The production of extracellular polysaccharide by Methylomonas mucosa (NRRL B-5696) was investigated. The microorganism uses methanol as the carbon source for their growth and produces exopolysaccharides. The productivity of exopolysaccharides was investigated under various culture modes: batch, fed-batch and continuous culture. In flask culture the growth of cell mass and the production of polysaccharide were inhibited at above 1% (v/v) methanol. At 1%(v/v) methanol maximum specific growth rate was obtained. As C/N ratio (g methanol/g ammonium sulfate) increased, polysaccharide production increased and cells mass decreased. Magnesium ion was also found to be essential for the polysaccharide production. In batch culture the production of polysaccharides was more affected by the specific growth rate than the cell concentration. In fed-batch culture the concentration of polysaccharide was 4 times higher than that of batch culture, but the yield was lower. The productivity of fed-batch with continuous feeding was higher than that of batch or fed-batch with intermittent feeding. This is due to no methanol limitation or inhibition that used to occur in fed-batch culture with intermittent feeding. In continuous culture pure oxygen was supplied to avoid the oxygen limitation. As the dilution rate in- creased up to 0.21 h-1, the yield and productivity increased. The solution viscosity of the produced polysaccharide obtained from above increased exponentially with the concentration of polysaccharide.

Optimal culture conditions for efficient in vitro propagation and polysaccharide production of Orostachys japonicus were established. The highest growth yield was achieved in 1/2 MS medium, while the lowest growth yield was obtained in 4 MS medium. The patterns of polysaccharide formation were a little similar in all cases, but on MB5 medium, the po]ysaccharide contents of plant were higher than others. Among the modified nitrate levels, effective growth level were obtained in 1/4 N and 1/2 N. High contents of polysaccharide were obtained in 4 N. Different concentration of potassium and calcium did not improve the growth and polysaccharide production. The micropropagated shoots were successfully acclimatized artificial soils.
The extraction conditions for the production of red ginseng polysaccharide were proposed. The crude fiber content of red ginseng marc (RGM) (15.3%) was much higher than that of white ginseng (WG) (2.1%) and red ginseng (RG) (0.5%). Thus, RGM was selected as the raw material for polysaccharide production. The correlation between the particle size of RGM and the polysaccharide extraction was investigated with a correlation analysis using the SPSS package. The two parameters were found to have a significant correlation (p<0.01). The polysaccharide extraction increased as the particle size of RGM decreased. The optimal concentration of RGM was 6.66% (w/v). The extraction yield increased as both the extraction temperature and the extraction time increased. Finally, the extraction temperature and time were selected as $85^{\circ}C$ and 5 hrs, respectively. Consequently, the extraction conditions for polysaccharide production were optimized and statistically confirmed.
The production of polysaccharide from Paecilomyces japonica was studied in the shake flask culture. For the cell growth and the polysaccharide production, the optimum synthetic medium was glucose peptone(YMP) medium. The flask culture conditions for the polysaccharide production were $27^{\circ}C$ and 200rpm with the initial pH 9 for 8days cultivation.
The fermentation conditions for the maximal production of the useful polysaccharides(water soluble polysaccharide and cell bound polysaccharide) from marine bacterium Zoogloea sp.(KCCM 10036) were investigated with a 5 L jar fermentor. The maximal production of these polysaccharides was obtained under the conditions of initial pH 7.8, 30$^{\circ}C$, 400 rpm of agitation speed, 2 Wm of aeration rate, 10%(w/v) of inoculum size and 2.5%(w/v) of glucose substrate and 10.38 g/L of total polysaccharide was produced. Apparent viscosity of the culture broth was increased with the production of these polysaccharides and the maximum value was reached to 22,500 cp.
For the efficient production of a new exo-polysaccharide from Ganoderma lucidum ASI 7004, the optimum conditions and methods in submerged cultivation were investigated with an airlift fermenter system. The optimum aeration rate was 2.5 Wm at the initial pH 5.0 and 28$^{\circ}C$. The increase of dissolved oxygen concentration by pure oxygen supply during cultivation did not improved the exo-polysaccaride production and the mycelial growth. The maximum exo-polysaccharide production and the mycelial growth under the optimum culture condition were obtained in media of glucose 60g/L, yeast extract 6g/L, (NH4)2HPO4 1g/L and KH2PO4 0.5g/L. Under these optimum medium and culture conditions, about 7.15g/L of exo-polysaccharide and 13.9g/L of mycelial growth were producted, respectively.
In order to maximize the cell growth and the polysaccharide production in Agaricus blazei, two kinds of fed-batch fermentation processes were performed with varying the feeding medium compositions and the feeding process. The relationship between dissolved oxygen and polysaccharide production in batch fermentation was applied to fed-batch fermentation. The biomasss concentration was 18.2 g/L and the polysaccharide production was 10.4 g/L.
Streptococcus pneumoniae (pneumococcus) is the most frequent causative agent of acute bacterial pneumonia. Outstanding characteristic of pneumococcus is an ample polysaccharide capsule that is highly anti-genic agent and is the major factor for classification of pneumococcus into more than 94 serotypes. In this study, production of capsular polysaccharide from Streptococcus pneumoniae type 23 was optimized by supplementation of metal ions or by modulation of culture conditions. When brain heart infusion (BHI) broth was supplemented with 1 mM concentration of $CaCl_2$, $CuSO_4$ and $MgSO_4$, growth of pneumococcus as well as polysaccharide yield were stimulated. Also slight aeration gave rise to better polysaccharide yield.
The effect of various nutrients for the production of the anti-complementary polysaccha- ride by Flammulina velutipes, was examined and the partial purification of the polysaccharide was carried out. The culture conditions and medium compositions considerably influenced the anti-complementary activity of the polysaccharides. When a culture was carried out at 23$\circ$C and pH 5.5 for 6 days in the synthetic medium supplemented with galactose and NaNO$_{3}$, the production of the anti-complementary polysaccharide was maximized. The crude polysaccharide, FV-1 was isolated from the culture broth and partially purified into two fractions, FV-1-II and FV-1-III by gel filtration using Sephadex G-100. FV-1-II was mainly consisted of xylose, mannose, galactose and glucose, and rhamnose, mannose and galactose, for FV-1-III. Also, the anti-complementary activity of FV-1-III was reduced partially in the absence of the Ca ion. When crossed immunoelect- rophoresis using anti-human C3 serum was carried out after incubation of normal human serum with the FV-1-III in the Ca ion free condition, a cleavage of C3 precipitin line was observed. These results indicate that the mode of complement activation by polysaccharide from Flammulina velutipes is via not only the classical pathway but also the alternative pathway.
The Study was carried out to investigate in the optimal mycelial growth and Exo-Polysaccharides of Fomitopsis pinicola. Jar fermentations were carried out to optimize the culture conditions for mycelial growth and exo- polysaccharide production. The optimal agitation speed and aeration rate were 200 rpm and 1.5 v.v.m., respectively. Under optimal culture conditions, the maximum mycelial growth and exo-polysaccharide production after 11 days with a 5 L jar fermenter containing the optimized medium were 10.21 g/L and 3.56 g/L, respectively. However, the fundamental information obtained this study is insufficient in the development of a efficient process for mycelial growth and exe-polysaccharide production from Fomitopsis pinicola.
