Effect of Testosterone Propionate and Estradiol -l7$\beta$ on the Biochemical Changes in the Fat Body and Haemolymph of the Bivoltine Silkworm Bombyx mori L.

  • Effect of topical application with 10, 20 and 30 ${\mu}g$/ml testosterone propionate and estradiol -17${\beta}$ on the fourth and fifth instar bivoltine NB18 silkworm larvae Bombyx mori, on the glycogen and protein contents of the Fat body and trehalose and protein contents of the haemolymph has been studied. Glycogen content of the fat body was significantly decreased in both testosterone propionate and estradiol -17${\beta}$ treatment groups except in the group treated with 30 ${\mu}g$ testosterone propionate where the increase was not significant when compared with those of carrier controls. The increase/decrease in haemolymph trehalose content did not show any significant difference in all the treated groups. Protein content of the fat body significantly increased in 10 and 20 mg testosterone propionate and estradiol -l7${\beta}$ treated groups but in 30 mg treated groups the increase was not significant when compared with those of carrier controls. There was no significant change in the haemolymph protein content in all the testosterone propionate and estradiol -17${\beta}$ treated groups except in group treated with 10 ${\mu}g$ estradiol -17${\beta}$ where it showed a significant decrease when compared with that of carrier control.

  • The commercial rearing of polyphagous Indian tasar silkworm, Antheraea mylitta Drury being practiced on naturally grown primary food plants like Terminalia arjuna, (Arjun) Terminalia tomentosa (Asan), and Shorea robusta (Sal) available in the tropical forests of central India, at times, is seriously affected by the disease- Pebrine, caused by Nosema sp., a microsporidian pathogen. The present investigation on comparative larval, silk gland weight and also cocoon parameters in Pebrine-free and Pebrine-infected ecorace of tasar silkworm Antheraea mylitta Drury (Daba TV), illustrates the tasar silkworm larvae infected with pebrine disease causing heavy losses to the economy of the silk industry.

Trichinosis Caused by Ingestion of Raw Soft-Shelled Turtle Meat in Korea

  • Reptiles, unlike mammals, have been considered to be unsuitable hosts of Trichinella spp., though larvae have been detected in their muscles and human outbreaks related to their consumption have, in fact, occurred. Herein we report 2 Korean cases of trichinosis, possibly transmitted via consumption of reptile meat. Both patients suffered from myalgia, headache, and facial edema. Laboratory examinations revealed leukocytosis with eosinophilia (54% and 39%, respectively) and elevated creatinine phosphokinase. ELISA was performed under the suspicion of trichinosis, showing positivity at the 29th and 60th day post-infection. Since they had consumed raw soft-shelled turtle meat, turtle was strongly suggested to be an infection source of trichinosis in Korea next to the wild boar and badger.

  • Fish larvae are immediately exposed to microbes from hatching to maturation of their lymphoid organs, therefore effective innate mechanisms is very important for survival in such an environment. The key component of innate immune system, C3 is central protein of all activation pathways of the complement system, leading to inflammatory reactions, such as opsonisation, chemotaxis, and cell lysis of pathogens. Although, innate mechanisms is essential for survival in the early stage of development, little is known about defence mechanisms. In this study, the alignment analysis showed that amino acid sequence of C3 from olive flounder liver EST homologous to other known C3 sequences with 73-99% identity. Also, we examined the tissue distribution of olive flounder C3 and analyzed expression pattern from the fertilized egg until 28 days post hatching. As a result, olive flounder C3 mRNA was expressed only in the liver and the mRNA level more increased as developmental proceed during the early stage. These results may suggest that olive flounder C3 plays an important function in the early immune response of olive flounder larvae.

  • Studies were carried out to investigate phenotypic expression, mortality and biochemical analysis of haemolymph proteins of nm-d, nm-f, nm-k and nmn bib-molting mutants of the silkworm, Bombyx mori. The non-molting mutants characters were expressed in the homozygote of each mutant genes. All strains of non-molting mutants were similar with each other in physiological characteristics, but the expression varied with each strains. The larvae of nm-d, nm-i and nmn died between day 5 and day 9 after hatching without the first molt. The nm-f and nm-k mutants died between day 5 and day 16 with a slight increase of body weight and, more than 90% of the mutants larvae died before the first molt and a few of them survived to the 2nd and the 3rd instar and died. The haemolymph protein components of nm-d, nm-i, and nmn were rapidly reduced, and on the other hand those of nm-f and nm-k consistently until they died. And there were no distinguishable difference in haemolymph components of non-molting mutants, as compared to those normals.

  • This study was carried out to observe the development of the autonomous skeletal development of the Tridentiger trigonocephalus. Seven days after hatching, the skeleton of the two line cranes began to ossify at premaxillary, maxilla and dentary when the average total length was 4.44 mm (n=5). At thirteen days after hatching, the frontal, parietal, and epiotic ossicles were observed in the cranium when the average total length was 5.62 mm (n=5). At thirty-two days after hatching, actinost and post cleithrum were ossified at the shoulder when the average total length was 11.8 mm (n=5). At forty days after hatching, the lateral ethmoid of the skull was ossified with an average total length of 13.3 mm (n=5) and all skeletal development was completed.

  • Triploid abalone, Haliotis discus hannai, was induced by the cold $(0^{\circ}C\;and\;3^{\circ}C)$ or the heat $(35^{\circ}C\;and\;40^{\circ}C)$ shock procedure with fertilized eggs, 12 min. or 32 min. post fertilization with the various time intervals of shock duration. Fertilization rate of each experimental group was not significantly different from that of corresponding diploid control (P>0.05). However hatching rates and normality rates of triploid larvae were significantly different from those of corresponding diploid control (P <0.05). In heat shock groups at $40^{\circ}C$, fertilization rate of eggs was extremely low $(0\~2.7\%)$ and hatched larvae were not detected in these treatment groups. Incidence of triploidy was confirmed by chromosome count and the highest rates of triploid $(84.0\%)$ revealed in cold $(3^{\circ}C)$ shock with 15 min. treatment duration 12 min. after fertilization. The number of diploid chromosome was 2n: 36, and that of tiploid was 3n=54.

Effects of histamine and antihistamine on the hard tick Haemaphysalis longicornis during blood sucking

  • At the time of host attachment, ticks are very sensitive to histamine, but during rapid blood sucking they paradoxically require histamine. Using a rabbit model, we studied the effects of histamine and antihistamine during attachment and fast-feeding in different life stages of Haemaphysalis longicorns. We examined how they responded to histamine and antihistamine by analyzing the detachment rate, histology of feeding lesions, and post-feeding behavior. A significant difference (P<0.01) was found in the detachment rate between experimental and control treatments throughout the observation period. Ticks exhibited a higher detachment rate (30.1%) at 12 h after histamine application during attachment time and on antihistamine-treated skin (25.4%) at 96 h during fast-feeding. After feeding on histamine-treated rabbits, the fully engorged body weights of larvae and nymphs were 0.7±0.36 mg and 3.5±0.65 mg, respectively. An average increase in body weight of 0.6±0.05 mg and 3.2±0.30 mg was observed for larvae and nymphs compared to the respective control weights. Nymphs and adults engorged after antihistamine treatment had an average body weight of 1.3±0.54 mg and 54±0.81 mg, respectively. An average decrease in body weight was observed in antihistamine-treated H. longicornis compared with control nymphs (3.3±0.42 mg) and adults (174±1.78 mg). Skin biopsies were collected after treatment, and differential histopathological characteristics were found between the treatment and control groups. Tick-infested skin collected from rabbits in the antihistamine-treated group lacked erythrocytes in the feeding pool, indicating that antihistamine impaired tick fast-feeding stage.

  • The ultrastructural changes of cuticular surface, excretory and digestive organs of Anisakis simplex larvae chronologically recovered from experimental cats were observed with a SEM and TEM. The larva recovered from an experimental cat at 3 days post-infection (PI) retained the cuticular surface with regular transverse striations and a longitudinal groove on the lateral side of body. This finding suggests that the molting of the 3rd stage larva of A. simplex to 4th one occurred from the 3rd day after infection in cats. The excretory organ (renette cell) consisted of a large cell with numerous ductules ramified from the main duct, mitochondria and secretory granules in cytoplasm. Secretory granules in the renette cell of larvae recovered at 24 hours PI were round whereas those of control and larvae recovered at 6 hours PI were amorphous. Muscular esophagus and ventriculus also retained many secretory granules in the cytoplasm. The secretory granules in these organs of larvae recovered at $6\sim24$ hours PI were electron-dense and widely distributed whereas those of control worm were packed in a pocket and retained various electron densities. In the cytoplasm of intestinal epithelial cells, numerous fine glycogen particles and mitochondria were distributed. The chronological changes of secretory granules in renette cell, muscular esophagus and ventriculus seem to be related with the worm penetration into host tissue.

  • Larvae of the freshwater shrimp, Macrobrachium nipponense(De Haan) were reared in the laboratory under constant conditions ($25^{\circ}C,\;7\%0$ S), and their feeding rate, oxygen consumption, ammonia nitrogen excretion, and growth were measured at regular intervals during development from hatching to post larval stage. Growth was measured as dry weight, carbon, nitrogen, hydrogen, protein and lipid. All these physiological and biochemical traits revealed significant changes from instar to instar. Average feeding rate was high in intermolt stage of the molt cycle and it showed a bell-shaped pattern. Respiration(R) increased from hatching to post larval stage. Excretion(U) increased in intermolt phase of larvae and it showed a bell-shaped variation pattern, in all larval instars with a maximum near the middle of the molt cycle. Regression equations describing rates of feeding, growth, respiration and ammonia excretion as functions of time during individual larval molt cycles were inserted in a simulation model, in order to analyse time-dependent patterns of variation as well as in bioenergetic efficiencies. Carbon was initially increased and nitrogen showed a tendency to increase in premolt phase during individual molt cycles. Protein remained clearly the predominant biochemical constituent in larval biomass.
