In this study, sediment transfer and precipitation analysis of the test bed watershed was conducted through the model for the application and practical use of the urban sediment disaster prevention technology, and used this as an aid to design to secure reliability. In addition, conducted the test bed monitoring with the defense technology, analyzed the effect, and established the maintenance plan. Analyzed the change of soil deposition volume through arbitrary slope adjustment for the currently installed stormwater conduit of the test bed watershed. As a result, it is important to reduce the total sedimentation amount in the adjustment of the slope of the entire pipeline, but it is important that the sedimentation depth of each sediment does not rise to such a degree as to threaten the performance of the pipeline. Considering these matters, it is necessary to design the pipeline to prevent the clogging of the soil from the viewpoint of the reliability of the entire pipeline. The sediment disaster defense technology test bed is divided into a new city and an old city, and old city test bed is under construction. The result obtained through the monitoring of the test bed in the new city, sediment disasters such as debris can delay the time to reach the downtown area, and it is possible to secure the golden time, such as evacuation and rescue through the warning system. Also, the maintenance of the test bed application was suggested. Continuous and systematic monitoring is required for securing the reliability of element technology and successful commercialization.
Phytic acid known as an antinutritional facfor was studied for its effect on the solubility and digestibility of high-phytate and low-phytate soy protein isolates (SPI) obtained by two different methods of pH adjustment. Phytic acid content was 2.48% in high-phytate SPI and 0.72% in low-phytate SPI. Solubility of soy proteins was higher in low-phytate SPI than in high-phytate SPI at all pH values tested and it was lowered by adding more phytic acid to result in precipitation of the proteins. The inhibitory effect of phytic acid toward pepsin digestion of SPI increased by the increasing amount of phytic acid added and its effect was slightly higher in high-phytate SPI than in low-phytate SPI.

Isolating total DNA from small samples using traditional methods is difficult and inefficient mainly due to loss of DNA during filtration and precipitation. With advances in molecular pathology, DNA extraction from micro-dissected cells has become essential in handling clinical samples. Genomic DNA extraction using small numbers of cells can be very important to successfully PCR amplify DNA from small biopsy specimens. We compared our experimental genomic DNA extraction method (A) with two other commercially available methods: using spin columns (B), and conventional resins (C), and determined the efficacy of DNA extraction from small numbers of cells smeared on a glass slide. Approximately 50, 100, 200, 500 and 1000 cells were isolated from fine needle aspiration biopsy (FNAB) slides aspirated from histologically proven papillary thyroid carcinoma masses. DNA was extracted using the three techniques. After measuring DNA quantity, PCR amplification was performed to detect the ${\beta}$-globin and $BRAF^{V600E}$ gene mutations. DNA extracted by method (A) showed better yield than the other methods in all cell groups. With our method, a suitable amount of genomic DNA to produce amplification was extracted from as few as 50 cells, while more than 100 to 200 cells were required when methods (B) or (C) were applied. Our genomic DNA extraction method provides high quality and improved yields for molecular analysis. It will be especially useful for paucicellular clinical samples which molecular pathologists often confront when handling fine needle aspiration cytology, exfoliative cytology and small biopsy specimens.
We studied a selective recovery condition of vanadium (V) from FALL (Fly Ash Leach Liquor) produced at a fossil fuel power station using heavy oil. By applying a spectroscopy to quantify the V in a sample, we identified a concentration range V interfered by on presence of metals such as Ni, Fe Also, the optimal vanadium precipitation rate according to the amount of 5.0M $NH_3$ loaded to the sample, solution pH and stirring time. As a result of the experiment, the maximum selective recovery ratio of V was achieved to be higher than 91.5% when the stirring duration was less than 1 minute at pH 7.0, and $25^{\circ}C$.
This study is intended to clarify the main microstructural factors that contribute to an increase of hardness during isothermal aging in Mg-Al alloy. For this work, Mg-9.3%Al alloy specimens were solution-treated at 688 K for 24 h followed by water quenching, and then aged at 473 K for up to 24 h. The aging at 473 K yielded nodular discontinuous precipitates (DPs) with (${\alpha}+{\beta}$) lamellar morphology at the grain boundaries, and the volume fraction of DPs increased from 0% to ~30% with increasing aging time up to 12 h. For the aging times longer than 12 h, further formation of DPs was substantially inhibited owing to the occurrence of significant continuous precipitation within the ${\alpha}-(Mg)$ matrix, and the density of continuous precipitates (CPs) becomes greater with increasing aging time. Hardness of the specimen was steadily increased with aging time up to 24 h. Microstructural examination on the aged specimens revealed that the increased overall hardness at the early stage of aging is associated with the increased volume fraction of DPs, but at the later stage of aging, where the amount of DPs was hardly changed, the increased hardness of the ${\alpha}-(Mg)$ matrix in response to the higher density of CPs within the matrix, plays a key role in increasing the overall hardness value.

To establish an aging management plan considering seawater influx and changes in groundwater within nuclear power plant sites, the characteristics of groundwater flow must be understood. This study investigated the characteristics of groundwater flow within the site and analyzed groundwater level recorded by monitoring wells to evaluate groundwater flow characteristics and elements that affected these characteristics for supplying the information to conduct the appropriate aging management for ensuring the safety of the safety-related structures in Shin Kori Unit 1 and 2. The increase in groundwater level during the wet season results from high sea-level conditions and the large amount of precipitation. As a result of the analysis of groundwater distribution and change characteristics, the site could be divided into a rainfall-affected area and a tide-affected area. First, the rainfall-affected area can further be divided into areas that are affected simultaneously by excavation, backfill, and a permanent dewatering system. Secondly, areas that are not affected by excavation, or the dewatering system, or by structure arrangement and excavation. Analysis of the spectrum for wells affected by tides resulted in confirmation of the M2 component (12.421 hr) and S2 component (12.000 hr) of the semidiurnal tides, and the O1 component (25.819 hr) of the diurnal tides. In the cross-correlation results regarding tides and groundwater levels, the lag time occurred diversely within 1-3 hours by the effect of the well location from sea, the distribution of the backfill material with depth, and the concrete structure.
Effects of climate change on groundwater system requires understanding the groundwater system in temporal and spatial scales through the long-term monitoring. In this study, the spatio-temporal variations of groundwater were analyzed through the continuous observation of water level, electrical conductivity (EC) and water temperature with automatic data-loggers and sampling in a Gwangneung catchment, Korea, for the four years from 2008 to 2011. Groundwater monitoring were performed at the nest-type wells, MW1 and MW2, located in upsteam and downstream of the catchment, respectively. During the survey period, both the total amount of annual precipitation and the frequency of concentrated rainfall have increased resulting in the elevation of runoff. Water level of MW1 showed no significant fluctuations even during the rainy season, indicating the confined groundwater system. In contrast, that of MW2 showed clear seasonal changes, indicating the unconfined system. The lag-time of temperature at both wells ranged from one to three months depending on the screened depths. Results of chemical analyses indicated that major water compositions were maintained constantly, except for the EC decreases due to the dilution effect. Values of the stable-isotope ratios for oxygen and deuterium were higher at MW2 than MW1, implying the confined system at the upstream area could be locally developed.
Chelating agents have been proposed to improve the efficiency of phytoextraction of heavy metal hyperaccumulator. However, little studies to elucidate mechanism of chelating agents to increase cadmium (Cd) extractability have been conducted. The objectives of this study were to evaluate effect of different chelating agents on Cd extractability and to determine mechanism of Cd mobilization affected by these agents. An arable soil was spiked with inorganic Cd ($CdCl_2$) to give a total Cd concentration of $20mgCdkg^{-1}$. Ethylenediaminetetraacetic acid (EDTA) and citric acid (CA) were selected and mixed with the arable soil at the rates of 0 and $5mmolkg^{-1}$. The mixture soils were incubated at $25^{\circ}C$ for 4 weeks in dark condition. Concentration of F1 Cd fractions (water soluble) significantly increased with addition of EDTA but did not changed with addition of CA. Especially; concentration of F5 Cd fractions (residual) significantly increased with addition of CA. Increase in water soluble with EDTA might be attributed to complexation of Cd and EDTA. Dissolved organic carbon concentration significantly increased with EDTA addition, but did not with CA implying that considerable amount of CA was decomposed to inorganic carbon by microorganism. Log activity of carbonate ($CO_3{^{2-}}$) which might be generated from CA increased with addition of CA. Increase in residual Cd fraction might be due to precipitation of Cd as $CdCO_3$. As a result, EDTA was effective in increasing Cd extractability, by contrast CA had significant effect in reducing Cd extractability.

Winter wheat is susceptible to several fungal pathogens throughout the growing season and foliar fungicide application is one of the strategies used in the management of fungal diseases in winter wheat. However, for fungicides to be profitable, weather conditions conducive to fungal disease development should be present. To determine if winter wheat yield response to fungicide application at the flowering growth stage (Feekes 10.5.1) was related to the growing season precipitation, grain yield from fungicide treated plots was compared to non-treated plots for 19 to 30 hard red winter wheat cultivars planted at 8 site years from 2011 through 2015. At all locations, Prothioconazole + Tebuconazole or Tebuconazole alone was applied at flowering timing for the fungicide treated plots. Grain yield response (difference between treated and non-treated) ranged from 66-696 kg/ha across years and locations. Grain yield response had a positive and significant linear relationship with cumulative rainfall in May through June for the mid and top grain yield ranked cultivars ($R^2=54%$, 78%, respectively) indicating that a higher amount of accumulated rainfall in this period increased chances of getting a higher yield response from fungicide application. Cultivars treated with a fungicide had slightly higher protein content (up to 0.5%) compared to non-treated. These results indicate that application of fungicides when there is sufficient moisture in May and June may increase chances of profitability from fungicide application.
Mobility reduction of 2,4,6-trinitrotoluene (TNT) was tested by amending monopotassium phosphate (MKP) and montmorillonite to a firing range soil contaminated with TNT. While addition of MKP enhanced sorption of TNT on soil matrix, and combined use of MKP with montmorillonite significantly decreased desorption of TNT as well as remarkably increased the TNT sorption. Montmorillonite amendment by 5% of soil mass resulted in TNT desorption of 0.12 mg/kg from soil loaded with 9.93 mg/kg-TNT. The decrease of TNT desorption was proportional to the amount of montmorillonite amended. At 10 and 15% amendment, only 0.79 and 1.23 mg/kg-TNT was desorbed from 29.33 and 48.80 mg/kg-TNT. In addition, the leaching of TNT with synthetic precipitation leaching procedure (SPLP) and hydroxypropyl-${\beta}$-cyclodextrin (HPCD) decreased, indicating that TNT in MKP/montmorillonite-treated soil became more stable and less leachable. The results demonstrate that addition of MKP and montmorillonite to TNT-contaminated soil reduces the mobility of TNT from soil not only by increasing TNT sorption, but also decreasing TNT desorption. It was found that MKP and montmorillonite amendments by 5 and 10% of soil mass, respectively, were optimal for reducing the mobility of soil TNT.
