약배양에 대한 반응이 비교적ㆍ높은 것으로 알려진 고추의 두 품종 계룡산재래와 밀양재래의 약을 2,4-D와 kinetin이 각각 0.1 mg/L씩 첨가된 배지에 치상한 후 4$^{\circ}C$와 32$^{\circ}C$의 온도 전처리가 소포자배 발생에 미치는 영향을 조사하였다. 소포자배의 발생은 사용한 두 품종 중 밀양재래에서 높았다. 고온처리는 저온처리에 비해 소포자 배의 발생이 많았고 캘러스의 발생은 거의 없었으며 발생한 배는 동일 배지에서 또는 호르몬이 없는 배지에 옮겼을 때 대부분이 정상적인 유식물로 발달하였다. 저온처리에서는 캘러스의 발생이 많았고 발생한 배는 대부분이 비정상적으로 발달하였다. 소포자배 발생에 효과적인 고온처리 기간은 밀양재래에서는 3일, 계룡산재래에서는 6일이었다. 고추의 약배양 시 고온처리는 소포자배의 유기뿐만 아니라 유기된 배의 생장에도 중요한 영향을 미치는 것으로 나타났다.
강원도에서 확보한 폐옥수수대는 글루칸 44.6 %, 자일란 23.8 %, 리그닌 23.8 %, 회분 4.5 %, 기타 8.1 %로 이루어져 있다. 묽은 황산을 이용한 전처리 공정을 통계적 방법인 완전요인배치으로 분석한 결과 온도가 가장 중요한 변수로 나타났으며 자일로스 수율 기준으로 처리 시간과 농도에 비해 각각 3.5배와 3.2배 큰 영향력을 보여주었다. 포도당 수율에 대해서도 온도의 영향력이 가장 큰 것으로 나타났지만 포도당 수율은 전체적으로 5 % 이내의 낮은 값을 나타내었다. 마이크로웨이브와 초음파 각각을 이용한 전처리 공정은 그 효과가 크지 않거나 미미하였으나 묽은 황산처리와 복합적으로 적용했을 때는 그 효과는 상당히 큰 것으로 나타났다. 특히 마이크로웨이브와 묽은 황산처리를 순차적으로 적용했을 때 자일로스 수율은 2배 넘게 향상되었다. 향후에는, 다양한 복합 전처리 공정에 대한 심도있는 연구와 더불어 전처리와 효소당화 공정의 최적화에 대한 연구를 수행할 계획이다.
본 연구에서는 버섯 폐배지의 혐기성소화 효율 향상을 위해 수열전처리를 실시하고, 리그노셀룰로오스계 물질의 고온 가수분해 과정에서 생성될 수 있는 중간산물이 기질의 생분해도와 바이오가스 전환 효율에 미치는 영향을 함께 판단하였다. 수열전처리 온도의 범위를 150, 180, 210℃로 설정하였으며, 모든 수열전처리 온도에서 기질의 가용화율이 향상되는 결과를 확인할 수 있었다. 추가적으로, 150℃로 버섯 폐배지를 전처리한 경우에는 혐기성소화 효율에 영향을 미칠 수 있는 C/N 비가 개선되는 효과를 함께 확인하였다. 다만 전처리 온도가 180, 210℃인 경우에는 오히려 150℃로 전처리를 수행한 경우에 비해 메탄 생성량이 저하되는 경향을 보였는데, 이는 리그노셀룰로오스 물질의 중간분해 산물인 퓨란유도체의 형성으로 인해 메탄생성균이 영향을 받은 것으로 판단된다. 결국, 수열전처리를 통해 리그노셀룰로오스계 바이오매스의 가용화율 향상을 통한 메탄 생성 향상을 기대할 수 있으나, 혐기성소화 효율을 저해할 수 있는 중간산물이 생성되지 않는 적정 전처리 온도의 확인과 적용이 중요할 것으로 판단된다.
The effect of thermal pretreatment conditions on hydrolysis characteristics of dairy manure and sawdust mixtures has been evaluated. Thermal pretreatment temperature varied between 35 and $120^{\circ}C$ and the period of the treatment changed between 30 and 1440min (24h). As thermal pretreatment temperature and duration increased, organic material solublization rates were improved. Maximum solubilizations of chemical oxygen demand (SCOD), carbohydrates, and volatile fatty acids(VFAs) were observed when dairy manure treated for one day at $120^{\circ}C$. Although one day treatment duration at $120^{\circ}C$ showed the highest SCOD, soluble carbohydrates, and VFAs concentration, its hydrolysis rate was only about 12%. The results reveal that the thermal pretreatment conditions tried in this study are not enough to solubilize the organic matter contained in dairy manure and sawdust mixtures. In order to maximize hydrolysis performance, the further research needs to determine the factors influences on organic material solubilization in addition to thermal pretreatment temperature and duration.
The aim of present study is to investigate the effect of naringin on the pharmacokinetics of verapamil and its major metabolite, norverapamil in rabbits. The pharmacokinetic parameters of verapamil and norverapamil were determined after administering verapamil (9 mg/kg) orally to rabbits in the pretreated with naringin (1.5, 7.5, and 15 mg/kg). Naringin pretreatment significantly altered the pharmacokinetic parameters of verapamil. Compared with the control group (given verapamil alone), the $K_a,\;C_{max}$ and AUC of verapamil were significantly (p<0.05 or p<0.01) increased in the pretreatment of naringin, However there were no significant change in $T_{max}\;and\;t_{1/2}$ of verapamil. Consequently, pretreatment of naringin significantly (p<0.05, p<0.01) increased the AB% of verapamil significantly in a dose dependent manner (p<0.05 or p<0.01 ), and elevated the RB% of verapamil by 1.26- to 1.69-fold. the MR of verapamil were significantly (p<0.05) increased in the pretreatment of naringin, implying that pretreatment of naringin may effectively inhibit the CYP3A4-mediated metabolism of verapamil. In conclusion, pretreatment of naringin enhanced the oral bioavailability of verapamil. Based on these results, the verapamil dosage should be adjusted when given with naringin or a naringin-containing dietary supplement.
The efficiency of acid, enzyme and microbial pretreatment of rice bran was compared based on the content of cellulose, hemicellulose, reducing sugars and xylose in the substrate. An isolate of Aspergillus niger or a strain of Trichoderma viride(MTCC 800) was employed for microbial pretreatment of rice bran in solid state. Acid pretreatment resulted in the highest amount of reducing sugars followed by enzyme and microbial pretreatment. A. niger showed a higher rate of hydrolysis than T. viride. The rice bran hydrolysate obtained from the different methods was subsequently fermented to ethanol either by Zymomonas mobilis(NCIM 806) or by Pichia stipitis(NCIM 3497). P. stipitis fermentation resulted in higher ethanol(37% higher) and biomass production($76{\sim}83%$ higher) than those of Z. mobilis. Maximum ethanol production resulted at 12h in Zymomonas fermentation, while in Pichia fermentation, it was observed at 60h. Microbial pretreatment of rice bran by A. niger followed by fermentation employing P. stipitis was more efficient but slower than the other microbial pretreatment and fermentation.
To evaluate an effect of ethanol pretreatment on the toluene metabolism, toluene (50% in olive oil) was given three times at 0.2 ml/100g body weight at the interval of one day to the rats fed with 5% ethanol during two months. The ethanol pretreated rats were not identified particular liver injury by the histopathologic findings. In case of toluene treatment, the ethanol pretreatment to the rats led to more increased concentration of urinary hippuric acid than those treated with only toluene. The ethanol pretreatment to the rats led to the increased activities of hepatic aniline hydroxylase and these enzyme activities were higher both in toluene treated and those pretreated with ethanol, but no differences were found in two groups. Ethanol pretreated rats showed the more increased activities of benzylalcohol dehydrogenase than control group. Moreover, the ethanol pretreatment to the toluene treated rats led to significantly more increased activities of benzylalcohol dehydrogenase compared with those treated with toluene only. Furthermore, the alcohol pretreatment to the toluene treated rats also led to somewhat higher activities of benzaldehyde dehydrogenase than those treated with toluene. In conclusion, these results indicate that the chronic pretreatment of ethanol at not so much liver damage as normal may rather activate the toluene metabolism.
It is well known that the uterine contractility is affected by sexual hormone. In this experiment, the authors attempted to study the influences of testosterone and estrogen or the uterine contractility to oxytocics. The contractile sensitivity of the excised uterine muscle of non-castrated and castrated rabbits with testosterone and estrogen 24 hours before experiment is observed respectively. And the cholinesterase activity and electrolytes (Na, K, Ca and Mg) in the uterine muscle are measured in order to study the relationship with contractile sensitivity and those changes. The results obtained were summarized as follows: 1. The contractile effect of spareng on the excised uterine muscle of non-castrated rabbits pretreated with estrogen was markedly increased in small dose, but that of rabbits pretreated with testosterone was significantly increased in large dose, comparing with that of the control group. In castrated rabbits, the contractile sensitivity of the uterine muscle to spareng was significantly increased by pretreatment with estrogen in large dose but it was markedly decreased by pretreatment with testosterone in small dose. 2. The contractile effect of quinine on the excised uterine muscle of non-castrated rabbits pretreated with estrogen was significantly decreased but that of castrated rabbits pretreated with both estrogen and testosterone were markedly increased comparing with that of the control group. 3. The cholinesterase activity in the uterine muscle of non-castrated rabbits was significantly increased by pretreatment with small dose of estrogen or large dose of testosterone, but that of castrated rabbits was markedly decreased by pretreatment with large dose of estrogen. 4. Na and K contents in the uterine muscle of non-castrated rabbits were markedly increased by pretreatment with both estrogen and testosterone, but that of castrated rabbits was significantly increased by pretreatment with small dose of estrogen. 5. Ca content in uterine muscle of non-castrated rabbits was significantly decreased by pretreatment with both large dose of estrogen and testosterone but increased by pretreatment of testosterone. In castrated rabbits, Ca content was significantly decreased by pretreatment with both estrogen and testosterone. 6. Mg content in the uterine muscle of non-castrated rabbits was markedly increased by pretreatment with estrogen and small dose of testosterone, but that of castrated rabbits was significantly decreased by pretreatment with both large dose of estrogen and testosterone.
본 연구의 목적은 탈지미세조류(LEA) 세포벽 분해를 통한 바이오당 생산에 있어 당화효소 사용없이 마이크로파 전처리만을 이용하여 글루코오스와 자일로오스를 생산하는 것이다. LEA의 주성분인 셀룰로오스와 헤미셀룰로오스의 무효소 당화를 위해 산 가수분해 기반의 마이크로파 전처리 조건을 반응표면분석법을 이용하여 최적화하였다. 마이크로파를 이용한 무효소 당화 공정의 주요 변수는 마이크로파 출력(198~702 W), 전처리 시간(39~241 sec)와 황산 농도(0~0.1 mol)로 최적 조건 예측을 위해 중심합성계획법을 이용하여 2차 회귀함수를 도출하였다. 마이크로파 출력과 전처리 시간이 LEA로부터 육탄당(C6)과 오탄당(C5) 생산에 유의한 영향을 주는 변수이며 증가에 따라 육탄당과 오탄당 당화율이 증가하는 경향을 확인하였다. 육탄당과 오탄당 당화율 최대화를 위한 산 가수분해를 적용한 마이크로파 전처리 최적 조건은 마이크로파 출력 700 W, 전처리 시간 185.7 sec와 황산 0.48 mol으로 육탄당 당화율 92.7%와 오탄당 당화율 74.5%가 예측되었으며 확인 실험을 통해 육탄당 당화율 94.2%와 오탄당 당화율 70.8%가 확인되어 예측의 유효성을 확인할 수 있었다. 이는 LEA의 셀룰로오스와 헤미셀룰로오스 당화를 위해 산 가수분해 적용 마이크로파 전처리만을 이용한 무효소 당화 공정 적용과 $100^{\circ}C$ 이하의 낮은 온도와 짧은 전처리 시간 적용을 가능하여 기존 전처리 대비 효과적인 공정 임을 입증했다.
