Molecular Detection of Catabolic Genes for Polycyclic Aromatic Hydrocarbons in the Reed Rhizosphere of Sunchon Bay

  • This study focused on detecting catabolic genes for polycyclic aromatic hydrocarbons (PAHs) distributed in the reed rhizosphere of Sunchon Bay, Korea. These marsh and mud environments were severely affected by human activities, including agriculture and fisheries. Our previous study on microbial roles in natural decontamination displayed the possibility that PAH-degrading bacteria, such as Achromobacter sp., Alcaligenes sp., Burkholderia sp. and Pseudomonas sp. play an important decontamination role in a reed rhizosphere. In order to gain further fundamental knowledge on the natural decontamination process, catabolic genes for PAH metabolism were investigated through PCR amplification of dioxygenase genes using soil genomic DNA and sequencing. Comparative analysis of predicted amino acid sequences from 50 randomly selected dioxygenase clones capable of hydroxylating inactivated aromatic nuclei indicated that these were divided into three groups, two of which might be originated from PAH-degrading bacteria. Amino acid sequences of each dioxygenase clone were a part of the genes encoding enzymes for initial catabolism of naphthalene, phenanthrene, or pyrene that might be originated from bacteria in the reed rhizosphere of Sunchon Bay.

Trickle Bed Reactor에서 Pt/Kieselguhr 촉매를 이용한 다환방향족 탄화수소 수소화 반응 (Hydrogenation of Polycyclic Aromatic Hydrocarbons Over Pt/Kieselguhr Catalysts in a Trickle Bed Reactor)

  • 본 연구의 목적은 열분해연료유(pyrolysis fuel oil, PFO)에 포함된 다환 방향족 탄화수소(polycyclic aromatic hydro, PAHs) 수소화 반응용 촉매로서 Pt(1wt%)/Kieselguhr 비드 촉매 및 펠렛 촉매를 제조하는 것이다. Trickle-bed 반응기에서 PFO-cut 수소화 반응을 통한 포화 고리 화합물(saturated cyclic compound)의 수율을 최대화하기 위한 최적의 반응 온도 및 수소/PFO-cut 유량비는 각각 250℃와 1800으로 결정하였다. PFO-cut의 공간속도(LHSV)가 감소할수록 포화 고리 화합물의 수율이 증가하였다. 펠렛 촉매와 비드 촉매의 수소화 반응 성능 차이는 크지 않았다. Kieselguhr 지지체를 성형한 후에 Pt를 담지한 촉매(AI 촉매)가 kieselguhr 분말에 Pt를 담지한 후에 성형한 촉매(BI 촉매)에 비해 수소화 활성이 높았으며, 이러한 경향은 펠릿 촉매와 비드 촉매에서 공통적으로 나타났다. 이는 AI 촉매의 Pt 활성점 수가 BI 촉매 보다 많기 때문이다. 본 연구에서 제조한 촉매 중에서 AI법으로 제조된 펠렛 촉매가 제조된 촉매 중 반응 활성이 가장 우수한 것을 확인하였다. PFO-cut 수소화 반응 생성물 중 C8~C15 범위의 고리 화합물이 대부분을 차지했으며, C11 고리 화합물의 분포도가 가장 높았다. 수소화 반응과 더불어서 분해 반응도 함께 촉진되어 생성물의 탄소수 분포가 경질 탄화수소 쪽으로 이동함을 확인하였다.

Accumulation and Characterization of Polycyclic Aromatic Hydrocarbons in Seafood from the Coastal Areas of Korea

  • Twenty seafood samples, which are common edible species and commercially important items in Korea, were purchased at the local fisheries markets and were analyzed for polycyclic aromatic hydrocarbons (PAHs) using gas chromatography coupled to mass spectrometer detector (GC/MSD). The levels of sixteen PAHs in seafood from Korean coasts were 161 to 2,243 pg/g wet weight. The highest concentration was found at saury (Coloabis saira) and the lowest level was found at jacopever (Sebastes schlegeli). The concentrations of potentially carcinogenic PAHs of six species were in the range of 9 to 123 pg/g wet weight. The residues of PAHs in fishes from Korean coasts were slightly low or relatively moderate to other countries. There was no correlation between PAH residues and lipid contents in seafood samples. The predominant contributors in fish samples were lower-molecular-weight two and three ring aromatic PAHs such as naphthalene, acenaphthene, fluorene and phenanthrene. Filter-feeding organisms like shrimp, crab and topshell were dominated by three- and four-ring aromatic PAHs. The PAH profiles in marine sediments, bivalves, fishes, shrimp, crab and topshell according to exposure pathway were compared through factor analysis. The PAH profiles were clearly classified by the difference of species or environmental matrices. This result suggests that most of PAHs within the same samples behave identically in marine environment.

Determination of Normal Saturated- and Polycyclic Aromatic Hydrocarbons in the River Water of Bangladesh by Liquid-Liquid Extraction and Gas Chromatography

  • A liquid-liquid extraction followed by evaporative concentration method was used to determine the concentration of normal, or straight chain, saturated hydrocarbons (NSH) $(C_{10}\;to\;C_{24})$ and polycyclic aromatic hydrocarbons (PAH) here defined as: fluorene, anthracene, pyrene, chrysene and perylene, in the Buriganga River water of Bangladesh. Samples were collected from 5 and 25 cm depth of water at the southern, middle and northern parts of the river at Postogolla, Sadarghat and Sowarighat stations. Hydrocarbons were extracted from 450 mL of water into 75 mL n-hexane and then concentrated into 1 or 2 mL solution by evaporation. These solutions were analyzed by gas chromatography. The highest and lowest concentrations were determined as $257\;{\mu}gL^{-1}\;for\;C_{13}\;and \;0.24\;{\mu}g\;L^{-1}\;for\;C_{22}$ at 5 ㎝ depth of water, at the northern part of the Sowarighat and southern part of the Postogolla, respectively. This method could allow the analysis of water for $C_{22}$ as low as $0.24\;{\mu}g\;L^{-1}$.

Evaluation of Chemical Analysis Method and Determination of Polycyclic Aromatic Hydrocarbons Content from Seafood and Dairy Products

  • This study was carried out to investigate contents of 8 polycyclic aromatic hydrocarbons (PAHs) from frequently consumed seafood and dairy products and to evaluate their chemical analysis methods. Samples were collected from markets of 9 cities in Korea chosen as the population reference and evaluated. The methodology involved saponification, extraction with n-hexane, clean-up on Sep-Pak silica cartridges and gas chromatograph-mass spectrometry analysis. Validation proceeded on 2 matrices. Recoveries for 8 PAHs ranged from 86.87 to 103.57%. The limit of detection (LOD) 8 PAHs was $0.04{\sim}0.20{\mu}g/kg$, and limit of quantification (LOQ) of 8 PAHs was $0.12{\sim}0.60{\mu}g/kg$. The mean concentration of benzo[a]pyrene (BaP) was $0.34{\mu}g/kg$ from seafood and $0.34{\mu}g/kg$ from dairy products. The total PAHs concentration was $1.06{\mu}g/kg$ in seafood and $1.52{\mu}g/kg$ in dairy products.
