충남지역 두류작물 재배지 식물기생선충 감염현황 (Incidence of Plant-parasitic Nematodes from Legume Fields in Chungnam Province)

  • To survey the incidence of plant-parasitic nematodes, 187 soil samples from leguminous plant fields in Chungnam province, Korea were assessed from June to August 2018. As the result, four nematode genera - the cyst nematode (Heterodera spp.), root-knot nematode (Meloidogyne spp.), root-lesion nematode (Pratylenchus spp.), spiral nematode (Helicotylenchus spp.) - were detected. Among them, the incidence of cyst nematode was the highest in Taean (73%) and lowest in Hongseong (10%). The cyst nematode was detected from the adzuki and kidney bean fields as well as soybean fields. The incidence of cyst nematode in upland fields was higher than in paddy fields. In addition, the extracted cyst nematodes consisted of two species, which was identified as Heterodera glycines and H. sojae. The incidence of H. glycines (22%) was higher than H. sojae (10%), and a few of the fields (5%) was mixed with two cyst nematodes. This result suggested that we should consider that the cyst nematode is the major plantparasitic nematode when controlling the nematodes in soybean fields in Chungnam province.

Effects of Arugula Vermicompost on the Root-Knot Nematode (Meloidogyne javanica) and the Promotion of Resistance Genes in Tomato Plants

  • Root-knot nematodes are the most important plant-parasitic nematodes worldwide. Many efforts have been made to find non-chemical, risk-free, and environmentally friendly methods for nematode control. In this study, the effects of compost and vermicompost of arugula (Eruca sativa) on Meloidogyne javanica were investigated in three glasshouse experiments. In addition, the expression of the defense-related genes nonexpressor of pathogenesis-related 1 (NPR1) and lipoxygenase 1 (LOX1) was detected in tomato plants treated with vermicompost of arugula at 0, 2, 7, and 14 days after nematode inoculation. The result showed that the vermicompost of arugula significantly reduced the reproduction factor of the nematode by 54.4% to 70.5% in the three experiments and increased the dry weight of shoots of infected tomato plants. Gene expression analysis showed that LOX1 expression increased on the second and seventh day after nematode inoculation, while NPR1 expression decreased. The vermicompost of arugula showed stronger nematode inhibitory potential than the vermicompost of animal manure. The vermicompost of arugula is superior to arugula compost in suppressing the activity of M. javaniva and reducing its impact. It manipulates the expression of resistance genes and could induce systemic resistance against root-knot nematodes.

Survey of Egg- and Cyst-parasitic Fungi of Potato Cyst Nematode in Indonesia

  • Twelve fungal isolates out of 123 isolates obtained from cysts and soils of potato cyst nematode (PCN)-infested fields in Central Java, Indonesia had parasitic abilities of over 50% on PCN eggs or females (cysts) in vitro pathogenicity tests. Cultural and morphological characters and DNA sequences of ribosomal genes in ITS region revealed that they were four isolates of Gliocladium (Trichoderma) virens, three isolates of Fusarium oxysporum, one of F. lateritium, one of Penicillium tritinum and two of Taralomyces spp. A hundred percent infections occurred in eggs or cysts by three fungal isolates G. virens, F. oxysporum and P. oxalicum, suggesting that these fungi may have a good potential for the PCN biocontrol. Especially, G. virens isolates, which occurred most frequently in the PCN-infested potato fields and are known to be highly adaptable to varying habitats, may be developed as reliable agents for controlling PCN with both egg- and cyst-parasitic capabilities and with high ecological adaptabilities.

국내 주요 참다래 재배지에 발생하는 뿌리혹선충 종류 및 분포 (Occurrence and Distribution of Root-Knot Nematodes in Kiwifruit Orchard)

  • 국내 참다래 주산지인 전라남도, 경상남도, 제주도 지역의 참다래 재배지 총 102개 지점을 대상으로 식물기생선충 종류와 밀도를 조사하였다. 검출된 식물기생선충은 뿌리혹선충(Meloidogyne spp.), 나선선충(Helicotylenchus spp.), 침선충(Paratylenchus spp.) 등 9개의 속이었으며, 102개 재배지 중 56%의 재배지가 뿌리혹선충에 감염되어 있는 것으로 나타나 국내 참다래 재배지에서 가장 중요한 식물기생선충은 뿌리혹선충이었다. 뿌리혹선충의 평균 밀도는 97마리/300 cm3 토양으로 참다래의 수량 감소 피해가 우려된다. 뿌리혹선충의 종을 동정한 결과, M. arenaria, M. hapla, M. incognita, M. javanica 4종이 확인되었으며, 그 중 M. arenaria가 가장 우점하고 있다. 그 중 M. arenaria가 가장 우점하고 있다. 과수류에서의 식물기생선충 감염 경로는 감염된 묘목의 이식을 통한 확산이 많음으로 참다래 묘목 생산 및 보급단계에서 뿌리혹선충 무균묘 생산에 주의를 기울여야 할 것이다.

Citrus 속 과수재배지의 식물기생선충 조사 (Survey of Plant Parasitic Nematode in Citrus Fruit Orchards in Korea)

  • 이 연구는 Citrus속 과수재배지에서 식물기생선충의 종류와 분포를 조사하기 위해 수행하였다. 토양 샘플은 주요 재배지에서 2008년 4월부터 8월에 채집하였다. 식물기생선충은 전체 178개 샘플 가운데 139개 샘플에서 검출되었다. 감귤재배지에서 Tylenchulus semipenetrans가 178토양샘플 중 139 샘플에서 검출되었고(90.4%), 유자재배지는 94 토양샘플 중 85토양 샘플에서 T. semipenetrans가 검출되었다(71.4%). 그러나 주요한 식물기생선충의 종류는 다소 차이가 있었다. 국내 Citrus속 과수 재배지에는 T. semipenetrans가 널리 분포하고 있었으며, 감귤과수에 가장 중요한 식물기생선충임을 확인하였다. 재배 년수가 증가할수록 T. semipenetrans의 검출빈도가 증가하였다. 또 토양산도가 알칼리성으로 갈수록 감귤선충의 밀도는 증가하였으나, 상관계수는 높지 않았다.

하우스시설 재배지에서 발생하는 주요 선충 (The major plant-parasitic nematodes in plastic vinyl house field)

  • To know how much damages occurred by nematodes in plastic vinyl house field, soil samples were collected from strawberry cultivation areas at Buyeo and Nonsan in Chungnam and Jinju in Kyeongnam, melon cultivation area at Gocksung in Junnam and cucumber field at Gongju in Chungnam. And then, nematode samples were separated from each soil sample, and identified the kind of plant-parasitic nematodes. Plant-parasitic nematodes were separated from 52 soil samples. Among samples, Meloidogyne spp., Pratylenchus spp. and Helicotylenchus spp. were isolated from 45 (86.5%%) and 33 (63.5%) and 47 collected soil samples (90.4%), respectively. As a result of identification of plant-parasitic nematodes from regional collected soil samples, distribution of Helicotylenchus spp. was higher than any other plant-parasitic nematode. And the population of Meloidogyne spp. and Pratylenchu spp. were also higher, and a occurrence ratio of Meloidogyne spp. is higher than Pratylenchu spp. except the cucumber growing area at Gongju.

Occurrence of Plant-Parasitic Nematodes of Turfgrass in Korea

  • Plant-parasitic nematodes are not only an important constraint on agricultural crop production, but also cause both direct and indirect damage to turfgrass, which is a ground cover plant. However, studies on plant-parasitic nematodes of turfgrass in Korea are scarce. A survey for plant-parasitic nematodes was carried out on 13 golf courses in Korea. The results yielded 28 species/taxa belonging to 16 genera and 12 families of plant-parasitic nematodes. Among the isolated species, Helicotylenchus microlobus, Mesocriconema nebraskense, Tylenchorhynchus claytoni, Mesocriconema sp., and Meloidogyne graminicola were the most prevalent species in all management zones. Twelve species were new records of plant-parasitic nematodes in Korea. Highest maximum densities were showed by T. claytoni, Paratylenchus nanus, M. nebraskense, M. graminicola, and H. microlobus. Diversity (H'), was significantly higher in fairways compared to tees and greens, though species evenness (J') and dominance (D) showed no statistically significant differences. This information is crucial in nematode problem diagnosis, and the subsequent formulation of management strategies.

Survey of Nematodes in Coniferous Bonsai in Korea

  • As preventive measures for bonsai exports, nematodes were isolated from 55 bonsai samples of five coniferous species (Chamaecyparis pisifera, Juniperus chinensis, Pinus densiflora, Pinus thunbergii, and Taxus cuspidate) from all 15 bonsai gardens in Korea. Nematodes belonging to 21 genera in 7 orders were isolated from the 55 bonsai samples. Among plant-parasitic nematodes, Tylenchus spp. was the most frequently isolated (14.9%), followed by Ditylenchus spp. (10.5%), Aphelenchoides spp. (9.5%), Aphelenchus sp. (5.5%), Criconemoides sp. (4.0%), Helicotylenchus sp. (0.7%), Hemicycliophora sp. (0.7%), Mesocriconema sp. (0.7%), Tylenchorhynchus sp. (0.7%), and Paratylenchus sp. (0.4%). Among nonparasitic nematodes, Cephalobina was the most frequently isolated nematodes (26.5%), followed by Rhabditida (19.3%), Dorylaimida (17.8%), Pangrolaimida (14.5%), Plectida (6.5%), Tryphylida (6.2%), Mononchida (3.3%), Alaimida (2.9%), Monhysterida (2.5%), and Triplonchida (0.4%). Based on these results, we conclude that there is no problematic plant-parasitic nematode in bonsai gardens of Korea.

뿌리혹선충 유전자의 RNA 간섭 억제에 의한 선충저항성 식물 개발 및 선충방제의 최근 연구 동향 (Recent Studies on Development of Transgenic Plants Induced Root-Knot Nematode Resistance by RNA Interference Suppression of Nematode Genes and Nematode Prevention)

  • Root-knot nematodes cause billions of dollars in crop losses annually have a broad range of host over 2,000 species of plants. These nematodes are known as obligate, sedentary endo-parasites in a plant host to feed upon to complete their life cycle. To prevent the plant parasitic nematode, methyl bromide was widely applied as a soil fumigant. Other strategies to prevent or control nematodes involve RNAi-mediated suppression, R gene transformation, natural products or chemical treatments, the expression of peptide or proteins in susceptible plants, and others. Over the last decade, the entry in GenBank for Meloidogyne reveals 73,340 ESTs and recently two complete Meloidogyne spp. genomes sequences have simultaneously been presented by two groups. Recent works have demonstrated the effect of RNAi suppression to nematode target genes. These results will provide novel members of genes as a foundation for studies focused on understanding the function of M. incognita nematode genes as well as for the development of novel target genes for parasite control. Thus the successful development of biotechnology-derived plants with nematode resistance will result in large yield benefits for producers as well as environmental benefits and will accelerate the research related to pathogensresistant crops.

Plant-Parasitic Nematode Genera Associated with Turfgrass in Maryland Golf Courses and Athletic Fields

  • Field surveys were conducted to assess the occurrence and diversity of plant-parasitic nematodes (PPNs) in golf courses and athletic fields across Maryland, USA, during 2022 and 2023. A total of 28 golf courses and ten athletic fields were surveyed, revealing the prevalence and abundance of 13 PPNs taxa in the region. Criconemoides was identified as the most prevalent (94.9%) and Tylenchorhynchus as the most abundant (2.3) across all samples. Central golf courses (west side of the Chesapeake Bay) exhibited a high prevalence of Criconemoides and Tylenchorhynchus, while Eastern Shore golf courses and athletic fields displayed a higher prevalence of Helicotylenchus and Criconemoides. Further, Belonolaimus longicaudatus was reported for the first time from turfgrass in Maryland, raising concerns due to its potential to cause severe damage on both cool- and warm-season turfgrass. Biodiversity analysis indicated that richness (R2) was higher in athletic fields, while diversity (H') and evenness (J') were significantly greater in golf courses. This study provides baseline information for monitoring PPNs distribution in Maryland and also for the development of effective nematode management approaches in turfgrass ecosystems.
