgall. Lc. lactis subsp. lactis IS-11857 and IS-29862 each has high BHS activity, Lc. lactis subsp. lactis IS-10285 and IS-16183 each had a positive spectrum of bacteriocin activity against E. coli 3301 and Lysteria monocytogenes ATCC 19112, while L. brevis IS-26958 has high BHS activity as well as positive spectrum of bacteriocin against E. coli 3301, Lysteria monocytogenes ATCC 19112, and S. aureus IFO 3060. All of the ten dadih lactic strains performed in vitro acid and bile tolerance, indicating a possibility to reach the intestine alive, and display probiotic activities.
