The Effect of the Compound of Tomato Extract to the Prostatic Cancer Cell and the Prostate of the Rat Model of Benign Prostatic Hyperplasia (토마토 추출액 복합체가 전립선 암 세포와 전립선 비대증에 미치는 영향)

  • Benign prostatic hyperplasia (BPH) is one of the common disease in elderly men. Recently old-age population is increased and we are growing more and more interested in clinical importance of BPH. In this study, the effect of PLX, which was the mixture of tomato extract (including 2% of lycopene) and chitooligosaccharide, on prostatic cancer cell and testosterone-induced BPH in adult rats of the Sprague Dawley strain was determined. The cell viability was evaluated by MTT method using L929 and LNCaP cell line, pretreated with various concentrations of PLX. The expression of prostatic specific antigen (PSA) and 5${\alpha$}$-reductase genes were evaluated by realtime PCR using LNCaP cell line and compared various concentrations of PLX with 50 ${\mu}$M of finasteride. An experimental prostatic hyperplasia was induced in male Sprague Dawley rats by giving testosterone for 8 weeks. After 2 weeks from start of giving testosterone, PLX and finasteride were administered orally once a day. The results were analyzed with prostate weight per body weight at 8 weeks. Cell viability of L929 cell line decreased specifically at the concentration of 2000 ${\mu}$g/mf of PLX. The cytotoxicity of PLX to the LNCaP cell line was shown at above 500 ${\mu}$g/ml of PLX. The inhibitory effect of PLX to the expression of PSA and 5${\alpha$}$-reductase genes in LNCaP cell line increased with the concentration of PLX. In vivo study, the results of PLX and finasteride administered group were 3.75${\pm}$0.60 and 3.49${\pm}$0.49 prostate weight ${\times}10^3$/body weight, which were lower than the result of BPH induced group (4.74${\pm}$0.58). These results suggested that PLX may be an effective material in BPH by having the role of the 5a-reductase inhibitor.

Bio-Soda Pulping of Rice Straw with Pleurotus cornucopiae under Atmospheric Pressure

  • This study was carried out to develop the bio-chemical pulping method to enhance the energy saving and decrease the capital cost through the soda pulping under atmospheric pressure ($100^{\circ}C$). Nonwood substrates, rice straw, were pretreated by white-rot fungi, Pleurotus cornucopiae. Several basic pieces of data that can be applied in soda pulping were acquired. The results of this study were as follows. Under the conditions without any nutrients or with glucose, N and glucose + N, the weight losses of rice straws inoculated by Pleurotus cornucopiae were 12.1∼32.6 %, 12.0∼26.3 %, 13.0∼25.4 % and 15.3∼24.7 % for 5, 10, 15, 20, 25 and 30 days incubation periods respectively. The more the fungal incubation was extended, the more the weight losses were gained. The yield of untreated rice straw was 54.8 % after pulping. When any nutrients was not added or glucose, N and glucose + N were added for the pretreatment, the total yields were ranged to 57.3∼42.9 %, 51.0∼43.3 %, 51.7∼43.9 % and 52.1∼46.1 % for 5 different incubation periods respectively. The yields were gradually decreased based on the extending of the incubation periods. The physical properties of the rice straw soda pulp without fungal treatment, the density, breaking length, burst index, tear index and folding endurance were 0.24g/㎤, 2.32 Km, 0.91 kPaㆍ$m^2$/g, 46.7 mNㆍ$m^2$/g and 21 times, respectively. In the case of pretreatment without any nutrients or with glucose, N and glucose + N as nutrients, the density was 0.24g/㎤, the breaking length was 3.30∼6.46 Km, the burst index was 1.36∼3.01 kPaㆍ$m^2$/g, the tear index was 33.0∼57.0 mNㆍ$m^2$/g and the folding endurance was 14∼381 times at most incubating periods, when pulping was done. The physical properties were increased as the incubation duration was extended. Especially, when N and glucose + N were added, the physical properties showed superior results during each incubation period.

Correlation of $^{137}Cs/^{60}Co$ Activity Ratio in Radwaste with Primary Coolant (원자로 냉각재와 방사성폐기물 내 $^{137}Cs/^{60}Co$ 핵종비)

  • In order to compare the correlation of radioactivity ratio between the radwaste streams and the primary coolant of PWR NPPs, A RCS sampling kit was installed to primary coolant system for the collection of the radionuclides during the normal operation of NPPs. RCS samples were collected from PWR type of domestic NPPs through 2004 to 2005, and pretreated with acid microwave digestion or leaching method to assay quantitatively of several interesting radionuclides. The radioactivity ratios of $^{137}Cs\;to\;^{60}Co$ in a filter cartridge and a resin cartridge were 2.3E-2 and 7.3E-1, respectively. At a same period of the reactor operating cycle, the radioactivity ratios of $^{137}Cs\;to\;^{60}Co$ were 6.3E-1 for a evaporator bottom, 6.7E-1 for a spent resin, and 5.6E-2 for a dry active waste, so that these radwaste streams were identified as having similar characteristics with the corresponding RCS samples.

Screening test for Dendropanax morbifera Leveille extracts: in vitro comparison to ox-LDL-induced lipid accumulation, ethanol-induced fatty liver and HMG-CoA reductase inhibition (황칠나무 추출물의 고지혈증 완화 효과 스크리닝)

  • The objective of this study was to compare the antihyperlipidemic effects of different Dendropanax morbifera leaf extracts in vitro. The extracts differed in terms of specimen age, harvesting season, and extraction method. RAW 264.7 cells were pretreated with these extracts and stimulated by oxidized low-density lipoprotein. Ethanol was used to induce toxicity in HepG2 cells. Cellular lipid accumulation was quantified using oil red O staining in both these cells. The extracts were evaluated for their inhibitory effects on 3-hydroxy-3-methylglutaryl coenzyme A (HMG-CoA) reductase. RAW 264.7 cells treated with the 60% ethanol extract of an 8-year-old specimen harvested in November exhibited the lowest lipid accumulation. The 30% ethanol extract of a 5-year-old specimen harvested in May exhibited the greatest protection from ethanol-induced cytotoxicity in HepG2 cells. The hot water extract of an 8-year-old specimen harvested in May showed the greatest inhibition of HMG-CoA reductase. These results showed that D. morbifera extracts prepared from leaves that are harvested in May possess the highest antihyperlipidemic effects.

Characterization of a Multimodular Endo-β-1,4-Glucanase (Cel9K) from Paenibacillus sp. X4 with a Potential Additive for Saccharification

  • An endo-${\beta}$-1,4-glucanase gene, cel9K, was cloned using the shot-gun method from Paenibacillus sp. X4, which was isolated from alpine soil. The gene was 2,994 bp in length, encoding a protein of 997 amino acid residues with a predicted signal peptide composed of 32 amino acid residues. Cel9K was a multimodular enzyme, and the molecular mass and theoretical pI of the mature Cel9K were 103.5 kDa and 4.81, respectively. Cel9K contains the GGxxDAGD, PHHR, GAxxGG, YxDDI, and EVxxDYN motifs found in most glycoside hydrolase family 9 (GH9) members. The protein sequence showed the highest similarity (88%) with the cellulase of Bacillus sp. BP23 in comparison with the enzymes with reported properties. The enzyme was purified by chromatography using HiTrap Q, CHT-II, and HiTrap Butyl HP. Using SDS-PAGE/activity staining, the molecular mass of Cel9K was estimated to be 93 kDa, which is a truncated form produced by the proteolytic cleavage of its C-terminus. Cel9K was optimally active at pH 5.5 and $50^{\circ}C$ and showed a half-life of 59.2 min at $50^{\circ}C$. The CMCase activity was increased to more than 150% in the presence of 2 mM $Na^+$, $K^+$, and $Ba^{2+}$, but decreased significantly to less than 50% by $Mn^{2+}$ and $Co^{2+}$. The addition of Cel9K to a commercial enzyme set (Celluclast 1.5L + Novozym 188) increased the saccharification of the pretreated reed and rice straw powders by 30.4% and 15.9%, respectively. The results suggest that Cel9K can be used to enhance the enzymatic conversion of lignocellulosic biomass to reducing sugars as an additive.

Survey of the oil contaminated level and preliminary field bioremediation test in the Mountain Baegun at Uiwang city (의왕시 백운산 주변 유류 오염도 조사 및 현장 복원 기초실험)

  • The objective of this study was to survey the oil contamination around the Mountain Baegun at Uiwang city to obtain the preliminary data for bioremediation. For measuring the oil concentrations and physical properties from soil, we analyzed BTEX. TPH and pH, organic content, water content, pormeability coefficient, gravity, porosity and used the purge & trap method for analyzing BTEX. Using the Accelerated Solvent Extractor, we pretreated the samples and then analyzed TPH using GC-FID as soon as possible. From the analysis results, maximum concentration of TPH was 24.773mg/kg and BTEX was 101.7mg/kg. The results of TPH at the Mountain Baegun were higher than the enforcement standard of soil contamination(Korea) and the BTEX concentrations were also higher than the advisory standard of soil contamination(Korea). From these results, the Mountain Baegun may requires to remedy the oil-contaminated soil. In addition, we performed the field bioremediation test for five weeks at the Mountain Baegun using the microbial additives that were developed by our laboratory. From the results of the field test, we could find the about 95% of the oil was removed from the contaminated soil in five weeks. So we consider that it is the one of the useful solutions to remedy the oil-polluted site.

  • Background: Nicotinamide adenine dinucleotide glycohydrolase(NADase) is located on the surface of the cells. It is bound by glycosylphosphatidylinositol(GPI)-linkage, which can be cleaved by bacterial PI-specific phospholipase C(PI-PLC). Recently, it was studied that NADase was increased in infected tuberculosis animal, but absolute NADase is uncertainly increased because of high NADase in Mycobacterium tuberculosis. Therefore, we studied pure NADase activity in red blood cells of normal person and patients of tuberculosis. Method: We evaluated the 19 healthy adults and 16 tuberculosis infected patients, and then, the latter cases were evaluated after 3 months antituberculosis therapy. NADase activity was calculated by scintillated counting of cleaved radioactive [carbonyl-$^3H$] nicotinamide Result: NADase activity was $2021.1{\pm}824.0\;pmol/min/10^6$ erythrocytes in healthy adults vs. $3339.0{\pm}1568.0$ in tuberculosis infected patients, and was $3339.0{\pm}1568.0$ in pretreated patients vs. $2238.6{\pm}1013.1$ in same 3 months treated patients. Conclusion: NADase activity of erythrocytes is elevated in tuberculosis infection, and normalized afer antituberculosis therapy. Therefore, we suggested NADase activity as the new diagnostic and therapeutic indicator.

Protective Effect of Jinmu-tang on $H_2O_2$-induced Cell Death in C6 Glial Cells (진무탕(眞武湯)이 $H_2O_2$로 유도된 C6 Glial 세포사에 미치는 영향)

  • Objectives : The purpose of this study was to investigate the mechanism of protective effect of Jinmu-tang (JMT, Zhenwu-tang) extract on $H_2O_2$-induced cell death in C6 glial cells. Methods : Cultured C6 glial cells of white mice were pretreated with JMT extract and exposed to $H_2O_2$ for inducing cell death. We measure the cell viability by 3-[4,5-dimethylthiazol-2-yl]-2,5-diphenyltetrazolium bromide (MTT) assay and investigate the cell morphology using a light microscope after crystal violet (CV) staining. Reactive oxygen species (ROS) formation was analyzed using a flow cytometer and a fluorescent microscope after staining with 2'7'-dichlorofluorescein diacetate (DCF-DA). DNA fragmentation was analyzed using a flow cytometer after propidium iodide (PI) staining and nuclei morphology was investigated using a fluorescent microscope after 2-[4-amidinophenyl]-6-indo-lecarbamidine dihydrochloride (DAPI) staining. We analyzed expression of Bax, processing of procaspase-3 and poly (ADP-ribose) polymerase (PARP), and activation of nuclear factor-${\kappa}B$ (NF-${\kappa}B$) by western blot method. Tumor necrosis factor-${\alpha}$ (TNF-${\alpha}$) secretion was analyzed using Quantikine kit. Results : We determined the elevated cell viability by JMT extract on $H_2O_2$-induced C6 glial cell death. ROS formation, DNA fragmentation, $I{\kappa}B{\alpha}$ phosphorylation, NF-${\kappa}B$ activation, and secretion of TNF-${\alpha}$ induced by $H_2O_2$ are inhibited by JMT extract pre-treatment. JMT extract inhibits Bax expression, processing of caspase-3 and PARP that are critical biochemical markers of apoptotic cell death. Conclusions : These results suggest that JMT extract has a protective effect on $H_2O_2$-induced C6 glial cell death in various pathways.

Thalidomide Combined with Chemotherapy in Treating Patients with Advanced Colorectal Cancer

  • Objective: To assess the safety and effectiveness of thalidomide (produced by CHANGZHOU PHARMACEUTICAL FACTORY CO.LTD) combined with chemotherapy in treating patients with advanced colorectal cancer. Method: A consecutive cohort of pretreated patients with advanced colorectal cancer were treated with thalidomide combined with chemotherapy. And chemotherapy for patients with advanced colorectal cancer were administered according to the condition of patients. Thalidomide was orally administered at a dosage of 50mg/day to 150mg/day before sleeping for at least 14 days. After at least 14 days of treatment, safety and side effects were evaluated. Results: There were 12 female and 3 male patients with advanced cancer recruited into this study, including 9 patients with colon, 6 patients with rectal cancer. The median age of patients was 57(41-82) years. Partial response was observed in 2 patients (2/15), and stable disease in 3 patients(3/15). Incidences of Grade 1 to 2 myelosuppression was observed in 1/15 patients, and Grade 1 to 2 elevation of hepatic enzyme was recorded in 1/15 patients. Adverse effects on the gastrointestinal tract were documented in 1/15 patients, and were Grade 1. No Grade 3-4 toxicities were diagnosed. No treatment related death was found. Conclusions: Thalidomide combined with chemotherapy was safe and mildly effective in treating patients with advanced colorectal cancer. However, further study should be conducted to clarify the effectiveness of this combination.

Effects of Yijintang-gamibang on Reflux Esophagitis Induced by Pylorus and Forestomach Ligation in Rat (역류성식도염 유발 흰쥐에 대한 이진탕가미방(二陳湯加味方)의 효과)

  • Purpose : The object of this study was to observe the suppressive effects of Yijintang-gamibang (YJGMB), Yijintang with Atractylodis Rhizoma, Massa Medicata Fermentata, Hordei Fructus Germiniatus, and Coptidis Rhizoma. YJGMB has been traditionally used in Korean medicine for treating various digestive diseases. We tested it on the rat reflux esophagitis (RE) induced by pylorus and forestomach ligation in rats as compared with omeprazole, a well-known proton pump inhibitor. Method : Three different dosages of YJGMB 200, 100 and 50mg/kg, were orally pretreated once a day for 28 days before pylorus and forestomach ligation. Seven groups, each of 8 rats per group were used in the study. Six hours after pylorus and forestomach ligation, changes of the stomach and esophagus lesion areas, gastric volumes, acid and pepsin outputs, invasive lesion percentages, fundic mucosa and total thicknesses were measured as histomorphometry. The results were compared with omeprazole, antioxidant and proton pump inhibitor, and 30 and 10mg/kg treated groups in which the effects on RE were already confirmed. Results : As results of pylorus and forestomach ligation, marked increases of esophageal and gastric mucosa lesion areas, gastric volumes, acid outputs, pepsin outputs were observed with histopathological changes of RE, such as hemorrhages, ulcerative lesions and edematous changes on the fundic mucosa. However, these pylorus and forestomach ligation-induced RE were dose-dependently inhibited by treatment of 200, 100 and 50mg/kg of YJGMB. YJGMB 200mg/kg showed similar protective effects as compared with 30mg/kg of omeprazole in the present study, and more favorable effects were observed in 50mg/kg of YJGMB treated rats as compared with omeprazole 10mg/kg in the present study. Conclusion : The results obtained in this study suggest that YJGMB has favorable protective effects on the RE induced by pylorus and forestomach ligation. Therefore, it is expected that YJGMB will also show favorable effects on RE corresponding well to the suggestion of traditional Korean medicine. However, more detailed mechanism studies should be conducted in future with the screening of the biological active chemical compounds in herbs.
