Characterization of Mitochondrial Plasmids from Pleurotus spp. (Pleurotus속 균주들의 미토콘드리아 플라스미드 특성)

  • Plasmid DNAs were detected from the mitochondrial fraction of four strains of whiterot fungus, Pleurotus ostreatus. The size of the plasmids were 10.2 and 7.2 kb in strain NFFA 2, 10.2 kb in NFFA 4001, 11.2 kb in NFFA 4501, and 10.2 and 11.2 kb in KFCC 11635. The two strains,NFFA 2ml and NFFA 2m2, which are mutant derivatives of NFFA 2, did not contain any plasmids. The cleavage by proteinase K indicated that these plasmids have DNA ends associated with proteins. In digestion with proteinase K all the plasm ids remained resistant to lambda exonuclease which hydrolyzes DNA from 5' ends and were sensitive to exonuclease III which hydrolyzes DNA from 3' ends. This suggests that the plasmids are linear double-stranded DNA and the terminal proteins are covalently linked to 5' ends of plasm ids. In order to find relationship between these plasmids, hybridization of plasm ids by each separate plasmid DNA was done. The result indicated that the plasmids can be classified into at least 3 groups. Plasmids of group I were present in all the P ostreatus. More mitochondrial plasmids were detected in P cornucopiae. P ,florida, P pulmonarius, P sajor-caju, and P spodoleucus. The size of plasmids ranged between 7.2 kb and 14 kb. All the species except P cornucopiae contained plasmids of approximately 10 kb which hybridized with the 10.2 kb plasmid (group I) of P ostreatus NFFA 2.

Isolation of antibiotic resistant Lactobacillus spp from fermented milk and starters, and detection of transmissible R plasmids (유산균 발효유 및 starter에서 항생제내성 유산균 분리 및 전달성 R plasmids 분포조사)

  • Plasmids are covalently closed circular molecules of DNA that are stably inherited and replicate somewhat independently of the bacterial chromosome. Genes carried on plasmids can mediate a wide variety of important functions, including antibiotics (R plasmids) and heavy metals resistance, toxins production, cell penetration, iron chelation, complement resistance, and metabolic characteristics such as sucrose and lactose fermentation. Fifty strains of lactobacilli were isolated from 26 staters and 29 fermented milk products. They were classified 27 strains as Lactobacillus paracasei subsp paracasei, 11 stains as Lactococcus lactis subsp cremoris, 6 strains as L delbrueckii subsp lactis, 4 strains as L acidophius, and 2 strains as L delbrueckii subsp bulgaricus. All of these strains were examined for drug resistance and transferability of R plasmids. All of the isolates were sensitive to Am, C, CF, E, NB, P, T, and Te. But resistant to SXT 94% (47 strains), K 66% (33 strains), S 56% (28 strains), ENR 50% (25 strains), NOR 38% (19 strains) CIP 38% (19 strains), GM 16% (8 strains), and N 14% (7 strains), in order. And 32 different resistant patterns were found. The most frequently encountered patterns were CIP-ENR-K-NOR-S-SXT (5 strains). In vitro R plasmids transfer experiment, 57 antibiotic resistant strains which were not transfer to the recipient 2 Escherichia coli strains by conjugation, These results indicate that Lactobacillus in internal trade market' stater recognize R factor but transmissible R plasmid is not existed.

Dynamics of Supercoiled and Relaxed pTZ18U Plasmids Probed with a Long-Lifetime Metal-Ligand Complex

  • $[Ru(bpy)_2(dppz)]^2+$ (bpy=2,2'-bipyfidine, dppz=dipyrido[3,2-a:2',3'-c]phenazine) (RuBD), a long-lifetime metal-ligand complex, displays favorable photophysical properties. These include long lifetime, polarized emission, but no significant fluorescence from the complex that is not bound to DNA. To show the usefulness of this luminophore (RuBD) for probing the bending and torsional dynamics of nucleic acids, its intensity and anisotropy decays when intercalated into supercoiled and relaxed pTZ18U plasmids were examined using frequency-domain fluorometry with a blue light-emitting diode (LED) as the modulated light source. The mean lifetimes for the supercoiled plasmids (< $\tau$ >=148 ns) were somewhat shorter than those for the relaxed plasmids (< $\tau$ >=160 ns). This suggests that the relaxed plasmids were shielded more efficiently from water. The anisotropy decay data also showed somewhat shorter slow rotational correlation times for supercoiled plasmids (288 ns) than for the relaxed plasmids (355 ns). The presence of two rotational correlation times suggests that RuBD reveals both the bending and torsional motions of the plasmids. These results indicate that RuBD can be useful for studying both the bending and torsional dynamics of mucleic acids.

Drug Resistance and R. Plasmids of Enterobacteria Isolated from Bovine Udder Infections (젖소 유방염유래(乳房炎由來) 장내세균(腸內細菌)의 약제내성(藥劑耐性) 및 R. Plasmids)

  • A total of 250 enteric bacteria (148 Escherichia coli, 41 Klebsiella pneumoniae, 46 Enterobacter spp. and 15 Proteus spp.) isolated from bovine udder infections in 1979 through 1980 were examined for drug resistance and prevalence of R. plasmids. The drug tested were streptomycin (SM), kanamycin (KM), ampicillin (AP), chloramphenicol (CP), tetracycline (TC), gentamicin (GM), oxolinic acid (OA) and nalidixic acid (NA). The detection of R. plasmids was performed with Escherichia coli ML 1410 NAr as the recipient. Of the 148 Escherichia coli isolated, 68(45.9%) were found to be resistant to one or more drugs tested, and about 50% of the resistant strains were multiply resistant. of the 68 drugresistant strains, 13(19.1%) were found to carry R. plasmids which were capable of performing a conjugal transfer. CP resistance was transfered together with the other resistance. Of 41 strains of Klebsiella pneumoniae isolated, 90.2% were resistant to the drugs, alone or in combination thereof. Strains resistant to AP and TC were 63.4%, and 48.8%, respectively. R. plasmids were detected in 78.4% of the drug-resistant strains, and these strains transfered all or a part of their drug resistance pattern. AP and CP resistance were transfered in 100% of AP and CP-resistant strains. Eleven (37.9%) of 29 R. plasmids showed a thermosensitive transfer. Of the 46 strains of Enterobacter spp. isolated, 37(80.4%) were resistant to the drugs tested. A high percentage of resistance was noted for AP(65.2%). All strains resistant to four or more drugs transferred their resistances to Escherichia coli ML 1410, but strains resistant to three or fewer drugs did not transfer the resistances. All of the 15 Proteus strains isolated were resistant to more than two drugs. of them, 6 were quadruple resistance to SM, KM, CP and TC, and 9 were double one to AP and TC. Three (20.0%) of the drug-resistant isolates had R.plasmids conferring AP and TC resistance. GM, OA and NA of the drugs tested were very active to all of 250 Gram-negative enteric bacteria isolated from bovine udder infections.

Characterization of Tetracycline Resistance Plasmids of Staphylococcus aureus (황색포도상구균의 테트라사이클린 내성 플라스미드 동정)

  • Plasmids were isolated from 15 tetracycline (Tc) resistant S. aureus. Two small tetracycline resistance plasmids, pKH16 and pKH17, have been isolated from Staphylococcus aureus JY10 and Staphylococcus aureus JY22, respectively and the complete nucleotide sequences of those plasmids have been determined. pKH16 consisted of 4,442 bp and showed high identity to pKH6 (99% matching percentage) isolated in 1989 from S. aureus SA2. pKH17 consisted of 4,441 bp and showed less identity to pKH6 (95% matching percentage) than pKH16. PCR analysis showed that tetK and tetM did not exist in ten large plasmids isolated from ten Tc resistant S. aureus. Twelve Tc resistant S. aureus showed reistance both to Tc and Mn and we might analogize that twelve Tc resistant S. aureus had tetM in their chromosome.

Effects of Phenolic Compounds and Hosts on the vir Gene Expression of Various Ti Plasmids

  • The vir genes expression of Ti plasmid is induced by a family of related phenolic compounds. We investigated the effects of various phenolic compounds, Ti plasmids and hosts on the expression of the vir genes in the same type of octopine Ti plasmids, pTiKU12, pTiAch5 and pTiA6. The vir gene induction of pTiKU12 was remarkably stimulated by p-coumaric acid in relation to acetosyringone, but those of pTiAch5 and pTiA6 were more stimulated by acetosyringone than by p-coumaric acid. The effect of phenolic compound on the vir gene induction was different according to the kind of Ti plasmids. Also, the vir gene expression of A. tumefaciens KU913, which has pTiKU12 was about 6.2 times as much as that of A. tumefaciens KU915, which has pTiKU12 in KU12 host, in the presence of ferulic acid. But no difference was shown in the presence of p-coumaric acid. The vir gene induction abilities of phenolic compounds are different according to the kinds of phenolic compounds, Ti plasmids and hosts.

  • Previously, we reported on the antibiotic resistance patterns of 50 strains of Staphylococcus aureus which were isolated from a hospital in Busan, Korea from July 2005 to December 2006. We have isolated small plasmids and classified plasmid types by agarose gel electrophoresis. We have selected 5 plasmids and determined complete nucleotide sequences of those plasmids. The aim of this paper is to report on the characteristics of cadmium, erythromycin, lincomycin resistance plasmids and a cryptic plasmid based on the sequence analysis obtained by using the BLAST program.

Genetic Transformation of Geobacillus kaustophilus HTA426 by Conjugative Transfer of Host-Mimicking Plasmids

  • We established an efficient transformation method for thermophile Geobacillus kaustophilus HTA426 using conjugative transfer from Escherichia coli of host-mimicking plasmids that imitate DNA methylation of strain HTA426 to circumvent its DNA restriction barriers. Two conjugative plasmids, pSTE33T and pUCG18T, capable of shuttling between E. coli and Geobacillus spp., were constructed. The plasmids were first introduced into E. coli BR408, which expressed one inherent DNA methylase gene (dam) and two heterologous methylase genes from strain HTA426 (GK1380-GK1381 and GK0343-GK0344). The plasmids were then directly transferred from E. coli cells to strain HTA426 by conjugative transfer using pUB307 or pRK2013 as a helper plasmid. pUCG18T was introduced very efficiently (transfer efficiency, $10^{-5}-10^{-3}\;recipient^{-1}$). pSTE33T showed lower efficiency ($10^{-7}-10^{-6}\;recipient^{-1}$) but had a high copy number and high segregational stability. Methylase genes in the donor substantially affected the transfer efficiency, demonstrating that the host-mimicking strategy contributes to efficient transformation. The transformation method, along with the two distinguishing plasmids, increases the potential of G. kaustophilus HTA426 as a thermophilic host to be used in various applications and as a model for biological studies of this genus. Our results also demonstrate that conjugative transfer is a promising approach for introducing exogenous DNA into thermophiles.

  • The effect of stb locus of E. COLI IncFII plasmid NR1 on the stability of chimeric plasmids was investigated. First, we have isolated the stability locus (stb) from E. coli NR1 plasmid and then inserted into the three different vectors, pUC8, YRp17 and YEp24. By examining their stability in E. coli and yeast, we showed that the recombinant plasmids containing stb locus were resonably stable. Also, by comparing the amounts of the rDNA fragments per haploid genome with those of the plasmid fragments, we showed they copy number of recombinant plasmids was not increased. Consequently, the stb locus of E. coli IncFII plasmid NR1 stabilized the chimeric plasmids but did not affect the replication or copy number of plasmids.

Characterization of Plasmids from Bifidobacterium sp.

  • Ten strains of plasmid-harboring Bifidobacterium sp. were isolated from the feces of adults and children, and named as Bifidobacterium sp. GE1-GE8, ST, and SH5. These plasmids were categorized into three homologous groups (pKJ50-homologous, pKJ36-homologous, and non-homologous groups) according to Southern hybridization patterns using the formerly characterized bifidobacterial plasmids, pKJ50 and pKJ36, as probes. nine strains harboring the plasmids were shown to accumulate single-stranded DNA as a replication intermediate, based on the S1 nuclease treatment and Southern hybridization. These results suggest that the strains replicate by a rolling circle mechanism. Minimal inhibitory concentrations (MIC) of the isolated bifidobacteria against several antibiotics were determined. Two strains, GE2 and GE3, showed relatively high MiC values against tetracycline ($793.6{\mu}g/ml$) and erythromycin ($153.6{\mu}g/ml$), respectively. The tetracycline resistance of GE2 disappeared when the resident plasmid of GE2 was cured by ethidium bromide. These results show that pKJ36-homologous and pKJ50-homologous plasmids are prevalent among various Bifidobacterium strains and some Bifidobacterium plasmids appear to code for antibiotic resistance.
