석류피 에탄올 추출물의 항치아우식 활성 (Anticariogenic Properties of the Ethanol Extract of Pomegranate (Punica granatum) Husk)

  • Dental caries is a major worldwide oral disease problem. Streptococcus mutans (S. mutans) plays an important role in the information of dental plaque and it is being noticed as major causative bacteria of dental caries. Therefore, the development of more effective, substantial and safe preventive agents against dental caries is strongly required. In the present study, inhibitory effects of the ethanol extract of the husk of pomegranate (Punica granatum L.) on the growth, acid production, adhesion and water-insoluble glucan synthesis of Streptococcus mutans (S. mutans) were examined. The ethanol extract of pomegranate husk (250 - 4000 ${\mu}$g/ml) significantly lowered the growth of S. mutans in a dose dependent manner. The acid production of S. mutans were inhibited by the presence of ethanol extract of pomegranate husk (500 - 4000 ${\mu}$g/ml) significantly. The ethanol extract of pomegranate husk (5000 - 4000 ${\mu}$g/ml) also significantly lowered the adherence of S. mutans. In water-insoluble glucan synthesis assay, 1000 - 4000 ${\mu}$g/ml of the ethanol extract of pomegranate husk significantly inhibited the formation of water-insoluble glucan. These results suggest that pomegranate husk may inhibit the caries-inducing properties of S. mutans. Further studies are necessary to clarify the active constituents of pomegranate husk responsible for such biomolecular activities.

Estrogenic activity of Pomegranate extract in MCF7-ERE cells

  • Pomegranate, a small tree originating in Orient, belongs to Punicaceae family. The seeds contain an oil of which about 80% is rare trans 18 carbon fatty acid (punicic acid), and have highest botanical concentration of a sex steroid, estrone. Pharmacological properties of pomegranate extract have been studied, with anti-microbial, anti-parasitic, ant-viral, and anti-cancer effects. We have examined the estrogenic activity of the pomegranate extracts using MCP-7-ERE cells. MCF-7-ERE cells, stably transfected with pERE-Luc were treated various amount of pomegranate extract and after overnight treatments, luciferase activity were measured. Estradiol(E2) dose dependently induced luciferase activity in this cell and maximal response is obtained at 100pM E2.

석류가 뇌혈류 및 비만에 미치는 실험적 효과 (The Experimental Effects of Pomegranate on the Cerebral Blood Flow and Obesity in Rats)

  • To examine the effects of pomegranate on cerebral blood flow and obesity, we observed regional cerebral blood flow in normal rats, as well as body weight, serum total cholesterol, serum HDL-cholesterol, serum LDL-cholesterol, serum triglycerides, serum total lipids, serum phospholipids and serum free fatty acid levels in rats fed a high fat diet. The results were as follows. Pomegranate seed extract significantly increased regional cerebral blood flow and the peel extract increased regional the cerebral blood flow in a dose dependent manner. In addition, the pomegranate extracts(seed, peel, juice) decreased body weight, serum total cholesterol, serum LDL-cholesterol, serum triglycerides, serum total lipids, serum phospholipids and serum free fatty acid levels, and increased serum HDL-cholesterol in the rats fed a high fat diet. More specially the seed extract significantly decreased serum total cholesterol, serum triglycerides and serum phospholipid levels.

Effect of Incorporation of Pomegranate Peel and Bagasse Powder and Their Extracts on Quality Characteristics of Chicken Meat Patties

  • This study was conducted to develop chicken meat patties by incorporating pomegranate peel and bagasse powders and their extracts. Patties were developed by incorporating pomegranate peel powder (PPP, 2 g), pomegranate aril bagasse powder (PABP, 4 g), pomegranate peel powder aqueous extract (PPAE, 6 g) and pomegranate aril bagasse powder aqueous extract (PABAE, 9 g) individually per 100 g of minced meat. Both types of powders and extracts treated patties had significantly higher total phenolic content than control and butylated hydroxytoluene (BHT) treated patties. Both types of powder (PPP and PABP) treated patties had significantly higher water holding capacity, ash, crude fibre content, and hardness values, and significantly lower moisture content and lightness values in comparison to control patties. Emulsion stability and cooking yield of PABP treated patties were significantly higher than control. Addition of extracts and BHT did not influence the physico-chemical properties and proximate composition of chicken patties. Both types of powders and extracts provided better protection to chicken meat patties against oxidative rancidity and microbial proliferation in comparison to control and BHT treated patties during refrigerated storage. It is concluded that pomegranate fruit byproducts in the form of peel powder, aril bagasse powder and their extracts can be successfully utilised in development of healthier chicken meat patties and these byproducts can also be effectively used as a replacement of synthetic antioxidants such as BHT.

석류 내피 용매별 추출물의 항산화 활성 (Antioxidant Activities of Solvent Extracts from Pomegranate Endocarp)

  • 본 연구는 가식부위가 20~30%로 과즙 착즙 시 주로 버려지고 있으나 석류의 부위별 추출물 중 가장 항산화성이 우수한 천연식품 소재인 석류 내피를 에탄올로 추출하여 천연항산화제로서의 활용 가능성을 조사하였다. 석류 내피 추출물을 극성이 낮은 용매에서 극성이 높은 용매로 순차적으로 계통 분획하여 항산화능을 분석하였다. 각 측정 방법 별로 항산화 활성에 있어 약간의 차이를 보였으나 전반적으로 ethyl acetate 분획의 항산화능이 높게 측정되었다. DPPH 라디칼 소거활성 측정 결과 ethyl acetate와 butanol 분획물에서 $IC_{50}$이 각각 2.40 ${\mu}g$/mL, 2.88 ${\mu}g$/mL로 기준 물질인 ascorbic acid와 유사한 항산화 활성을 나타내는 것으로 나타났다. Superoxide anion 소거활성에서 ethyl acetate과 butanol 분획물의 $IC_{50}$은 각각 58.94 ${\mu}g$/mL, 61.91 ${\mu}g$/mL로 기준물질인 tannin acid보다 항산화능이 우수하였다. $ABTS^+$ 소거활성에서 ethyl acetate와 butanol 분획물의 $IC_{50}$은 각각 39.26 ${\mu}g$/mL, 40.95 ${\mu}g$/mL로 나타났으며 환원력을 측정한 결과 ethyl acetate 분획물(0.1 mg/mL)의 흡광도가 1.404로 기준물질인 ascorbic acid보다 활성이 높게 측정되었다. SOD 유사활성과 아질산염 소거능에서는 유사한 결과를 나타냈는데 SOD 유사활성은 $42.88{\pm}0.66%$(dichloromethane), $15.43{\pm}0.85%$(butanol), $12.82{\pm}0.68%$(water)이었으며 아질산염 소거능에서 dichloromethane 분획물의 $IC_{50}$은 115.54 ${\mu}g$/mL로 기준물질인 ascorbic acid보다 높게 나타났다. 따라서 본 연구결과 석류 내피 추출물은 높은 항산화 활성을 가지고 있으며, 폐자원으로써 버려지는 석류 내피는 식품첨가물 등의 기능성 소재로의 이용가능성이 있을 것으로 여겨진다.

  • 석류는 오래전부터 임상치료 및 식품원료로 사용되었으며 많은 피토에스트로겐 성분을 함유하고 있다. 그 성분으로는 에스트론, 에스트라디올, 에스트리올, 베타-시토스테롤, 엘라그산 등이 였으며 이러한 성분으로 인해 과즙, 껍질, 오일 등은 약한 에스트로겐 활성을 나타낸다고 보고되었으며, 이로 인해 석류는 여성 갱년기 치료용 원료로 많이 사용되고 있다. 그러나 에스트로겐 성분의 존재 유무에 대한 견해의 차이가 나타나고 있어 본 연구는 LC/MS/MS를 이용하여 석류추출물 중의 에스트로겐 분석을 통해 이를 확인하고자 하였으며, 간편하고 정확한 분석법을 모색하고자 하였다. 3개 롯트 시료의 에스트로겐을 분석한 결과, 에스트론 83.5ppb, 에스트라디올 10.9ppb 및 에스트리올 38.6ppb로 검출되었다.

Effects of Punica granatum (Pomegranate) Extracts on the Menopause Women

  • A randomized, double blind, placebo-controlled human intervention study involving 58 menopause women was performed to investigate the effect of pomegranate extracts on menopausal symptoms. The volunteers were randomly distributed into two groups, one taking pomegranate extracts (1.5 g/day) and the other being a placebo by taking oral administration for 8 weeks. The results from this study showed that duration of hot flush and sweating were significantly decreased by taking pomegranate extracts. In addition, menopausal rating scales and the Kuppermann index were significantly decreased in pomegranate extract taking group. This result suggests a possibility that pomegranate extracts can be helpful for menopause women.

이란산 흑석류 농축액과 그 제품의 성분 및 함유된 Phyto 에스트로겐류에 관한 연구 (Proximate Compositions and Selected Phytoestrogens of Iranian Black Pomegranate Extract and Its Products)

  • 천연 호르몬 보충 제제의 개발을 위한 기초 연구로 이란산 흑석류 과즙 농축액과 이를 이용한 제품의 화학성분들을 분석하였다. 석류 농축액의 일반성분은 수분 39.3%, 조지방 0.4%, 조단백질 0.9%, 조회분 1.4%, 그리고 탄수화물은 42.0%이었다. 아미노산 함량은 글루탐산이 1310.0ppm, 아스파르트산이 896.2ppm, 아르기닌이 877.7ppm, 페닐알라닌이 57.5ppm순으로 무기 성분들은 철분 6640.0ppm, 염소 3464.0ppm, 칼륨 2550.8ppm, 인 150.0ppm, 칼슘 80.0ppm순으로 많이 함유되어 있었다. 비타민은 5가지의 수용성 비타민이 함유되어 있었으며 그 중 비타민 C(20mg/100g)를 제외한 나머지 비타민들이 아주 적은 양이 있었다. 주요 지방산은 palmitic acid(8.3%)와 stearic acid(69.4%)로 전체 지방산의 약 60~80%를 차지하고 있었다. 또한 6종의 Phyto 및 에스트로겐류가 들어 있었으며 각각 daidzein 23.72ppm, quercetin 9,75ppm, catechin 1.48ppm, genistein 0.29ppm, 2,3-di-MeO-estradiol이 0.04ppm, 그리고 17$\beta$-에스트라디올이 0.15ppm이 함유되어 있었다. 이상의 결과들을 석류 농축액으로 제조한 제품 포에버 120과 칡과 대두 isoflavon 농축분말과 비교하였다.
