  • Supercritical Fluid Chromatography(SFC) has become a technique for solving problems that are difficult to be monitored by other chromatographic methods. However, the most widely used fluid, is no more polar than hexane. Polar samples which are difficult to be analyzed with pure supercritical $CO_2$ because of their high polarity can be separated by adding polar modifiers to supercritical $CO_2$. In this paper, a new method for monitoring the mobile phase composition in modified supercritical fluid chromatography was developed. The amount of water dissolved in supercritical $CO_2$ was measured by amperometric microsensor which is made of thin film of perfluorosulfonate ionomer(PFSI). The amount of water dissolved in supercritical $CO_2$ stayed constant for a much longer time than with a saturator column. With this new mixing device, we could get good resolutions for vitamins which are difficult to separate with pure $CO_2$.

Purification and Properties of Pectate Lyase Produced by Recombinant Strain -Containing pelK Gene from Alkalitolerant Bacillus sp. YA- 14 (알칼리내성 Bacillus sp. YA-14 유래의 Pectate Lyase 유전자를 함유한 재조합균주로부터 효소의 정제 및 특성)

  • Pectate lyase produced by recombinant strain containing pectate lyase gene from alkalitolerant Bacillus sp. YA-14 was succesively purified with 257.6 purification folds and a 10.2% yields by the affinity method, eM-cellulose column chromatography followed by gel filtration on Sephadex G-I00 column. The optimal pH and temperature for pectate lyase activity were 10.0 and $60^{\circ}C$, respectively. The enzyme was stable between pH 4.0 and 10.0, and up to $50^{\circ}C$. The molecular weight of this enzyme was estimated to be 43,000 daltons by SDS-PAGE. Amino acid analysis showed that the enzyme contained more polar and basic amino acids, especially serine, glycine and tyrosine, than that of various pectate lyase from other strains. The N-terminal amino acid sequence was Ala-Asp-Leu-Gly-His-Gln-Thr.

Effect of Proline on First Polar Body Formation in Porcine Primary Oocyte

  • Follicular fluid (FF) contains an oocyte maturation inhibitor with unknown chemical properties. This study was carried out to chemically define the factor(s) inhibiting cumulus cell denudation (CD) and first polar body formation (PBF). Porcine FF (PFF) was extracted with methanol and the extract was serially separated using gel filtration on Superose 12 and Superdex columns. A Superdex fraction was derived with PITC and analyzed with an amino acid analysis column. The results obtained are as follows; PFF had an activity inhibiting both CD and PBF of porcine primary oocytes. Superdex fractions RV2.11 prepared from PFF exhibited an activity inhibiting CD and PBF. By amino acid analysis, the fraction RV2.11 appeared to be proline having the same activity inhibiting CD and PBF. In conclusion, PFF had oocyte maturation inhibitors, of which proline should inhibit CD and PBF.

Cryoprotective Properties of Exopolysaccharide (P-21653) Produced by the Antarctic Bacterium, Pseudoalteromonas arctica KOPRI 21653

  • Twenty-five bacterial strains that secrete mucous materials were isolated from sediment obtained from King George Island, Antarctica. Seven of these strains proved capable of producing cryoprotective exopolysaccharides. The strain KOPRI 21653 was selected for the further study of an anti-ice-nucleating polysaccharide (ANP), which originated from a polar region. KOPRI 21653 was identified as Pseudoalteromonas arctica as the result of 16S rRNA analysis. The exopolysaccharide, P-21653, was purified completely from the KOPRI 21653 cell culture via column chromatography and protease treatment. The principal sugar components of P-21653 were determined to be galactose and glucose, at a ratio of 1:1.5, via GC-MS analysis. The cryoprotective activity of P-21653 was characterized via an E. coli viability test. In the presence of 0.1% (w/v) P-21653, the survival ratio of E. coli cells was as high as 82.6% over three repeated freeze-thaw cycles. The survival ratio decreased drastically to 71.5 and 48.1 %, respectively, in five and seven repeated cycle conditions; however, the survival ratios were greater over three (96.6-92.1%) to seven (100.5-91.6%) freeze-thaw cycles in the presence of 0.5 and 1.0% (w/v) P-21653. In addition, at much lower concentrations (0.1-1.0%), P-21653 resulted in survival ratios (83.1-98.4%) similar to those of two commercially available cryoprotectants ($V_{EG}$ plus X-1000, 92.9% and $V_{M3}$, 95.3%), which were utilized at the recommended concentrations (90%). The biochemical characteristics of exopolysaccharide P-21653 reflect that this compound may be developed as a useful cryoprotectant for use in medical applications and in the food industry.

  • Flavours in kimchee are the result of unique combination of various sugars, organic acids and amino acids as well as various volatile organic compounds including sulfur-containing compounds, terpenes, alcohols, and some volatile organic acids. In the experiment for the flavour extracting methods, dynamic headspace(DHS) is more effective for collection of volatile flavour than simultaneous distillation extraction(SDE). The best polarity available at the moment is 5% phenyl methyl poly-siloxane which will separate non-polar, intermediate and polar components with good resolution.

  • Five compounds of terpenoids and coumarins were isolated from the non-polar fraction of Artemisia apiacea by open column chromatography. Their structures were elucidated as $\alpha$-amyrin (1), $\beta$-amyrin (2), $\beta$-sitosterol (3), 5,6,7-trimethoxycoumarin (4) and 6-methoxy-7,8-methylenedioxycoumarin (5) by chemical and spectroscopic analysis. This is the first report of the isolation of $\alpha$-amyrin, $\beta$-amyrin, 5,6,7-trimethoxycoumarin and 6-methoxy-7,8-methylene-dioxycoumarin from this plant.

Water Column Structure and Dispersal Pattern of Suspended Particulate Matter (SPM) in a floating ice-dominated fjord, Marian Cove, Antarctica during Austral Summer (유빙이 점유한 남극 마리안 소만의 하계기간 수층 구조와 부유물질 분산)

  • Vertical measurement of CTDT at about 30 min intervals and spatial surface temperature, salinity, and concentration of suspended particulate matters were conducted to elucidate the character of water column and the dispersal pattern in a floating ice-dominated fjord, Marian Cove, West Antarctica. Marian Cove showed two distinct water layers in terms of turbidity; 1) cold, fresh, and turbid surface plume in the upper 2 m,2) warm, saline, and relatively clean Maxwell Bay inflow between 15-45 m in water depth. Thermal melting of Maxwell Bay inflow and tidewater glacier/floating ices developed the surface mixed layer and the activity of floating ices cause Maxwell Bay inflow to be unstable. Due to the unstable water column, the development of Maxwell Bay inflow and subsequent surface plume are not influenced by tidal frequency. Coastal current generated by strong northwesterly wind may extend warm, saline, and turbid surface plume into the central part of the cove along the northern coast via the western coast of Weaver Peninsula. Terrigenous sediments of meltwaters from the glaciated ice cliffs near the corner of tidewater glacier and some coasts enter into the cove and their dispersion depends upon the hydrographic regimes (tide, wind, wave etc.). At the period of spring tide, the strong wind stress with the northwesterly wind direction reserve suspended sediment-fed surface plume and so allow the possibility of deposition of terrigenous sediments within the basin of cove.

Gas chromatographic profiles of rose essential oils: a round-robin test on oil of rose, Chinese Kushui type (Rosa sertata × Rosa rugosa) (장미 정유의 기체 크로마토그래피 분석표: 중국산 쿠스이형 장미유에 대한 재현정밀도 시험)

  • Analysis of the aroma constituents present in the rose essential oil of Chinese Kushui type (Rosa sertata ${\times}$ Rosa rugosa) by GC-FID and GC-MS was performed independently as an expert for the inter-laboratory round-robin test to verify reproducibility according to the decision of the preliminary meeting of ISO/TC-54 (Shanghai, Sep. 14-15, 2010). Total 179 peaks (using SPB-1 apolar column), 165 peaks (using DB-624 intermediate polar column), and 162 peaks (using Supelcowax-10 polar column) were separated by GC-FID, respectively. Major constituents (over 5%) by GC-FID were ${\beta}$-citronellol (41.6~46.7%), geraniol (9.7~11.0%), and nerol (3.4~4.5%). ${\beta}$-Citronellol peak was overlapped with nerol peak on SPB-1 and DB-624 columns, whereas the two peaks were separated each other on Supelcowax-10 column. Our results were generally consistent with Chinese data (ISO/DIS 25175); however, a peak of phenethyl alcohol separated by using PEG (Supelco wax) column was found at the quite different retention time. Comparative analysis was conducted using Bulgarian rose (Rosa damascena Miller) oil and perfume. Bulgarian rose oil showed rich amounts of characteristic aroma constituents than the essential oil of Chinese Kushui type.

Lipid Composition of Barley Flour Produced in Korea (한국산(韓國産) 보리가루의 지방질(脂肪質) 조성(組成))

  • Grains of naked barley (Baekdong cultivar) were polished, powdered and subjected to the successive extraction into free and bound liquid fractions. These were further fractionated into lipid classes and quantified by means of thin layer chromatography, column chromatography and gas-liquid chromatography. Contents of free and bound lipids in barley flour were 2.27% and 1.01%, which were decreased to 2.12% and 0.76%, respectively, after purification. Free and bound lipids were consisted of monoglycerides, diglycerides, triglycerides, free sterols, sterol esters, free fatty acids and polar lipids. Major constituents of free lipids were 56.2% triglycerides, 14.9% free fatty acids and 13.4% sterols while those of bound lipids were 73.8% polar lipids, 8.4% free fatty acids and 5.2% triglycerides. The content of non-polar lipids in free lipids was 93.6% as compared with 26.2% in bound lipids. However, phospholipids content in bound lipids was 55.5% as compared with 2.5% in free lipids, and glycolipids content in bound lipids was 19.4% as compared with 3.9% in free lipids. Major fatty acids in the free and bound lipid fractions were linoleic acid 52.1%, 54.8%, palmitic acid 24.8%, 30.0% and oleic acid 15.6%, 8.8%, respectively, showing similar patterns in both fractions. The amount of unsaturated fatty acids in free lipids was 72.8% as compared with 68.0% in bound lipids. In comparing the fatty acid composition of non-polar lipids, glycolipids and phospholipids, no difference was observed between free and bound lipid fractions while a slight difference was found among the lipid constituents.
