Effect of diluent variation on cryopreservation of large yellow croaker Larimichthys crocea

  • The aim of this research was to investigate different factors, including cryoprotective agents (CPAs), diluents, dilution ratios, equilibrium times, freezing rates, and thawing methods to optimize cryopreservation protocols for large yellow croaker (Larimichthys crocea). The parameters evaluated were sperm motility, sperm activity index (SAI), survival rate, and DNA damage. Different types of CPAs, such as dimethyl sulfoxide (DMSO), propylene glycol (PG), ethylene glycol (EG), methanol, and glycerol, were tested for sperm preservation. The highest motility, SAI, and survival rate were observed when EG was used. Different diluents such as Stein's solution, Hank's balanced salt solution, marine fish Ringer's solution, artificial seminal plasma (ASP) of small yellow croaker, and Cortland solution were investigated. The highest post-thaw motility was observed upon using ASP as the diluent. Different concentrations of EG were then mixed with ASP to identify the optimal EG concentration. Experimental results showed that the motility (70.33 ± 1.20%), SAI (5), and survival rate (78.30 ± 0.42%) of post-thaw sperm were optimum when 10% EG and ASP were used as the CPA and diluent of cryopreservation, respectively. Post-thaw sperm motility was high at equilibration times below 150 s and at an optimum dilution ratio of 1:1 (sperm: CPA + diluent) and was not significantly different compared with fresh sperm motility. The freezing rate was found to be slow below -10℃/min. The thawing temperature of 45℃ was identified as ideal. The percentage of tail DNA in post-thaw sperm at 10% EG and ASP was also investigated and was found to have more significant DNA damage than that in fresh sperm but significantly lower damage than that in post-thaw sperm at EG concentrations of 5%, 15%, and 20% (p < 0.05). The cryopreservation protocols obtained in this study will be useful in large yellow croaker hatcheries.

Knowledge and Perceptions of the End of Life among Tunisian Medical and Paramedical Staff

  • Purpose: End-of-life (EOL) care is a vulnerable period in an individual's life. Healthcare professionals (HPs) strive to balance the preservation of human life with respect for the patient's wishes. The aims of our study were to assess HPs' knowledge and perceptions of EOL care and to propose areas of improvement to improve the quality of care. Methods: We conducted a single-center, cross-sectional study involving HPs from a university hospital who encountered EOL care situations. We used a questionnaire divided into four sections: knowledge, practice, perception, and training. We calculated the rate of correct answers and the collective competence index. Results: Eighty-six questionnaires were analyzed, with 82.5% (71/86) completed by medical respondents and 17.5% (15/86) by paramedical respondents. Most of the respondents, 71.8% (51/71), were interns and residents. The study focused on palliative care, medical assistance in dying, aggressive medical treatment, and euthanasia, finding adequate knowledge in the first three areas. Respondents assigned to the intensive care unit and those with more than 8 years of experience had significantly higher correct answer rates than their counterparts. Seventy-five percent of respondents (65/86) reported feeling that they had little or no mastery of EOL care, primarily attributing this to insufficient training and the unavailability of trainers. Conclusion: Based on the findings of our study, which we believe to be the first of its kind in Tunisia, we can conclude that HPs possess an acceptable level of knowledge regarding EOL care. However, they require more exposure and training to develop expertise in this area.

Manufacturing of Wine with Korean Figs(Ficus carica L.) and Quality Improvement by Adding Fig Leaves (국내산 무화과를 이용한 와인 제조 및 품질 개선 효과)

  • Fig(Ficus carica L.) which belongs to Moraceae family has been used as digestion promoter, cure for ulcerative inflammation and eruption in Korea. Figs have very short preservation period and can be kept only 1 week even in the refrigeration system. The possibility of wine-making from Korean figs was tested by analyzing the fermentation characteristics, proximate analysis and free sugar contents of wine made of slices with 20% sugar addition. Ethanol fermentation of the figs by Saccharomyces cerevisiae was done rapidly. The ethanol contents, pH, total acidity and sugar content were 12.1%, 3.91, 0.42% and 9.9 °Brix, respectively. The results from the sensory evaluation were as follows. The wine made from the sliced figs were better than that from fig juices in their sensory scores. The wine made with frozen figs was not different from that made with fresh ones. To improve the quality of wine, the addition of dried leaf powder in the concentration of 100 ppm were tried and found it was superior to the one made without leaf.

  • This paper researches, by looking into visitors of the Garden of Morning Calm, Pyunggang Botanical Garden, Jangheung Natural Arboretum and the Garden of Wild Flower, how they use these arboretums and what are the facilities they like most to provide directions in formation and planning of domestic private arboretums. The followings are executive summery of this research. It was researched that most of the them visited arboretums for emotional life (42.93%). They answered that the major reason that they go to arboretums is rest and service functions (42.97%). Such number suggest arboretums that they should provide, along with their original purpose of biological preservation, practical purposes, including rest and service functions. When asked facilities they preferred most, most visitors answered that they are most satisfied with facilities like restaurants and cafeterias. Information desk, management office, forest museum, wild life zoo, green house, exhibition room, rest area, pond and fountains influenced the visitor satisfaction level as well. Regarding to facility references, visitors most preferred information desk, management office, restaurant and cafeteria.

How to Keep the Sustainability of the Landscape Resources of the East Coast in South Korea

  • The purpose of this study is to present the conditions and methods for keeping the sustainability of the landscape resources of the East Coast of Gangwon province, the Republic of Korea by investigating the landscape resources management in the area and pointing out its problems. The unit of analysis in this study is four cities and two counties located along the national road route seven in Gangwon province. The classification and characteristics of the landscape resources in this area was analyzed by a literature review, and we surveyed the tourists visiting the area and statistically analyzed the data in order to examine their satisfaction with the landscape resources management and make recommendations. The problems of the landscape resources management are: 1) the disturbance of the persistence of life by reclamation, the population reduction in the ecosystem due to the overload in environmental capacity, and the severance of space between land and water. 2) the reduction of the benefits from indirect experience by interfering with the conservation of fluxes --- the manipulation of horizontal arrangement of the landscape resource, the visual disturbance by the construction of high-storied buildings, and the disharmony between the color/image and the environment. The means for keeping sustainability of the landscape resources include the regulations of development and use, the change in the recognition of the value of landscape resources and the moral system, and the improvement of resource management skills.

Optimization of Pan Bread Prepared with Ramie Powder and Preservation of Optimized Pan Bread Treated by Gamma Irradiation during Storage

  • This study was conducted to develop an optimal composite recipe for pan bread with ramie powder that has high sensory approval with all age groups and to estimate the DPPH radical scavenging activity and the pan bread shelf life after gamma irradiation. The sensory evaluation results showed significant differences in flavor (p<0.05), appearance (p<0.01), color (p<0.01), moistness (p<0.01), and overall quality (p<0.05) based on the amount of ramie powder added. As a result, the optimum formulations by numerical and graphical methods were calculated to be as follows: ramie powder 2.76 g (0.92%) and water 184.7 mL. Optimized pan bread with ramie powder and white pan bread were irradiated with gamma-rays at doses of 0, 10, 15, and 20 kGy. The total bacterial growth increased with the longer storage time and the least amount of ramie powder added. Consequently, these results suggest that the addition of ramie powder to pan bread provides added value to the bread in terms of increased shelf life.

  • In this study, we examined the effects of different concentrations of red pepper seed (0%, 1%, 3%, 5%, and 7%) on pH, total acidity, color, reducing sugar content, total microbes, lactic acid bacteria, and sensory quality, in terms of prolonging the shelf-life of kimchi. The additions of red pepper seed resulted in higher pH levels, while total acidity was lower than that of the control. The kimchi with red pepper seed had higher reducing sugar contents than the control. Furthermore, the kimchi with red pepper seed showed higher b-values than the control. During fermentation, the amounts of lactic acid bacteria and total microbes were higher in the kimchi with red pepper seed than in the control, and the higher ratios resulted in higher quality kimchi. The additions of red pepper seed also resulted in changes of color, flavor, taste, and texture, having a large influence on overall product quality. In conclusion, in terms of preservation and consumption, the 3% and 5% red pepper seed concentrations offered enhanced shelf-life and better quality kimchi products.

  • The purpose of this study is to find out the color preference on rural rooftop color. Today, public profit value of a rural community has been heavily recognized in the areas of preservation of pro-environmental and traditional life styles and foundation of collective community. Comparatively with urban, rural town conserves the scenic of those days when the historic landscape existed. Therefore, elements of historic and tradition in rural town are interesting resources to people who want traditional experience. Color is one of the most influenced elements for the image of Landscape. On the other hand, radical modernization has brought conflicting color element that does not blend with existing environmental color. Among others, roof represents one of the important traits of rapidly developing rural community due to vast amount of space it covers. In order to come up with more appropriate coloring scheme, it is necessary determine color that forms a balance with present conditions of rural life and historic landscape. This study provides more objective rooftop coloring selection process by employing additional surveys regarding color image adjective. I justified necessity of the balance between environmental color and roof color through analyzing correlation between environmental color of the rural town and Color Image Scale of the preliminary selection of roofs.

Therefore, it is required more safety medical power distribution system in order to protect patients and physicians more securely from unexpected leakage of current.   • The annals of Joseon Dynasty has been known as one of the UNESCO's Memory of the World Heritage. The annals of Joseon Dynasty composed with two kind of books, wax coated book and non coated book. Especially the waxed book have been severely damaged by various reason. For the safety preservation of the annals of Joseon Dynasty waxed book, the thermal ageing mechanism of beeswax coated Hanji paper has been evaluated. The weight loss of waxed Hanji were rapidly increased until 30 days in three temperatures(105, 120 & $150^{\circ}C$) and stabilized after 30 days. The acid value and relative intensity of carbonyl groups in beeswax were increased with strong conditions. This means that the deterioration of beeswax should be progressed with oxidizing reaction. The physical strength and the crystallinity of dewaxed Hanji were decreased with ageing time.
