Plasma Concentrations of Vitamins E and A, and Effects of Vitamin E Supplementation on Oxidative Stress and Immune Status in Korean Non-Insulin Dependent Diabetic Patients

  • Plasma concentrations of Vitamins E and A were measured in 15 non-insulin dependent Korean female subjects and 15 age-matched normal subjects using reversed-phase high-performance liquid chromatography. No differences were found in plasma Vitamin E concentrations between the 2 groups. Plasma Vitamin A concentrations were higher in subjects with non-insulin dependent diabetes melitus (NIDDM). The effects were evaluated of 4 weeks of daily supplementation of 400 mg Vitamin E on plasma levels of these two vitamins. In addition, the effects were observed for Vitamin E supplementation on oxidative stress and immune-related compound productions in non-insulin dependent diabetic patients and control subjects. After treatment with Vitamin E, plasma Vitamin E concentrations were significantly elevated in both groups. Basal plasma thiobarbituric acid reactive substances (TBABS) were identical, and a decreased level of TBARS caused by Vitamin E was observed only in the diabetic group (0.02739$\pm$0.0024 versus 0.01814$\pm$0.0008 nmols malondialdehyde equivalents/dl plasma ; p<0.05). The basal and after-treatment levels of immunoglobulins A, G, M were identical in control and diabetic groups, indicating that Vitamin E did not appear to alter gross humoral responses in this study. However, elevation of Complement 3 ($C_3$) was noticed due to Vitamin E supplementation, revealing a possible effect of vitamin E on one aspect of humoral immunity, Furthermore, an increase in prostaglandin E_2 ($PGE_2$) levels in diabetic patients was normalized by Vitamin E supplementation. This suggests indirectly that the depressed cell-mediated response due to elevated $PGE_2$ could be normalized. For the definitive antioxidant intake recommendations for prevention and treatment of adverse effects of non-insulin dependent diabetes, evidence from intervention trials like this study should be collected. The present data suggests that Vitamin E may oxen some protective effects against oxidative damage and might have beneficial effects of partial immune-stimulation.

Nutritional Status of Antioxidant Vitamins in the Elderly Living in Ulsan Metropolitan City

  • Body antioxidant status is an important factor for the prevention of many chronic diseases in the elderly. This study was done to evaluate the nutritional status of antioxidant vitamins of the elderly by determining their intakes and plasma levels. It was also aimed to compare daily intakes and plasma levels of antioxidant vitamins by sex of age. Subjects were 225 elderly persons aged over 60 years old (63 males, 162 females) living in Ulsan area. Subjects were divided by groups according to age(< 65, 65-74, 75) and sex. Dietary Intakes were assessed by semi-quantitative food frequency questionnaires(FFQ). Plasma Vitamin C level was measured by 2,4-dinitrophenylhydrazine method and plasma levels of vitamin E, A and ${\beta}$-carotene were measured by HFLC. The average intakes of vitamin C were 104.9g(150% of RDA) and 104.4g(149% of RDA) in the elder1y males and females, respectively. Vitamin C intake of the elderly was significantly decreased by aging but not different by sex. The average intakes of vitamin A were 678$\mu\textrm{g}$RE(96.9% of RDA) and 604$\mu\textrm{g}$:RE(86.2% of RDA) in elderly males and females, respectively. The average levels of Plasma vitamin C were 6.22mg/L and 11.45mg/L in the elderly males and females, respectively. Those levels are within normal range(6-20mg/L). However the percentage of the elderly males with deficiency(< 2mg/L) and marginal level(2-4mg/L) of vitamin C were 27.4% and 16.1%, respectively. Plasma retinaol levels were 0.39mg/L for the elderly males and 0.37mg/L for the elderly females, which were within normal range. But the percentage of subjects with marginal level were 28% ill both males and females. Plasma ${\alpha}$-tocopherol levels of the elderly were lower than normal range(5-12mg/L). Plasma levels of vitamin C, E and ${\beta}$-carotene, except retinol, were significantly higher in the elderly females compared to males and showed significant decrease by aging.

노인의 혈중 호모시스테인, 엽산, 비타민 $B_{12}$ 수준 및 영양소 섭취 상태와 신경인지기능과의 관련성 (Relationship among Plasma Homocysteine, Folate, Vitamin $B_{12}$ and Nutrient Intake and Neurocognitive Function in the Elderly)

  • This study examined the relationship among plasma homocysteine, folate, and vitamin $B_{12}$ levels and neurocognitive function in 118 community-dwelling elderly subjects (mean age, $75.1{\pm}6.7$ years). The Mini-Mental State Examination (MMSE-KC) was used to screen and assess neurocognitive function in the participants. Dietary intake data including the use of dietary supplements were obtained using the 24-hour recall method by well-trained interviewers. Plasma folate and vitamin $B_{12}$ concentrations were analyzed by radioimmunoassay, and homocysteine was assessed by a high performance liquid chromatography-fluorescence method. The proportions of participants with suboptimal levels of plasma folate (< 3 ng/mL), vitamin $B_{12}$ (< 221 pmol/mL), and homocysteine (> $15{\mu}mol/L$) were 16.1%, 5.9%, and 21.2%, respectively. A multiple regression analysis showed that plasma homocysteine was negatively associated with plasma folate and vitamin $B_{12}$ levels. The MMSE-KC test scores were significantly associated with plasma homocysteine and folate, but not with vitamin $B_{12}$, after adjusting for age, gender, body mass index, living with spouse, education, current smoking, energy intake, and chronic diseases such as hypertension, diabetes, thyroid disease, dyslipidemia, stroke, and cardiovascular disease. A general linear model adjusted for covariates revealed that MMSE-KC test scores increased from the lowest to the highest quartiles of vitamin $B_1$, vitamin $B_2$, vitamin $B_6$, vitamin $B_{12}$, and vitamin C intake (p for trend = 0.012, 0.039, 0.014, 0.046, 0.026, respectively). These results indicate that the problem of folate inadequacy and hyperhomocysteinemia are highly prevalent among community-dwelling elderly people and that dietary intake of the B vitamins and vitamin C is positively associated with cognitive function scores.

소아 장중첩증과 혈중 비타민 C의 관계에 관한 연구 (Relationship between Intussusception and Vitamin C Concentrations of Whole Blood and Plasma)

  • 목적: 장내 장중첩증 원인 바이러스의 침습에 의한 임파선 비대 및 장중첩증으로 인한 교액성 장폐색증은 혈중 비타민 C의 감소를 동반할 수 있으며, 또한 체내 비타민 C가 부족할 경우 장내 adenovirus 감염 및 침습을 유발할 수 있을 것이다. 본 연구에서는 장중첩증과 전혈과 혈장의 비타민 C 농도와의 관계에 대해 알아보고자 하였다. 방법: 1995년 5월부터 1998년 12월까지 경상대학병원 응급실로 내원하여 진단받았던 장중첩증 환아 57명과 장중첩증 연구군과 연령 분포가 동일한 진주시에 거주하는 건강한 소아 256명을 대조 군으로 2, 4-dinitrophenylhydrazine 방법을 이용하여 전혈과 혈장 비타민 C를 측정하였고 장중첩증 환아의 나이, 성별, 백혈구 수, 발열 유무, 발병에서 진단까지의 시간, 혈변의 유무와 수술 유무 등을 후향적으로 조사, 비교하였다. 결과: 장중첩증으로 내원한 환아들의 평균 나이는 1년 3개월이었으며 남아가 39명, 여아가 18명이었다. 장중첩증 환아의 전혈 비타민 C농도는 $1.49{\pm}0.64$ mg/dL로 대조군의 $2.18{\pm}0.49$ mg/dL에 비해 낮았고(P=0.0001), 혈장 비타민 C농도는 $0.59{\pm}0.36$ mg/dL로 대조군의 $1.47{\pm}0.56$ mg/dL 보다 낮았다(P=0.0001). 장중첩증 환아군에서 전혈 비타민 C 농도 중앙값이 여아 1.56 mg/dL, 남아 1.34 mg/dL로, 남아가 여아에 비해 낮았다(P=0.003). 그러나 장중첩증 환아의 연령, 백혈구수, 발열, 발병에서 진단까지의 시간, 혈변과 수술의 유무에 따라서는 전혈 및 혈장 비타민 C의 농도는 차이가 없었다. 결론: 장중첩증이 있는 환아는 건강 대조군에 비해 전혈과 혈장 비타민 C의 농도가 낮았지만 이것이 원인인지 또는 결과인지는 본 연구로 알 수 없었다. 장중첩증과 비타민 C의 관계를 정확하게 알기 위해서는 전향적인 연구가 필요할 것으로 사료된다.

캐모마일, 세이지, 녹차 건분의 섭취가 노령흰쥐의 항산화능에 미치는 영향 (Effect of Dried Powders of Chamomile, Sage, and Green Tea on Antioxidative Capacity in 15-Month-Old Rats)

  • This study was performed to investigate the effect of chamomile, sage, and green tea intakes on anti oxidative capacity in 15-month-old rats. Dried powders of three plants were analyzed to determine the amount of total flavonoids, $\beta$-carotene, vitamin C, vitamin E, and dietary fibers. In order to examine the change of antioxidative capacity in old rats, forty-eight Sprague-Dawley male rats weighing 621.2$\pm$9.5 g were divided into four groups according to body weight and fed for four weeks with each experimental diet of three dried powders and control diet. Plasma and liver thiobarbituric acid reactive substances (TBARS) levels and xanthine oxidase (XO) activities, erythrocyte and liver superoxide dismutase (SOD) activities and plasma vitamin A, C, E, and total carotenoids levels were measured. The total flavonoids and vitamin C contents were the highest in green tea powder. Beta-carotene and vitamin E contents were not significantly different among all three dried powders. Total dietary fiber contents also were not different among all three dried powders, but soluble dietary fiber contents of chamomile was higher than other two dried powders. Plasma TBARS level was found to be significantly lower in all the experimental groups as compared to control group. Chamomile powder group, especially, showed the lowest level among all experimental groups. Liver TBARS levels of experimental groups were also lower than that of control group and significant differences were observed in chamomile and green tea groups compared to control. Plasma XO activity of green tea group was significantly lower than control group. Liver XO activities of green tea and chamomile groups were significantly lower than control group. Erythrocyte SOD activity was not significantly different among all the groups. However, liver SOD activities of sage and green tea groups showed a significant increase as compared to control group. Plasma vitamin A level was not significantly different among all the groups. Plasma total carotenoids levels were found to be significantly higher in experimental groups as compared to control group. Plasma vitamin E level of chamomile group and vitamin C level of green tea group showed the highest level among all the groups. In conclusion, chamomile, sage, and green tea intakes had an effect on improving antioxidative capacity in 15-month-old rats. Effects of green tea and chamomile powders, especially, were higher than sage powder.

Pharmacokinetic Characterization of Nano-emulsion Vitamin A, D and E (LaVita) in Rats

  • BACKGROUND: Bioavailability enhancement by solubilization of lipophilic drugs in nano-emulsion has been reported and it may be useful in pharmaceutical and nutraceutical products. This study was performed to compare in vivo bioavailability of nano-emulsion formulation with that of the general product as control. METHODS AND RESULTS: The pharmacokinetics assessment of Vitamin A, D and E complex of nanoemulsion formulation (LaVita), in comparison to the general product, was performed in the male rat plasma by a single oral dose at 20 mL/kg body weight (n=3/group). For nano-emulsion formulation (LaVita), $C_{max}$ of vitamin A and E in plasma were much higher and the area under the curve (AUC) of vitamin A, D and E were 14-63% higher, and the half-life of vitamin E was 2-fold longer than the general product. According to statistical analysis, each $C_{max}$ of vitamin A, D & E was significantly higher (p<0.01, 0.05 and 0.01, respectively) than that of general product. Half-life of vitamin A was significantly higher (p<0.01) and AUC of vitamin A and D were also significantly higher than the general product. CONCLUSION(s): Considering significant increment of $C_{max}$ and AUC, LaVita made of nano-emulsion could be more effective the absorption rate and extent for bioavailability of vitamin A, D & E than those of general product.

Long-Term Intake of High Doses of Vitamin C Down-regulates Anti-oxidant Enzymes in Human Erythrocytes

  • We located a group of healthy young males (aged $20{\sim}30$) who had been taking a high dose (more than 5 g) of vitamin C daily for more than one year. We observed that this vitamin C group had plasma levels of vitamin C that were more than three times that of the control group. The control group had not taken any additional vitamin C except for that included in their diets. But the vitamin C group showed significantly lower amounts of Cu/ZnSOD, catalase and glutathione-s-transferase and lower activities of glutathione peroxidase and glutathione reductase in erythrocyte lysates than the control group. However, there was no difference in the plasma levels of lipid peroxides between the two groups. These results suggest that vitamin C offsets its own contribution to anti-oxidant activity by repressing the expression of anti-oxidant enzymes and also excludes the possibility that vitamin C acts as a pro-oxidant in vivo.

The Effect of Vitamin {TEX}$B_{6}${/TEX} Deficiency and Age on Plasma Cholesterol Profile in Intensely Exercised Rats

  • The purpose of this study was to determine whether vitamin {TEX}$B_{6}${/TEX} deficiency and age affect the blood cho-lesterol profile in exercising rats. Fifty four rats were fed either a viramin {TEX}$B_{6}${/TEX} deficient dief(-{TEX}$B_{6}${/TEX}) of a control diet(+{TEX}$B_{6}${/TEX}) for 6 weeks, then subdivided into 3 groups:non-exercise group(NE), exercise and sacrifice group(ES), exercise and recuperation group(ER). ES group was exercised in treadmill({TEX}$10^{o}${/TEX}, 0.5~0.8km/h) for 2 hours and sacrifice. ER group was recuperated three days with respective diet after exercise. At week 3 and 6, and level of plasma total cholesterol(TC), high density lipoprotein cholesterol(HDL_C) and low density lipoprotein cholesterol(LDL_C) were compared. In NE group, there was no difference in the levels of TC, HDL_C and LDL_C between +{TEX}$B_{6}${/TEX} rats and -{TEX}$B_{6}${/TEX} rats. The plasma levels of TC and LDL_C of 6 weeks were higher than those of 3 weeks and on difference in HDL_V between 3 weeks rats and 6 weeks rats. In ES group, there was also no difference in the levels of TC, HDL_C and LDL_C between +{TEX}$B_{6}${/TEX} rats and -{TEX}$B_{6}${/TEX} rats and there was no difference in TC, LDL-cholesterol between 3 weeks rats and 6 weeks rats. The level of HDL_C pf 6 weeks was lower than that of 3 weeks rats. In ER group, there was no difference in the levels of TC and LDL_C not only between +{TEX}$B_{6}${/TEX} rats and-{TEX}$B_{6}${/TEX} rats but also between 3 weeks rats and 6 weeks rats. The level of HDL_C was lower in -{TEX}$B_{6}${/TEX} rats than in +{TEX}$B_{6}${/TEX} rats and higher in 6 weeks rats than in 3 weeks rats. These results suggest that vitamin {TEX}$B_{6}${/TEX} deficiency may affect the HDL_C during exercise and after recuperation. The desirable effect of exercise on plasma Cholesterol profile is strengthened in adult age than young age.

A Concentration-Function Basis for Ideal Vitamin C Intake

  • Vitamin C is an essential nutrient involved in many functions. Humans are unable to synthesize vitamin C de novo, because they lack the last enzyme in the biosynthetic pathway. Previous Recommended Dietary Allowances (RDAs) for vitamin C were based on prevention of deficiency with a margin of safety. However preventing deficiency may not be equivalent to ideal nutrient intake. Recommendation should be based on vitamin function in relation to concentration. For this goal, data set of the relationship between wide-range of vitamin C dose and resulting concentrations in plasma and tissues and characterization of functional outcomes in relation to these concentrations should be acquired. This article reviews the current knowledge in these areas and suggest how this knowledge may contribute toward establishing dietary guideline for ideal vitamin C intake.
