Use of Androgenesis in Haploid Breeding

  • Haploids are plants with a gametophytic number of chromosomes in their sporophytes. Androgenesis occurs from asymmetric division of pollen grains into generative cells and vegetative cells, followed by re-entry of the vegetative cell during S-phase, which causes microspores progress into G2/M transition in culture. One of the most interesting features of haploids is the possibility to produce doubled haploid (DH) individuals. Doubled haploidy is extremely useful to plant breeders because it enables shortened breeding periods and efficiency in selection of useful recessive agronomic traits. Doubled-haploid technology is not only applicable to breeding, but also to transformation programs of desired genes. In addition to practical breeding programs, DH lines provide useful materials of fundamental genetics including exploitation of QTLs and genes conferred with various agronomic traits by establishing DH populations. This paper provides historical overviews on androgenesis and describes several mechanisms associated with pollen embryogenesis, including mode of actions in pollen embryogenesis, mechanisms of chromosome doubling and factors affecting androgenesis. We also discuss recent progress in application of haploids to breeding, genes associated with in vitro response and drawbacks to anther culture for application of doubled haploids in crop breeding.

Epidermal growth factor 발현을 위한 화분립의 이용 (Utilization of pollen grains for the expression of epidermal growth factor)

  • 개화시기의 수술로부터 수집한 백합화분을 aluminum oxide 미세입자와 섞어 교반에 의해 상해를 발생시켰다. 이어서 이들 화분은 signal peptide-fused epidermal growth factor (EGF) DNA를 지니는 Agrobacterium 세포로 vacuum infiltration을 시켰으며 24 hr 화분신장을 통한 배양을 실시하였다. 이들 신장 화분에서의 EGF mRNA 및 단백질 발현은 성공적으로 확인되었으며 이는 cDNA blot hybridization 및 immuno-blotting의 분석결과이다.

기내 화분배양을 이용한 Tissue Plasminogen Activator 발현분석 (Analysis of Tissue Plasminogen Activator Expression using Pollen Culture in vitro)

  • 백합(Lilium longiflorum)으로부터 수집한 화분에 대하여 기내배양, Agrobacterium 및 vacuum infiltration과정을 이용한 형질전환 그리고 kanamycin배지에서의 선별배양을 통하여 PCR에 의하여 증폭된 1.7 kb의 human tissue plasminogen activator(tPA) cDNA의 발현을 분석하였다. 16시간 정도 배양한 신장화분관의 western blotting결과, human standard와 유사한 크기로의 발현을 확인하였다. 이로써 백합화분은 신속한 단백질발현의 분석을 위한 일회성 생산숙주로서의 가능성을 제시하고 있다.

Genetic Variation of a Single Pollen-derived Doubled Haploid Population in Rice

  • Somaclonal variation was observed in the field on doubled haploid plants derived from single pollen of a rice cultivar "Hwaseongbyeo". The variations of seven quantitative traits including plant height and one qualitative trait (pubescence) in 436 lines ($R_2$ generation) were analyzed. The number of lines which fell beyond the boundaries of the 95% confidence intervals of the check variety, Hwaseongbyeo was checked for each quantitative trait, and of those fertility showed the highest variation frequency (85.6%), followed by plant height (77.5%), flag leaf length (66.5%), grains per panicle (42.2%), days to heading (34.5%), panicle length (30.7%) and panicles per hill (22.7%). And the variations of quantitative traits except days to flowering appeared to move in the negative direction compared to "Hwaseongbyeo". Variability within lines was also observed for quantitative and qualitative traits. Twenty-nine $R_2$ lines (7%) segregated for pubescence and 130 $R_2$ lines (30%) showed variation with regard to fertility. This suggests that mutations usually occur before diploidization. Twenty-nine $R_2$ lines representing a wide spectrum of variation were chosen for RAPD analysis. The number of lines showing DNA polymorphism compared to Hwaseongbyeo ranged 0 from to 10 according to the primer used and this seems to indicate that specific loci have highly mutable genomic site.utable genomic site.

아시아틱 백합 ( Lilium asiatic hybrid 'Gran Paradiso' )의 약배양에 의한 식물체 재생 (Plant Regeneration by Anther Culture of Lilium asiatic hybrid 'Gran Paradiso')

  • 아시아틱백합 'Gran Paradiso'의 약유래 식물체를 획득하기 위해 auxin과 cytokinin을 다양하게 조합한 MS배지에 약배양을 실시하였다. 배양 약은 초기 및 후기 2핵기인 개화 3일 전의 약이 캘러스 유도에 가장 적합하였다. MS배지에 5.0mg/L 2,4-D 단용처리나 1.0mg/L 2,4-D와 1.0 mg/L kinetin이 혼용처리되었을 경우 기관분화성 캘러스가 발생되었으나 NAA와 BA 혼용처리가 2,4-D와 kinetin 혼용처리보다 기관분화를 통한 식물체 재생에 효과적이었다. 캘러스에서 shoot는 0.5mg/L NAA와 1.0mg/L BA혼용처리에서 배양 180경에 분화되었으며 3~4개의 잎이 부착된 다아체와 뿌리 및 소자구는 2.0mg/L NAA와 2.0mg/L BA혼용처리에서 배양 250일 후에 관찰되었다. 재생된 식물체의 근단조사 결과 2배체 (2n=2x=24)로 확인되었으며 식물체를 분에 옮긴 후 2년이 경과되었을 때 모두 개화되었다.

벼의 소포자 배양을 통한 식물체 재분화 (Plant Regeneration from Rice Microspore Cultures)

  • Efficient plant regeneration system was established through the culture of rice (Oryza sativa L.) microspores. Microspores released by anther shedding culture developed into proembryos and calluses in B5 medium within two weeks of culture. The optimal hormone and carbon sources were dependent on the genotypes used. Microspore's viability decreased rapidly in culture time, therefore less than 3% of the total microspores were viable at the 9th day when the first microspore division was observed. Of two types of microspores (pollen dimorphism) observed in culture, only the P-grain, larger microspores than normal one was able to divide. Using 4',6-diamidino-2-phenylindole (DAPI) fluorescence staining, it was confirmed that the symmetrical division of uninucleate microspore was the first step leading to continuous microspore development. Microspore-derived proembryos and calluses were regenerated to plants in N6 medium containing 1 mg/L NAA and 5 mg/L kinetin, and 78.3% of the regenerated plants were haploids.

딸기 내저온성 품종육성에 적합한 교배조합 선발 (Selection of Cross Combination for Development New Cultivar of Low Temperature Tolerant in Strawberry)

  • 딸기의 내저온성 품종육성을 위한 우수 교배친 선발을 위해 '다홍', '감홍', '매향', '설향'을 자방친으로 이용하고 '다홍', '감홍', '매향', '설향', '수경', '선홍', '원교3111호' 등을 화분친으로 하여 24조합을 작성하고 조합능력 검정을 실시하였다. 그 결과, 자방친을 '다홍'으로 하였을 경우에는 전체조합에서 과중이 작아지는 경향이 있었고, '감홍'은 생존율이 낮아 두 품종은 자방친으로 적합하지 않은 것으로 나타났다. 화분친에 있어서도 '선홍'을 화분친으로 이용한 경우 경도가 낮아지는 경향이 있었고, '수경'은 기형과가 다소 발생하는 경향이 있어 화분친으로 적합하지 않았다. 그러나 '매향'과 '설향'을 자방친으로 이용할 경우 발아율과 생존율이 상대적으로 높았고, 과실품질이 우수하고 수량성도 높은 것으로 나타났다. 따라서 '매향'과' 설향'이 자방친으로 우수하였고, 화분친으로는'매향', '설향', ' 원교3111호' 등이 우수하였다. 특히, '매향' × '설향', '설향' × '매향', '설향' × '원교3111호' 조합은 당도, 과실크기, 경도, 수량성 측면에서 우수성이 인정되기 때문에 내저온성 교배조합으로 적합하다고 판단된다.
