Effect of Gelatinase and Chitinase Producing Microorganism on the Growth of Soybean and Control of Stink Bug in Field

  • This study was conducted to investigate the application effect of Lysobacter antibioticus HS124 (gelatinase and chitinase producing microorganism; GCM) for the improvement of soybean yield and control of stink bug. Our results showed that the yield of GCM-treated soybean increased by 17.0, 20.3, 19.0, and 25.6% in the experimental field sites of Muan-gun, Sunchang-gun, Gwangju 1, and Gwangju 2, respectively, compared to the yield of the soybean obtained by conventional practice treatment (CPT); however in the Gimjae-si, the yield decreased by 10.6%. Results in both pods and seeds $plant^{-1}$ were significantly increased in Gwangju 2 by the GCM cultured broth treatment (GCMT). Ratio of 3 seeds $pod^{-1}$ in Sunchang-gun was statistically significant between GCMT and CPT, however, the result in the other field showed no significance. Germination rate was only statistically improved by GCMT in Gwangju 2 field site. GCMT reduced the appearance of stink bug in all experimental field sites except in Gimjae-si. The soybean seed damage by stink bug was no significance in all of treatments. Therefore, GCMT could improve the productivity of soybean and also control the infestation of stink bug.

Effects of Hexaconazole on Growth and Antioxidant Potential of Cucumber Seedlings under UV-B Radiation

  • The present study was conducted to determine the effect of hexaconazole (HEX), a triazole fungicide, on the growth, yield, photosynthetic response and antioxidant potential in cucumber (Cucumis sativus L.) plants subjected to UV-B stress. UV-B radiation and HEX were applied separately or in combination to cucumber seedlings. The growth parameters were significantly reduced under UV-B treatment, however, this growth inhibition was less in HEX treated plants. HEX caused noticeable changes in plant morphology such as reduced shoot length and leaf area, and increased leaf thickness. HEX was quite persistent in inhibiting shoot growth by causing a reduction in shoot fresh and dry weight. HEX noticeably recovered the UV-B induced inhibition of biomass production. Significant accumutation in anthocyanin and flavonoid pigments in the leaves occurred as a result of HEX or UV-B treatments. HEX permitted the survival of more green leaf tissue preventing chlorophyll content reduction and higher quantum yield for photosystemII under UV-B exposure. HEX treatment induced a transient rise in ABA levels in the leaves, and combined application of HEX and UV-B showed a significant enhancement of ABA content which activates $H_2O_2$ generation. UV-B exposure induced accumulation of $H_2O_2$ in the leaves, while HEX prevented UV-B induced increase in $H_2O_2$, indicating that HEX serves as an antioxidant agent able to scavenge $H_2O$ to protect cells from oxidative damage. An increase in the ascorbic acid was observed in the HEX treated cucumber leaves affecting many enzyme activities by removing $H_2O_2$ during photosynthetic processes. The activities of antioxidant enzymes including catalase(CAT), ascorbate peroxidase(APX), superoxide dismutase(SOD) and peroxidase(POD) in the leaves in the presence of HEX under UV-B stress were higher than those under UV-B stress alone. These findings suggest that HEX may participate in the enhanced tolerance to oxidative stress. From these results it can be concluded that HEX moderately ameliolate the effect of UV-B stress in cucumber by improving the components of antioxidant defense system.

Effects of Flooding Treatment on Physiological Characteristics of Rice Cultivars (수도 관수처리에 의한 생리적 특성의 품종간 차이)

  • To clarify the varietal differences of resistance to complete flooding, physiological characteristics in relation to flooding resistance in each growth stage of rice plants, the present study was carried out. The rate of survival after flooding at seeding stage of rice plants of the cultivar 'FR 13A' and Ind. X Jap. cultivars were 91.8% and 33.8% to 40%, respectively, while the Japonica varieties were completely dead. The tolerant varieties which has higher rate of survival showed the higher $O_2$ release, the higher catalase activity and lower peroxidase activity according to flooding treatment. Japonica type varieties showed the higher elongation of plant during flooding, lower recovery of flooding damage and the abrupt decrease of yield according to the increase of flooding periods in comparision to Ind. X Jap. crossed varieties when they were flooded at the active tillering stage. When rice plants were flooded at the booting stage, Ind. X Jap. crossed varieties had the higher photosynthesis, respiratory rate, root oxidizing power, ethylene evolution and lower yield reduction in comparision to japonica varieties.varieties.

Feasibility Study on the Utilization of EMAT Technology for In-line Inspection of Gas Pipeline

  • If gas is leaking out of gas pipelines, it could cause a huge explosion. Accordingly, it is important to ensure the integrity of gas pipelines. Traditionally, over the years, gas-operating companies have used the ILI system, which is based on axial magnetic flux leakage (MFL), to inspect the gas pipelines. Relatively, there is a low probability of detection (POD) for the axial defects with the axial MFL-based ILI. To prevent the buried pipeline from corrosion, it requires a protective coating. In addition to the potential damage to the coating by environmental factors and external forces, there could be defects on the damaged coating area. Thus, it is essential that nondestructive evaluation methods for detecting axial defects (axial cracks, axial groove) and damaged coating be developed. In this study, an electromagnetic acoustic transducer (EMAT) sensor was designed and fabricated for detecting axial defects and coating disbondment. In order to validate the performances of the developed EMAT sensor, experiments were performed with specimens from axial cracks, axial grooves, and coating disbondment. The experimental results showed that the developed EMAT sensor could detect not only the axial cracks (minimum 5% depth of wall thickness) and axial grooves (minimum 10% depth of wall thickness), but also the coating disbondment.

Economic Injury Levels for the Common Cutworm, Spodoptera litura (Fabricius) (Lepidoptera: Noctuidae) on Soybean (콩 담배거세미나방(Spodoptera litura)의 경제적 피해수준)

  • The damage aspects of soybean by common cutworm, Spodoptera litura (Fabricius) (Lepidoptera: Noctuidae) at different larval density and different growth stage of soybean were studied in soybean field. The percent yield reduction(Y) of soybean infested by different densities of S. litura (X, no. of larvae/plant) under outdoor conditions for a three week period were estimated by the following equations: (1) Y = 1.655X - 6.025 ($R^{2}=0.952$) for the R1 (flowering stage); (2) Y = 0.725X - 0.475 ($R^{2}=0.986$) for the R3 (beginning pod stage); and (3) Y = 0.635X - 1.325 ($R^{2}=0.986$) for the R5 (beginning seed stage). Based on the relationships between the densities of S. litura larvae and the yield index of soybean, the number of larvae (2nd-3rd instar) which caused 5% loss of yield (Tolerable injury level) was estimated to as approximately 6.7 for the R1, 7.5 for the R5, and 10.0 per plant for the R5, respectively. Average soybean leaf areas consumed by 1st, 2nd, 3rd, 4th, 5th and 6th larvae of Spodoptera litura during 24 hr at $28^{\circ}C$ was 0.3, 0.7, 2.6, 4.0, 20.1, and $55.8\;cm^{2}$, respectively.

Studies on Dry Matter Production and Variation of Agronomic Characteristics of Determinate and Indeterminate Types of Soybean Cultivars (Glycine max L.) Under Different Growing Condition (유ㆍ무한형대두품종의 재배조건에 따른 건물생산 및 형질변이에 관한 연구)

  • To provide useful information for developing new high yielding soybean varieties and for improving cultural practices, an investigation was made on variation of dry matter production and on relationship among several agronomic characters of soybean plants grown under different planting times and densities as well as under different fertilizer levels, using Kwang-kyo, Dong puk-tae, and Suke # 51 as determinate types and Shelby, SRF-300 and Harosoy as indeterminate types at the Crop Experiment Station during the period of 1972 and 1973. The results obtained were summarized as follow: 1. The dry weight, CGR and LAI at the initial flowering stage were high in the high plant population irrespective of varieties, planting times, and fertilizer levels. However, those characters of the indeterminate type were lower than those of the determinate types. The same characters of the indererminate type at the terminal leaf stage were either same or higher than those of the determinate types. 2. The dry weight of the determinate type at the initial flowering stage was similar to the indeterminate, type, when planting times were May 21 or June 15. The dry weights of both types of varieties were low when planted on July 10. When fertilizer levels were increased, the CGR, dry weight and LAI at the initial flowering stages were also increased. 3. Even though significant differences of LAI were obtained among the varieties within the same plant type, the indeterminate type was in general lower than that of the determinate type regardless of planting time and densities, or fertilizer levels, while the yield of the indeterminate type was comparable to the yield of the determinate type. 4. The high degree of leaf- and petiole-fall at the greenbean stage was highly associated with early planting and high levels of fertilizers. However, less amount of leaf- or petiole-fall was found when planted on July 10 or under low plant population. 5. The percent of stem weight was high under higher plant population, while the percent of leaf weight was high under lower plant population. When planting time was late, the percent of stem and petiole weight were reduced, while the leaf weight was increased. 6. The percent of pod weight of the determinate type at the terminal leaf stage was about 2% when planted on May 21, about 8% when planted on June 15, and about 9% when planted on July 10. The percent of pod weight of the indeterminate type at the terminal leaf stage were about 6 % when planted on May 21, 14% when planted on June 15 and 21% when planted on July 10. 7. Kwang kyo showed less degree of leaf-fall even when lodged due to high levels of fertilizer applied, while SRF-300 showed great damage due to lodging. 8. High yields were obtained when planted on May 21, but there were little yield differences between yields from May 21 and June 15 plantings. The reduction of yield due to late planting of July 10 was less apparent in the determinate type of varieties, while it was high in the indeterminate type. 9. The optimum plant population per are for high yield was 1, 250 to 2, 500 plants when planted on May 21, 2, 500 plants when planted on June 15, and 3, 333 plants when planted on July 10. 10. High correlation coefficients were obtained between dry matter weight and LAI at the terminal leaf stages, and between the dry matter weight and yield at the greenbean stages. The optimum dry weight for high yield in the determinate type was expected to be 25 kg. per are at the initial flowering stage and 50 kg. per are at the terminal leaf stage. In the indeterminate type the LAI and dry weight at the greenbean stage were 4 to 5 and 80 kg. per are, respectively. 11. Under the high plant population plant height was increased, while the stem diameter and the number of nodes and branches were reduced. Consequently, the percent of mainstem to main stem plus branches were increased, and the length of internode was also elongated. The ratios of stem weight, number of nodes and pods, and yield of main stem were increased when high plant population was associated with the early planting. The percent of main stem to branches for the indeterminate type was higher than that of the determinate type. 12. Under the high plant densities and late planting, the percent of the pod number and yields of main stem were increased, indicating that varieties with no or less branches were better adaptable under such conditions. 13. High degree of simple correlation coefficients was obtained between the LAI at the initial flowering stage and terminal leaf stage, and the total node number, dry matter and dry stem weight of both determinate and indeterminate types. Even though no significant correlation was found between the LAI at the initial flowering stage of the determinate type and the stem length and pod number per are, highly significant correlation coefficients were obtained between such characters in the indeterminate type of varieties. 14. The dry matter was positively correlated with the LAI, CGR, stem length, and pod number, node number and dry stem weight per are, while no significant correlation was found between the dry matter and stem diameter. 15. The correlation coefficients between lodging index and the LAI, dry weight, stem length and dry stem weight were highly significant. Negative correlation was obtained for the indeterminate type between the stem diameter and lodging index. The correlation coefficient between the stem diameter and lodging index was non-significant for the determinate type, while positive correlation was obtained between the yield and lodging index in the determinate type. The lodging index was also positively correlated with average length of internode of main stem. 16. The 100 seed weight appeared to be lowered under the high plant population and no fertilizer condition, and when planted late. Apparent differences of 100 seed weight were found between main stem and branches, being higher for the main stem than for the branches. 17. No variation of protein content was found due to different cultural practices. However, the oil content was apparently reduced when planted late.

Screening for Genotypes Lacking Lipoxygenase from Germplasm Collection of Korean Soybean Land Races (한국 재래종 콩집단에서 비린내 없는 콩품종 육성을 위한 Lipoxygenase 결실인자 변이 연구)

  • Soybean seeds contain lipoxygenase, which is responsible for the objectionable beany flavors in soybean seeds. The isozymes of lipoxygenase (1$\times$1, 1$\times$2, 1$\times$3) were discovered in United States of America, Japan, and Korea, and the mode of inheritance of the mutant genes was determined. This investigation was conducted to screen lipoxygenase-1, 2, and 3 lacking soybean lines from the Korean soybean land race population. Two lipoxygenase-1lacking lines, KAS 610-8 and KAS 621-8 were found in this investigation. In general, lipoxygenase acking varieties were small in seed size and low in oil content. A severe pod borer damage was observed in the two selected lipoxygenase-1 lacking lines. Lipoxygenase lacking line was not found in Korean wild soybean population used in this study and the lipoxygenase lacking lines were found only in Kyung-Nam province and the results imply that lipoxygenase lacking mutants were induced recently in cultivars.

Growth and Yield Components Responses to Delayed Planting of Soybean in Southern Region of Korea (남부지역 콩 만파에 따른 품종별 생육 및 수량반응)

  • Double cropping system including paddy field soybean is widely adopted nationwide, due to rise in market price and its higher income than paddy field rice. Sowing date of soybean as a second crop is being delayed depending on first crop's growth period and harvesting time. Due to the increased temperature in October and delayed first frost date, soybean could be harvested without frost damage even in late-plating. Therefore, selection of soybean cultivar which is appropriate for this environment is very important. The effect of sowing date and genotype of soybean on growth and yield was investigated for three planting dates (June 20, July 5, and July 20) with ten cultivars developed for soy-pate production, to figure out plant development and yield pattern in delayed planting. As planting date is delayed, plant height and pod number was decreased and this pattern was more clearly detected in mid-late maturity cultivars. Hundred-seed weight did not show significant changes even in late planting, due to compensations between yield components. Yield reduction of July 20 in contrast to that of June 20 showed that Nampung (9.6%) showed the least yield decline. Maximum yield was achieved from Daepung, Taekwang, and Uram among other soybean cultivars in late planting. Shortening of growth period was strongly detected in reproductive stage while length of vegetative stage was regularly maintained in both early and mid-late maturity cultivars.

Morphological Characteristics and Effects of Temperature on the Development of Piezodorus hybneri (Gmelin) (Hemiptera: Pentatomidae) on Soybean (콩 가로줄노린재(Piezodorus hybneri)의 형태적 특징 및 온도별 발육특성)

  • This study was conducted to investigate morphological characteristics and effects of temperature on the development of Piezodorus hybneri on soybean. The unibanded stink bug, Piezodorus hybneri, is a serious insect pest in soybean fields giving damage to seeds in pod and leaves of soybean lowering both quality and yield. Eggs were spherical and laid in two raws on the leaves and pods of soybean plants. Body lengths of females and males were 9.8 mm and 8.7 mm, respectively. Egg hatch rates were better in higher temperature within the range of examined temperatures, which ranged in $81.2{sim}93.2%$. The development periods of eggs at the temperatures of 20, 25, 30 and $35^{\circ}C$ were 10.7, 5.0, 4.0, and 3.0 days, respectively. Mean developmental periods of 1st, 2nd, 3rd, 4th and 5th nymphs at $25^{\circ}C$ were 3.2, 3.4, 3.4, 3.3 and 5.9 days, respectively. Development threshold and effective accumulative temperature were $13.3^{\circ}C$ and 65.5 DD (day degree) for egg stage, $9.9^{\circ}C$ and 322.8 DD for nymph stage, $10.7^{\circ}C$ and 386.4 DD from egg to adult, respectively. Oviposition began from 10 days after emergence at $25^{\circ}C$, and the longevity of female and male were 52.5 and 38.2 days, respectively. Total number of eggs and egg masses laid by a female at $25^{\circ}C$ were 496 and 21.3, respectively, The longevity of adult female was shortened with increasing temperature, whereas the total numbers of eggs laid by a female were decreased.
