肛腸療法에 의한 枳實의 卽刻型 알레르기 反應 抑制 效果 (Inhibitory Effect of Immediate-Type Allergic Reaction of Poncirus trifoliata L. by Anal Therapy)

  • Anal therapy is another way of taking medicine It is a traditional pathway but not available in common situation. Nevertheless. It has many benefit and usefulness, it has not treated so much. The dried immature fruit of Poncirus trifoliata L(Ji-Sil) is widely used to treat urticaria, itch and indigestion in traditional Korean Medicine. So this study was carried out to examine the effect of an aqueous extract from immature fruit of Poncirus trifoliate L. on immediate-type allergic reaction by anal administration. Anal administration of Poncirus trifoliata L. showed a marked inhibition rate in systemic immediate-type allergic reaction with a dose of 0.1 - 1 g/kg 1 hr before intraperitoneal injection of compound 48/80. Anal administration of Poncirus tnjoliata L. (0.1 -1 g/kg) significant1y reduced plasma histamine contents induced by compound 48/80. Anal administration of Poncirus trifoliata L. (10 g/kg) also inhibited to $44.6\%$ (P<0.01) local allergic reaction activated by anti-dinitrophenyl (DNP) IgE. In addition, Syzygium aromaticum dose-dependently inhibited the histamine release from the peritoneal mast cells by compound 48/80 or anti-DNP IgE. These results provide evidence that anal therapy of Poncirus trifoliata L. may be beneficial in the treatment of systemic and local immediate-type hypersensitivity by inhibition of histamine release from mast cells in vivo and in vitro.

Antiallergic Action of Magnolia Officinalis on Immediate Hypersensitivity Reaction

  • We studied the effect of aqueous extract of Magnolia officinalis bark (Magnoliaceae) (MOAE) on the immediate hypersensitivity reaction. MOAE (0.01 to 1g/kg) dose-dependently inhibited compound 48/80 induced systemic anaphylaxis in rats. MOAE (0.1 and 1g/kg) also significantly inhibited local immunoglobulin E (lgE)-mediated passive cutaneous anaphylactic reaction. When MOAE was pretreated at concentrations ranging from 0.01 to 1g/kg, the levels of plasma histamine were reduced in a dose-dependent manner. MOAE (0.001 to 1 mg/ml) dose-dependently inhibited the histamine release from rat peritoneal mast cells (RPMC) activated by compound 48/80 or anti-dinitrophenyl (DNP) Igl. The level of cyclic AMP (CAMP) in RPMC, when MOAE was added, significantly increased compared with that of the normal control. Moreover, MOAE (0.01 to 1 mg/ml) had a significant inhibitory effect on anti-DNP Igl-induced tumor necrosis factor-$\alpha$ production from RPMC. These results indicate that MOAE inhibits immediate hypersensitivity reaction in vivo and in vitro.

혈당측정을 위한 폴리포스파젠 진단막의 제조 (Preparation of Polyphosphazene Diagnostic Membranes for Blood Glucose Measurements)

  • 당뇨병 환자의 혈당치 측정을 위하여 메톡시에톡시기와 트라이플루오르에톡시기가 함께 치환된 포스파젠 고분자를 기초로 한 새로운 형태의 진단막을 제조하였다. 플라즈마와 혈액속의 글루코우즈의 농도를 변화시켜가며 활성화 된 폴리포스파젠 진단막을 가지고 680 nm에서의 최종흡광도를 측정하였다. 시간에 따른 흡광도 변화량(K/S)의 최종 결과치가 글루코우즈의 농도가 증가함에 따라 직선적으로 증가하였다. 친수성기와 소수성기의 치환율이 글루코우즈의 농도 측정에 미치는 영향을 조사하였다. 친수성기의 치환율이 증가함에 따라 글루코우즈의 농도와 K/S와의 기울기 값(Dose-Response Slope : DRS)이 점차 증가하였다. 그러나 친수성기의 치환율이 30% 이상일 경우에는 DRS가 급격히 증가하여 정확한 글루코우즈의 농도 측정이 어려웠다.

분리형 펄스 레이져 증착법을 이용한 ZnO 박막의 특성에 관한 연구 (Preparation of High Quality ZnO Thin Films by Separated Pulsed Laser Deposition)

  • The Separated Pulsed Laser Deposition (SPLD) technique uses two chambers that are separated by a conic metallic wall with a central orifice. The pressures of ablation chamber and deposition chamber were controlled by the differential vacuum system. We deposited zinc oxide (ZnO) thin films by SPLD method to obtain high quality thin films. When the bias voltage of +500 V was applied between a substrate and an orifice, the ZnO thin film was deposited efficiently. The deposited ZnO thin film at ablation chamber gas pressure of Ar 5 mTorr showed the sharpest ultraviolet absorption edge indicatory the band gap of about 3.1 eV. ZnO thin films were deposited using effect of electric and magnetic fields in the SPLD method. E${\times}$B drift happened by an electric fields and a magnetic fields, activated plasma plume, as a result the film surface became very smooth. When the bias voltage of +500 V and magnet of 0,1 T were applied the ZnO thin films surface state showed high quality. Grain size was 41.99 nm and RMS was 0.84 nm.

Evaluation of Hemostatic Function with Thromboelastography in Dogs with Hypercoagulable Diseases

  • Thromboembolic complications are increasing in veterinary medicine. Thromboelastography (TEG) is a more comprehensive method for assessing the clotting process than standard plasma-based coagulation tests. This study compared the ability of TEG and standard coagulation tests to analyze the overall hemostatic state of dogs. The study involved 40 dogs with underlying diseases that predispose to hypercoagulability, including neoplasia, hyperadrenocorticism, immune-mediated diseases, gastrointestinal diseases, and protein-losing nephropathies and enteropathies, and 20 healthy dogs. Their overall hemostatic functional state was evaluated by TEG and routine coagulation assays, including activated partial thromboplastin time, prothrombin time, platelet count, and D-dimer concentration. TEG analysis showed significant differences in clot formation time, ${\alpha}$ angle, and maximum amplitude (MA) between diseased and control dogs (P < 0.001 each). Increased MA was the most frequent abnormality on TEG and was indicative of hypercoagulability. TEG was useful in detecting hemostatic dysfunction in dogs with diseases associated with hypercoagulability. Dogs with TEG tracings indicative of hypercoagulability are likely to be in procoagulant states. Future prospective studies are needed to evaluate whether TEG tracings indicative of hypercoagulability are predictive of thrombosis in dogs.

자하거(紫河車)약침이 흰쥐의 혈액성상과 항산화효소의 활성에 미치는 영향 (Effects of Hominis placenta pharmacopuncture on the blood picture and antioxidative activity in rats)

  • Objectives: To investigate the effects of Hominis placenta pharmacopuncture on the blood picture and antioxidative activity in rats. Methods: Sprague-Dawley rats were divided into 3 groups; normal control (n=5), pharmacopuncture at CV12 (CV12 group, n=5), and pharmacopuncture at ST36 (ST36 group, n=5) once every other day for 4 weeks. Blood cell counting was performed and liver superoxide dismutase (SOD), catalase (CAT) and glutathione peroxidase (GSH-Px) activities were analyzed. Results: Values of red blood cell and plasma cell volume were significantly higher in the ST36 group than the normal control. Values of hematocrit, total protein, and albumin were not significantly different among groups. White blood cell count and the percentage of neutrophils, lymphocytes, and eosinophils were not significantly different among groups. However, monocytes and basophils were significantly increased in the ST36, and CV12 groups, respectively. SOD and CAT in the CV12 and ST36 groups were significantly activated than in the normal control group, while the activity of GSH-Px showed no significant difference among groups. Conclusions: Based on the findings of this study, Hominis placenta pharmacopuncture may have positive impact on antioxidative capacity, thus activate various functions of the body.

식물성 산성당으로부터 헤파리노이드의 제조 (Preparation of Heparinoids from Acidic Plant Polysaccharides)

  • Anticoagulant activities were tested for the fifteen kinds of medicinal plants by measuring activated partial thromboplastin time (aPTT). Of them five kinds or species (Artemisia princeps, Sanguisorba officinalis, Artemisia apiacea, Eclipa alba, Schizonepeta tenuifolia) were selected and fractionated for the preparation of acidic polysaccharides. They were extracted with water by refluxing and the extracts were precipitated with ethanol. The precipitates were separated based on charge using a DEAE-Sephadex. The low salt and high salt fractions were sulfated with anhydrous pyridine and chlorosulfonic acid complex. In vitro anticoagulant activities of sulfated polysaccharides were tested by measuring aPTT, prothrombin time (PT), and factor Xa clotting time using normal human plasma. No relationship was found between the amount of uronic acids and anticoagulant activities, but the sulfated ones show the increase of activities. In vivo anticoagulant properties of the sulfated polysaccharide from Artemisia apiacea were also tested by the intraveneous administration of three different doses (3,5 and 10 mg/kg) to rats. APTT and PT were increased significantly and the action of factor Xa and thrombin mediated through antithrombin III were inhibited slightly.

Performance assessment of HEPA filter against radioactive aerosols from metal cutting during nuclear decommissioning

  • Radioactive aerosols are produced during the cutting of contaminated and activated metals. They must be collected and removed by a high-performing filtration system before releasing to the environment from the decommissioning workplace. The filtration system requires regular replacement to ensure the sufficient removal of radioactive aerosols because its filtration efficiency gradually decreases. This study evaluates the efficiency and lifetime of filters while cutting metals by using a plasma arc cutter. Particularly, this study considers the aerodynamic diameter distribution of number and mass concentrations for aerosols from 6 nm to 10 ㎛ when evaluating the performance of filters. After 20 time reuses for cutting operation performed in a cutting chamber, the removal efficiency is reduced from over 99 to below 93% at 2 ㎛. The results are used to analyze the lifetime of filters, the frequencies of their replacements, and impact on internal radiation dose.

Staurosporine Enhances Parathyroid Hormone-Induced Calcium Signal in UMR-106 Osteoblastic Cells

  • Parathyroid hormone (PTH) treatment of bone and kidney-derived cells not only activates adenyly cyclase buy also increases intracellular free calcium, and translocates protein kinase C (PKC) from cytosol to plasma membranes. We have found that acute phorbol ester pretreatment significantly decreases PTH-induced calcium transients and the effect of phorbol ester was antagonized by staurosporine (ST). Although the major effect of ST in that study was the reversal of the action of phorbol ester, it appeared that ST may also have promoted the effect of PTH directly. To further investigate the observation, we examined the effect of ST on the intracellular calcium transients induced by PTH and $\alpha$-thrombin ($\alpha$-TH). For calcium transient experiments, UMR-106 cells were loaded with 2 mM fluo-acetoxymethylester for 30 min at room temperature. The cells were then washed and suspended in buffer containing 1 mM calcium. Fluorescence was detected at 530 nm, with excitation at 505 nm. ST alone did not cause calcium transients, but enhanced the transients elicited by PTH response. added 5 min before the hormone. Another protein kinase inhibitor H-7 likewise enhanced the calcium responses elicited by PTH, while genistein did not affect PTH response. Calcium transients elicited by $\alpha$-TH were also enhanced by ST. The results suggest that there might be tonically activated endogenous protein kinase(s) which inhibit calcium signaling of some calcemic agents.

Cytosolic domain regulates the calcium sensitivity and surface expression of BEST1 channels in the HEK293 cells

  • BEST family is a class of Ca2+-activated Cl- channels evolutionary well conserved from bacteria to human. The human BEST paralogs (BEST1-BEST4) share significant amino acid sequence homology in the N-terminal region, which forms the transmembrane helicases and contains the direct calcium-binding site, Ca2+-clasp. But the cytosolic C-terminal region is less conserved in the paralogs. Interestingly, this domain-specific sequence conservation is also found in the BEST1 orthologs. However, the functional role of the C-terminal region in the BEST channels is still poorly understood. Thus, we aimed to understand the functional role of the C-terminal region in the human and mouse BEST1 channels by using electrophysiological recordings. We found that the calcium-dependent activation of BEST1 channels can be modulated by the C-terminal region. The C-terminal deletion hBEST1 reduced the Ca2+-dependent current activation and the hBEST1-mBEST1 chimera showed a significantly reduced calcium sensitivity to hBEST1 in the HEK293 cells. And the C-terminal domain could regulate cellular expression and plasma membrane targeting of BEST1 channels. Our results can provide a basis for understanding the C-terminal roles in the structure-function of BEST family proteins.
