The Removal of Algae by Pre-oxidation (전산화 공정을 이용한 조류제거)

  • The blue-green algae which caused odor problem in the tap water are difficult to precipitate in sedimentation basin and clogged the filter void rapidly. The studies of this paper were not only oxidation, coagulation and sedimentation processes for effectively removing blue-green algae but yellow clay and polyamine for verification as coagulants aids. The results of this research are summarized as follows: Higher ozone dose(C) and longer contact time(T) were needed for a high degree of removing blue-green algae efficiency. the removal rate of blue-green algae was proportional to the $C\times T$ value. The removal percent of chlorophyll-a by sedimentation and filter without pre-ozonation was about 75% but 1 mg/L pre-ozonation could increase the removal percent of chlorophyll-a to 99% and more pre-ozonation could remove completely. Though the removal efficiency of turbidity could increased by high dose of chlorination, the dissolved organic carbon was increased. More chlorine dose from 4 to 10 mg/L dissolved organic carbon was decreased. Using yellow clay as coagulant aids increased density of floc so the settling velocity of floc become rising but polyamine could not increase settling velocity of floc though it could formated large floc.

Chemopreventive Effect of Gamdutang Aqua-Acupuncture Solution (감두약침액의 암예방 효과)

  • Gamdutang aqua-acupuncture solution (GAS), Gamdutang water-extracted solution (GWS) and Dae-Gamdutang aqua-acupuncture solution (DGAS) were prepared and tested for chemopreventive potentials. GAS was potent inducer of quinone reductase (QR) activity in Heapa1c1c7 murine hepatoma cells in culture, whereas GWS is less potent. GAS, GWS and DGAS were significantly induced QR activity in cultured rat normal liver cell, Ac2F. Glutathione (GSH) levels were increased about 1.8, 1.0 and 1.1 fold with GAS, GWS and DGAS in Hepa1c1c7 cells, respectively. In addition glutathione S-transferase (GST) activity was increased with GAS, GWS and DGAS. The effects of GAS, GWS and DGAS on the growth of Acanthamoeba castellanii were tested. Proliferation of A. castellanii was inhibited by GAS, GWS and DGAS at concentrations of 1 $\times$ and 5$\times$. These results suggest that GAS has chemopreventive potential by inducing QR and GST activities, increasing GSH levels and inhibition of polyamine metabolism.

Effects of Polyamines and Methylglyoxal bis(guanylhydrazone) on Activity of Diamine Oxidase in Soybean (Glycine max) Seedlings without Cotyledons (자엽을 제거한 대두 유식물에서 Polyamine과 Methylglyoxal bis(guanylhydrazone)가 Diamine Oxidase의 활성에 미치는 영향)

  • The effect of polyamines and methylglyoxal bis(guanylhydrazone)(MGBG) on the activity of diamine oxidase was studied in soybean (Glycine max) seedlings. $10^{-2}\;M$ of putrescine, spennidine, and spermine inhibited diamine oxidase activity, whereas $10^{-6}\;M$ putrescine increased enzyme activity. These results suggest that diamine oxidase can be induced by a specific substrate, putrescine. The content of putrescine was increased in response to the increase in concentratioin of MGBG. In vitro, 40% of the diamine oxidase activity was inhibited by $10^{-3}\;M$ MGBG. In vivo, the diamine oxidase activity was increased by a low concentrration of MGBG. It was suggested that MGBG inhibited the formation of spermidine and that the accumulated putrescine induced diamine oxidase, whereas the diamine oxidase acitivity was inhibited by a high concentration of MGBG. It is suggested that a high cocentration of MGBG increases the putrescine content by inhibiting diamine oxidase activity which is responsible for putrescine degradation.dation.

Testing of Cancer Chemopreventive Potential of Prunella vulgaris L. Aqua-acupuncture Solution Using Biochemical Markers of Carcinogenesis (발암과정 생화학적 표식자를 이용한 하고초 약침액의 암예방 활성 측정)

  • Prunella vulgaris L. aqua-acupuncture solution (PVAS) was tested for cancer chemopreventive activity using chemoprevention-associated biochemical end points. The following effects were measured. : (a) inhibition of 7,12-dimethylbenz[a]anthracene (DMBA)-induced cytochrome P4501A1 activity (b) inhibition of $[^3H]B[a]P-DNA$ binding (c) inhibition of phorbol 12-myristate 13-acetate (TPA)-induced free radical formation in HL-60 cells (d) inhibition of polyamine metabolism. PVAS inhibited cytochrome P4501A1-mediated ethoxyresorufin-O-deethylase activity. The binding of $[^3H]B[a]P$ metabolites to DNA of NCTC-clone 1469 cells was inhibited significantly by PVAS. There is 22% inhibition of TPA-induced free radical formation in human leukemic cells with 5 mg/ml PVAS. Proliferation of Acanthamoeba castellanii was inhibited by PAVS at concentration of 30 mg/ml. PAVS positive in these assays may inhibit the carcinogenesis process and is considered very promising cancer-preventing agent because of its multiple activities.

Insight Into Genes Involved in the Production of Extracellular Chitinase in a Biocontrol Bacterium Lysobacter enzymogenes C-3

  • The chitinase producing Lysobacter enzymogenes C-3 has previously been shown to suppress plant pathogens in vitro and in the field, but little is known of the regulation of chitinase production, or its role in antimicrobial activity and biocontrol. In this study, we isolated and characterized chitinase-defective mutants by screening the transposon mutants of L. enzymogenes C-3. These mutations disrupted genes involved in diverse functions: glucose-galactose transpoter (gluP), disulfide bond formation protein B (dsbB), Clp protease (clp), and polyamine synthase (speD). The chitinase production of the SpeD mutant was restored by the addition of exogenous spermidine or spermine to the bacterial cultures. The speD and clp mutants lost in vitro antifungal activities against plant fungal pathogens. However, the gluP and dsbB mutants showed similar antifungal activities to that of the wild-type. The growth of the mutants in nutrient rich conditions containing chitin was similar with that of the wild-type. However, growth of the speD and gluP mutants was defective in chitin minimal medium, but was observed no growth retardation in the clp and dsbB mutant on chitin minimal medium. In this study, we identified the four genes might be involved and play different role in the production of extracellular chitinase and antifungal activity in L. enzymogenes C-3.

IAA Synthesis and Polyamine Metabolism in Higher Plants: Effects of IAA on the Activity of Diamine Oxidase in Seedlings of Soybean (고등식물에서 IAA합성과 Polyamine 대사에 관한 연구: 대두의 유식물에서 IAA가 Diamine Oxidase의 활성에 미치는 영향)

  • Vie investigated the effect of indole acetic acid (IAA) on diamine oxidase (DAO; EC 1.4.3.6) in the seedling of soybean (Glycine max L. forma Paldalkong). DAO activity was not detected in the resting soybean seeds. During germination it appeared in the elongating zone on day 3, increased up to day 4 and decreased thereafter. Endogenous IAA content has been shown the same pattern as DAO activity. However, cadaverine content was reduced on day 4. To investigate the effect of IAA on DAO activity, the segments of hypocotyl on day 3 were soaked in various concentration of IAA. The activity increased at low concentrations ($10^{-7}-10^{-6}M$) of 1M as compared to that of control but not at high concentrations ($10^{-5}-10^{-4}M$) of IAA. Differing from DAO activity, cadaverine content increased as concentrations of IAA increased. On the other hand, ethylene was induced by IAA at high concentrations. To study the effect of ethylene on DAO activity, we cotreated IAA and 2,S-Norbonadiene (NDE). In this case, DAO activity was not affected.fected.

Effect of Ultraviolet (UV-B) on Antioxidants and Antioxidative Enzymes in Garden Balsam(Impatiens balsamina L.) (자외선(UV-B)이 봉선화(Impatiens balsamina L.)의 항산화제 및 항산화 효소에 미치는 영향)

  • To investigate the effects of ultraviolet(UV-B) on growth and biochemical defense responses of plant, garden balsam (Impatiens balsamina L.) was subjected to enhanced UV-B irradiation [daily dose: 0.02 (No UV-B) and 11.34 (enhanced UV-B) kJ $m^{-2}$ ; $UV-B_{BE}$] for 3 weeks. Enhanced UV-B drastically inhibited leaf area as well as dry weight of garden balsam. The content of malondialdehyde was significantly increased by about 50% after 3 weeks of UV-B irradiation. The ratios of dehydroascorbate/ascorbate and oxidized glutathione/reduced glutathione were also considerably increased by UV-B irradiation. Three major polyamines of garden balsam leaves: putrescine, spermidine and spermine were observed. All polyamine contents were increased with UV-B irradiation. The enzyme (superoxide dismutase, ascorbate peroxidase etc.) activities of garden balsam were increased by the UV-B enhancement. Based on the results, enhanced UV-B caused oxidative stress in garden balsam and biochemical protection responses might be activated to prevent from damaging effects of oxidative stress generated by UV-B irradiation.

The Role of Sympathetic Activity in the Early Phase of Liver Regeneration after Partial Hepatectomy (간-부분절제(肝-部分切除) 후 나타나는 재생과정(再生過程)에서 교감신경계(交感神經系)의 역할(役割)에 관(關)한 연구(硏究))

  • This study was undertaken to confirm whether or not the sympathetic nervous system takes part in the liver regeneration after partial hepatectomy. The male Sprague-Dawley rats were pretreated with I.P. injection of guanethidine 25 mg/kg: single dose (G-1); multiple doses once a day for 3 days (G-3), for 5 days (G-5), or for 6 days (G-6). The rats were subjected to partial hepatectomy $(70.4{\pm}1.99%)$ under light anesthesia of diethyl ether. 1) The systolic blood pressure of control rat was $98.0{\pm}3.9\;mmHg$ and was not affected by G-1. But after the pretreatment with G-3, G-5 or G-6, the pressure was markedly decreased by over 25 %. 2) Both of plasma norepinephrine and epinephrine levels showed the marked increases 3 hrs after the hepatectomy. However, the increases are entirely inhibited by G-1 or G-6. 3) All the liver contents of putrescine, spermidine and spermine showed the significant increases 6 hrs after the hepatectomy and were not affected by G-1 or G-6 with the exception of the inhibition of putrescine increase by only G-6. The present results suggest that the sympathetic activation appeared after partial hepatectomy seems not to play an important role in rat liver regeneration.

Development of TPA-induced Ornithine Decarboxylase (ODC) Inhibitors from Plants as Cancer Chemopreventive Agents

  • Chemical carcinogenesis is associated with the increase of intracellular polyamine levels, and 12-O-tetradecanoylphorbol-13-acetate (TPA)-induced mouse epidermal ODC activity are essential to skin tumor promotion by TPA. Therefore, for the discovery of new cancer chemopreventive agents, we have evaluated about 73 kinds of natural products to study inhibitory effects against ODC activity induced by TPA in T24 cell culture system. The total methanol extracts of plants fractionated into three layers (hexane, ethyl acetate and water layer) were tested and the hexane fraction of Angelica gigas $(root\;bark,\;IC_{50}:\;7.4\;{\mu}g/ml)$ and the ethyl acetate fraction of Corydalis ternata $(root,\;IC_{50}:\;7.5\;{\mu}g/ml)$ were the most effective on the inhibition of TPA-induced ODC activity, These active fractions are under investigation with further sequential fractionation using column chromatography.

Characteristics of Alkaline Sizing Associated with Pulp and Size Fixing Agent (펄프와 정착제에 따른 중성사이징 특성)

  • AKD sizing behavior of handsheets, which were prepared from the different pulps with various AKD fixing agents, was studied in this investigation. AKD sizing performance could be improved by removal of fines in the stock, minimizing damage of fiber, using a suitable fixing agent and heat treatment. Fillers with low surface energy, like calcium carbonate and talc, showed quite positive effect in AKD sizing. When PAM was added to the DIP stock as fixing agent, it showed lower sizing degree than polyamine did. But in case of NBKP system, PAM was more effective than polymine.
