Acute carbohydrate engorgement in the Korean indigenous cattle, a case report (한우에서 급성탄수화물 과식증에 의한 폐사 증례)

  • This case occurred following accidental consumptions of toxic levels of feedstuffs in the Korean indigenous cattle which were pregnant for 3-6 months. Grossly, poorly defined bluish coloration in the ventral sac of the rumen, reticulum and omasum was visible on the surface of the stomach. The severely affected ruminal walls were perforated and blood-tinged exudate was observed and death followed due to acute diffuse peritonitis. There were copious amount of porridgelike rumen contents, which has a distinct fermentative odor. The epithelia were easily wiped off and hyperemic in patch. Histopathologically, chemical rumenitis with neutrophilic infiltration and opportunistic fungal infection was evident in the mucosa and submucosa. Multifocal necrotic foci with fungal hyphae on the liver and severe congestion on the renal medulla were noted. There was no evidence of heavy metal and aflatoxin Bl in the suspected feedstuffs. This case was diagnosed as acute carbohydate engorgement in cattle.

Brucellosis outbreak of Korean indigenous cattle at Yeongwol and Pyeongchang county in Korea

  • This study was attempted to investigate the properties of brucellosis in Korean indigenous cattle at the Yeongwol and Pyeongchang county. Brucella spp was differentiated and identified from cotyledons, amniotic fluids and supramammary lymph nodes which confirmed with clinical, serological, epidemiological evidences (69 cases) from January to June, 2004. Isolation frequency of this causative agent from supramammary lymph nodes, cotyledons and amniotic fluids from 38 pregnant Korean indigenous cattle were $39.1\%,\;87.5\%,\;and\;63.2\%$, respectively, and finally confirmed with Brucella abortus biotype 1 through biochemical and serological test. A Brucella specific DNA with 711bp band was detected by PCR assay using BCSP primer. The two cases were definite epidemiological evidences that infected Korean indigenous cattle acrossed the border to Yeongwol and Pyeongchang from near two provinces. Effective prevention programs are urgently needed for further spreading this epidemics.

Proteomic Analysis of Bovine Pregnancy-specific Serum Proteins by 2D Fluorescence Difference Gel Electrophoresis

  • Two dimensional-fluorescence difference gel electrophoresis (2D DIGE) is an emerging technique for comparative proteomics, which improves the reproducibility and reliability of differential protein expression analysis between samples. The purpose of this study was to investigate bovine pregnancy-specific proteins in the proteome between bovine pregnant and non-pregnant serum using DIGE technique. Serums of 2 pregnant Holstein dairy cattle at day 21 after artificial insemination and those of 2 non-pregnant were used in this study. The pre-electrophoretic labeling of pregnant and non-pregnant serum proteins were mixed with Cy3 and Cy5 fluorescent dyes, respectively, and an internal standard was labeled with Cy2. Labeled proteins with Cy2, Cy3, and Cy5 were separated together in a single gel, and then were detected by fluorescence image analyzer. The 2D DIGE method using fluorescence CyDye DIGE flour had higher sensitivity than conventional 2D gel electrophoresis, and showed reproducible results. Approximately 1,500 protein spots were detected by 2D DIGE. Several proteins showed a more than 1.5-fold up and down regulation between non-pregnant and pregnant serum proteins. The differentially expressed proteins were identified by MALDI-TOF mass spectrometer. A total 16 protein spots were detected to regulate differentially in the pregnant serum, among which 7 spots were up-regulated proteins such as conglutinin precursor, modified bovine fibrinogen and IgG1, and 6 spots were down-regulated proteins such as hemoglobin, complement component 3, bovine fibrinogen and IgG2a three spots were not identified. The identified proteins demonstrate that early pregnant bovine serum may have several pregnancy-specific proteins, and these could be a valuable information for the development of pregnancy-diagnostic markers in early pregnancy bovine serum.

Studies on Embryo Cryopreservation and Twinning by Embryo Transfer of Korean Native Cattle I. Transfer of Frozen-thawed Embryos and Production of Calves (한우 수정란의 동결보존 및 쌍자생산에 관한 연구 I. 동결 수정란의 이식과 자우 생산)

  • This study was carried out to establish the techniques for producing the calves of genetically superior Korean Native cattle by transfer of frozen-thawed embryos. The effects of some factors related to embryo recovery following superovulation and pregnancy rate following transfer of frozen-thawed embryos were evaluated. Also calving state was investigated. The results obtained were as follows ; The mean number of total and transferrable embryos recovered per superovulated cow was 8.72 and 4.90, respectively, from a total of 72 superovulations using 34 donor cows. There were no significant differences in the number of total or transferrable embryos recovered per superovulated cow between products of follicle stimulating hormone (FSH), years, seasons, and collection numbers. The pregnancy rate was found 44.44% following transfer of frozen-thawed embryos of Korean Native cattle to a total of 180 recipient cows including 82 Angus, 27 Charolais, 62 Hereford and 9 Korean Native cows. The pregnancy rate was significantly (P<0.05) higher in the transfer of excellent (42.99) and good embryos (40.17%), compared with fair (5.90%) grade embryos. And the pregnancy rate was significantly (P<0.05) higher in the transfer of embryos of morula stage (43.86%) than blastocyst stage (15.51%). But there were no significant differences in pregnancy rates between natural and induced estrus estrus asynchrony of 1 days, breeds, and parities of recipient cows. The normal calving rate of 80 pregnant cows following transfer of frozen4hawed em-bryos was 87.5% and the other 10 pregnant cows showed abortion during the period from pregnancy diagnosis at 50~60 days to calving. The average gestation length of normally delivered recipients was 288.50 days and the average birth weight of 70 calves born was 24.22 kg. The gestation length was significantly (P<0.05) shorter in the recipients delivering female calves (286.70 days) than males (289.39 days). But there were no significant differences in gestation tength and birth weight of calves born between the recipient breeds.

Transfer of Korean Native Cattle Embryos to Beef Recipients (한우 수정란의 육우 이식에 관하여)

  • This study was carried out to propagate Korean native cattle using beef recipients by embryo transfer. Seven Korean native cattle donors were superovulated with FSH 32mg and Embryos collected from donors were frozen and preserved in National Animal Breeding Institute. Frozen-thawed embryos were transferred to synchronized 40 beef recipients nonsurgically in Daekwanryeong Branch of National Animal Breeding Institute. The results obtained were as follows : 1. Total ova and transferable embryos per donor were 11.4 and 11.1 from 7 donors, respectively. 2. Among 40 recipients transferred with frozen-thawed embryos, 20 were pregnant(50.0%). 3. The pregnancy rate according to time from embryo thawing to transfer was higher when transferred within 3 hours than after 3 hours(57.6% vs. 14.3%). 4. The cow recipients showed slightly higher pregnancy rate than the heifer(53.3% vs. 48.0%). 5. Two grade embryos showed higher pregnancy rate than 1 grade(66.7% vs. 45.2%).

Ovarian potential of Cameroonian Zebu cattle (Bos indicus) slaughtered in Etoudi - Yaoundé

  • Background: This study helps to evaluate the Ovarian potential of Cameroonian Zebu cattle (Bos indicus) slaughtered in Etoudi - Yaoundé for implementing Assisted Reproductive Techniques (ARTs). The aim was to enhance the productivity of the cattle sub-sector in Cameroon while conserving local genetic resources. Methods: A total of 144 cows, including 94 cycled cows and 50 pregnant cows, were included in the study. Live weights were determined by measuring the thoracic perimeter of each cow using a Rondo measuring tape. Age was determined postmortem through examination of dentition and horns, while the uterus and ovaries were removed, weighed, and classified based on physiological status (pregnant or nonpregnant). Follicles were counted, and their diameters were measured and categorized into small (Ø < 3 mm), medium (Ø 3-8 mm), and large (Ø > 8 mm). Results: The results revealed an average follicular population of 32.02 ± 9.31 per cow, with 22.43 ± 8.45 small follicles, 8.42 ± 3.87 medium follicles, and 0.76 ± 0.34 large follicles. The weight of the right ovary was significantly higher than that of the left ovary (p < 0.05), and cows aged 6 to 9 years exhibited a higher number of follicles compared to other age groups. Cows with medium (BCS = 3) and large (BCS = 4-5) body reserves had the heaviest ovaries. Additionally, pregnant cows had heavier uteri compared to non-pregnant cows, and cows with a body condition score of 3 or higher had higher uterine weights than lean cows (BCS = 1-2). Conclusions: This study demonstrates that age, body condition score, and pregnancy status influence ovarian weight. Body Condition Score serves as a reliable indicator of cow nutritional status, and cows with BCS of 3-5 are excellent candidates for in vitro production of embryos.

A histological study on the development of scapula in the Korean native cattle (한우 태자의 견갑골 발생에 관한 조직학적 연구)

  • This study was undertaken to establish the sequence of development of ages and its time of the fetal endochondral ossification in the scapula of the Korean native cattle. This study was also designed to confirm through histological observation the earliest stages of both chondrification and ossification. Thirty eight scapulae, a series of embryos and fetuses from the pregnant Korean native cattle ranging from 11 to 110mm in crown-rump (C-R) length, were used. The following results were obtained. The ossification center was observed in the supra- and infra- spinous fossa in the 5th group (CRL 51-60mm), that was markedly ossified in the 6th group (CRL 61~70mm) by Alizarin red S stain. The chondrogenic center of scapula was observed in the 1st group (CRL 11~20mm). The primary ossification center was presented in the 4th group (CRL 41~50mm). In the 5th group(CRL 51~60mm), the endochondral ossification progressed actively. Alcianophility was markedly increased in the interterritorial matrix in the 3rd group (CRL 31~40mm. However this reaction was markedly decreased in the interterritorial matrix the adjacent portion to the marrow cavity and trabecula in the 5th group (CRL 51~60mm).

Ultrasonographic Diagnosis for the Treatment of Genital Disease and Early Pregnancy Diagnosis in Korean Native Cattle (한우에서 생식기질환의 치료 및 조기임신진단을 위한 초음파영상진단)

  • Ultrasonographic diagnosis of genital disease and early pregnancy diagnosis was performed in Korean native cattle. The size of ovarian follicle in preovulation, luteal stage and follicular cyst was 18.9, 9.2 and 27.6 mm, respectively, and the thickness of follicular wall was 2.3, 1.8 and 2.8 mm, respectively. The size of corpus luteums in formation stage, activity stage, regression stage, cystic corpora lutea and luteal cyst was 6.2, 11.3, 8.6, 26.7 and 25.9 mm, respectively. The thickness of luteal wall in cystic corpora lutea and luteal cyst was 8.4 and 4.9 mm, respectively. The size of embryo or fetus on day 25, 27, 30, 35, 40, 45 and 50 was 0.8, 0.9, 1.3, 1.5, 2.2, 2.8 and 3.8 cm, respectively. The size of amniotic vesicle on day 25, 27 and 30 was 1.2, 2.1 and 3,0 cm, respectively. The diameter of pregnant uterus on day 25 and 27 was 7.0 and 7.8 cm, respectively. It was concluded that the ultrasonographci values determined in this study can be used as references for the treatment of genital disease and early pregnancy diagnosis in Korean native cattle.

Studies on the Improvement of Precision in Early Pregnancy Diagnosis by Milk Progesterone Test(EIA) in Dairy Cows (Milk Progesterone Test(EIA)에 의한 소의 임신조기판단 정확도 향상에 관한 연구)

  • These experiment was carried out to improve the precision of early pregnancy diagnosis in dairy cattle. Changes in progesterone concentration of milk were measured by Enzyme Immunoassay(EIA) in 73 cows up to 21 days after insemination. The average concentraton of progesterone in milk was 1.9ng/ml at eatrus ; it increased to 17.8ng/ml on day 14, and thereafter it declined to 4.3ng/ml on day 21 in nonpregnant cows. Whereas in pregnant animals, it was maintained and elevated further to 22.2ng/ml on day 21. The accuracy of the pregnancy diagnosis for animals classified as positive (pregnant ; over 10ng/ml and decreasing rate<1.5) and negative (non-pregnant ; under 7ng/ml and decreasing rate>1.5) were 95% and 100% respectively. The samples ranging between 7ng/ml and 10ng/ml were classified as positive (decreasing rate<1.5) and negative(>1.5), which accuracy was 54.5% and 100% respectively. However this range appears to be of the most interest for the veterinary practitioner since cows in proestrus or early interestrus tend to have milk progesterone levels within this value. Causes for the insufficient precision of false pregnancy diagnosis are discussed.

Proteomic Analysis of Plasma Protein during Early Pregnancy in Korean Native Cattle (Hanwoo) (한우의 임신 초기 혈장단백질 발현 양상 분석)

  • This study was performed to identify and analyze the specifically expressed plasma proteins during early pregnancy in both pregnant and non-pregnant Hanwoo. Blood samples were collected at 0 (the day of AI), 2, 3, 4, 7, and 11 weeks after AI from pregnant (n=3) and non-pregnant (n=4) Hanwoo, respectively. The hematological parameters were measured. After 2-dimensional electrophoresis using serum, normalized protein spots were selected for the significant expression variation deviated over two fold in its expression level between two groups. Among 17 spots selected, 15 were identified as albumin, IgG1 heavy chain constant region, haptoglobin, ferrochelatase, fibrinogen, hemopexin. 5 spots were expressed only in non-pregnant specific. The spot identification of 1105 and 6106 was decreased after 3 weeks from AI. However, 2/17 spots were still unidentified. Further studies are needed to analyze the function of the proteins associated with early pregnancy.
