Effect of Korean Native Plant Diet on Lipid Metabolism, Antioxidative Capacity and Cadmium Detoxification in Rats (한국 고유의 산채류 첨가 식이가 흰쥐의 지방 대사 및 항산화능과 Cadmium 제독에 미치는 영향)

  • This study was performed to investigate the effects of Korean native plant diets on lipid and cadmium (Cd) metabolisms and also antioxidative enzyme activities in rats. Seventy male Sprague-Dawley rats weighing 210.3$\pm$2.7g were blocked into ten groups according to body weight. Rats were raised for four weeks with diets containing 0 or 0.04%(w/w) cadmium chloride and 5%(w/w) plant powder-Ssook(Artemisia princeps var. orientalis HARA), Chamchwi(Aster scaber THUNB), Gomchwi(Lingularia fischeri TUBCZ). or Solbeerum (Portulaca oleracea LINNE). Food intake, weight gain, food efficiency ratio, and weights of liver, kidney, spleen and epididymal fat were significantly lower in Cd-exposed groups, food efficiency ratio, and weights of liver, kidney, spleen and epididymal fat were significanlty lower in Cd-exposed groups. Plasma total lipid level, liver total lipid, cholesterol and triglycerde concentrations, and fecal total lipid, cholesterol and triglyceride excretions were decreased by Cd administration. Total lipid and triglyceride concentration in plasma and liver were lower in Chamchwi groups together with increased fecal total lipid and triglyceride excretions. Cholesterol concentration in plasma and liver were lower in Gomchwi groups with increased fecal cholesterol excretions. Activities of red-blood-cell superoxide dismutase(SOD), and liver catalase, SOD and glutathione peroxidase (GSH-px) were decreased significantly by Cd administration. Thiobarbituric acid reactive substance(TBARS) concentrations of plasma and liver were increased by Cd administration. Thiobarbituric acid reactive substance(TBARS) concentration of plasma and liver were increased by Cd administration. Administration of plant diets decreased plasma and liver catalase, SOD and GSH-px activities in Cd-free diet groups and increased them in Cd-administered groups. Plasma and liver TBARS concentrations were decreased in animals fed plant diets, with Chamchwi showing the most effective antioxidative capacity. The concentration of Cd accumulated in blood and tissue decreased in Cd-exposed groups fed plant diets. Liver Cd concentration were lower in the Chamchwi and Gomchwi groups, and kidney Cd levels were lower in the Ssook and Chamchwi groups. Among them, Chamchwi was the most effective in Cd detoxification. In conclusion, Chamchwi and Gomchwi were effective in lowering tissue lipid levels, with Chamchwi having the greatest antioxidative and detoxifying effects.

Effect of Water and Ethanol Extracts of Persimmon Leaf and Green Tea Different Conditions on Lipid Metabolism and Antioxidative Capacity in 12-month-old Rats (추출 조건을 달리한 감잎과 녹차의 물 및 에탄올 추출물이 노령쥐의 지방대사와 항산화능에 미치는 영향)

  • This study was performed to investigate effects of dried leaf powders, water, 75% and 95% ethanol extracts of persimmon leaf and green tea on lipid metabolism, lipid peroxidation and antioxidative enzyme activity in 12-month-old rats. Fifty-four male Sprague-Dawley rats weighing 542$\pm$4.5g were blocked into groups according to their body weight and were raised for four weeks with the diets containing 5%(w/w) dried leaf powders of persimmon(Diospyros kaki Thunb) and green tea(Camellia Sinensis O. Ktze), water or 75% and 95% ethanol extracts from same amount of each dried tea powder. Food intake was not significantly different among all groups, but weight gain of green tea powder group was significantly lower than that of control group. Plasma and liver lipid levels of all the tea diet groups were lower than those of control group. Especially, 75% ethanol extract of persimmon leaf decreased total lipid and triglyceride concentrations in plasma and 95% ethanol extract of persimmon leaf decreased liver total lipid level. However, there was no difference between 75% ethanol extracts groups and 95% ethanol extracts groups in lipid metabolism. Superoxide dismutase(SOD) and catalase activities in erythrocyte were remarkably increased by all the green tea diets. SOD, catalase and glutathione peroxidase activities in liver were increased by the feeding of ethanol extracts from green tea and persimmon leaf powder. Liver xanthine oxidase activity was not different among all groups. Plasma Thiobarbirutic acid reactive substance(TBARS) concentrations of all the green tea diet groups were significantly low. It was thought that high flavonoids in green tea inhibited plasma lipid peroxidation by promoting SOD, catalase activities in erythrocyte. 95% ethanol extract of persimmon leaf also inhibited plasma lipid peroxidation by high vitamin E and beta-carotene. Persimmon leaf powder decreased liver TBARS concentration by vitamin E, betacarotene and vitamin C and by increasing activities of antioxidative enzymes with flavonoids. In conclusion, dried leaf powders, water, 75% and 95% ethanol extracts of persimmon leaf and green tea were effective in lowering lipid levels and inhibiting lipid peroxidation in 12-month-old rats. Above all, ethanol extracts of persimmon leaf decreased plasma and liver lipid levels and persimmon leaf powder effectively inhibited liver lipid peroxidation. Extracts of green tea leaf inhibited plasma lipid peroxidation. In lowering lipid levels and inhibiting lipid peroxidation, ethanol extracts were more effective than water extracts, but there was no difference between 75% ethanol extracts and 95% ethanol extracts in lipid metabolism.   • The level of long-chain n-3 fatty acids in chicken and pork can be increased by changing the diet of the animals. Increased levels of these essential fatty acids improve cardiovascular health in humans. The purpose of this study was to study the effects of the consumption of pork and chicken enriched in docosahexaenoic acid (DHA) on plasma lipids. The consumption of these products decreased the levels of two cardiovascular risk factors, LDL-cholesterol and triacylglycerols, in the plasma of female college students. The effect on LDL-cholesterol differed from that of fish oil, which does not affect the level of LDL-cholesterol. The proportions of DHA in the triacylglycerols and the glycerophospholipids were increased markedly. The greatest changes in the glycerophospholipids were in the ether types of the ethanolamine glycerophospholipids. Dietary DHA appears to be incorporated preferentially into the plasma ethanolamine plasmalogens, which can act as antioxidants. This agrees with our hypothesis that DHA stimulated the transcription of the genes for peroxisomal enzymes that are required for plasmalogen synthesis.

  • Lipid peroxidation induces functional deterioration of cell membrane and induces cell death in extreme cases. These phenomena are known to be related generally to the change of physical properties of lipid membrane such as decreased lipid order or increased water penetration. Even though the electric property of lipid membrane is important, there has been no report about the change of electric properties after lipid peroxidation. Herein, we demonstrate the molecular energy band change in red blood cell membrane through peroxidation by air-based atmospheric pressure DBD plasma treatment. Ion-induced secondary electron emission coefficient (${\gamma}$ value) was measured by using home-made gamma-focused ion beam (${\gamma}$-FIB) system and electron energy band was calculated based on the quantum mechanical Auger neutralization theory. The oxidized lipids showed higher gamma values and lower electron work functions, which implies the change of surface charging or electrical conductance. This result suggests that modified electrical properties should play a role in cell signaling under oxidative stress.

  • Objectives : To investigate the effects of acupuncture at Gansoo (B-18) and Chungwan (CV-12) on lipid metabolism in rats fed peroxidized lipid diet. Methods : Effects of acupuncture at Gansoo and Chungwan on plasma and liver lipid composition and antioxidative capacity were investigated in rats fed peroxidized lipids. Results : Although the level of plasma total cholesterol and triglyceride showed a tendency to decrease in the acupuncture group, the plasma HDL-cholesterol concentration showed no significant difference. The level of liver total cholesterol and triglyceride showed no significant difference in all treatment groups. Thiobarbituric acid( TBARS ) values in plasma and liver showed a tendency to decrease in the acupuncture groups. The plasma GOT and GPT activities showed low values in the acupuncture groups. The liver glutathione peroxidase, superoxide dismutase and catalase activity showed high values in the acupuncture groups. Conclusions : The results suggest that acupuncture at Gansoo (B-18) and Chungwan (CV-12) may have an influence on lipid metabolism via enhancing antioxidative capacity.

Effects of Yellow and Black Soybeans on Plasma and Hepatic Lipid Composition and Fecal Lipid Excretion in Rats (노란콩과 검정콩 식이가 흰쥐의 체내 지질 함량과 분변으로의 지질 배설에 미치는 영향)

  • This study was undertaken to determine the effect of yellow and black soybeans on plasma and hepatic lipid composition and fecal lipid excretion in Sprague Dawley rats. Animals were fed with diets containing 52% yellow soybean, 50% black soybean, or 20% casein for 7 weeks. Feeding efficiency was significantly increased in the animals fed soybeans(p<0.05). Plasma cholesterol concentration was significantly lowered in the group fed yellow soybean compared with black soybean and casein-fed control (p<0.05). Hepatic triglyceride concentration was significantly lowered in soybean groups compared with casein-fed control(p<0.05), whereas hepatic cholesterol concentration was not affected by diet treatments. Soybean feeding significantly increased fecal weight, triglyceride and bile acid contents compared with casein feeding(p<0.05). It is concluded that soybean feeding in rats affects plasma and hepatic lipid levels by increasing the excretion of triglyceride and bile acids.

LDL Oxidation, Total Radical Trapping Antioxidant Potential and Plasma Antioxidant Vitamin Systems in Obese School Children (비만아동의 지질과산화물 형성과 항산화 체계에 관한 연구)

  • The purpose of this study was to examine the lipid peroxidation, plasma antioxidant status and insulin resistance in childhood obesity. To this end, we measured blood lipid profiles, glucose, insulin concentrations, plasma antioxidant vitamins, baseline conjugated diene formation as a measure of LDL oxidation in vivo and TRAP (total radical trapping antioxidant potential) of 93 school children (58 nonobese, 35 overweight-obese). Insulin resistance was estimated by homeostasis model assessment of insulin resistance (HOMA-IR). The overweight-obese children showed significantly higher levels of leptin (p < 0.0001) and triglyceride (p < 0.05) and significantly lower level of plasma Iycopene (p < 0.001) and $\gamma$-tocopherol (p < 0.05) compared with the normal weight children. Furthermore, the levels of TRAP were significantly lower in overweight-obese children (p < 0.05). Significant positive relationships between plasma leptin and conjugated dienes formation (p < 0.005) and inverse relationship between plasma leptin and lipid corrected levels of $\beta$-carotene (p < 0.05), Iycopene (p < 0.05) were observed. Our results showed an increased lipid peroxidation and decreased antioxidant capacity in childhood obesity which could be involved in the atherosclerotic process.

  • This experiment was conducted to examine the effects of fish oil supplementation with low does on the lipid concentration and fatty acid composition of plasma and the fatty acid composition of plasma phospholipid and erythrocyte of infants. Among 18 breast-fed infants, 6 were in control group and 12 were in fish oil groups. The subjects in fish oil groups were nursed by their mothers who supplemented with fish oil 1.96g/d or 3.92g/d, respectively for 2 weeks from 10 to 12 weeks postpartum. The nursing mothers consumed their usual diets at home. Blood samples were collected at the final day of experiment. There were no significant changes in daily intakes of total lipid, triglyceride, free fatty acid, phospholipid and cholesterol of infants by fish oil supplementation. However, the content of EPA (eicosapentaenoic acid)increased and that of ARA (arachidonic acid) decreaed significantly in plasma PC(phophatidylchline). And also, there were tendencies to increase triglyceride concentration and to decrease cholesterol and phopholipid concentrations of plasma. As the above results, atherogenic index (AI) showed a tendency to decrease, but not significant. DHA (docosahexaenoic acid) and EPA contents in plasma PC and PE (phosphatidylethanolamin) as well as those of erythrocyte tended to increase. In these results, we concluded that fish oil supplementation with low dose to lactating women does not obviously affect of the plasma lipid concentrations and fatty acid composition of plasma PC and PE as well as erythrocyte. However the increase of EPA content of plasma PC and the tendency to increase DHA and EPA contents of plasma as well as erythrocyte membrane indicate that there may be some beneficial effect on infant lipid metabolism of fish oil intake of nutsing mother were increased.

  • Effects of cheungho aqua-acupuncture at gansoo$(BL_{18})$ and illweul$(GB_{14})$ on plasma lipid composition were investigated in rat fed high fat diet. Body weight gain showed a tendency to decrease in aqua-acupuncture groups and in aqua-acupuncture groups, gansoo and illweul join aqua-acupuncture group showed a lower values than other aqua-acupuncture groups. Concentrations of plasma triglyceride, free fatty acids, lipoprotein and glucose showed a tendency to decreasea in the aqua-acupuncture groups. However in aqua-acupuncture groups, these values showed no significantly different. In plasma lipid composition, total cholesterol and LDL- cholesterol showed a tendency to decrease in aqua-acupuncture groups, however the values of LDL-cholesterol showed no significantly different in the aqua-acupuncture groups. HDL-cholesterol concentration showed a tendency to increase in aqua-acupuncture groups and in aqua-acupuncture groups, gansoo aqua-acupuncture group showed a high value.

  • The present study was conducted to evaluate the effect of dietary levels of vitamin A on the lipid composition in rats fed Zn-deficient diets. Fifty male Sprague-Dawley rats were fed 5 experimental diets for 7 weeks. Rats were fed Zn-and vitamin A-deficient diet or the same diets supplemented with various levels of vitamin. A liver total lipid, cholesterol and triglycerides contents tended to decrease in Zn-deficient rats, but the supplementation of excess vitamin A enhanced those contents. Plasma cholesterol content was significantly higher in feeding Zn-deficient and excess vitamin A diet. Plasma triglyceride and phospolipid contents were higher in Zn-deficient groups, but not influenced by dietary levels of vitamin. A . Plasma HDL-cholesterol content was not changed by Zn-deficient diets. Plasma alkaline phosphatase activity was significantly reduced in Zn-deficient groups. Zinc contents of plasma and liver were influenced by the dietary level of zinc, but not changed by supplementation of vitamin A.
