Application of Gel-based Proteome Analysis Techniques to Studying Post-mortem Proteolysis in Meat

  • This study was conducted to evaluate the possible application of 2 D-SDS-PAGE (2 DE)-based proteome analysis techniques to the assessment of extreme proteolysis in postmortem skeletal muscle. Eight Hanwoo longissimus muscles were incubated immediately after slaughter for 24 h at 5$^{\circ}C$, 15$^{\circ}C$ or 36$^{\circ}C$. Warner Bratzler (WB)-shear force and ultrastructural configuration were determined at 24 h, and rate of proteolysis to 24 h was determined by 1 D-SDS-PAGE (1 DE) and 2 DE. In addition, tentative protein identification was performed from peptide mass fingerprints of MALDI-ToF analysis of major protein groups on 2 DE profiles. The result showed that although ultrastructural configuration was similar between the 5$^{\circ}C$ and 36$^{\circ}C$ treatments, meat at 5$^{\circ}C$ had higher WBshear force (approximately 5 kg greater). A higher rate of protein degradation at 36$^{\circ}C$ was observed based on Troponin-T degradation, 1 DE, and 2 DE analysis. This indicates that proteolysis during the early postmortem period was a significant determinant of shear force at 24 h. Little difference in proteolysis between 5$^{\circ}C$ and 15$^{\circ}C$ treatments was found based on classic 1 DE profile assessment. Meanwhile, considerable differences in the 2 DE profiles between the two treatments were revealed, with substantially higher rate of proteolysis at 15$^{\circ}C$ compared to 5$^{\circ}C$. Nuclease treatment improved 2 DE profile resolution. 400 ${\mu}$g and 600 ${\mu}$g of sample loading appeared to be appropriate for 24 cm pH 3-10 and pH 5-7 IPG strips, respectively. Protein detection and quantification of the 5$^{\circ}C$, 15$^{\circ}C$ and 36$^{\circ}C$ 2 DE profiles revealed 78, 163 and 232 protein spots respectively that were differentially modified in terms of their electrophoretic properties between approximately pI 5.3-7.7 with the molecular weight range of approximately 71-12 kDa. The current results demonstrated that 2 DE was a superior tool to 1 DE for characterising proteolysis in postmortem skeletal muscle.

Comparisons of Handling Practices of Culled Sheep Meat for Production of Mutton Curry

  • In most developing countries consumers purchase retail cuts from hot carcasses and prepare traditional meat products as per their convenience and requirements. In this study, effects of different post mortem handling practices on quality of meat curry from culled sheep meat have been studied. After slaughter, leg cuts were subjected to nine commonly prevalent handling conditions in India viz. deboning (boning out) and cooking within 2-3 h (1), deboning immediately and cooking after 5-6 h (2), deboning after 5-6 h and cooking (3), deboning immediately, storage at $4^{\circ}C$ for 24 h and cooking (4), chilling for 24 h at $4^{\circ}C$, deboning and cooking (5), deboning after 5-6 h, storage for 24 h at $4^{\circ}C$, and cooking (6), deboning after 5-6 h, storage for 48 h at 4??C and cooking (7), deboning after 5-6 h, freezing and cooking (8), deboning after 5-6 h, storage for 24 h at $4^{\circ}C$, freezing and cooking (9). Significant differences were observed in pH, water-holding capacity, cooking loss and shear force values. Sensory scores were significantly higher in conditions (1), (5) and (9), and significantly lower in conditions (4) and (6). From the results, it was concluded that, to have the best quality product, meat should be cooked either immediately after slaughter or should be deboned just before cooking. Storage of deboned meat at refrigerated temperature must be avoided.

  • Incident X-ray angles of weight bearing foot AP radiography which differ in each medical institution were compared. Optimal angles were derived from the study and suggested as basic data for clinical application. 19 patients with no abnormality on right foot took part in the study from June 2013 to July 2013. Weight bearing foot AP radiography in the correct position was examined with the incident X-ray angles of $10^{\circ}$, $15^{\circ}$, $20^{\circ}$, $25^{\circ}$, $30^{\circ}$, and $35^{\circ}$. All the images were evaluated subjectively by 5 radiological technologists of over 15 year work experience using Likert scale as 1 for 'very poor', 2 for 'poor', 3 for 'fair', 4 for 'good', 5 for 'very good', 6 for 'excellent', and analyzed with ANOVA and Scheffe. Image quality was the most optimal at $20^{\circ}$ as 4.53, and relatively good at $25^{\circ}$ as 4.38 and at $30^{\circ}$ as 4.18. $35^{\circ}$ and $10^{\circ}$ were relatively poor. The average points of 6 groups indicated statistically significant difference (p<.01). In the post-mortem, 3 subgroups showed no statistically significant difference, and the average point of the groups of $20^{\circ}$, $25^{\circ}$, $30^{\circ}$was evaluated relatively higher than the other 2 groups. An optimal incident X-ray angle within the range from $20^{\circ}$ to $30^{\circ}$ for weight bearing foot AP radiography could make it possible to obtain the image of high diagnostic value and would be useful for clinical application in the future.

Relationship between Myosin Isoforms and Meat Quality Traits in Pig Semitendinosus Neuromuscular Compartments

  • The aim was to determine the relationship between muscle structure and meat quality traits in neuromuscular compartments (NMCs: R1, R2, R3, R4) of pig semitendinosus muscle. Barrows from the INTA-MGC genetic line (Argentina) were slaughtered at 100 kg body weight. In each NMC the following parameters were determined: the fibre types I, IIA, IIX and IIB by immunohistochemistry, the fibre cross sectional area (FCSA), the pH of meat after 24 h post-mortem ($pH_{24}$), instrumental meat tenderness (WB) and colour ($L^*$, $a^*$, $b^*$). There were significant differences in the following: $L^*$ (R1 = R4$a^*$ (R1>R4>R2 = R3), $b^*$ (R1 = R4R1 = R3 = R4), $pH_{24}$ (R1 = R4>R2 = R3). The relative percentages of FCSA were as follows: I (R4>R1>R3>R2), IIA (R1>R4>R3>R2), IIX (R1 = R2 = R3 = R4) and IIB (R2>R3>R1>R4). The correlation values were statistically significant between IIB and WB (R1 and R4, $r_s$ = 0.66), (R2 and R3 $r_s$ = 0.74), IIB and $L^*$ (R1 and R4 $r_s$ = 0.84), IIX and $L^*$ without discriminating NMCs. Our data suggest that the NMC where the sampling takes place is important for determining meat quality traits because of the heterogeneity of the whole muscle.

  • Progress in medical science and cell biology has resulted in the transplantation of human cells and tissues from on human into another, facilitating reproduction and the restoration of form and function, as well as enhancing the quality of life. For more than 40 years, society has recognized the medical and humanitarian value of donation and transplanting organs and tissues. The standard operating procedures of hard tissues reflect the collective expertise and conscientious efforts of tissue bank professionals to provide a foundation for the guidance of tissue banking activities. Procurement of allograft tissues from surgical bone donors is a part of tissue banking. During the past decades the use of bone allografts has become widely accepted for the filling of skelectal defects in a variety of surgical procedures. In particular in the field of orthopaedic and oral and maxillofacial surgery the demand for allografts obtained from either living or post-mortem donors has increased. Hospital-based tissue banks mainly retrieve allografts from living donors undergoing primary total hip replacement for osteoarthritis or hemi arthroplasty for hip fractures and orthgnatic surgery such as angle reduction. Although bone banks have existed for many years, the elements of organized and maintaining a hospital bone bank have not been well documented. The experience with a tissue bank at Korea Tissue Bank(KTB) between 2001 and 2004 provides a model of procurement, storage, processing, sterilization and documentation associated with such a facility. The following report describes the standard operating procedures of hard tissues such as femoral head obtained from living donors.

Occurrence, clinical signs, postmortem lesions and etiology of enterotoxaemia in Black Bengal goats

  • A year round study was carried out to investigate the etiology, clinical signs, postmortem lesions and occurrence of naturally occurring enterotoxaemia in Black Bengal goats. Sixteen goats of different age and sex died in different seasons with sigh associated with enterotoxaemia made the materials of this study. Accidental access to large amount of concentrate was noted as one of the predisposing factors although few cases were reported to occur without known diet change. Younger animals(50%) and males(62.50%) were found more prone to the disease and it was likely to be more prevalent during winter(50%) followed by at rainy season(31.25%) and summer(18.75%). Diarrhoea(81.25%), dullness(56.25%), drooping of the ears(50%), anorexia(43.75%) were recorded as major clinical signs whereas enterocolitis(100%), lung edema(87.50%), fluid filled intestines(87.50%), enlarged mesenteric lymph nodes(56.52%) etc. were most common post mortem lesions found. A few cases showed lesions on heart(31.25%), brain(25%) and/or liver/spleen(18.75%) but no lesion was found on kidney. Thus the so called 'pulpy kidney' lesion was absent. Intestinal contents were subjected to conventional bacteriological culture based methods to identify the causal agents. Based on the morphological, cultural and biochemical properties the causal agent was identified as Clostridium perfringens. Despite the study was carried out at certain area it showed a clear picture of goat enterotoxaemia in terms of etiology, clinical signs, postmortem lesions and occurrence of goat enterotoxaemia in Bangladesh.

색채프로그램과 예능프로그램을 활용한 대학생의 심리변화 (Psychological Changes of Undergraduate using Color Programs and Entertainment Programs)

  • 본 연구는 예능프로그램을 시청한 후 색채프로그램을 실시한 실험집단과 실시하지 않은 통제집단의 심리상태(강박증, 불안 및 적대감)에 어떠한 영향을 미치는지 확인하기 위해 실시하였으며, 결과는 다음과 같다. 첫째, 색채프로그램에 참여한 실험집단의 심리상태(강박증, 불안 및 적대감)는 사전 검사치보다 사후 검사치 모두 95% 통계수준에서 유의적으로 낮아졌으나 통제집단을 그런 변화가 나타나지 않았다. 둘째, 예능프로그램을 시청한 후 색채프로그램을 참여한 대학생들의 색채표현에서 빨강색과 파랑색의 지각이 폭넓게 나타났다. 이러한 변화는 흰색의 이성과 검정의 억압에서 현재 해야 할 일에 대한 책임감을 갖는 변화과정이라고 해석할 수 있었다. 결론적으로, 예능프로그램을 시청한 대학생들의 심리상태에 긍정적인 영향을 줄 수 있도록 다양한 색채 프로그램을 교육과정에 도입하는 것은 의미 있다고 본다.

고분자전해질 연료전지 내구성 향상을 위한 시동 기술 개발에 관한 연구 (Development of a Durable Startup Procedure for PEMFCs)

  • Various polymer electrolyte membrane fuel cell (PEMFC) startup procedures were tested to explore possible techniques for reducing performance decay and improving durability during repeated startup-shutdown cycles. The effects of applying a dummy load, which prevents cell reversal by consuming the air at the cathode, on the degradation of a membrane electrode assembly (MEA) were investigated via single cell experiments. The electrochemical results showed that application of a dummy load during the startup procedure significantly reduced the performance decay, the decrease in the electrochemically active surface area (EAS), and the increase in the charge transfer resistance ($R_{ct}$), which resulted in a dramatic improvement in durability. After 1200 startup-shutdown cycles, post-mortem analyses were carried out to investigate the degradation mechanisms via various physicochemical methods including FESEM, an on-line $CO_2$ analysis, EPMA, XRD, FETEM, SAED, FTIR. After 1200 startup-shutdown cycles, severe Pt particle sintering/agglomeration/dissolution and carbon corrosion were observed at the cathode catalyst layer when starting up a PEMFC without a dummy load, which significantly contributed to a loss of Pt surface area, and thus to cell performance degradation. However, applying a dummy load during the startup procedure remarkably mitigated such severe degradations, and should be used to increase the durability of MEAs in PEMFCs. Our results suggest that starting up PEMFCs while applying a dummy load is an effective method for mitigating performance degradation caused by reverse current under a repetition of unprotected startup cycles.

MREIT Conductivity Imaging of Pneumonic Canine Lungs: Preliminary Post-mortem Study

  • In magnetic resonance electrical impedance tomography (MREIT), a current-injection MR imaging method is adopted to produce a cross-sectional image of an electrical conductivity distribution in addition to MR images. The purpose of this study was to test the feasibility of MREIT for differentiating the canine lung parenchyma without and with pneumonia. Three normal healthy beagles and two mixed breed dogs with pneumonia were used. After attaching electrodes around the chest, we placed the dog inside our MR scanner. We injected as much as 30 mA current in a form of short pulses into the chest region. Reconstructed conductivity images of normal canine lungs exhibit a peculiar pattern of a relatively coarse salt and pepper noise. On the contrary, conductivity images of pneumonic canine lungs show significantly enhanced contrast of the lesions while the corresponding MR images show a little bit of contrast in the middle and caudal lung parenchyma due to the accumulation of pleural fluid. This preliminary study indicates that MREIT imaging of the chest may deliver unique new diagnostic information.

수행시간 중심의 초등 EPL 교육이 초등학생 컴퓨팅 사고력에 미치는 효과 (Effect of Execution Time-oriented the Elementary EPL Education on Computational Thinking Ability of Elementary School Students)

  • 본 연구는 초등학생의 컴퓨팅 사고력 신장을 위한 교육 방법으로 수행시간 중심의 초등 EPL 교육 프로그램을 개발하여 적용한 후 그 효과를 검증하였다. 교육 프로그램은 초등학교 학생 48명을 대상으로 실시한 사전 요구 분석 결과를 바탕으로 하여 개발하였다. 개발한 교육 프로그램의 효과를 검증하기 위해 OO대학교에서 실시한 초등학생 교육 기부 프로그램의 지원자 표집에 의한 지원자 표본 25명을 대상으로 6일간 1일 7차시씩 42차시를 진행하였다. 한국정보과학교육연합회에서 주최한 '비버챌린지(Korea Bebras Challenge 2018)'를 활용하여 사전·사후 검사 결과를 통해 교육적 효과를 분석하였으며, 분석 결과 수행시간 중심의 초등 EPL 교육이 초등학생의 컴퓨팅 사고력 향상에 긍정적인 영향을 미친 것으로 나타났다.
