  • Plasma glucose levels before and after oral glucose administration have been compared in a group of 76 thyrotoxic subjects and a group of 8 normal control subjects in order to study the effect of glucose loading in thyrotoxicosis. Following were the results: 1. The mean fasting plasma glucose level was elevated in the thyrotoxic group(95.5mg%) compared to normal control group (88mg%). 2. The peak of glucose tolerance curve is at 30 minutes after glucose administration in both groups, but its mean value was 44mg% higher in thyrotoxic group than in control group. 3. The plasma glucose levels returned towards the fasting level in the later stage of the test more rapidly in thyrotoxic group than in control group. 4. 69.6% of oral glucose tolerance tests were impaired in the thyrotoxic group, and the occurance of abnormal glucose tolerance could be related to the degree of thyrotoxicity, sex and age. 5. The mechanisms of the impaired glucose tolerance in thyrotoxicosis are thought to be related to an increased rate of glucose absorption from gastrointestinal tract, abnormal liver function with decreased hepatic glycogenesis, increased glucose oxidation, decreased pancreatic release of insulin, and genetic relationship between diabetes and thyrotoxicosis.

Effect of dietary Platycodon grandiflorum on plasma glucose and lipid metabolism in $KK-A^y$ mice and streptozotocin-induced diabetic rats

  • This study was designed to investigate the effect of dietary Platycodon grandiflorum on plasma glucose and lipid metabolism in $KK-A^y$ mice and STZ-induced diabetic rats. Both plasma triglyceride and plasma cholesterol levels in STZ-induced diabetic rats were significantly decreased by dietary Platycodon grandiflorum feeding for 4 weeks compared to those of control rats, but there were no marked differences in $KK-A^y$ mice. However, for plasma glucose values, Platycodon grandiflorum feeding resulted in a significant decrease in both STZ-induced diabetic rats and $KK-A^y$ mice. Also, dietary Platycodon grandiflorum slightly decreased the postprandial glucose level at 30 and 60 mins during oral glucose tolerance test in $KK-A^y$ mice. Although there was no statistical significance, the fasting plasma insulin levels of Platycodon grandiflorum dieted $KK-A^y$ mice tended to decrease when compared to that of control mice. Therefore, the present results suggested that dietary Platycodon grandiflorum may have a beneficial effect on preventing hypercholesterolemia and hyperlipidemia.

당뇨환자의 식사 전후에 따른 혈중 Insulin-like growth factor(IGF-I), IGF-II 및 Insulin-like growth factor binding proteins(IGFBP)-3의 변화 (Changes of Insulin-like Growth factor-I, II and IGF-Binding Protein-3 on Fasting and Postprandial state in Diabetes)

  • IGFs and IGFBPs have an important role in controlling glucose homeostasis. This study was conducted to investigate the changes of insulin-like growth factor(IGF)-I. IGF-II and IGF binding proteins (IGFBPs) on fasting and postprandial state in Korean diabetes, Twenty eight healthy subjects and fifty seven diabetic patients participated in this study. The healthy subjects were not knowingly suffered from any disease and were not receiving any medical treatment, and diabetic subjects were undergo medical treatment, continuously. Weight and height were measured and body mass index (BMI) was calculated as weight (kg) divided by the square of height (m2). Blood pressure was measured. Plasma lipid profiles were analyzed by enzymatic methods, plasma Insulin and glucose levels were measured in fasting and postprandial state, respectively. The levels of serum IGFs and IGFBP-3 were measured by radioimmunoassay (RIA). The levels of glucose and insulin were significantly higher in diabetes than normal subjects on fasting as well as postprandial state (p<0.0l). The levels of IGF-I was significantly lower in diabetes than normal subjects, however in postprandial state, there was no significant difference between diabetes and control subjects, The levels of IGF-II were significantly lower in diabetes than control subjects both fasting and postpradial state, The level of IGFBP-3 were not significantly different between diabetes and normal subjects. Fasting IGF-I, IGF-II and IGFBP-3 levels were positively correlated with those levels on postprandial state, fasting IGe levels of IGF-I levels were positively correlated with fasting insulin levels, and postprandial IGF-I levels were positively correlated with fasting glucose, postprandial insulin and postprandial insulin levels, plasma triglyceride levels were correlated with plasma triglyceride levels. The IGFBP-3 levels were not correlated with IGF components, glucose, insulin and plasma lipids, These results demonstrate that in diabetes, the components IGF-I/IGFBPs system were significantly correlated with plsma glucose and insulin levels both fasting and postprandial state.

Effect of Oolong Tea Extracts on Plasma Glucose Level and Antioxidant System in Diabetic Rats

  • The present study was conducted to investigate the effect of oolong tea extract on blood glucose level and antioxidant system in diabetic rats. The Sprague-Dawley rats were fed on AIN-76 based experimental diets containing 1 % oolong tea extract for 6 weeks. They were induced to be diabetic by receiving streptozotocin (45mg/kg BW) intramuscularly. Blood glucose, blood and hepatic concentration of vitamins A and E, and antioxidant enzyme activities were measured. Oolong tea extract feeding decreased the plasma glucose in diabetic rats. Dietary supplementation of oolong tea extract did not affect antioxidative enzyme activities such as superoxide dismutase, glutathione peroxidase and catalase in diabetic rats. The plasma level of retinol was increased in diabetic rats by feeding oolong tea extract. Plasma and hepatic levels of ${\alpha}$-tocopherol were higher in diabetic rats fed oolong tea extract. In conclusion, these results suggest that oolong tea extract consumption might reduce the plasma glucose in diabetic rats and protect the oxidative damage from diabetic stress to some extent.

Quercetin attenuates fasting and postprandial hyperglycemia in animal models of diabetes mellitus

  • The objective of this study was to investigate the hypoglycemic effects of quercetin (QE) in animal models of diabetes mellitus (DM). A starch solution (1 g/kg) with and without QE (100 mg/kg) or acarbose (40 mg/kg) was orally administered to streptozotocin (STZ)-induced diabetic rats after an overnight fast. Postprandial plasma glucose levels were measured and incremental areas under the response curve were calculated. To study the effects of chronic feeding of QE, five-week-old db/db mice were fed an AIN-93G diet, a diet containing QE at 0.08%, or a diet containing acarbose at 0.03% for 7 weeks after 1 week of adaptation. Plasma glucose and insulin, blood glycated hemoglobin, and maltase activity of the small intestine were measured. Oral administration of QE (100 mg/kg) or acarbose (40 mg/kg) to STZ-treated rats significantly decreased incremental plasma glucose levels 30-180 min after a single oral dose of starch and the area under the postprandial glucose response, compared with the control group. QE (0.08% of diet) or acarbose (0.03% of diet) offered to db/db mice significantly reduced both plasma glucose and blood glycated hemoglobin compared to controls without significant influence on plasma insulin. Small intestine maltase activities were significantly reduced by consumption of QE or acarbose. Thus, QE could be effective in controlling fasting and postprandial blood glucose levels in animal models of DM.

아동기 비만이 혈장 지질, 혈압 및 혈당에 미치는 영향 (Relationships of Obesity in Childhood to Plasma Lipids, Blood Pressure and Blood Glucose)

  • In order to investigate the relationships of obesity in childhood to plasma lipid, blood pressure and blood glucose concentration, we selected 21 subjects for the moderate obese group(MO), 9 for mild obese group(MI), and 19 for the control group(C) among children aged 10~12. While the level of plasma triglyceride and VLDL-cholesterol of the MO group was much higher than that of group C, a significantly lower percentage of HDL-cholesterol was found in the MO compared to the percentage found in group C. Also the level of the total cholesterol and LDL-cholesterol of the MI group as well as the MO was much higher than that of C. The elevated total-cholesterol level of the Mi group was due to increased LDL-cholesterol and that of the MO was due to increases in both VLDL-cholesterol and LDL-cholesterol. As the result of these differences, the atherogenic index of the MO was significantly higher than that of C. The incidence of hypercholesterolemia( 200mg/dl) of the MI and MO was 60.0% and 77.8% respectively, All of the physical parameters and indexes except height were positively correlated with plasma lipid levels, systolic blood pressure, and blood glucose concentration. The analyses of the correlated with plasma lipid levels, systolic blood pressure, and blood glucose concentration. The analyses of the correlation indicated that central fat to peripheral fat ratio and waist to thigh girth ratio seemed to be closely associated to plasma lipid levels and atherogenic index. The MO had significantly higher systolic blood pressure than C and significantly higher blood glucose concentration was found in both MI and MO than in C. These results confirmed that obesity in childhood may be relevant to chronic metabolic diseases such as abnormal lipid metabolism, atherosclerosis, high blood pressure and diabetes mellitus.

참마 재분획물이 당뇨유발 흰쥐의 혈당 및 에너지원 조성에 미치는 영향 (The Effect of Dioscorea japonica Thunb Subfractions on Blood Glucose Levels and Energy Metabolite Composition in Streptozotocin Induced Diabetic Rats)

  • This study evaluates the effect of Dioscorea japonica Thunb subfractions on hyperglycemia and the composition of energy metabolites in diabetic rats. Diabetes emllitus was induced in male Sprague-Dawley rats by an injection of streptozotocin(STZ) dissolved in a citrate buffer into the tail vein at a dose of 45㎎/㎏ of body weight. Diabetic rats were assigned to 6 groups; STZ-control, subfraction A, B , C, D and E groups. All groups were fed an AIN-76 diet. The second butanol fraction of Dioscorea administered orally with carboxymethyl cellucose for 10 days after the STZ injection Body weight gain, diet intake and organ weights were monitored Levels of hematocrit, blood glucose, liver and muscle glycogen were measured. Levels of cholesterol, triglycerides and free fatty acids were also assayed. Body weight losses were observed by subfraction A group. Liver and kidney weights were not affected in any of the subgractioned groups. The decrease of blood glucose in daibetic rats which were fed Dioscorea japonica Thunb was significantly greater than the dicrease of blood glucose in the STZ-control group. cholesterol plasma level was not influenced in any subfraction of Dioscorea japonica Thunb. Liver triglyceride levels were significantly lowered in subfraction A compared with the STZ-control group. This study's results suggest that oral administration of subfraction C of Dioscorea japonica Thunb frction is capabl of reducing blood glucose, plasma triglyceride and free fatty acid levels, and therefore Dioscorea japonica Thunb may contain antihyperglycemic compounds.

Lotus leaf alleviates hyperglycemia and dyslipidemia in animal model of diabetes mellitus

  • The purpose of this study was to investigate the effects of lotus leaf on hyperglycemia and dyslipidemia in animal model of diabetes. Inhibitory activity of ethanol extract of lotus leaf against yeast ${\alpha}$-glucosidase was measured in vitro. The effect of lotus leaf on the postprandial increase in blood glucose levels was assessed in streptozotocin-induced diabetic rats. A starch solution (1 g/kg) with and without lotus leaf extract (500 mg/kg) was administered to the rats after an overnight fast, and postprandial plasma glucose levels were monitored. Four-week-old db/db mice were fed a basal diet or a diet containing 1% lotus leaf extract for 7 weeks after 1 week of acclimation to study the chronic effect of lotus leaf. After sacrifice, plasma glucose, insulin, triglycerides (TG), total cholesterol (CHOL), high-density lipoprotein (HDL)-CHOL, and blood glycated hemoglobin levels were measured. Lotus leaf extract inhibited ${\alpha}$-glucosidase activity by 37.9%, which was 1.3 times stronger than inhibition by acarbose at a concentration of 0.5 mg/mL in vitro. Oral administration of lotus leaf extract significantly decreased the area under the glucose response curve by 35.1% compared with that in the control group (P < 0.01). Chronic feeding of lotus leaf extract significantly lowered plasma glucose and blood glycated hemoglobin compared with those in the control group. Lotus leaf extract significantly reduced plasma TG and total CHOL and elevated HDL-CHOL levels compared with those in the control group. Therefore, we conclude that lotus leaf is effective for controlling hyperglycemia and dyslipidemia in an animal model of diabetes mellitus.

Effect of the Dietary Protein Level on Plasma Glucose, Lipids and Hormones in Streptozotocin-Diabetic Rats

  • Atherosclerotic vascular disease is a major cause of the increased morbidity and mortality assciated with diabetes mellitus. The prominent role of nutrition in hypercholesteolemia and atherosclerosis is generally accepted. Diet is a key element in the management of diabetes (type I-IDDM), yet the appropriate diet for patient with diabetes mellitus is not well known. Dietary protein has been shown to have a significant effect on plasma cholesterol levels in both experimental animals and humans. The present experiment was designed to determine the effect of the dietary protein level(20% vs 60%) on plasma glucose concentration, lipids profile, insulin and glucagon levels from non-diabetic and streptozotocin-induced diabetic rats. Results showed that a high protein diet decreased triglyceride concentration in diabetic rats. Also diabetic rats fed a high protein diet were hypocholesterolemic than rats fed a control diet. There were no effects by level of protein on fasting blood glucose concentration and insulin/glucagon ratio. Results from the present study suggest that a high protein diet may be beneficial to control pasma lipids in chemically-induced diabetic rats.

한국산 야생식용식물이 당뇨유발 흰쥐의 혈당 및 간과 근육내 에너지원 조성에 미치는 영향 (The Effects of Korean Wild Vegetables on Blood Glucose Levels and Liver-muscle Metabolism of Streptozotocin-induced Diabetic Rate)

  • Attempts were made to determine the effects of 5-Korean wild vegetabel consumptions on blood glucose leveles and orgen-energy metabolisms in streptozotocin-induced diabetic rats. The 5-Korean wild vegetables were : Cassia tora L.(C.t), Lycium chinese Mill(L.c), Trichosanthes kirilowii Max(T.k), Polygonatum odoratum var. Pluriflorum Ohwi(P.o) and Arctium lappa L(A.l). Sixty male Spargue-Dawley rats(160-220g) were induced diabetes mellitus by streptozotocin injection into the tail vein and were devided into 6 groups : a diabetic control and 5-experimental groups. All groups of the rats were fed on a AIN-76 diet, and the 5-experimental groups were fed with each wild vegetable (10%) for four weeks. An increased tendency in body weight of all the groups was observed and the tendency was more significant in L.c, T.k. and P.o. groups. The organ weight of liver and kidney were higher in L.c. and A.l. groups and lower in T.k. group which has shown the improvement from diabetes. Plasma glucose levels were markedly decreased from the 1st week in C.t, T.k. and P.o. groups and the tendency has lasted throughout the four weeks experimental period. The consumption of P.o. has decreased plasma cholesterol level while any significant difference was not seen in plasma protein levels from all the experimental groups. The level of plasma triglyceride was decreased in P.o. group and the levels of plasma free fatty acids were also significantly lower in P.o. and T.k. groups. The liver protein levels were significantly higher in P.o. and T.k. groups and these two groups also showed the negative or relatively small amount of urinary glucose excretion. The experimental group of T.k. has revealed the decreased level of muscles protein and the increased level of muscle glycogen. The 5-Korean wild vegetables contained dietary fiber and 9-analyzed minerals comparable to the ordinary use vegetables.
