Changes in Microflora, Enzyme Activities and Microscopic Structure of Waxy Rice and Steeping Water in Response to Different Steeping Conditions During Preparation of Gangjung (강정 제조시 찹쌀의 수침조건에 따른 찹쌀과 수침액의 미생물상, 효소활성 및 미세구조 변화)

  • This study was conducted to investigate the effects of steeping periods and temperatures of waxy rice on the microflora and enzyme activities of steeping water and waxy rice, as well as on the microscopic structures of waxy rice during the preparation of gangjung. When the steeping water was 15 and $35^{\circ}C$, yeast and lactic acid bacteria were the major microflora after 1 day. After 11 days, Lactobacillus spp. were the major bacteria, while Bacillus spp. were the primary microflora after 21 days. At $25^{\circ}C$ the primary species were Bacillus spp., Leuconostoc spp., and Bacillus spp. after 1, 11 and 21 days, respectively. The $\alpha$-amylase activity of steeping water increased proportionally with the increase of temperature and period, while the protease activity tended to increase for 11 days, but decreased at 21 days. The enzyme activities of the steeped waxy rice, especially the $\alpha$-amylase activity, decreased with higher steeping temperature, while the $\beta$-amylase and glucoamylase activity remained constant for up to 21 days. Observation using scanning electron microscopy indicated that intracellular cleavage of steeped waxy rice endosperm resulted in a rough surface morphology due to the exposure of polygonal starch granules. Tiny pinholes on the surface of waxy rice starch granules were also observed after steeping for 11 days.

  • The present study was designed to investigate the morphological developments of the stomach in the prenatal and postnatal rats. The gastric fundus of 16- to 22-day-old fetuses, neonates, 7-day-old, 14-day-old, 21-day-old, and adult rats were observed by light and scanning electron microscopy. Light microscopic studies. 1. In the 16-day-old fetuses, the walls of the gastric fundus were differentiated into epithelium, lamina propria, muscle layer, and serosa. The lamina propria was composed of mesenchymal connective tissue cells with sparse nucleus and their fibers. 2. In the 17-day-old fetuses, the muscular layer were differentiated into the circular and longitudinal muscle layers. 3. The epithelium on the fundus of stomach was stratified columnar cells at 16-, 17- and 18-day-old fetuses, but partly converted into simple columnar epithelium at 19-day-old fetuses. 4. The mucous cells were positive by PAS reaction at the 19-day-old fetuses and then these developed rapidly within 1 or 2 days before birth. 5. In the 20-day -old fetuses, the parietal cells were distinguished from other type cells and these cells were chiefly crowded in the middle parts of the gastric glands after 7-day-old. Scanning electron microscopic studies. 6. The surface of gastric mucosa was covered with forms of the various protrusions by forming villi at the last few days before birth and these protrusion forms were fused and folded each other. 7. The mucosal surface was closely packed by polygonal mucous cells. The openings of gastric glands were seen as slit-like round invagination and were more numerous and increased gradually in depth after the postnatal life. The above findings indicate that prominent changes occurred in the pattern of cellular proliferation in the stomach fundus at the end of gestation. The gastric epithelium had well-defined glands composed of parietal, chief and mucous cells just before the birth. The stomach fundus, therefore, were developed morphologically as those of normal adult at fourteen days after birth.

A Study on Paper of Old Monk by Yun Duseo (윤두서필 노승도의 바탕 종이 고찰)

  • Old Monk by Yun Duseo is realized to use silver paper which is made of rarity material. This sudy investigates reason why silver, that is easily discoloration in environmental condition, is susceptible in present. The method was the surface treatment materials and the base fiber through direct observation, optical instruments and XRF analysis. Based on these results, reproducing experiment was conducted to research materials in paper of Old Monk and the surface treatment technique. The results, the paper was treated 'Dochim' with a mineral having polygonal crystals for improving smoothness and density. XRF analysis result showed high calcium content, therefore, the paper was not used silver, it guess pulverized shells of abalone or other types of shellfish. Accordingly, the research was narrowed down to shells with shiny, reflective surfaces which at the same time contain high amounts of calcium. A new experiment was conducted using unprocessed abalone shells. Through the reproducing experiment, shellfish has high possibility as used mineral in paper. Notwithstanding, this experiment was unable to precisely reproduce the smoothness and shine of the original paper. Further research may therefore be needed.

Mineralogical Characteristics of Tachylite occurring in Basic Dike, Basaltic Agglomerate Formation, Ulleung Island and Its Implications of Volcanic Activity (울릉도의 하부층 현무암질 집괴암 층내 염기성 암맥에서 산출되는 타킬라이트의 광물학적 특성과 화산학적 의미)

  • Tachylite, black basaltic glass formed by the rapid cooling of molten basalt, locally occurs at the Basaltic Agglomerate Formation (BAF), the lowest formation of Ulleung Island. The purposes of this study are to characterize the occurrence and mineralogy of tachylite and to elucidate its formation condition, with emphasis on its fracture pattern, which can serve as implications for the early volcanic activity of Ulleung Island. To this end, we investigated the occurrence pattern of tachylite in the field and carried out mineralogical analyses using optical microscope, XRD, EPMA, and SEM. Tachylite occurs at the chilled margin of basic dikes which are distributed in Naesujeon, Dodong and Jeodong seasides, Turtle Rock, and Yaerimwon, whose widths vary from several cm to 10 cm. It is evident that the outer surface of tachylite is dense and smooth, whereas the inner surface, if fractured, is characterized by conchoidal fracture. The matrix of tachylite consists of amorphous, glass and some fine-grained phenocrysts present in tachylite include biotite, anorthoclase, sanidine, plagioclase, hornblende, and Fe-Ti oxides. The fracture patterns characteristic of tachylite are subrounded, oval, or less commonly polygonal, bounded by joints to form globule or lump. Taking into account texture and mineralogy, tachylite is interpreted to have undergone little subsequent alteration at low temperature via hydration or hydrolysis that could form clay minerals after it was formed. Because tachylite with peculiar fractures occurs as dikes in a close association with BAF, its presence is considered as reliable evidence that when tachylite formed, the most part of BAF was still under subaqueous conditions, or at least saturated with seawater.

Characteristics of Pearlescent Pigment using in Make-up Cosmetics (색조화장에 사용되는 진주광택 안료의 특성)

  • We investigated the morphological characteristics of the pearlescent pigment by using scanning electron microscope, energy dispersive X-ray spectrometry and thermal analyzer. The result is that the shape of pigment is platy polygonal form through observing the pearlescent pigment by the scanning electron microscope. The size of pigment is various and not formed in standardized size or shape. The pigment flakes were measured about from $30{\mu}m$ to $300{\mu}m$. The tip of the piece of pigment is pointed shape or angled. The result of observing them by the scanning electron microscope in magnifying high power is that the edge and the lateral face of them is an round form and the measurement of thickness is about $9{\mu}m$. As well using the high magnification scanning electron microscope, the surface of the pigment flake observed like rugged as coating with the $TiO_2$ element, the diameter of the coating particle is around 60 nm, then the coating particle consists of granular substance. Analysis of the configuration elements of pearlescent pigment using by the energy dispersive X-ray spectrometry is that O, Si, C, Na, Ca, Ti, Zn detected in the surface of pigment and its lateral face identifies similar components. In thermal analysis, there are no contained quantity differences between them in beginning from $100^{\circ}C$ to $800^{\circ}C$ showing thermal analysis, 1.1% out of contained quantity reduced at $115^{\circ}C$, 1.7% dropped at $416^{\circ}C$, and 1.9% decreased at $797^{\circ}C$.

  • The characteristics of deposits formed by the Daejon-cheon and Soobuk-cheon, dissecting the mountains such as Byungpung Mt. and Samin Mt. in western part of Damyang county, Jeonmam province. Results from field survey and bore hole logging by KIGAM are used in interpreting depositional environment, in this study. By the result of deposits near of the channels Daejon-cheon and Soobuk-cheon, and main channel of Youngsan River, the depth of sediment layers in this area is 4~7m, far thinner than formerly estimated. Weathered material of local rocks forms the base of the sedimentary layers. It can be assumed that the location channel of the Youngsan river has been stable ever since the start of the sedimentary events. Sediment particles of tributaries are angular than those of Youngsan River. Particles are larger and sorting is poor. It is interpreted as mount flash flood deposits. Main sources of sediments at the valley bottom or deposition dominated area are the terrace deposits or slope deposits over the gentle foot-slope or front of surrounding mountains. Some particles show polygonal cracking on the surface originated from the strong chemical weathering, while most of these has high angularity. It means various geomorphic processes operate to produce and transport the particles in this area.Isolated hills within the sedimentary plains are made with weathered materials of local bedrock. In the case of foot-slope of the hills, thin sedimentary layers are found. So it can be concluded that surface features of deposition zone of the Daejon-cheon and Soobuk-cheon is formed by the filling of lower part of the valley and its feature partly controlled by the relief of the weathering front.

Characterization of MACS Isolated Cells from Differentiated Human ES Cells (인간 배아줄기세포로부터 분화된 세포에서 MACS 방법을 이용하여 분리한 세포의 특성에 대한 연구)

  • Objective: Human embryonic stem (ES) cells have a great potential in regenerative medicine and tissue engineering. The human ES cells could be differentiated into specific cell types by treatments of growth factors and alterations of gene expressions. However, the efficacy of guided differentiation and isolation of specific cells are still low. In this study, we characterized isolated cells from differentiated human ES cells by magnetic activated cell sorting (MACS) system using specific antibodies to cell surface markers. Methods: The undifferentiated hES cells (Miz-hESC4) were sub-cultured by mechanical isolation of colonies and embryoid bodies were spontaneously differentiated with DMEM containing 10% FBS for 2 weeks. The differentiated cells were isolated to positive and negative cells with MACS system using CD34, human epithelial antigen (HEA) and human fibroblast (HFB) antibodies, respectively. Observation of morphological changes and analysis of marker genes expression were performed during further culture of MACS isolated cells for 4 weeks. Results: Morphology of the CD34 positive cells was firstly round, and then it was changed to small polygonal shape after further culture. The HEA positive cells showed large polygonal, and the HFB positive spindle shape. In RT-PCR analysis of marker genes, the CD34 and HFB positive cells expressed endodermal and mesodermal genes, and HEA positive cells expressed ectodermal genes such as NESTIN and NF68KD. The marker genes expression pattern of CD34 positive cells changed during the extension of culture time. Conclusion: Our results showed the possibility of successful isolation of specific cells by MACS system from undirected differentiated human ES cells. Thus, MACS system and marker antibodies for specific cell types might be useful for guided differentiation and isolation of specific cells from human ES cells.

Cellular activities of osteoblast-like cells on alkali-treated titanium surface (알칼리 처리된 타이타늄 표면에 대한 골아 유사세포의 세포 활성도)

  • To improve osseointegration at the boneto-implant interface, several studies have been carried out to modify titanium surface. Variations in surface texture or microtopography may affect the cellular response to an implant. Osteoblast-like cells attach more readily to a rougher titanium surface, and synthesis of extracellular matrix and subsequent mineralization were found to be enhanced on rough or porous coated titanium. However, regarding the effect of roughened surface by physical and mechanical methods, most studies carried out on the reactions of cells to micrometric topography, little work has been performed on the reaction of cells to nanotopography. The purpose of this study was to examme the response of osteoblast-like cell cultured on blasted surfaces and alkali treated surfaces, and to evaluate the influence of surface texture or submicro-scaled surface topography on the cell attachment, cell proliferation and the gene expression of osteoblastic phenotype using ROS 17/2.8 cell lines. In scanning electron micrographs, the blasted, alkali treated and machined surfaces demonstrated microscopic differences in the surface topography. The specimens of alkali treatment had a submicro-scaled porous sur-face with pore size about 200 nm. The blasted surfaces showed irregularities in morphology with small(<10 ${\mu}m$) depression and indentation among flatter-appearing areas of various sizes. Based on profilometry, the blasted surfaces was significantly rougher than the machined and the alkali treated surfaces (p$TiO_2$) were observed on alkali treated surfaces, whereas not observed on machined and blasted surfaces. The attachment morphology of cells according to time was observed by the scanning electron microscope. After 1 hour incubation, the cells were in the process of adhesion and spreading on the prepared surfaces. After 3 hours, the cells on all prepared surfaces were further spreaded and flattened, however on the blasted and alkali treated surfaces, the cells exhibited slightly irregular shapes and some gaps or spaces were seen. After 24 hours incubation, most cells of the all groups had a flattened and polygonal shape, but the cells were more spreaded on the machined surfaces than the blasted and alkali treated surfaces. The MTT assay indicated the increase on machined, alkali treated and blasted surfaces according to time, and the alkali treated and blasted surfaces showed significantly increased in optical density comparing with machined surfaces at 1 day (p<0.01). Gene expression study showed that mRNA expression level of ${\alpha}\;1(I)$ collagen, alkaline phosphatase and osteopontin of the osteoblast-like cells showed a tendency to be higher on blasted and alkali treated surfaces than on the machined surfaces, although no siginificant difference in the mRNA expression level of ${\alpha}\;1(I)$ collagen, alkaline phosphatase and osteopontin was observed among all groups. In conclusion, we suggest that submicroscaled surfaces on osteoblast-like cell response do not over-ride the one of the surface with micro-scaled topography produced by blasting method, although the microscaled and submicro-scaled surfaces can accelerate osteogenic cell attachment and function compared with the machined surfaces.

Epidermal Structure and Stomatal Types in Some Species of Korean Crassulaceae (한국산 돌나물과(Crassulaceae) 수종의 표피구조와 기공유형)

  • This study was carried out to investigate the epidermal structure and types of stomata on the upper and lower surfaces of the leaves in six species and one variety belonging to Korean Crassulaceae. The shape of epidermal cells was polygonal, isodiametric, or elongated. The cell wall was thick, sinuous or deeply sinuous. The subsidiary cell wall was thin or mostly arched. The cytolith-containing cells were found in Orostachys japonicus. The distribution of stomata was more on the upper surface than on the lower surface in the leaves of O. japonicus and Sedum sarmentosum. The stomata in the other species were less on the upper surface than on the lower surface, and stomatal size was different in each species. The great majority of stomatal types was the helico-eumesogenous type. This type was subdivided into two, parahelico-eumesogenous type and diahelico-eumesogenous type by us on the basis of the angle of division of guard mother cell and the long axis of the last-formed subsidiary cells. Sometimes allelo-eumesogenous type as well as aniso-eumesogenous type was found. The tetra-eumesogenous type with four mesogenous subsidiary cells was rarely observed. It was observed that the varieties of aniso-eumesogenous type with one to three subsidiary cells and one helix of subsidiary cells were developed by the secondary division of subsidiary cells of aniso-eumesogenous stoma. The compound shape of helico-eumesogenous stomata was found. Five new kinds of stomatal types were observed as follows; the helico-tetra-eumesogenous type, the coallelohelico-eumesogenous type, the cohelico-allelo-eumesogenous type, the duplotetra-eumesogenous type, and the aniso-euperigenous type. These types were reported here in the vascular plants for the first time.

Studies on the mass mortality of the cultured grouper, Epinephelus septemfasciatus (양식(養殖) 능성어, Epinephelus septemfasciatus 대량(大量) 폐사(斃死)에 관(關)한 연구(硏究))

  • In the late summer of 1990 and 1991, mass mortality occured among cage-cultured grouper, Epinephelus septemfasciatus in south cost of Korea. The moribund fish didn't feed and became pale or dark chestnut colour and irregularly swimmed due to the loss of equilibrium, finally the diseased fish fell down side away on the bottom or the surface of cage showing the bent of body and died. The diseased fish showed the extensive hemorrahge in brain, the swelling of spleen and bile duct as the specific syptoms of internal organs. So the gill, skin and other organs of the diseased fish were examined for the presence of pathogenic parasites and bacteria. The parasitic Trichodina sp. were detected only from the gill lamella of the diseased fish, but these parasites seemed to be not a direct causative agents that induced the gross mortality of the cultured grouper. because these parasites were also observed in normal grouper, yellowtail, red seabream and rock bream co-cultured with the diseased grouper in same or near cages. In the viral examination, although isolation of the causative agent by the use of estabilshed cell Lines, RTG-2 and CHSE-214, was not succeed, the normal grouper inoculated intramuscularly with the filtered homogenate of the organs of the diseased fish showed the same external and internal signs with the naturally infected grouper. They died within a week. By using the naturally and the artificially infected fishes, electron microscopic observation revealed numerous hexagonal or polygonal particles in the cytoplasm of liver cells. Based on the these results, we suggest that the mass mortality of the cultured grouper would be occurred by the infection of a viral agent.
