  • Poly(lactic acid) is a biodegradable themoplastic based on the renewable resources to substitute for petrochemical plastics. Most of PLA is produced by ring opening polymerization from lactide. However, pyrolysis process in the lactide synthesis is expensive, we studied lactide synthetic process for more economical preparation of PLA. In this research was tried to minimize the pyrolysis time, and obtained L-lactide from lactic acid without any catalyst.

  • A new local drug delivery device to treat otitis media (OM) has been developed. This device consists of a biodegradable poly(L-lactic acid) (PLLA) film containing antibiotic (ampicillin, AMP), which can be placed into the middle ear cavity and release the therapeutic concentration of AMP for prolonged period. Biodegradable films containing AMP (10 w/w%) were prepared by solution casting method using a suspension of the drug in a $PLLA/CH_{2}Cl_{2}$ solution (molecular weight of PLLA, 100,000 (100 K) and 300,000 (300 K), respectively). PLLA-AMP films were characterized by FTIR, DSC, and SEM. In vitro release of AMP from AMP-PLLA films were examined. The release pattern of AMP from AMP-PLLA films remained consistent from 1 day to 14 days, and the release rates of AMP from AMP-100K-PLLA film and AMP-300K-PLLA film were $0.7384\;{\mu}g/ml/day$, $0.4107\;{\mu}g/ml/day$, respectively.

  • For an ideal extractive bioconversion in aqueous two-phase systems, the product has to be preferentially partitioned into the phase opposite to the one in which the biocatalyst is located. Partitioning behaviors of Lactobacillus helveticus IAM 11090 and lactic acid in aqueous two-phase systems composed a polycation, poly(ethylenimine) (PEI), and an uncharged polymer (hydroxyethyl)cellulose (HEC) were investigated. L. helveticus cells were preferentially partitioned to the HEC-rich top phase while about 85% of lactic acid was partitioned to the PEI-rich bottom phase. These results indicate that extraction of charged, low molecular weight products in an aqueous two-phase systems can be promoted by using an oppositely charged polymer as one of the phase-forming polymer. By the ideal partitioning of the cells and lactic acid, an aqueous PEI/HEC two-phase system can be used as a potential system for the extractive lactic acid fermentation of cheese whey.

The Effect of Nucleating Agent for Improving Heat Resistance Properties of L-Lactide Polymer (L-Lactide 폴리머의 내열성 향상을 위한 핵제의 효과 연구)

  • This paper is to review and study about the crystallization nucleating agent to improve the heat-resistance properties of poly(lactic acid). Four sub-micron sized nucleating agents, metallic salts of 2,2'-methylene bis(4,6-di-tert-butylphenol), were prepared and used as a crystallization nucleating agent. Thermal and mechanical properties of polymer compounds were investigated by DSC, HDT and UTM. As the results, While the heat-resistance properties of the polymer compound samples were increased linearly with the contents of nucleating agent as well as their smaller size. Among them, the highest heat-resistance property of compound was observed with 2 wt% of MPZ2. HDT values of PL98Z2 compound was $116^{\circ}C$ at 0.455Mpa.

Polydopamine-mediated surface modifications of poly ʟ-lactic acid with hydroxyapatite, heparin and bone morphogenetic protein-2 and their effects on osseointegration

  • Surface modified poly ${\text\tiny{L}}$-lactic acid (PLLA) samples with hydroxyapatite (HA), heparin and bone morphogenetic protein-2 (BMP-2) mediated by polydopamine (pDA) coating (PLLA/pDA/HA/Hep/BMP-2) were prepared, and their effects on the enhancements of bone formation and osseointegration were evaluated in vitro and in vivo as compared to PLLA, PLLA/pDA/HA, and PLLA/pDA/Hep/BMP-2. The changes in surface chemical compositions, morphologies and wettabilities were observed by X-ray photoelectron spectroscopy (XPS), field-emission scanning electron microscopy (FE-SEM), atomic force microscopy (AFM) and water contact angle measurements. Pre-coating of HA particles with pDA provided uniform and homogeneous anchoring of particles to PLLA surface. In addition, the strong ionic interaction between heparin and pDA led PLLA surface readily heparinized for loading of BMP-2. In vitro experiments revealed that the levels of alkaline phosphatase (ALP) activity, calcium deposition, and osteocalcin (OCN) gene expression were higher in MG-63 human osteosarcoma cell lines grown on PLLA/pDA/HA/Hep/BMP-2 than on control PLLA, PLLA/pDA/HA, and PLLA/pDA/Hep/BMP-2. In vivo studies using micro-computed tomography (micro-CT) also showed that PLLA/pDA/HA/Hep/BMP-2 screw exhibited greatest value of bone volume (BV) and bone volume/tissue volume (BV/TV) among samples. Histological evaluations with H&E and Von Kossa staining demonstrated that a combination of HA and BMP-2 contributed to the strong osseointegration.

Effects of Solvent on the Fabrication of Poly(L-lactide) Scaffold Membranes through Phase Inversion (상전이를 통한 Poly(L-lactide) 스캐폴드 막의 제조에서의 용매의 효과)

  • Porous poly(L-lactic acid)(PLLA) scaffold membranes were prepared via. phase separation process. Chloroform, dichloromethane and 1,4-dioxane were used as solvent and, ethyl alcohol was used as non-solvent. Morphologies, mechanical properties and mass transfer characteristics of the scaffold membranes were investigated through SEM, stress-strain test and glucose diffusion test. The scaffold membranes obtained from the casting solutions with chloroform and with dichloromethane showed similar morphologies. They showed sponge-like porous structure with the pore size in the range of $3-10{\mu}m$ and, their porosities were in 50-80% range. Using 1,4-dioxane as solvent, nano-fibrous scaffold membranes with porosities over 80% were fabricated. When the polymer content in the solution with 1,4-dioxane was lowered to 4%, highly porous, macroporous and nano-fibrous scaffold membranes were obtained. The size of the macropore was tens of the microns and the porosity was around 90%. These results indicate that the solvent has significant effect on the scaffold membrane structure and, that scaffold membranes with various structures can be fabricated through phase separation method by choosing solvent and by controlling polymer concentration in the casting solution.

Fabrication of Poly(L-lactide) Scaffold Membranes through Phase Inversion with Pure and Mixed Solvents (순수용매와 혼합용매를 이용한 상전이를 통한 Poly(L-lactide) 스캐폴드 막의 제조)

  • This paper reports a fabrication of poly(L-lactic acid) (PLLA) scaffold membranes through phase separation process using pure and mixed solvents. Chloroform and 1,4-dioxane were used as pure solvents and mixed solvents were obtained by mixing the pure solvents together. Morphologies, mechanical properties and mass transfer characteristics of the scaffold membranes were investigated through SEM, stress-strain test and glucose diffusion test. Scaffold membranes from the solution with pure chloroform showed solid-wall pore structure. In contrast, nano-fibrous membranes were fabricated from the solution with pure 1,4-dioxane. In case of mixed solvents, the scaffold membranes showed various structures with changing composition of the solvents. When 1,4-dioxane content was lower than 20 wt% in the solvent, scaffold membrane showed solid-wall pore structure. When the content was 20 wt%, scaffold membranes with macropores with the maximum size of $100{\mu}m$ was obtained. In the concentration range of 1,4-dioxane over 25 wt%, the scaffold membranes showed nano-fibrous structures. In this range, the fibers showed different diameters with changing composition of the solvent. The minimum fiber diameter was about $15{\mu}m$, when 1,4-dioxane composition was 80 wt%. These results indicate that the composition of the solvent showed a significant effect on the structure of scaffold membrane.

  • Poly(lactic acid) (PLA) is a highly versatile, biodegradable, aliphatic polyester derived from 100% renewable resources, such as corn and sugar beets. Because of the degradation mechanism, PLA is ideally suited for many applications in the environment where recovery of the product is not practical, such as agricultural mulch films and bags. Composting of post consumer PLA items is also a viable solution for many PLA products. (omitted)

Biodegradable sheath-core biphasic monofilament braided stent for bio-functional treatment of esophageal strictures

  • In this study, a polydioxanone (PDO) and poly(L-lactic acid) (PLLA) sheath-core biphasic monofilament was designed to develop an esophageal stent with improved mechanical properties and controlled biodegradability. The radial force of PDO/PLLA sheath-core stent was 10.24 N, while that of PDO stent was 5.64 N. Deteriorations of tensile strength, elastic modulus and elongation during degradation test were also delayed on PDO/PLLA group. Hyaluronic acid-dopamine conjugate and $BaSO_4/PDO$ conjugate coating layers provided improved tissue adhesion strength and reasonable X-ray contrast, respectively. Taken all together, the sheath-core filaments with tissue adhesive and radiopaque properties will be useful in designing esophageal stents.

Synthesis of Biodegradable Aliphatic Polyester with Amino Group in the Side Chain (곁사슬에 아미노기를 도입한 생분해성 지방족 폴리에스테르의 합성)

  • Aiphatic diester monomer, 3-[(benzyloxycarbonylamino)butyl]-1,4-dioxane-2,5-dione (BABD), was synthesized with the N-$\varepsilon$-benzyloxy-carbonyl-L-lysine as starting material. This monomer was synthesized to add the functionality to poly(lactic acid)s. BABD unit was successfully incorporated into the PLLA chain which was confirmed by $^1H$ NMR. The copolymer composition could be controlled by the feed ratios of monomer. The $M_n$ of this resultant polymer is expected to reach high molecular weight after the purification of monomer and optimization of polymerization time, though the polymer showed relatively low degree of polymerization ($M_n$=3300). The copolymer is expected to possess the enhanced hydrophilicity and the possibility of chemical modification on amino group.
