Paleoecological Aspect using Palynology since 4,000 Year B. P. in the Lowland of Western Central Korea (화분분석에 의한 한국 중서부 저지대의 4,000년전 이후 고환경)

  • A palynological investigation of the Annyeong-ri, Hwasung-gun Gyeonggi-do, in where Yung-Nung and Gun-Nung are located, was carried out for the first time. From the pollen analysis, two pollen zones are established: Pollen Zone I : Ainus-Quercus-Pinus and Pollen Zone Ⅱ : Pinus. Pollen Zone I is characterized by Mixed coniferous and deciduous broadleaved vegetation during about 3,500 yr. B.P.-2,000 yr. B.P., while after 2,000 yr. B.P. Pollen Zone Ⅱ appears to be Coniferous vegetation and Evergreen broadleaved vegetation. The former reflects there was a lowland fluvio-lacustrine environment before fully lacustrine one which represents the latter. Pollen analysis indicates the vegetation before 2,000 yr. B.P. was controled by environmental factor such as climate conditions, whereas after that time vegetation was strongly effected by human's impact. It is concluded that pollen analysis is useful to find out the relationship between vegetation history and man's interference. In addition freshwater algae play to tell the lake environments.

Utilization of Pollen Grain from Liluim cv. Casablanca as a Transient Gene Expression Host (유전자의 일시발현 분석용 숙주개발을 위한 카사블랑카백합(Lilium cv. Casablanca) 화분립의 이용)

  • Lilium cv. Casablanca pollen grains stored at $-70^{\circ}C$ were grown in pollen germination medium with Agrobacterium tumefaciens LBA4404 cells harboring pBI121 for 18 hr at $27^{\circ}C$. Following this, cefotaxime (250 mg/L) was treated for 6 hr to eradicate the bacterial cells. Histochemical GUS analysis revealed that the transgenic pollen displayed deep blue color mostly from 12 hr after the co-cultivation. Presence of $200\;{\mu}M$ acetosyringone was determined not to be more effective for GUS transformation than its absence. GUS DNA integration in the transgenic pollen genomic DNA was clearly demonstrated by Southern blot analysis.

Effect of Pine Pollen on Rat Liver(II) (송화분이 흰쥐의 간장에 미치는 영향(II))

  • This study was conducted to investigate the nutritional constituents of pine pollen and the effects of pine pollen on rat liver. Using the chemical components analysis method, the constituents of pine pollen are crude protein(13.97%). crude lipid (6.50%), crude ash (3.25%). Detectable in pine pollen are main fatty acids and non-essential amino acids such as glutamic acid and aspartic acid such as oleic and linoleic acid and 18kinds of amino acid including essential amino acids. The effects of pine pollen on rat liver fed with a high fat diet for 6 weeks showed significant decrease in total lipid and triglyceride as compared with the group fed with a fat diet only. Total cholesterol content decreased by 20.33%. No significant difference was found in total protein content of each group. Pine pollen Is efficacious in preventing geriatric diseases (arteriosclerosis, hyper-lipidemia and cardiovascular system-related diseases).

Relationship between Pollen Concentration and Meteorological Condition in an Urban Area (도시지역 공중화분 농도와 기상조건과의 관계)

  • This study attempted to determine important meteorological parameters related to airborne pollen concentrations in urban areas. Hourly pollen measurement data were prepared from a regular sampling with a volumetric Burkard spore trap at a site in the Ulsan city, during the spring season (March~May) of 2011. Results showed that the daily mean and maximum concentrations for total pollen counts during the spring season were statistically significantly correlated with both air temperature and wind speed; daily mean pollen concentration was the most highly related to daily maximum temperature (r=0.567, p<0.001). It was also identified that pollen concentration has a stronger relationship with wind speed at the rural site than at the urban one, which confirms that strong wind conditions over the pollen sources area can be favorable for pollen dispersal, resulting in increases in airborne pollen concentrations downwind. From the results of an oak-pollen episode analysis, it was found that there was a significant relationship between hourly variation of oak pollen concentrations and dynamic meteorological factors, such as wind and mixing height (representing the boundary layer depth); especially, a strong southwestern wind and elevated mixing height was associated with high nocturnal concentrations of oak pollen. This study suggests that temperature, wind, and mixing height can be important considerations in explaining the pollen concentration variations. Additional examination of complex interactions of multiple meteorological parameters affecting pollen behavior should be carried out in order to better understand and predict the temporal and spatial pollen distribution in urban areas.

  • Using a sampling device of our own making, airborne pollen has been monitored in Kyoto, Japan from the middle of February to the end of May 2004. From the morphological analysis of pollen grains by Scanning Electron Microscope (SEM), it was possible to identify some pollen types like Cryptomeria, Pine, Alder, Cyclobalanopsis, Chamaecyparis, and Equisetum. Daily average airborne pollen counts show strong variations from the day to day which makes the appropriate daily forecasts that could be of practical use for patients difficult. Diurnal variation of airborne pollen grains at our local sampling site is very irregular and shows no similarity between pollen types. The highest concentrations of Cryptomeria and Alder pollens in the south -west wind directions might be attributed to the airborne pollen transport, while the increase in Pine pollen grain in the southern wind direction was probably due to the local spread. Prevailing wind direction (SW) during the pollinating periods of Cryptomeria and Alder pollens could suggest a long-distance transport from a distant mountain.

A Marine Terrace Correlated to MIS 5e on the Basis of Pollen Analysis at Sanha-Dong, Ulsan, Southeastern Coast of the Korean Peninsula (화분분석에 의한 한국 남동부 해안 산하동 일대의 MIS 5e 대비 해성단구 동정)

  • The pollen analysis on the deposits of the lower marine terrace I of the estimated paleoshoreline height of 18m was performed in order to estimate the formation age of this terrace developed at the Sanha-dong coast, Ulsan, southeastern coast of the Korean peninsula. The pollen assemblage of the peat layer of SH-1 pollen zone (Quercus-Ulmus/Zelkova zone), lying directly on the marine rounded pebble layer of this terrace, shows that the climatic environment of the deposition period of SH-1 pollen zone was almost similar to that of the Postglacial climatic optimum period, but slightly cooler than that of the late warm stage of Last Interglacial(MIS 5a) in the eastern coast of Korea. This heightens the possibility that the deposition period of the marine rounded pebble layer which was covered by the above SH-1 peat layer is the MIS 5e which has been estimated by a previous study of the sedimentary facies of this terrace deposits (Choi, 2016). The pollen assemblage of SH-2 pollen zone (Pinus-Quercus zone) shows that the climate of this period was almost similar to that of the late Postglacial, but slightly cooler than that of the period of SH-1 pollen zone. This means that the climate around the Sanha-dong was still warmer in the deposition period of the peat layer of SH-2 pollen zone. Thus, the peat layer of SH-2 pollen zone was considered to have been deposited during the period from the early regression stage of the MIS 5d which is the estimated final stage in the deposition period of the above peat layer of SH-1 pollen zone to any stage in which the warmer environment of MIS 5 has still lasted. The humic silt layer of SH-3 pollen zone (Pinus-Ulmus/Zelkova-Abies zone) is assumed to have been deposited during the interstadial of the Last Glacial (MIS 3).

Pollen analysis of the Lake Hane, Ohda-city, Shimane Prefecture, Southwest Japan (남서 일본 도근현 大田市 파근호 퇴적물의 화분분석)

  • The pollen of the sediments from the Lake Hane were analysed for reconstruction of vegetation history around Ohda-City in Southwest Japan, and the correlation with the pollen zone reported for the Lake Shinji. From the pollen assemblages of 29 samples from core OH94 (19.7 m), two pollen zone were established, and they were subdivided into five subzones in ascending order as following; Cyclobalanopsis-Catanopsis pollen zone (Cyclobalanopsis subzone, Catanopsis subzone and, Podocarpus subzone), and Gramineae pollen zone (Cryptomeria subzone and Cyclobalanopsis-Quercus subzone). The K-Ah volcanic ash is identified in the Cyclobalanopsis-Catanopsis pollen zone. We estimated the beginning of the Cyclobalanopsis-Catanopsis pollen zone was before c.a. 6,300 BP.

PRK1, a Receptor-like Kinase from Petunia inflata, is Essential for Post-meiotic Development of Pollen and Embryo Sac

  • We previously identified and characterized a predominantly pollen-expressed gene of Petunia inflata that encodes a receptor-like kinase named PRK1. The extracellular domain of PRK1 contains leucine-rich repeats which have been implicated in protein-protein interactions, and the cytoplasmic domain was found to autophosphorylate on serine and tyrosine. To investigate the function PRK1 in pollen development, we transformed P. inflata plants with a construct containing the promoter of a predominantly pollen-expressed gene of tomato, LAT52, fused to an antisense PRK1 cDNA corresponding to part of the extracellular domain of PRK1, There transgenic plants were found to each produce approximately equal amounts of normal and aborted pollen. Analysis of the inheritance of the transgene inserts in two of the transgenic plants, ASRK-13 and ASRK-20, to their progeny revealed that certain transgene inserts cosegregated with the pollen abortion phenotype. Microscopic examination of the aborted pollen grains showed that their outer wall, the exine, was essentially normal, but that their cytoplasm contained only starch-like granules. Staining of the nuclei of the microspores at different stages of uninucleate stage. However, at subsequent stages half of the microspores completed mitosis and developed into normal binucleate pollen, but the other half initially remained uninucleate, then lost their nucleio. Analysis of the amounts of PRK1 mRNA and the antisense PRK1 transcript suggested that the pollen abortion phenotype most likely resulted from down-regulation of the PRK1 gene by the antisense PRK1 transgene. These results suggest that PRK1 plays an essential role in a signal transduction pathway that mediates post-meiotic development of microspores.

Storage of Pollen Biofactory for in vitro Growth and rProtein Synthesis (기내배양과 재조합 단백질 생산을 위한 화분 Biofactory의 저장기술의 개발)

  • A method of collection and long-term storage of viable lily (Lilium longiflorum) pollen grains were developed for their in vitro growth and transformation in consistency. Petroleum ether, n-heptane, cyclohexane and benzene, as pollen collection medium, were determined less toxic to pollen growth in vitro than others tested. Pollen grains, however, lost their growth activity if stored in these solvents more than a week, So, a serial performance, that is, pollen grain collection in these solvents, air-drying and immediate transfer to low temperature condition was determined desirable for keeping the viability much longer. Pollen grains from this storage showed a successful transformation in vitro with a cDNA encoding tissue plasminogen activator (TPA) protein using Agrobacterium via vacuum infiltration according to western blotting analysis.

Analysis of UreB Protein Synthesis from Transgenic Lily Pollen (형질전환 백합화분을 이용한 UreB단백질의 발현분석)

  • In an attempt to produce recombinant proteins using the pollen enriched in some plant species, a 1.7 kb DNA encoding urease subunit B (UreB) amplified by PCR from Helicobacter pylori urease gene cluster in pH808 plasmid was cloned to be expressed under CaMV35S promoter in lily (Lilium longiflorum) pollen tubes elongated in vitro. Lily pollen at early germinating stage was transformed with the ureB DNA using Agrobacterium via vacuum infiltration and, incubated for a full pollen tube growth 16 - 24 h in the dark in the presence of kanamycin. DNA integration and expression in the transgenic pollen were analyzed by the standard molecular techniques and the results suggest that the pollen in vitro may be employed as a protein factory in a disposable fashion.
