Feeding Unprotected CLA Methyl Esters Compared to Sunflower Seeds Increased Milk CLA Level but Inhibited Milk Fat Synthesis in Cows

  • An experiment was conducted to compare the effect of the same amount of 18:2 offered either as 18:2n-6 or as a mixture of unprotected 18:2c9t11 and 18:2t10c12 on feed intake, milk components as well as plasma and milk fatty acid profile. Fifteen cows were blocked by milk yield and milk fat percentage and within block assigned randomly to 1 of 3 treatments (n = 5). Each cow passed a 12-d adjustment period (AP) on a basal diet. After the AP cows received 1 of 3 supplements during an 18-d experimental period (EP). The supplements contained either 1.0 kg ground sunflower seeds (S), 0.5 kg conjugated linoleic acid (CLA)-oil (C) or 0.75 kg of a mixture of ground sunflower seeds and CLA-oil (2:1; SC). All 3 supplements contained the same amount of 18:2 either as CLA (${\Sigma}18$:2c9t11+18:2t10c12, 1:1) or as 18:2c9c12. During the last 2 d of AP and the last 4 d of EP feed intake and milk yield were recorded daily and milk samples were collected at each milking. Blood samples were collected from the jugular vein on d 11 of AP and d 15 and 18 of EP. The 18:2 intake increased in all treatments from AP to EP. Regardless of the amount of supplemented CLA, the milk fat percentage decreased by 2.35 and 2.10%-units in treatment C and SC, respectively, whereas in the treatment S the decrease was with 0.99%-unit less pronounced. Thus, C and SC cows excreted daily a lower amount of milk fat than S cows. The concentration of trans 18:1 in the plasma and the milk increased from AP to EP and increased with increasing dietary CLA supply. While the concentration of 18:2c9t11 and 18:2t10c12 in the plasma and that of 18:2t10c12 in the milk paralleled dietary supply, the level of 18:2c9t11 in the milk was similar in C and CS but still lower in S. Although the dietary concentration of CLA was highest in treatment C, the partial replacement of CLA by sunflower seeds had a similar inhibitory effect on milk fat synthesis. Comparable 18:2c9t11 levels in the milk in both CLA treatments implies that this isomer is subjected to greater biohydrogenation with increasing supply than 18:2t10c12. The fact that unprotected 18:2t10c12 escaped biohydrogenation in sufficient amounts to affect milk fat synthesis reveals opportunities to develop feeding strategies where reduced milk fat production is desirable or required by the metabolic state of the cow.

Effect of Thyroid Hormones on the Redox Balance of Broiler Chickens

  • In the present study, two trials were conducted to evaluate the effects of hyper- and hypothyroid status on the redox balance of broiler chickens. In Trial 1, 3 groups of broiler chickens were randomly subjected to one of the three treatments: subcutaneous administration of triiodothyronine (T3, $150{\mu}g/kg$ BW), methimazole (MMI, 150 mg/kg BW), or saline. The blood, liver and heart were sampled at 3 h after injection. In Trial 2, three groups of 20 broiler chickens were randomly fed with one of the three diets: control, dietary supplementation of T3 (1.5 mg/kg diet) or MMI (1 g/kg diet) for 7 days. In trial 1, the plasma concentrations of T3 and T3 to thyronine ratio (T3/T4) were significantly increased by T3 injection. Plasma levels of thiobarbituric acid reacting substances (TBARS) tended to be increased (p = 0.067) by both T3 and MMI treatments while the ferric reduced/antioxidant capacity (FRAP) was increased only by MMI treatment. Acute T3 treatment had no significant effect on the activities of superoxide dismutase (SOD) and the concentrations of FRAP and TBARS in either liver or heart tissue. In contrast, the hepatic activities of SOD were decreased (p<0.05) while the cardiac levels of FRAP were significantly increased (p<0.0001) by MMI treatment. In chronic treatments, the rectal temperature of chickens was significantly decreased (p<0.05) by MMI treatment. The circulating T3 levels were significantly increased (p<0.05) by long-term T3 treatment, and showed a trend to decrease in MMI treatment. The plasma concentrations of TBARS were significantly (p<0.05) increased by MMI treatment. All the redox parameters measured in either liver or heart were not significantly altered by either long-term T3 or MMI treatment except that the hepatic SOD activities were significantly augmented by T3 treatment. The result showed that neither acute nor long-term elevation of circulating T3 levels induced lipid peroxidation in broiler chickens. The enhanced enzymatic antioxidant system (SOD in cardiac tissue) may be involved in the protection of the bird to increased oxidative challenge. The responses of redox balance to changed thyroid state seem to be tissue specific.

망상 폴리우레탄 폼의 물성 및 표면처리를 통한 미생물 고정화 특성의 향상 (Physical Properties of Reticulated Polyurethane Foams and the Enhancement of Microbial Adhesion through their Surface Treatments)

  • 각 샘플의 공극사이즈 (PPI)에 따른 미생물 고정화 능의 관계는 대체로 공극의 크기가 줄어들수록 미생물 고정화 량이 증가하는 경향을 보여주었다. 일반적으로 공극의 크기가 작을수록 비표면적이 증가하기 때문에 미생물 고정화 능이 증가할 것으로 사료되지만, 본 실험에서는 각 시편의 표면적을 측정 한 것이 아니고, 각 시편의 질량을 기준으로 미생물 점착량을 산출하였기 때문에 이와 같은 결과를 나타낸 것으로 사료된다. 특히 폴리우레탄 오픈 셀의 경우 담체의 PPI가 커질 경우 일반적으로 그 단위 부피당 표면적은 증가되지만, 단위 질량당 표면적은 이에 비례하여 증가하지 않음을 시편 관찰을 통하여 확인하였다. 따라서 이와 같은 이유로 인하여 각 시편의 PPI에 대한 점착율의 경향성은 발견하기 어려웠다. 각 시편의 비표면적 (단위질량당 표면적)을 실험을 통하여 결정한 후, 각 시편의 단위 면적 당 미생물 고정화 율을 산출한 결과, 단위 면적당 고정화 된 미생물의 수는 PPI 증가에 따라서 오히려 감소하였다. 따라서, 겉보기 밀도를 비롯하여 유실율, 점착율의 실험을 통한 모든 결론을 종합하여 고려해볼 때 45 PPI 담체가 가장 우수한 것으로 나타났다. 또한 고분자의 표면처리를 통해 미생물 점착율을 증가시킬 수 있다는 것을 실험으로 확인할 수 있었다. 특히 PEI 처리 담체의 경우 건조 무게의 증가량이 미처리 담체에 비하여 약 3 배정도 향상되는 것을 알 수 있었다. 또한 플라즈마 처리 실험 결과에서 알 수 있듯이 폴리우레탄 폼 담체에 미생물의 점착율이 플라즈마 표면처리에 의해 표면 친수도의 향상으로 인하여 증가되었다. 미생물의 점착율은 플라즈마 처리를 했을 때 가장 증가했고, Chitosan 처리를 한 경우를 제외하고 PEI를 처리했을 때도 처리하지 않았을 때보다 증가했다. 본 실험을 통하여 미생물 고정화 실험을 통하여 각 PPI별로 폴리우레탄 담체의 미생물 고정화 능력의 차이를 비교할 수 있음을 알았다. 따라서 앞으로의 실험은 고정화된 미생물을 현미경 관찰을 통하여 확인하고, 플라즈마 처리 후 미생물의 화학적 고정화(공유결합이나 이온결합체 형성을 통한 고정화 과정에서의 관능기 및 미생물의 유실이 없는 미생물 고정화)를 통한 미생물 고정화 율의 증가 방법을 개발하는데 주안점을 두고자 한다 한편, 다른 형태의 고분자나 미생물 종류를 달리하여 본 연구를 확장시킴으로써 환경 및 생물 산업에 유용한 소재로 사용되는 최적의 미생물 고정화 담체를 개발할 수 있으리라 사료된다.

콜드 플라즈마 처리를 이용한 분유와 양파분말 살균 (Cold Plasma Treatment Application to Improve Microbiological Safety of Infant Milk Powder and Onion Powder)

  • 분유 분말의 C. sakazakii와 양파 분말의 B. cereus의 포자를 저해하는데 있어 가장 효과적인 플라즈마 형성 가스는 각각 He-$O_2$와 He이었다. 분유 분말에 대한 콜드 플라즈마 처리시간이 길어질수록 그리고 형성 전력이 높아질수록 C. sakazakii의 저해도가 커졌으며, 900W, 40분 처리시 가장 큰 저해율을 나타내었다. 양파 분말에 접종된 B. cereus 포자는 콜드 플라즈마 단독 처리와 마이크로파와 병합된 콜드 플라즈마 처리에 대하여 강한 저항력을 가지고 있었으나 열처리($90^{\circ}C$, 1분)를 두 형태의 플라즈마 처리에 선병합시켰을 때 약 90%가 저해되었다. 병합 처리한 결과, 각각의 방법을 단독으로 처리한 것보다 병합 처리시 B. cereus 포자의 저해율(약 1.0 log spores/g)이 높아졌다. 본 연구는 분체 식품에 존재하는 식중독 균의 효율적인 살균 방법을 제시하였고, 분체 식품의 새로운 살균 공정으로서의 콜드 플라즈마의 적용 가능성을 보여주었다.

셀레늄강화 버섯폐배지의 급여기간이 비육후기 거세한우의 혈중 글루타치온 과산화효소 활성 및 조직내 셀레늄축적에 미치는 영향 (Effects of the Supplementation Period of Spent Composts of Selenium-Enriched Mushrooms on Plasma Glutathione Peroxidase Activity and Selenium Deposition in Finishing Hanwoo Steers)

  • 본 연구는 셀레늄강화 한우고기 생산을 위하여 셀레늄강화 버섯폐배지(Se-SMC)가 함유된 셀레늄사료(사료내 셀레늄함량: 0.9ppm)를 활용하여 급여사양기간(2개월, 3개월, 4개월)을 달리 하였을 때, 비육후기 거세한우의 조직 내 셀레늄 함량과 혈중 글루타치온 과산화효소(GSH-Px) 활성에 미치는 영향을 조사하여 최대 포화축적기간을 제시하고자 실시하였다. 본 연구에 사용된 비육후기 거세한우는 30두로 각 급여기간(2개월: 평균 체중 677kg, 3개월: 평균 체중 610kg 및 4개월: 평균 체중 524kg)에 대한 대조구 및 Se-SMC 급여구로 나누어 처리구당 5두씩 배치하여 총 6처리구로 구분하여 실시하였다. 대조구와 Se-SMC 급여구에 사용된 실험사료는 0.1과 0.9ppm의 셀레늄을 각각 함유하였다. 각 급여기간이 종료되면 도축하여 후지와 간을 채취하였으며 이를 셀레늄 함량분석에 이용하였다. 그리고 혈중 셀레늄 농도 및 글루타치온 과산화효소활성을 분석하기 위하여 도축 시에 채혈을 실시하였다. 건물섭취량은 Se-SMC 급여 및 급여기간에 의하여 유의한 차이가 나타나지 않았다. 또한 일당증체량은 대조구 및 Se-SMC 급여구간 그리고 급여기간 간에 유의한 차이가 나타나지 않았을 뿐만 아니라 총증체량 또한 각 급여기간내 대조구와 Se-SMC 급여구 간에 유의한 차이가 나타나지 않았다. Se-SMC 급여는 혈중 셀레늄농도를 유의하게 증가시켰으나(P<0.001), 급여기간에 따른 혈중셀레늄 농도에는 유의한 영향을 미치지 않았다. 아울러 혈중 GSH-Px 활성은 혈중 셀레늄농도에서 나타난 양상과 유사한 경향을 나타내었고, 급여기간에 의한 효과는 나타나지 않았다. 후지내 셀레늄 함량은 급여기간(P<0.05)이 증가함에 따라, 그리고 Se-SMC 급여(P<0.001)에 의하여 유의하게 증가하였다. 하지만, Se-SMC의 3개월 급여군과 4개월 급여군 간에는 유의한 차이가 나타나지 않았다. 간내 셀레늄 함량은 Se-SMC의 급여기간이 증가함에 따라 다소 증가하는 경향을 나타내었으나 유의한 효과는 나타나지 않았다. 하지만, 동일한 급여기간 내 Se-SMC는 대조구에 비하여 유의하게 높았다(P<0.01).이상의 결과로부터 근육내 셀레늄 축적을 위한 적정 Se-SMC 급여기간은 후지와 간내 셀레늄 함량을 고려할 때 약 2 내지 3개월의 급여가 충분할 것으로 사료된다.

실란 프라이머를 이용한 PDMS-PMMA 접착 (A study on PDMS-PMMA Bonding using Silane Primer)

  • In this paper, we present surface treatments for achieving bonds between PMMA and PDMS substrates. Silane primer is used for the formation of hydroxyl group on PMMA surfaces. The formed hydroxyl groups enhance the bonding strength of PDMS-PMMA substrates without channel clogging and structure deformation. The bonding strength on the different surface treatments (include oxygen plasma, 3-APTES, and corona discharge) is evaluated to find optimal bonding condition. The maximum bonding strength at the optimal surface treatment is over 300 kPa. The surface treatment using silane primer can be used to the bonding process of Micro-TAS and Lab-on-a-Chip.

Impacts of Photoperiod and Maternal Pineal Gland on Pre- and Post-natal development of Indian palm Squirrel F. pennanti

  • Studies till date suggest the existence of a fetal biological clock in suprachiasmatic nuclei entrained by the circadian signal from mother. Melatonin from maternal pineal gland reaches to the fetus by crossing every biological barrier including placenta, hence fetuses were exposed to similar melatonin variation as their mother. Experimental modulations of maternal pineal gland activity of pregnant females either by exposing the them to different photoperiodic schedules or by exogenous melatonin treatments till the date of parturition, regulated the fetal plasma level of melatonin, thereby the prenatal (fetal) growth and development. This clearly suggests the maternal transport of melatonin to their fetus through placenta since fetal retina-hypothalamic tract was incomplete. An extension of experimental schedules till 60 days of post-partum period regulated the neonatal pineal gland activity and gonadal maturation along with their plasma levels of melatonin and sex steroids suggesting clearly the phenomenon of maternal transfer of melatonin to their young ones during the post-natal period, when the neonates were solely dependent on the mother's milk for their nutrition and energetic demands. On the basis of above observations we may suggest that the maternal pineal gland activity regulate the prenatal development by passing its melatonin to fetus via placenta and post-natal growth and sexual maturation by passing maternal melatonin to neonates via milk. Hence, the photoperiod perceived by mother is translated into the maternal plasma level of melatonin which not only regulates the prenatal but also the post-natal growth and sexual maturation of neonates.

  • The influence of the subsequent-annealing (SA) treatment on plasma electrolytic oxidation (PEO)-treated Mg-based alloy was investigated and the dependence of the dehydration reaction on the SA temperature was also studied. For this purpose, a series of the SA treatments were carried out on the coated samples at two different temperatures, i.e. 423 and 523 K for 10 h. In contrast to the sample without SA treatment, the sample annealed at 523 K exhibited a significant difference in term of surface morphology since the MgO content in the oxide layer increased with increasing SA temperature. With increasing SA temperature, the dehydration of $Mg(OH)_2$ led to the increase in the relative amount of the MgO, which was a hard phase. From the nano-indentation results, the applied loads of the samples were seen to increase as SA temperature increased. However, the corrosion resistance of the sample annealed at 423 K was higher than that of the samples annealed at 523 K.

  • Plasma techniques permit modification of the surface layers of the substrate while maintaining its bulk properties. The use of plasma treatment on textile fibers and fabrics is very limited, however, the limitations, plasma treatments have been used to modify surface properties of textiles. In this study, multifunctional finish was performed by corona discharge technique for surface functionalization. Electron spectroscopy for chemical analysis(ESCA) was used to determine the ratios of carbon, oxygen, nitrogen, and phosphor at a 20$\AA$ sampling depth. K/S value and limiting oxygen index(LOI) value were used to obtain information on the effect of the finished fabrics. The mechanism of the flame retardancy was analyzed by the thermogravimetry-and the residue number. In ESCA, relative N1s intensity increased in case of mixtrure. The flame retardancy of the polyester and polyester/cotton fabrics treated with JA-6034 and JA-6050 was found to be operative mainly in the condensed phase mechanism. The multifunctional finish was better effect for the post treated finish agent.

A Study on the Optimal Amino Acid Pattern at the Proximal Duodenum in Growing Sheep

  • Nine crossbred castrated lambs fitted with rumen and duodenum cannula and fed a diet of hay and concentrate formulated with ground corn and soybean meal as main ingredients were used to assess the duodenal ideal amino acid pattern. Three synthetic amino acid mixtures with different profile of essential amino acids were duodenally infused in order to get three different amino acid patterns flowing into the duodenum. The mixtures were designed to have similar amino acid profile as rumen microbial protein (Pm), casein (Pc) and modified muscle amino acid (Pmm). Results showed a lower urine nitrogen excretion (p=0.05), a higher nitrogen retention (p=0.04) and bodyweight gain with treatment Pmm. The modified muscle amino acid pattern also promoted a lower ratio of Gly to other amino acids in plasma (Gly/OAA) and a higher RNA and RNA/DNA concentration in the liver of the sheep. Meanwhile, the urea concentration in plasma was reduced and the insulin concentration was increased with Pmm treatment. No differences in glucose and growth hormone concentration in plasma were found among three treatments. All results obtained indicate that the modified muscle amino acid pattern (Lys 100%, Met+Cys 39%, Thr 76%, His 41%, Arg 72%, Leu 158%, Ile 81%, Val 105%, Phe 81% and Trp 13%) was the best for growing sheep.
