Electrochemical reduction of nitrate was studied using Zn, Cu and (Ir+Ru)-Ti cathodes and Pt/Ti anode in a cell divided by an ion exchange membrane. During electrolysis, effects of the different cathode types on operating parameters (i.e., voltage, temperature and pH), nitrate removal efficiency and by-products (i.e., nitrite and ammonia) formation were investigated. Ammonia oxidation rate in the presence of NaCl was also determined using the different ratios of hypochlorous acid to ammonia. The operating parameter values were similar for all types of cathode materials and were maintained relatively constant. Nitrate was well reduced and converted mostly to ammonia using Zn and Cu cathodes. Ammonia, produced as a by-product of nitrate reduction, was oxidized in the presence of NaCl in the electrochemical process and the oxidation performance was enhanced upon increasing the hypochlorous acid-to-ammonia ratio to 1.09:1. Zn and Cu cathodes promoted the nitrate reduction to ammonia and the produced ammonia was finally removed from solution by reacting with hypochlorite ions. Using Zn or Cu cathodes, instead of noble metal cathodes, in the electrochemical process can be an alternative technology for nitrate-containing wastewater treatment.
Intraspecific genetic relationship of 19 variation types of the Yam (Dioscorea alata) classified by their external morphological characteristics such as leaf and tuber shape were assessed by DNA using random and specific primer. Twenty two out of 113 primers (100 random[10-mer] primers, two 15 mer [M13 core sequence, and (GGAT)$_4$ sequence]) had been used in PCR-amplification. Only 12 primers, however, were success in DNA amplification in all of the analyzed plants, resulting in 93 randomly and specifically amplified DNA fragments. The analyzed taxa showed very high polymorphisms(69 bands, 71.0 %), allowing individual taxon to be identified based on DNA fingerprinting. Monomorphic bands among total amplified DNA bands of each primer was low under the 50%. Similarity indices between accessions were computed from PCR(polymerase chain reaction) data, and genetic relationships among intraspecific variations were closely related at the levels ranging from 0.66 to 0.90. These DNA data were not matched well with those of morphological characters since they were divided into two major groups at the similarity coefficient value of 0.70. Therefore, Grouping of species into variation types by mainly morphological charactistics was suggested unreasonable.
Controlling the production of diverse cell/tissue types is essential for the development of multicellular organisms such as animals and plants. The Arabidopsis thaliana root, which contains distinct cells/tissues along longitudinal and radial axes, has served as an elegant model to investigate how genetic programs and environmental signals interact to produce different cell/tissue types. In the root, a series of asymmetric cell divisions (ACDs) give rise to three ground tissue layers at maturity (endodermis, middle cortex, and cortex). Because the middle cortex is formed by a periclinal (parallel to the axis) ACD of the endodermis around 7 to 14 days post-germination, middle cortex formation is used as a parameter to assess maturation of the root ground tissue. Molecular, genetic, and physiological studies have revealed that the control of the timing and extent of middle cortex formation during root maturation relies on the interaction of plant hormones and transcription factors. In particular, abscisic acid and gibberellin act synergistically to regulate the timing and extent of middle cortex formation, unlike their typical antagonism. The SHORT-ROOT, SCARECROW, SCARECROW-LIKE 3, and DELLA transcription factors, all of which belong to the plant-specific GRAS family, play key roles in the regulation of middle cortex formation. Recently, two additional transcription factors, SEUSS and GA- AND ABA-RESPONSIVE ZINC FINGER, have also been characterized during ground tissue maturation. In this review, we provide a detailed account of the regulatory networks that control the timing and extent of middle cortex formation during post-embryonic root development.
A technical approach to design and carry out an experiment to determine the uptake of selected radionuclides in site-specific conditions in Kuwait was developed and successfully executed for developing a radioecological decision support system. The radionuclides from soil-to-plant transfer factors have been obtained for leafy and non-leafy vegetables, and root crops cultivated in Kuwait. Two types of vegetated soils were selected and spiked with high concentrations of three relatively short-lived selected radionuclides (85Sr, 134Cs, and 133Ba). The highest strontium and barium transfer factors were found in the order: leafy vegetables > root crops > non-leafy vegetables. The approximate range of radiocesium transfer factor was found to be low in all plant groups and was comparable to those reported elsewhere in different soil types of temperate and tropical environments. A strong negative correlation between the obtained transfer factors and the distribution coefficient of the radionuclide in soil was found. It is recommended to adopt the newly derived parameters for the sensitive areas in Kuwait and other Gulf countries instead of using the generic parameters, whenever dose calculation codes are used. This will help to more accurately assess and predict the end results of the committed effective dose equivalent through ingestion pathway.
Analyses of now karyotypes using different maize (Zea mays L.) inbred lines have been performed. The accumulation and isolation of high quality and quantity metaphase chromosomes from root tips can be achieved from many kinds of maize lines. The chromosome suspensions were prepared by a simple slicing method from synchronized maize root tips and analyzed with a now cytometry. The variations of experimental now karyotypes were detected among inbred lines in terms of the positions and/or the numbers of chromosome peaks. The 2C DNA amount among 8 inbred lines ranged from 5.09 to 5.52 pg. The variability of DNA content in maize chromosome 1 was 9.1 % ranging from 0.685 to 0.747 pg. The selection of appropriate maize lines is critical for sorting specific single chromosome types. At least five different chromosome types can be discriminated and sorted from five maize lines.
The phenotype of Rhizoctonia solani KR-13 was randomly segregated to both melanin-producing (M+) and non-producing (M-) types through successive cultures on PDA. M+type with dark melanin showed strong pathogenicity to rice and self-anastomosis. Meanwhile, M- type with white or less-melanized mycelia showed very weak pathogenicity and non-self-anastomosis. Melanin production of R. solani was affected by incubation temperature in both M+ and M- types, but not by light treatment. The application of tricyclazole, an inhibitor of fungal melanin biosynthesis, showed no controlling effect on R. solani causing rice sheath blight. Results of this study showed that melanization of mycelia of R. solani is an important pathogenicity factor in rice.
Mitochondrial DNA restriction fragment length polymorphisms are convenient markers for identifying cytoplasmic variation among plants. We have collected 212 wild soybeans (Glycine soja Sieb. et Zucc) from all over Korea, and classified mitochondrial genome types based on hybridization patterns in DNA gel-blot analyses using two mitochondrial DNA clones, cox2 and atp6, as probes. Korean wild soybean was classified with eight-mtDNA types, and some of the mtDNAs showed geographical clines among the regions. The diversity index of the mtDNA was much higher in the western and southern regions than in the eastern and northern regions of Korea, respectively. Dissemination and distributive characteristics of wild soybeans in Korea were discussed.
Cosmarium candianum and C. obtusatum were sampled at 11 stations in Korea from October 1987 to June 1988. The samplings were cultured for study of the character variations in colonies. As a result, C. candianum var. candianum f. minutum, C. candianum var. depressum and C. candianum var. latius need not be separated from C. candianum, because 4 types in C. candianum complex occurred simultaneously in the same colony. It was changed the species reported as C. circulare Reinsch in Korea presently into C. candianum Delponte according to Compere's review. Because 2 types of C. obtusatum complex occurred at the same colony, C. obtusatum var. beanlandii need not separated from C. obtusatum.
There are 474 species of native vascular plant distributed in offshore islands in Shandong Province, which belong to 262 genera and 82 families. The distribution types of genera of seed plant were classified into 15 types. Temperate characteristic is dominant in the flora of offshore islands and tropical characteristic ranks secondary. Therefore, some islands play the role of stepping islands. Furthermore, 98.85% of the total genera are monotypic or bitypic which shows the offshore habitat heterogeneity in Shandong Province.
Recently, many researches for plant alkaloids, one of the largest groups of natural products, are reported because of their various pharmacological activity. This study was carried out to clone putrescine N-methyltransferase (PMT) gene which is a key enzyme in diverting polyamine metabolism towards the biosynthesis of nicotine and related alkaloids from Burley tobacco. To induce expression of PMT gene in tobacco plant, the floral meristem was removed and then mRNA was purified from root. cDNA encoding PMT gene was isolated by RT PCR and cloned. Three different groups of clones were screened by PCR and restriction enzyme digestion analysis and were characterized. The data of these screening revealed that three types of PMT are present in Burley tobacco. Comparison of the nucleotide sequence of this three genes encoding putative PMT with those of other tobaccos revealed that two types of PMT are newly discovered from Nicotiana tabacum cv. Br21 tobacco and they were same as PMT2, PMT3 of N. tabacum cv. Xanthi.
