Induction of Symbiosis between Nostoc muscorum and Cultured Plant Cells II. Changes of nitrogen fixation ability and morphology by association of N. muscorum with cultured tobacco cells (Nostoc muscorum과 식물배양세포의 공생유도에 관한 연구 II. N. muscorum과 담배배양세포의 혼합배양에 따른 질소고정능과 형태적 변화)

  • Investigation on the ability of nitrogen usage by N2-fixing Nostoc muscorum and cultured tobacco cells when they were associately cultured on nitrogen-free media was carried out. Also, effect of polyamines on the associated cultured condition was carried out. In addition, morphological changes of N. muscorum and cultured tobacco cells in associate culture were observed to detect the possibility of induction of nitrogen fixing ability on cultued plant cells. The activity of nitrogenase increased markedly when N. muscorum was grown exclusively on nitrogen-free media. When N. muscorum was cultured associately with cultured tobacco cells on nitrogen-free media containing polyamines, high activity was detected in 10-4 M spermine treated group. Investigation on the change of polyamine amounts showed two times increase in spermidine and eight times increase in spermine on a associate culture. These effects of associated culture were shown through morphological change such as dense loclization of N. muscorum around the cultured tobacco cells as well as inside the cells. These results indicate the viability of N. muscorum in cultured tobacco cells and possible induction of nitrogen fixation ability by symbiosis.

  • PU block polymers are constituted by sequences of constitutional units CU which are : a diisocyanate T, a polyol or a polyamine V, an extensor E .Due to the CU different solubility parameters ,hard and soft micro domains are formed;they can aggregate into different phases. A linear PU can be described by two parameters R=T/N and $R_{E}$=(R-1)V/E.A stoichiometric model is proposed which gives the general formula of all the possible PU: X((TV)$_{k}$(TE)$_{n}$)$_{m}$X , where k and m can be calculated from the experimental molecular weights respectively of pre polymer and polymer and n= R-1.Physical properties depend on PU composition and on nature of CU. I.R. analysis is useful for discovering the hydrogen bonds location : DSC gives informations on the presence of the different phases .PALS analysis shows that nano cavities become tighter on increasing n. A series of particular Pus ,obtained from :TDI, a low molecular weight polyol, a cyclic extensor, at different R and RE was prepared and analysed. These PU are amorphous, monophasic and appear to be good candidates for membranes.nes.nes.

  • Rice (Oryza sativa L.) plants were cultivated to examine changes in antioxidative defence mechanism induced by elevated ozone levels. Catalase activities in tolerant Jinpumbyeo and susceptible Chucheongbyeo under ozone fumigation were reduced at 5 hrs and 3 hrs after ozone fumigation, respectively. With the increased ozone supply, peroxidase activity in Jinpumbyeo was steadily enhanced whereas in Chucheongbyeo it was not changed. Four SOD-isozymes were detected by NBT staining of native-PAGE. Two isozymes of them were obviously induced by ozone supply, particularly in Jinpumbyeo. The continuous ozone fumigation increased remarkably putrescine levels in leaves whereas it did not affect the levels of spermidine and spermine. In this study, it was implied that ozone in cell inhibits strongly diamine oxidase and thus promotes ethylene biosynthesis which will cause the senescence in rice plants.

Effect of Cnidii Rhizoma on Phase II Enzyme and Ornithine Decarboxylase Activities (천궁이 Phase II 효소 유도와 Ornithine Decarboxylase 활성에 미치는 영향)

  • Water extract from Cnidii Rhizoma (CRW) was tested for colon cancer chemopreventive activity by measuring the induction of phase II detoxification enzyme activity [quinone reductase (QR) and glutathione S-transferase (GST)] and glutathion (GSH) levels and ornithine decarboxylase (ODC) activity in cultured human colorectal adenocarcinoma HT-29 cells. CRW inhibited cell proliferation in cultured HT-29 cells. CRW induced QR activity in a dose-dependent manner in a concentration range of 0.1${\sim}$5.0 $mg/m{\ell}$. GST activity was also induced with the treatment of CRW in HT-29 cells. In addition GSH levels was increased with CRW. CRW inhibited ODC activity, a key enzyme of polyamine biosynthesis, which is enhanced in tumor promotion. These results suggest that CRW has colon cancer chemopreventive activity by increasing phase II enzyme activity and GSH levels and inhibiting ODC activity in vitro.

  • This study was carried out to investigate the relationship between endogenous polyamines (PAs) and floral bud differentiation in chrysanthemum (Chrysanthemum morifolium). In this study, PA content (free, bound, and conjugated) in apical buds, leaves, and roots changed appreciably during floral bud differentiation. PAs accumulated during series of processes such as floral induction, differentiation of floret primordia, and crown formation in apical buds; changes in PAs in apical buds may have a relationship with those in leaves and roots. The levels of free PAs and conjugated PAs [putrescine (Put) and spermine (Spm)] in apical buds rapidly increased during the initiation stage of floral bud differentiation, while free and conjugated spermidine (Spd) reached their highest levels at the stage of floret primordium differentiation. In the free, conjugated, and bound PA fractions, the changes in Spm content were negligible compared to those of Put and Spd throughout the experiment. These findings indicate that PAs participate in regulating the process of flower bud differentiation in chrysanthemum.

  • The new unsymmetric $N_6$ ligand 1-amino-13-(2-pyridyl)-3,6,9,12-tetraazatridecane (aptatd) containing one pyridyl group has been synthesized and characterized by EA, IR, and NMR. Its proton association constants $(log K_H^n)$ and stability constants $(log K_{ML})$ for Co(Ⅱ), Ni(Ⅱ), Cu(Ⅱ), and Zn(Ⅱ) ions were determined at 298.1 K and ionic strength 0.100 mol $dm^{-3}$ (KNO₃) in aqueous solution by potentiometry: log $K_H^1$=8.80, log $K_H^2$=8.49, log $K_H^3$=6.84, log $K_H^4$=4.17, log $K_H^5$=3.47; log $K_{ML}(Co^{2+})$=18.00, log $K_{ML}(Ni^{2+})$=21.31, log $K_{ML}(Cu^{2+})$=23.62, log $K_{ML}(Zn^{2+})$=15.60. The X-ray structure of its nickel(Ⅱ) complex [Ni(aptatd)]$(ClO_4)_2$ are reported: orthorhombic space group Pbca, a=15.715(1) Å, b=14.280(2) Å, c=19.443(2) Å, V=4363.4 (9) ų with Z=8. The geometry around nickel is a distorted octahedron with the pyridine nitrogen atom being cis to the nitrogen atom of the terminal primary amine.

Experimental and numerical study of the behavior of fiber reinforced concrete beams with nano-graphene oxide and strengthening CFRP sheets

  • In many fiber concrete beams with Carbon Fiber Reinforced Polymer (CFRP), debonding occurs between the carbon sheets and the concrete due to the low strength of the bonding resin. A total of 42 fiber concrete beams with a cross-section of 10×10 cm with a span length of 50 cm are fabricated and retrofitted with CFRP and subjected to a 4-point bending test. Graphene Oxide (GO) at 1, 2, and 3 wt% of the resin is used to improve the mechanical properties of the bonding resins, and the effect of length, width, and the number of layers of CFRP and resin material are investigated. The crack pattern, failure mode, and stress-strain curve are analyzed and compared in each case. The results showed that adding GO to polyamine resin could improve the bonding between the resin and the fiber concrete beam. Furthermore, the optimum amount of nanomaterials is equal to 2% by the weight of the resin. Using 2% nanomaterials showed that by increasing the length, width, and number of layers, the bearing and stiffness of fiber concrete beams increased significantly.

Ginsenoside Rk1 is a novel inhibitor of NMDA receptors in cultured rat hippocampal neurons

  • Background: Ginsenoside Rk1, a saponin component isolated from heat-processed Panax ginseng Meyer, has been implicated in the regulation of antitumor and anti-inflammatory activities. Although our previous studies have demonstrated that ginsenoside Rg3 significantly attenuated the activation of NMDA receptors (NMDARs) in hippocampal neurons, the effects of ginsenosides Rg5 and Rk1, which are derived from heat-mediated dehydration of ginsenoside Rg3, on neuronal NMDARs have not yet been elucidated. Methods: We examined the regulation of NMDARs by ginsenosides Rg5 and Rk1 in cultured rat hippocampal neurons using fura-2-based calcium imaging and whole-cell patch-clamp recordings. Results: The results from our investigation showed that ginsenosides Rg3 and Rg5 inhibited NMDARs with similar potencies. However, ginsenoside Rk1 inhibited NMDARs most effectively among the five compounds (Rg3, Rg5, Rk1, Rg5/Rk1 mixture, and protopanaxadiol) tested in cultured hippocampal neurons. Its inhibition is independent of the NMDA- and glycine-binding sites, and its action seems to involve in an interaction with the polyamine-binding site of the NMDAR channel complex. Conclusion: Taken together, our results suggest that ginsenoside Rk1 might be a novel component contributable to the development of ginseng-based therapeutic treatments for neurodegenerative diseases.

  • Application methods of chemicals were investigated tn minimize strength reduction of corrugated boards under the high humidity environment encountered in the cold chain system. Starch insolubilizers were introduced in the starch solution preparation of the Stain hall method and their insolubilization effect of starch binder were estimated. The performance of water repellent agents(WRA) and moisture proof agents(MPA) were evaluated in terms of water and moisture resistance. And effects of the combination of the chemicals and the coating method were also examined. Addition of the polyamine polyamide insolubilizer to the main part in the Stain hall process improved the binding force and water resistance of starch, which contributed to minimize the strength reduction of paper under the high humidity environment. AZC and Glyoxal type insolubilizers could not be used in the experiment due to an excessively increased viscosity of starch solution and the poor stability. Conventional WRA treatment to the base paper enhanced water and moisture resistance very slightly even though water repellency of the paper reached R10 by the treatment. MPA showed excellent performance than WRA not only in water and moisture resistance but in water repellency. Double coating on paper with MPA was more effective than the single coating at the same coating weight. A newly developed MPA showed excellent performance and runnability only by a single coating instead of a double coating.

  • Nitrogen compounds of Panax ginseng and their biological activities in plant and animal were reviewed. Major nitrogen combounds found in P. ginseng are free amino acids, Water soluble teins, insoluble proteins and peptides. Minor nitrogen compounds are dencichine. glycol)roteins. amines, alkaloides, methoxy or alkyl pyrazine derivatives. free nucleosides and nllrleir arid bases. 4-me- thymi-5-thiazoleethanol and pyroglutamic acid. The contents of total nitrogen and protein in root increased until 13 years old rvhich was the highest age tinder investigation. Soluble protein content increased With the root weight and was higher in xylem pith than cortex-epidermis indicating the rlosc relation with root growth. Arginine which covered 58% of total free amino aroids may serve as a storage nitrogen. Arginine seems to be changed into proline in rhizome, threonine in stem and again threoning and arginine in leaf. The greater the root weight the higher the polyaminc content. Polyamine stimulated the growth of root callus. Physiological roles of other minor nitrogen compounds are unknown although dencichine content is relatively high (0.5% d.w.). biochemical and pharmatological activities of some nitrogen compounds for animal were more investigated than physiological roll iota plant itself. Radiation and U.V. protective function (heat stable protein), insulin-like activity in lipogenesis and lipolysis (adenosine and pyroglutamic acid), depression of blood sugar content (glycopeptide). hemostatir and nellrotoxic activity (denrichine) and. sedative and hypnotic activity (4-methyl-5-thiazoleethilnol) are reported. Heat stable protein increased with root age. The traditional quality critsria appear to be well in accordance with biological activities of nitrogen compounds. Chemical stlldies of nitrogen compounds seem relatively rare, probably dole to difficulty of isolation, subsequently the investigations of biological activities are little.
