Differential responses of two rice varieties to salt stress

  • Two rice varieties, viz. Nonabokra and Pokkali, have been evaluated for their responses to salinity in terms of some physiological and biochemical attributes. During the exposure to salinity (200 mM concentration of sodium chloride for 24, 48, and 72 h), a significant increase in sodium was recorded which was also concomitant with the changes of other metabolic profiles like proline, phenol, polyamine, etc. The protein oxidation was significantly increased and also varied between the two cultivars. The changes in activities of anti-oxidative enzymes under stress were significantly different to the control. The detrimental effects of salinity were also evident in terms of lipid peroxidation, chlorophyll content, protein profiles, and generation of free radicals; and these were more pronounced in Pokkali than in Nonabokra. The assessment and analysis of these physiological characters under salinity could unravel the mechanism of salt responses revealed in this present study and thus might be useful for selection of tolerant plant types under the above conditions of salinity.

Effects of Cultivation Conditions on the Growth and Polyamine Composition in Methylobacterium extorquens AM1 Growing on Methanol (Methylobacterium extorquens AM1의 메탄올을 이용한 성장과 세포내 폴리아민 구성에 미치는 배양조건의 영향)

  • Methylobacterium extorquens AM1 growing on methanol as a sole source of carbon and energy was found to grow most rapidly (t$t_{d}$ =6h) at 30.deg.C in a mineral medium (pH 7.0) containing 0.5% (v/v) methanol which was agitated at 200 rpm (optimal cultivation condition). Cells grown under the optimal cultivation condition contained more spermidine, but less putrescine, than the cells grown on 2.5%(v/v) ( $t_{d}$ =8h ) or at 20.deg.C ( $t_{d}$ =8h ). Cells cultivated under the optimal condition was found to contain more spermidine than the cells grown at pH 6.0 (( $t_{d}$ =7h ) and pH 8.0 ($t_{d}$ =7.3h). the cells growing at the stationary phase under the optimal condition accumulated more spermine or putrescine than the cells growing at the same phase on 2.5%(v/v) methanol or at pH 6.0 and pH 8.0, respectively. M. extorquens AM1 grown in a medium agitated at 100 rpm ( $t_{d}$ =8.8h ) contained less spermidine and spermine than the cells grown under the optimal cultivation condition.

Physiological and Biochemical Responses of Fifteen Rice Cultivars to UV-B Radiation

  • This study was conducted to examine the physiological and biochemical responses against UV-B radiation in the seedling of 15 different rice cultivars, having the different physiological sensitivities. Out of 15 rice cultivars tested, moderate and susceptible groups showed significant decreases in biomass and RGR (relative growth rate). Contents of total chlorophyll were reduced remarkedly by irradiation of UV-B. In all rice cultivars tested, the content of chlorophyll a was strongly decreased, while the contents of chlorophyll b were slightly reduced without showing clear different among three groups and 15 cultivars. Carotenoid content was largely reduced by UV-B radiation, whereas polyamine content was moderately increased. The contents of MDA (malondialdehyde) that reflect the level of lipid peroxidation of cell membranes were clearly increased by UV-B stress, showing higher content in susceptible cultivars than moderate and torelant cultivars. The physiological important parameters highly related to visible injury were leaf color, chlorophyll, carotenoid, and lipid peroxidation, whereas biomass and polyamines were not closely correlated. Based on this results, it was concluded that changes of visible injury and the contents of chlorophyll and MDA could be adequately applied and utilized as physiological indicators to UV-B radiation.

Effects of Spermine on Quisqualate-induced Excitotoxicity in Rat Immature Cortical Neurons (흰쥐 미숙 대뇌피질 신경세포에서 Quisqualate로 유발된 흥분성 세포독성에 대한 spermine의 영향)

  • Glutamate (Glu) receptor-mediated excitoxicity has been implicated in many acute and chronic types of neurological disorders. Exposure of mature rat cortical neurons (15-18 days in culture) to the various concentrations of Glu resulted in a marked neuronal death, whereas immature rat cortical neurons (4∼5 days in culture) were resistant to the Glu-induced toxicity. Glu receptor subtype-specific agonists showed differential extent of toxicity in the immature neurons. The neurons treated with NMDA or kainate (KA) did not exhibit damage. However, quisqualate (QA) treatment induced a considerable cell death (36.1%) in immature enurons. The non-NMDA antagonist DNQX did not reduce this response. Interestingly, the QA-induced toxicity was potentiated by spermine in a concentration-dependent manner. Again, the spermine-enhanced damage was not altered by the polyamine antagonist ifenprodil. Taken together, unlike NMDA or KA, QA can induce neurotoxicity in immature rat cortical neurons and the QA-induced toxicity was potentiated by spermine. The lack of antagonizing effects of DNQX and ifenprodil on QA-induced toxicity and the potentiated toxicity by spermine, respectively, implies that both QA receptor and the polyamine site of NMDA receptor may not mediate the neurotoxicity observed in this study, and that a distinct mechanism(s) may be involved in excitotoxicity in immature neurons.

  • Investigations on the liability of nitrogen usuage by Nostoc muscorum that has nitrogen fixing ability, and cultured tobacco cells as they were associately cultured on nitrogen-free media and effects of polyamine on the associated culture condition were carried out. In addition, measurement on the activity of nitrate reductase, glutamine synthetase, glutamate dehydrogenase and glutamate synthase that take part in the metabolic pathway of nitrogen fixation product were performed. Among enzymes participating in the metabolic pathway of nitrogen fixation products, the activity of nitrogen reductase stimulated five times in associated culture, and that of glutamine synthetase of N. muscorum increased two times after heterocyst differentiated. Activity of glutamate dehydrogenase increased markedly when cultured tobacco cells were solely incubated on nitrogen-free media, but inhibited when cultured associately. And, glutamate synthase was showed the highest activity in 0.1 mM of spermine treated group.

Polyamine Induces Apoptosis Through the Calcium Signaling in Human Prostate Cancer Cells (전립선암세포에 있어서 폴리아민에 의한 칼슘신호와 세포사멸)

  • Polyamines are essential for the normal cell growth and differentiation. They are also known to have paradoxical dual effects on cell proliferation. In this paper we show that the excess amount of polyamines induces apoptosis through the modulation of calcium signaling in LNCaP human prostate cancer cells. Polyamines, particularly spermidine and spermine, stimulated cell proliferation at a lower concentration (under 10 ${\mu}M$), but it inhibited cell viability at a higher concentration (40 ${\mu}M$). The levels of intracellular $Ca^{2+}$ concentration were increased only at a high concentration of polyamines treatment without any noticeable changes at lower concentrations. Nifedipine did not alter the increase of polyamine-induced $Ca^{2+}$ levels, but flufenamic acid totally abolished the increase of intracellular $Ca^{2+}$ levels. These results mean that polyamines induce $Ca^{2+}$ influx from the surroundings through nonselective cation channels on the cell membrane. The expression of Bcl-2 protein was almost completely blocked, but the level of Bax protein was increased dramatically in the cells treated with high concentration of polyamine. The present study shows that polyamines at a high concentration induce apoptosis through the modulation of intracellular calcium signaling. The increase of intracellular calcium level induced by polyamines, was possibly a result from the extracellular calcium influx through the nonselective cation channels.

  • Aluminium based inorganic flocculants are extensively used in this country in the removal of fine particles present in the raw water for the production of drinking water. These inorganic flocculants, however, have potential hazard of high residual aluminium ions in the treated waters, which is known to be one of the reasons of alzheimer's disease. Inorganic flocculants alone are sometimes incapable of treating water when there are excessive turbidity in the raw water sources due to flooding. A polyamine type polymeric flocculant has long been used to treat raw water in the drinking water production in the European countries and United State of America. The optimum reaction conditions such as mole ratio of epichlorohydrin(EPI) to dimethylamine(DMA), reaction temperature and time for each stage for the pilot scale preparation of polyamine from EPI-DMA was studied in this work. The variation of intrinsic viscosity and flocculating efficiency in the water treatment of the synthesized polyamines were evaluated. The residual materials after polymerization reaction were analyzed by gas chromatography to study the effect of variation of reaction conditions.

Suppression of LH Concentration by Difluoromethylornithine in Gilts (Gilt에 있어서 Difluoromethylornithine에 의한 LH분비 억제)

  • While ornithine decarboxylase (ODC) is considered a key enzyme in the biosynthesis of polyamines, difluoromethylornithine(DFMO) acts as an inhibitor of polyamine synthesis. Cycling crossbred gilts were randomly assigned to one of two (treatment and control) groups (6/group). An indwelling silicone catheter was surgically implanted in the jugular vein of each animal. DFMO was dissolved in saline(200 mg/ml) and adminstered by i. m. injection at a dose of 80 mg/kg/day. The control group received an equivalent volume saline injection. DFMO was injected 3 times daily(08:00. 16:00. 24:00h) from day 16 of estrous cycle to 21 or until estrus. Once daily blood samples (10ml) were taken from day 14 until two days after the last DFMO treatment. Window blood samples were collected every 15 min for 8 h (from 08:00 to 16:00h) starting on day 16 and continuing until day 21 from one gilt per day. Serum progesterone (P$_4$), estradiol (E$_2$), LH and FSH were measured. Typical concentration profiles for P$_4$ and E$_2$ were seen during the follicular phase regardless of DFMO treatment. Injection of DFMO suppressed the preovulatory LH concentration in the serum(p<0.01) while having no effect on FSH profile. The present results indicate that DFMO had an inhibitory effect on LH secretion in the pig, but did not affect PI, E2 or FSH release.

Changes in Carbohydrate, Phenolics and Polyamines of Pepper Plants under Elevated-UV-B Radiation (자외선 조사에 의한 고추 유묘의 탄수화물 합성과 항산화물질 변화)

  • Pepper plants (Capsicum annuum, cv. Manitta) were subjected to different intensities of UV-B radiation to understand alterations of primary- and secondary-metabolism such as carbohydrates, phenolic compounds and polyamines. UV-B doses with a UV-B lamp ($1.2W\;m^{-2}$) were adjusted between 0 to 9 hr. The soluble sugars and starch contents in pepper leaves were highly influenced by UV-B treatment. The soluble sugars altered from $6.7mg\;g^{-1}\;fw$ to $5.2mg\;g^{-1}\;fw$ after 9 hrs of UV-B exposure. The starch contents after 3 hrs of UV-B exposure changed from $17.7mg\;g^{-1}\;fw$ to $12.3mg\;g^{-1}\;fw$ and then remained unchanged. The absorbance of UV-absorbing compounds reached initially maximum at all wavelengths read. On the basis of this result, we analyzed total phenolics, anthocyanin and simple free phenolic acids. Anthocyanin and free phenolic acids responded sensitively with a steady increase during UV-B treatment, although anthocyanin contents declined highly after 3 hrs of treatment. Whereas, there is no alteration of total phenolics (as gallic acid equivalent) caused by UV-B. Free polyamine levels in leaves increased rapidly and highly when UV-B was treated. The most prominent changes in polyamine induction were putrescine and spermidine (+ 70 %) after 3 hrs and spermine (+ 150 %) after 6 hrs.
