Anticandidal Activity of the Protein Substance from Coptidis Rhizoma (황련에서 분리된 단백질성분의 항진균효과)

  • Antimicrobial peptides are evolutionary ancient weapons for animal and plant species to depend themselves against infectious microbes. In the present study, we investigated if an antimicrobial peptide was produced from Coptidis Rhizoma. For the determination, protein substance from the medicinal plant was isolated by various preparations. Among the preparations, the protein portion dissolved in phosphate-buffered saline solution (CRP-DS) that contained the most amount of protein $(90\%)$ resulted in maximal inhibition of Candida albicans which causes local and systemic infections. Analyses by gel-electrophoresis and gel-permeation chromatography showed the CRP-DS formed a single band of approximately 11.8 KDa as molecular size. Antifungal activity of the CRP-DS was almost equivalent to antifungal activity by fluconazole, resulting in MIC (minimal inhibitory concentration) of approximately $50{\mu}g/ml$. The antifungal activity was a dose-dependent. The antifungal activity appeared to be inactivated by heat-treatment and ionic strength, respectively. In a murine model, the CRP-DS enhanced resistance of mice against disseminated candidiasis. The HPLC analysis demonstrated maximum $4\%$ of berberine as residual content in the CRP-DS preparation resulted in no influence on the antifungal activity. In addition, protein portion isolated from Phellodendri Cortex producing the alkaloid component like Coptidis Rhizoma had no such anticandidal effect. These results indicate that the protein substance from Coptidis Rhizoma was responsible for the antifungal activity.

A ROENTGENOGRAPHIC STUDY OF NORMAL SUBMANDIBULAR GLANDS USING ISOBARIC SIALOGRAPHY (등압수액선조영술을 이용한 정상성인의 악하선에 관한 방사선학적 연구)

  • The aim of this study was to establish the diagnostic criteria of normal submandibular glands in adults. Materials included 132 sialograms of selected person from 23 to 28 years of age. In subjects each variable was measured and evaluated statistically introducing 18 items respectively. Conclusions from this study were as follows. 1. The mean diameter of main duct was 1.96㎜ in distal end and 2.71㎜ in hilar end, so the diameter of distal end was smaller than that of hilar end in submandibular glands. 2. The mean length of main duct was 42.02㎜ in submandibular glands. 3. The mean angle of main duct was 35.67 degrees with inferior border of mandible and 103.05 degrees at posterior mylohyoid portion. 4. In configurations following shape of main duct, those revealed that 39.39% of cases of the curvilinear type, 16.67% of cases of the linear type, 22.73% of cases of the sigmiod type and 21.21% of cases of the angular type. And in configurations following course of main duct, those revealed that 83.33% of cases of the smooth type and 16.67% of cases of the tortuous type. 5. The mean caliber of submandibular glands was 45.59㎜ in superoinferior height, 43.56㎜ in anteroposterior width, 28.85㎜ in mediolateral width and 33.65㎜ in extent below mandibular angle. 6. Well-delineated acinar parenchyme was observed in 77.28% of cases and well-defined tertiary ducts in 50.06% of cases. 7. There were no differences between right and left submandibular glands in size and shape of main duct and parenchymal portion, but there were great variations in each individual.

Witches' broom of jujube tree(Zizyphus jujuba MILL. var. Inermis Rehd.) IV Effect of low temperature in winter upon the appearance of symptom (대추나무 미친병에 관한 연구 IV 동기저온과 병징발현과의 관계)

  • In an experiment to evaluate the various aspects of symptom of witches' broom of jujube tree with two different kinds of scions, taken from diseased plant and stem- grafted upon sound stocks-one cut before wintering(stored in cellar), the other after wintering, it was observed that the disease rate of those cut before wintering(Nov.) was $\98\%$ whereas those cut after wintering(March) showed much lower rate, $3\%$ initially and gradually increased up to $39\%$. In another experiment of diseased bud grafted into healthy seedling, the finding made in the following year was that 14 stocks(only one of which salt union by callusing) were infected out of the given 23- in the initial stage only 4, yet gradually increased to the number of 14. The shoots from the ground portion of the diseased stock were in general more quickly subjected to the disease than the others. Under natural condition, the diseased trees develop at first seemingly the same leaves as healthy ones; it is not until the branches and loaves grow to a considerable degree that the symptom appears. Once appearing, tile disease grows and the symptom continues to appear as late as in Sept. and early Oct., causing tile plant to develop the extraordinary branches and leaves of extremely reduced size, the typical symptom of witches' broom. Such phenomenon can be observed in the experiments of the foregoing paragraphs. And the suckers and roots of diseased plant are bound to be infected, it was found out with no exception. Viewing from the results of the above experiments and observation, it is believed that the low temperature during winter causes the virus in the above-ground portion of diseased plant to diminish or inactivated, and subsequently the virus in the roots moves up or multiply in the sprouting season.

Morphogenetical characteristics of Korean wild rat (Rattus norvegicus) (한국 야생 랫드의 형태 유전학적 특성)

  • The morphometrical characteristics such as external measurements and mandible size assessment in mice and rats have to be highly heritable and sufficiently variable between strains in order to calculate a strain specific profiles. The coat color of Korean wild rats were observed and morphometric analysis of external measurements were carried out on Korean wild rats compared to laboratory strains in order to clarify the genetic characteristics of Korean wild rats and to establish background data as a domestication of Korean wild rats for new laboratory strain. Korean wild rats were captured from Chunchon and Hoengsong. 4 inbred and 1 outbred strains of rats were used in this study for the comparison of genetic characteristic of Korean wild rats. Total body length, head length, tail length, hind foot length and ear length were measured and then statistical analysis were carried out by discrimiant analysis. The coat color of Korean wild rat showed golden white in ventral portion and dark agouti in dorsal portion. Korean wild rats could be distinguished from the other laboratory strains distinctly by morphogenetical analysis. There was significant variations among Korean wild rat compared to those of the other laboratory strains of rat. This study may provide that Korean wild rats have a unique genetic characterization compared to those of other inbred strains of rats based on morphogenetical characteristics by external measurements.

Strength and Pore Characteristics of Alkali-activated Slag-Red Mud Cement Mortar used Polymer According to Red Mud Content (레드머드 대체율에 따른 폴리머 혼입 알칼리활성화 슬래그-레드머드 시멘트모르타르의 강도 및 기공특성)

  • The alkali-slag-red mud(ASRC) cement belongs to clinker free cementitious material, which is made from alkali activator, blast-furnace slag(BFS) and red mud in designed proportion. This study is to investigate strength and pore characteristics of alkali-activated slag cement(NC), clinker free cementitious material, and ordinary portland cement(C) mortars using polymer according to red mud content. The results showed that the hardened alkali-activated slag-red mud cement paste was mostly consisted of C-S-H gel, being very fine in size and extremely irregular in its shape. So the hardened ASRC cement paste has lower total porosity, less portion of larger pore and more portion of smaller pore, as compared with those of hardened portland cement paste, and has higher strength within containing 10 wt.(%) of alkali-activated slag cement(NC) substituted by red mud.

Clinical Experience of Thoracodorsal Perforator Based Free Flap (흉배혈관 천공분지에 기초한 유리피판술의 임상적 이용)

  • Perforator flaps have become increasingly popular in microsurgery nowadays and are being used widely for many cases of reconstruction after trauma and cancer ablation. And thoracodorsal perforator based free flap is one of them having the merits of carrying a large skin paddle with leaving intact innervation and function of the remaining latissimus dorsi muscle. We made a homogeneous thin flap excluding the main muscle with a long vascular pedicle and tried to decrease the donor site morbidity. But, it needs a long learning-curve and we have met marginal flap necrosis frequently. Besides, prolonged operation time for complete perforator dissection may be a tedious job to the microsurgeon. To overcome these disadvantages, we usually included very small portion of the latissimus dorsi muscle during this flap elevation around the pedicled 2-3 thoracodorsal perforators during this flap elevation. We performed 3 cases of thoracodorsal perforator based free flap at Hallym university sacred heart hospital between May and August 2005 for the soft tissue defect of the scalp and feet. The average flap size was $8{\times}14\;cm$. Although it is not a true perforator flap, we can get the reliability for the flap survival with much better blood circulation and save the time of one or two hours to dissect the perforators completely. All cutaneous flaps survived completely without any complication except one fatty female who had the very small superficial fat necrosis due to flap bulkiness. We believe the thoracodorsal perforator based free flap can be extended its versatility and reliability by including the very small portion of the muscle around the perforators.

Euryhelmis squamula (Digenea: Heterophyidae) Recovered from Korean Raccoon Dog, Nyctereutes procyonoides koreensis, in Korea

  • In this study, we intended to describe an unrecorded species of heterophyid trematode recovered from the small intestine of a Korean raccoon dog, Nyctereutes procyonoides koreensis, in Korea. A total of 13 small flukes were collected from a deceased Korean raccoon dog which was found in Chuncheon-si, Gangwon-do, Korea in May 2017. The trematode body were covered with many small spines, rectangular, broader than long, 807-1,103 ㎛ long and 1,270-1,550 ㎛ wide. Oral sucker in the anterior end slightly smaller than acetabulum. Pharynx muscular and well developed. Esophagus relatively long and sigmoid. Acetabulum small and located at median in anterior 2/5 portion. Ceca bifurcated at the anterior of genital pore and acetabulum and terminated at testis level. Testes larger, deeply lobed and located at the near of posterior end of body. Ovary small, triangular and located at the slight left of median and the anterior of left testis. Vitelline follicles dendritic and extend from the middle level of esophagus to the posterior portion of body. Eggs embryonated, operculated, small and 33-35×15-16 ㎛ in size. Based on the morphological characteristics, the small heterophyid flukes recovered from the small intestines of Korean raccoon dog, N. procyonoides koreensis, were identified as Euryhelmis squamula (Digenea: Heterophyidae). Accordingly, this species of heterophyid flukes is to be a new trematode fauna in Korea by this study.

Developing a Cantilever-type Near-field Scanning Optical Microscope Using a Single Laser for Topography Detection and Sample Excitation

  • The capabilities of the near-field scanning optical microscope (NSOM) for obtaining high resolution lateral topographical images as well as for mapping the spectroscopic and optical properties of a sample below the diffraction limit of light have made it an attractive research field for most researchers dealing with optical characteristics of materials in nano scales. The apertured NSOM technique involves confining light into an aperture of sub-wavelength size and using it to illuminate a sample maintained at a distance equal to a fraction of the sub-wavelength aperture (near-field region). In this article, we present a setup for developing NSOM using a cantilever with a sub-wavelength aperture at the tip. A single laser is used for both cantilever deflection measurement and near-field sample excitation. The laser beam is focused at the apex of the cantilever where a portion of the beam is reflected and the other portion goes through the aperture and causes local near-field optical excitation of the sample, which is then raster scanned in the near-field region. The reflected beam is used for an optical beam deflection technique that yields topographical images by controlling the probe-sample in nano-distance. The fluorescence emissions signal is detected in far-field by the help of a silicon avalanche photodiode. The images obtained using this method show a good correlation between the topographical image and the mapping of the fluorescence emissions.

Effect of Flame Temperature on the Characteristics of Flame Synthesized TiO2 Nanoparticles (수소 확산화염에서 화염온도가 TiO2 나노입자의 합성에 미치는 영향)

  • In this work, $TiO_2$ nanoparticles were synthesized using a N2-diluted hydrogen coflow diffusion flame. The effects of flame temperature on the crystalline structure and the size of formed nanoparticles were investigated. The maximum centerline temperature of the flame ranged from 1,920K for $H_2-only$ flame to 863k for $81\%\;N_2-diluted$ flame. The morphology and the crystal structure of $TiO_2$ nanoparticles were analyzed by a TEM and a XRD, respectively. The particle size distribution was also measured by using a scanning mobility particle size. (SMPS). The mean particle diameter was calculated from the TEM images depended on the flame temperature, having minimum at about 1,look. Based on the SMPS measurements, the mean particle diameter of $TiO_2$ nanoparticles at flame temperatures > 1,300K was smaller than that at flame temperatures < 1,300K. From the XRD analysis, it was evident that the anatase fraction increased with decreasing the flame temperature. The portion of anatase phase in $TiO_2$ nanoparticles might be greater than $80\%$ when the flame temperature was lower than 1,000K.

A Case Report of an Advanced Gastric Cancer Patient Who Was Able to Undergo Conversion Surgery Treated with Chemotherapy and Korean Medicine (항암화학요법과 한방치료를 병행하여 전환수술이 가능하게 호전된 진행성 위암 환자 1례)

  • Objectives: The purpose of this study was to report a case of a patient with advanced gastric cancer with peritoneal metastasis treated with chemotherapy and Korean medicine Methods: A patient with advanced gastric cancer with peritoneal metastasis was treated with Xeloda/cisplatin since April 2019. The cycle was repeated every three weeks for a total of 11 times. At the same time, the patient was treated with Korean medicine. The tumor size was measured by computed tomography (CT) and esophagogastroduodenoscopy (EGD). Adverse events were evaluated by the National Cancer Institute Common Terminology Criteria for Adverse Events (NCI-CTCAE), version 5.0. Results: After treatment with Xeloda/cisplatin and Korean medicine for nine months, the extent of the proximal portion of the primary tumor and the size and number of multiple nodules around the stomach decreased and the cancer cells with peritoneal metastasis disappeared. The symptoms of discomfort and physical activity were gradually improved. As a result, the patient underwent conversion surgery. Conclusions: This case study suggests that the combination of chemotherapy and Korean medicine may contribute to the reduction in tumor size as well as the improvement in the quality of life.
