Simultaneous determination of preservatives in sausages using liquid chromatography with electrospray ionization tandem mass spectrometry (LC-MS/MS를 이용한 소시지 중 보존료 동시분석)

  • A simultaneous determination method was developed for nine preservatives (benzoic acid, sorbic acid, dehydroacetic acid, methyl-, ethyl-, isopropyl-, propyl-, isobutyl- and butyl-parabens) in sausage by liquid chromatography with electrospray ionization tandem mass spectrometry (LC/ESI-MS/MS). Each parameter was established by multiple reaction monitoring in negative mode. Separation was achieved on a phenyl-hexyl ($2.5{\mu}m$, $2.1{\times}150mm$, Waters) with A-20 mM ammonium acetate containing 0.1% acetic acid in water, B-Acetonitrile as mobile phase with gradient mode at a flow rate of 0.3 mL/min. The developed method was validated for specificity, linearity, accuracy and precision in sausages samples. Linearity was over 0.998 with calibration curve of the mixed standards. The mean recoveries from sausages fortified at the level of 2.0~10.0 mg/L were in range of 98.60~109.16% with RSDs lower than 8.93%. The limits of detection (LOD) and the limits of quantification (LOQ) were in the range between 0.0003~0.085 mg/L and 0.01~0.257 mg/L, respectively. Intra-day precision and inter-day precision were 0.45~6.16% and 2.81~13.33%, respectively. Using presently developed determination method, 33 field sausage samples from Gwangju city in Korea were screened over nine preservatives. As a result, no preservatives were detected in all samples.

  • This study aims to develop a new analytical method to detect zinc pyrithione, the ingredient of cosmetics appointed as restricted ingredients and used as preservatives.. The analytical method was based on data gathered from the relevant literature. Information about the amounts of these ingredients was researched in order to select the base-matrix materials used to validate the analytical method. After selecting and preparing the base-matrix materials, the analytical method was validated by method validation procedures. The analytical method was verified first by inter-laboratory validation and then through analyzing the cosmetics sold in the market. Based on the results of this study, guidelines are proposed for the analysis of restricted ingredients in cosmetics, which will provide a method to test the cosmetics circulating in the Korean market. The use of the proposed guidelines will increase the quality of the cosmetics as well as the safety of human health, which will enhance the competitiveness of the Korean cosmetics industry and lead to an increase in the exportation of cosmetics.

Effects of Applying Microbial Additive Inoculants to Spent Mushroom Substrate (Flammulina velutipes) on Rumen Fermentation and Total-tract Nutrient Digestibility in Hanwoo Steers (팽이버섯 부산물 발효에 따른 한우 거세우 반추위 성상 및 소화율에 미치는 영향)

  • We inoculated a spent mushroom substrate from Flammulina velutipes (SMSF) with a microbial additive and assessed the effects on chemical composition, ruminal fermentation parameters, and total-tract nutrient digestibility. In Exp. 1, three cannulated Hanwoo steers were used in an in situ trial to determine the degradation kinetics of dry matter (DM) and crude protein (CP). In Exp. 2, three Hanwoo steers were randomly assigned to experimental diets according to a $3{\times}3$ Latin square for a 3-week period (2 weeks for adaptation and 1 week for sample collection). Experimental diets included the control diet (3.75 kg/d formulated concentrate mixture + 1.25 kg/d rice straw), SMSF diet (3.19 kg/d formulated concentrate mixture + 1.25 kg/d rice straw + 0.56 kg/d SMSF), and inoculated SMSF (ISMSF) diet (3.19 kg/d formulated concentrate mixture + 1.25 kg/d rice straw + 0.56 kg/d ISMSF). The chemical composition of ISMSF did not differ from that of SMSF. Microbial additive inoculation decreased pH (P<0.05) and improved preservation for SMSF. The percentages of DM, neutral detergent fiber (NDF), and acid detergent fiber (ADF) in ISMSF were slightly lesser than those in SMSF. Ruminal fermentation characteristics and total-tract nutrient digestibility were not affected by diet. Overall, microbial additive inoculation improved preservation for SMSF and may allow improved digestion in the rumen; however, the total digestible nutrients (TDN) of SMSF and ISMSF diets were slightly lesser than the control diet. The ISMSF can be used as an alternative feedstuff to partially replace formulated concentrate feed.

Stability of Anti-Yeast Activities and Inhibitory Effects of Defatted Green Tea Seed Extracts on Yeast Film Formation (탈지 녹차씨 추출물의 항효모 활성 안정성 및 산막 형성 억제능 평가)

  • Water and 75% ethanol extracts were prepared from defatted green tea seeds and evaluated for their anti-yeast activities. The antimicrobial activities of defatted green tea seed extracts (DGTSEs) were tested against food-spoilage bacteria, yeasts, and molds. DGTSEs exhibited antimicrobial activities with minimum inhibitory concentrations of $39{\sim}1,250{\mu}g/mL$ against three bacteria, two molds, and all tested yeast strains. Ethanol extract showed higher antimicrobial activity than water extract. The stability of anti-yeast activities of DGTSEs was examined under different conditions of temperature, pH, and NaCl concentrations. The anti-yeast activities of DGTSEs were stable at pH 3~9, 0~20% NaCl, and $100^{\circ}C$ for 30 min. However, anti-yeast activities of DGTSEs decreased upon heating at $70^{\circ}C$ for 24 h or $121^{\circ}C$ for 15 min. DGTSEs were applied to food models to determine their inhibitory effects on yeast film formation. Water and 75% ethanol extracts were effective in preventing yeast film formation at concentrations more than 156 and $39{\mu}g/mL$ in soy sauce, 156 and $78{\mu}g/mL$ in pickle sauce, and 78 and $39{\mu}g/mL$ in kimchi, respectively.

Chlorphenesin Galactoside Production using Immobilized β-galactosidase-producing Escherichia coli (고정화된 β-galactosidase 생산 대장균을 이용한 chlorphenesin galactoside 생산)

  • Previous research showed that chlorphenesin galactoside (CPN-Gal), a preservative in cosmetics, was safer than CPN against human skin cells [9]. To establish a stable and long-term process for CPN-Gal production, we investigated the repeated-batch process. In this process, β-gal-producing recombinant Escherichia coli cells were immobilized in calcium alginate beads, and CPN was converted to CPN-Gal by the transgalactosylation reaction. The process was conducted in a 300 ml flask, which contained E. coli cell-immobilized alginate beads, 33.8 mM of CPN, and 400 g/l of lactose. The pH and temperature were 7.0 and 40℃, respectively. During the repeated-batch operation, four consecutive batch operations were conducted successfully until 192 hr. The conversion yield of CPN to CPN-Gal was 64% during 192 hr, which was higher than the values in previous reports [3, 13]. Thereafter, however, the conversion yield gradually decreased until the operation was finished at 336 hr. Western blotting of immobilized E. coli cells revealed that β-gal gradually decreased after 192 hr. In addition, alginate beads were cracked when the operation was finished. It is probable that, including this loss of E. coli cells by cracks, deactivation, and product inhibition of E. coli β-gal might lead to a gradual decrease in the production of CPN-Gal after 192 hr. However, as the purification of β-gal is not necessary with β-gal-producing recombinant E. coli cells, β-gal-producing E. coli cells might be a practical and cost-effective approach for enzymatically synthesizing CPN-Gal. It is expected that this process will be extended to long-term production process of CPN-Gal for commercialization.

RGP lens 다목적 용액의 습윤성에 관한 비교 연구

  • We measured the effect of wettability of six MPSs for RGP(rigid gas permeable) lens. The used MPSs(multipurpose solutions) were OPTI-SOAK(ALCON), SOLO care hard(CIBA Vision), Total care(ALLERGAN), Simplicity(BOSTON), Wetting and Soaking Sol.(Bausch & Lomb) and Aquas-multi(Saehan). These MPSs keeps hydrophilic property of lens surface and increase the effect of cleaning or increase the effect of preservative effect. To compare with the effect of wettability we followed the way of contact angle measurement which was general way to measure wettability and compared lens which was conducted by each MPS made by different companies. As a control, 0.9% NaCl solution and artificial tears were used. The degree of the effect of wettability was decided by contact angle. It is hydrophilic property nearby $0^{\circ}$ of contact angle and it is closed by hydrophobic property as it increases. The results showed that every lens was nearby hydrophilic property within $25\;^{\circ}-36\;^{\circ}$. Also, it was differed by various factors. The surface tension showed various differences between 19.8 and 31.3 mN/m. In the viscosity, MPSs represented the highest viscosity between $4^{\circ}C$ and $20^{\circ}C$. It was much higher than compared with the viscosity of soft lens MPS. This experiment could be used to grasping the interaction between solutions used to MPS and the natural endowments of lens and to considering the relations of different factors effecting the wettability.

Purification of Antimicrobial Compounds and Antimicrobial Effects of Schima wallichii subsp. liukiuensis against Candida sp. (Schima wallichii subsp. liukiuensis의 Candida종에 대한 항균효과 및 항균물질의 분리정제)

  • To develop natural antimicrobial substances from Theaceae, Schima wallichii subsp. liukiuensis was selected from 218 woody plants, and antimicrobial compounds against bacteria, fungi, and yeast were isolated. The antimicrobial activity of ethanol extracts proved higher than those of other organic solvents. The antimicrobial activity of S. liukiuensis extract showed no differences in sesonal variation, but, that of plant part was high in bark at autumn. An antimicrobial substance was isolated from the extract of Schima using column chromatography packed with silica gel and sephadex LH-20, and then a purified antimicrobial substance (Compound I) was obtained using HPLC analysis. The Compound I in the analysis of UV, IR, and GC-MS presumed a triterpene or steroidal saponin, ${\alpha}$-sitisterol as aglycon combined three sugars. The minimal inhibitory concentration (MIC) of the Compound I against a bacteria, fungi, and yeast were 1.25 g/L, 5.0 g/L, and 0.040 g/L, respectively. This is much lower than the MIC of hinokitiol, an natural antimicrobial compound used commercially, which suggests that Compound I could be developed as a natural preservative and pharmaceuticals.

  • In order to obtain yeast cells producing a bacteriocin, Subpeptin JM4-A or Subpeptin JM4-B, the 48 bp oligonucleotides corresponding to Subpeptin JM4-A and Subpeptin JM4-B genes including codon for start and stop were chemically synthesized and cloned into pAUR123, an yeast expression vector. Transformed yeast cells exhibited growth inhibition of Bacillus subtilis, Escherichia coli and Pseudonomas aeruginosa. This result indicates that yeast cells producing Subpeptin JM4-A or Subpeptin JM4-B possess bacteriocidal properties against both Gram positive B. subtilis and Gram negative E. coli and P. aeruginosa cells. The recombinant yeast strains constructed in this study can be applied in the food preservative or. animal foodfeed.

Studies on the Preservation of Korean Chestnuts (한국산(韓國産) 밤의 저장(貯藏)에 관(關)한 연구(硏究))

  • To elucidate the preservative characteristics of chestnuts, three varieties, Ungi, Okkang and Chuckpa, were stored in cellar, polyethylen film packing, and box with or without $\gamma-irradiation$, and chemical compositions, rates of sprouting and rotting were determined during the period of storage. The chemical compositions of raw chestnuts were: moisture, 59 to 63%; total sugar, 28 to 29%; reducing sugar, 0.1 to 0.2%; crude fiber, 0.6 to 0.9%; crude protein, 3.5%; vitamin C, 27 to 28mg%; and tannin, 50 to 58mg%. Total sugar and vitamin C were decreased during the period of storage, and of reducing sugar and tannin were increased. The rates after 6 months of storage after three vareitia were; 93 to 100% by the cellar storage; 35 to 57% by the box storage without $\gamma-irradiation$ and 1 to 4% with $\gamma-irradiation$; and none by the polyethylene film packing. The sprouting rate in Chuckpa decreased markedly as compared with the other two varieties. The rotting rates when stored for 6 months were: 4 to 6% by the box storage without $\gamma-irradiation$ and 7 to 12% with $\gamma-irradiation$; 5 to 8% by the polyethylene film packing; and 30 to 54% by the cellar storage. The rotting rate in Chuckpa was lower than the other varieties during the period.

Antimicrobial Activities of Botanical Antimicrobial Agent-Grapefruit Seed Extract Mixture for the Preparation of Anitimicrobial Packaging Paper (항균포장지 제조용 식물성 자몽종자추출물제재의 항균특성)

  • Botanical antimicrobial agent-grapefruit seed extract mixture(BAAG), which could be applied to the preparation of antimicrobial packaging paper, was investigated in order to prove the preservative function of fruits and vegetables. HAAG showed remarkable antimicrobial effects against Fusarium solani Botrytis cinerea, Pencillium crustosum, Erwinia carotovora, Phoma destructiva and Alternaria radicina causing the postharvest decay of fruits and vegetables. We have examined that HAAG could inhibit the growth of microorganims when treated with more than 500 $\mu$g/mL concentration. The activities of HAAG were stable in the wide spectrum of pH and temperature. Direct visualization of microbial cells by using scanning electron microscope showed the loss of microbial cell membrane function, which was destroyed by treating with the dilute solutions of HAAG. We could confirm that HAAG be an antimicrobial agent for the preparation of antimicrobial packaging paper.
