Pancreatic lipase inhibitory activity of taraxacum officinale in vitro and in vivo

  • Obesity has become a worldwide health problem. Orlistat, an inhibitor of pancreatic lipase, is currently approved as an anti-obesity drug. However, gastrointestinal side effects caused by Orlistat may limit its use. In this study the inhibitory activities of dandelion (Taraxacum officinale) against pancreatic lipase in vitro and in vivo were measured to determine its possible use as a natural anti-obesity agent. The inhibitory activities of the 95% ethanol extract of T. officinale and Orlistat were measured using 4-methylumbelliferyl oleate (4-MU oleate) as a substrate at concentrations of 250, 125, 100, 25, 12.5 and $4\;{\mu}g/ml$. To determine pancreatic lipase inhibitory activity in vivo, mice (n=16) were orally administered with com oil emulsion (5 ml/kg) alone or with the 95% ethanol extract of T. officinale (400 mg/kg) following an overnight fast. Plasma triglyceride levels were measured at 0, 90, 180, and 240 min after treatment and incremental areas under the response curves (AUC) were calculated. The 95% ethanol extract of T. officinale and Orlistat, inhibited, porcine pancreatic lipase activity by 86.3% and 95.7% at a concentration of $250\;{\mu}g/ml$, respectively. T. officinale extract showed dose-dependent inhibition with the $IC_{50}$ of $78.2\;{\mu}g/ml$. A single oral dose of the extract significantly inhibited increases in plasma triglyceride levels at 90 and 180 min and reduced AVC of plasma triglyceride response curve (p<0.05). The results indicate that T. officinale exhibits inhibitory activities against pancreatic lipase in vitro and in vivo. Further studies to elucidate anti-obesity effects of chronic consumption of T. officinale and to identify the active components responsible for inhibitory activity against pancreatic lipase are necessary.

Effect of the Ethanol Extract from the Aleurone Layer of Anthocyanin-Pigmented Rice on Blood Glucose and Lipid Metabolism in Streptozotocin Induced Diabetic Rats

  • The present study was designed to investigate the hypoglycemic activity and lipid metabolism of ethanol (EtOH) extract from the aleurone layer of anthocyanin-pigmented (AP) rice in streptozotocin (STZ)-induced diabetic rats. Sprague-Dawley male rats weighing 210~240 g were divided into 4 groups, normal, diabetic control, and two experimental groups, and diabetes in rats was induced by injection of STZ (45 mg/kg, body weight) into tail vein. The EtOH extract of the powdered aleurone layer of AP rice was administered orally in diabetic rats for 14 days. In order to find the hypoglycemic effects in the animal model, the body weight, plasma glucose levels, cholesterol, HDL-cholesterol, triglyceride (TG), free fatty acid (FFA), aspartate aminotransferase (AST) and alanine amino- transferase activities (ALT) were determined. Oral administration of 1.0 81kg on the EtOH extract for 14 days resulted in a significant reduction in blood glucose, ALT, TG and FFA. However, in the case of 2.0 g/kg, the hypo-glycemic effects were not considerable. This results suggest that the EtOH extract might induce hypoglycemic effects in STZ-induced diabetic rats due to some photochemical components in the aleurone layer of AP rice.

Nutritional Assessment of the Oldest-Old Population Aged 85 Over: Based on 2009 Korea National Health and Nutrition Examination Survey

  • As Korea is expected to become super high aged society in 2026, nutrition is important elements of health in the elderly and affects their life. In epidemiological surveys, anthropometric and biochemical measurements represent important components of nutritional assessment in elderly. In the present study, we investigated anthropometric index, biochemical markers, and nutrient intake for elderly aged over 85 by using data on 71 elderly (24 men, 47 women) over 85 years which was obtained from Korea National Health and Nutrition Examination Survey in 2009. According to BMI values, the prevalence of underweight and obesity was 9.7% and 34.5% in men, and 7.9% and 26.4% in women. The prevalence of sarcopenia was 70.7% in men and 25.0% in women. However, plasma biochemical data are normal range in the older population. In nutrients intake, daily Intake of energy, protein, dietary fiber, water, thiamin, riboflavin, niacin, calcium, and potassium was inadequate considering the Dietary Reference Intakes for Koreans(KDRIs). In conclusion, elderly need to be educated nutrition and their health dietary life to prevent malnutrition, and standard of nutrition intake should be rearranged in elderly aged 85 over considering their antrhopometric index.

  • Silicon nitride (SiNx:H) films are generally used as passivation layer on solar cell and they are usually made by plasma enhanced chemical vapor deposition (PECVD). In this study, we investigated the properties of silicon nitride (SiNx:H) films made by PECVD. Effects of mixture ratio of process gases with silane (SiH4) and ammonia (NH3) on the passivation qualities of silicon nitride film are evaluated. Passivation properties of SiNx:H are focused by making antireflection properties identical with thickness and refractive index controlled. The absorption coefficient of each film was evaluated by spectrometric ellipsometery and the minority carrier lifetimes were evaluated by quasi-steady-state photo-conductance (QSSPC) measurement. The optical properties were obtained by UV-visible spectrophotometer. The interface properties were measured by capacitance-voltage (C-V) measurement and the film components were identified by Fourier transform infrared spectroscopy (FT-IR) and Rutherford backscattering spectroscopy detection (RBS) - elastic recoil detection (ERD).

THE FE Kα EMISSION LINE OF INTERMEDIATE POLAR V1223 SAGITTARII

  • We present measurements of the Fe $K{\alpha}$ emission line of the intermediate polar V1223 Sagittarii observed with the Suzaku satellite. The spectrum is modeled with an absorbed thermal bremsstrahlung spectrum and three Gaussians for the three components of the Fe $K{\alpha}$ lines. We resolve the neutral or low-ionized (6.41keV), He-like (6.70keV), and H-like (7.00keV) iron lines. We also obtain a thermal continuum temperature of 25 keV, which supports a thermal origin of the hard X-rays observed from the shock heated layers of gas between the white dwarf and the shock front. Hence, we believe that the He-like and H-like lines are from the collisional plasma. On the origin of the Fe $K{\alpha}$ fluorescence line, we find that it could be partly from reflections of hard X-rays from the white dwarf surface and the $N_H$ absorption columns. We also discuss the Fe $K{\alpha}$ emission line as veritable tool for the probe of some astrophysical sites.

Development of Membrane Strip Assay System for Lipoprotein Cholesterol (Membrane strip을 이용한 지질단백질 Cholesterol 측정시스템의 개발)

  • To develop a home-version assay system for plasma lipoprotein cholesterol, variables that can control the assay performance were optimized. The system was constructcd by using two major components: nitrocellulose membrane strip with immobilized enzymes (cholesterol esterase, cholesterol oxidase, and horseradish peroxidase); and sample carrier solution containing non-ionic detergent (Triton X-100) and chromogen (3,3'-diaminobenzidine). Once a sample combined with the carrier was absorbed from the bottom of the strip, cholesterol was delivered by capillary action to the immobilized enzymes and a sequential reactions took place. In the final reaction, the chromogen was oxidized and then generated a color as signal that was proportional to the concentration of cholesterol. The signal intensity was enhanced by optimizing conditions for the immobilization of enzymes and the chemical composition of carriel. Under these conditions, a dose-response curve was obtained and revealed a high sensitivity enough to measure the cholesterol in blood.

Change in the Conformation of $p47^{phox}$ by Sodium Dodecyl Sulfate, an Activator of the Leukocyte NADPH Oxidase

  • The leukocyte NADPH oxidase of neutrophils is a membrane-bound enzyme that catalyzes the production of $O_2^-$ from oxygen using NADPH as an electron donor. Dormant in resting neutrophils, the enzyme acquires catalytic activity when the cells are exposed to appropriate stimuli. During activation, the cytosolic oxidase components $p47^{phox}$ and $p67^{phox}$ migrate to the plasma membrane, where they associate with cytochrome $b_{558}$, a membrane-bound flavohemoprotein, to assemble the active oxidase. The oxidase can be activated in a cell-free system; the activating agent usually employed is an anionic amphiphile such as sodium dodecyl sulfate (SDS). Because $p47^{phox}$ can translocate by itself during activation, the conformational change in $p47^{phox}$ may be responsible for the activation of NADPH oxidase. We show here that the treatment of $p47^{phox}$ with SDS leads to an increase in the reactivity of the sutbydryl group of cysteines toward N-ethylmaleimide, indicating that the conformational change occurs when $p47^{phox}$ is exposed to SDS. We propose that this change in conformation results in the appearance of a binding site through which $p47^{phox}$ interacts with cytochrome $b_{558}$during the activation process.

  • A two-step oxidation method was applied on Al6061 to debate the growth mechanism of plasma electrolytic oxidation (PEO) coating. The specimens were first oxidized in the primary electrolyte solution {$Na_3PO_4$ (8g/l), NaOH (2g/l), consequently, the specimens were transferred into a different electrolyte {$K_2ZrF_6$ (8g/l), NaOH (2g/l), $Na_2SiF_6$ (0.5g/l)} for further oxidation. The processes was conducted for various processing times. It was found the second step electrolyte component were reached to inner layers, in contrast to the primary step components which were thrustle to the outer layer. The presence of the secondary component in the inner layers were significantly varied with processing time which suggest the change in growth properties with processing time. further more the inside growth of the secondary component confirmed the increasing trend in the downward growth of the coating layer. The corrosion and hardness properties of the coatings were found highly improved with change in growth features with increasing the processing time.

C 1s photoelectron energy loss spectra of organic electroluminescent materials

  • The C 1s photoelectron energy loss spectra of tris (8-hydroxy-quinoline) aluminum (Alq$_3$) and N,N'-diphenyl-N,N'-bis (3-methyl phenyl)-1,1'-bi-phenyl-4,4'-diamine (TPD) thin films have been investigated. Two major loss structures, namely the plasmon dominated loss lines and shake-up satellites, have been observed. The shake-up spectrum of the C 1s photoelectron line is directly related to the $\pi$-$\pi$$\^$*/ energy gap of the molecule which plays an important role in organic electroluminescent materials. The molecular orbitals of Alq$_3$ and TPD and their major components, quinolime and benzene, have been calculated with the AMI semi-empirical method. The amount of the plasma-dominated loss of Alq$_3$ and TPD, which has to do with the delocalization of electrons through the molecule, was about 24 eV, alike in both cases. The main peak of the C 1s shake-up spectrum of Alq$_3$ and TPD, however, was 5.2 eV and 6.8 eV respectively. It was found that the main shake-up peak reflects more the local $\pi$\longrightarrow$\pi$$\^$*/ transition of quinoline and benzene component rather than the excitation of the whole molecule of Alq$_3$ and TPD. The C 1s shake-up spectra, however, revealed some correlation with the optical energy gap of the organic eletroluminescent materials.

Development of Powder Injection Molding Process for Fabrication of Glass Component

  • Powder injection molding (PIM), which combines the advantages of powder metallurgy and plastic injection molding technologies, has become one of the most efficient methods for the net-shape production of both metal and ceramic components. In this work, plasma display panel glass bodies are prepared by the PIM process. After sintering, the hot isostatic pressing (HIP) process is adopted for improving the density and mechanical properties of the PIMed glass bodies. The mechanical and thermal behaviors of the prepared specimens are analyzed through bending tests and dilatometric analysis, respectively. After HIPing, the flexural strength of the prepared glass body reaches up to 92.17 MPa, which is 1.273 and 2.178 times that of the fused glass body and PIMed bodies, respectively. Moreover, a thermal expansion coefficient of $7.816{\times}10^{-6}/^{\circ}C$ is obtained, which coincides with that of the raw glass powder ($7.5-8.0{\times}10^{-6}/^{\circ}C$), indicating that the glass body is fully densified after the HIP process.
