  • Codonopsis lanceolata (Campanulaceae) has been used in traditional medicines, as its roots contain several kinds of triterpenoid saponin with high medicinal values. In this work, we induced transgenic hairy roots of C. lanceolata and analyzed triterpenoid saponins from the hairy roots and hairy root-derived transgenic plants. Hairy roots were obtained from leaf explants by the transformation of Agrobacterium rhizogenes R1000. Transgenic hairy root lines were confirmed by the transcriptional activities of rolA, B, C, and D genes by RT-PCR. Transgenic root lines actively proliferated on hormone-free medium but not in nontransformed roots. Hairy roots contained richer triterpenoids (lancemaside A, foetidissimoside A, and aster saponin Hb) than nontransformed roots. Transgenic plants were regenerated from the hairy roots via somatic embryogenesis. They showed phenotypic alterations such as shortened shoots and an increased number of axillary buds and adventitious roots. The transgenic plants also contained higher triterpenoid levels than wild-type plants. These results suggest that hairy roots and transgenic plants of C. lanceolata could be used as medicinal materials for the production of triterpene saponins.

  • Methanol extract of Ilex pubescens roots prolonged bleeding time threefold, and inhibited the generation of malondialdehyde released during platelet aggregation inducted by thrombin. Through several purification procedures, its saponin, named ilexoside, was proved to be responsible for the antithrombotic activities of the plant. Ilexosides A, -D and -J and 24-carboxypomolic acid showed strong inhibitory activities on platelet aggregation induced by thrombin.

  • These studies were conducted to know the any differences in growth characteristics and root quality due to flower color and culture periods in Chinese bellflower. White flowered lines had tendency of increasing in growth characters such as stem numbers, capsule numbers and seed yield per plant, plant height, and root characters such as root weight, root length and root diameter even though statistically not significant. No differences were observed in saponin and essential oil contents between them. Three years old plants had more stems and capsules per plants, root weight and root length than two years old plants while no difference in saponin and essential oil contents between them. Saponin content was gradually decreased while essential oil content was clearly lowed in the roots from plants cultured more than 3 years even though no differences in stem and capsule numbers per plant and plant height.

  • 1. To know the site of saponin synthesis in this plant, 4-years old Panax ginseng C.A. Meyer was administered with 1, 2-l4C-acetate (Na salt, 10 ucilplant) by stem injection and was continued to grow for 3 weeks and the distribution of the radioactivity in leaf, stem and root part was identified. The percentage of radioactivity recovered was about 3.99%. 2. The sliced roots or leaf discs (2g) were bathed in the reaction mixture containing sugar, ATP, NADPH, and the distribution of the radioactivity of the fractions (sugar, saponin, sapogenin) was identified. 3. It seemed that major synthesized saponins in roots and leaves are dial and triol-type, respectively. Although both types of saponins are synthesized in roots, the main saponins seemed to be dial saponins and a significant portion of triol saponins are supplied from leaves through stem.

  • Saponin was distributed in all tissues of Phytolacca americana and P. esculenta. Phytolaccoside E was a major saponin, but the callus tissues induced from stems of P. americana contained phytolaccoside B as a major component. Saponin contents in leaves and stems were almost similar to those in roots. Two unknown compounds, $C_{32}H_{50}O_{4}$ (triterpenoid) and $C_{18}H_{22}O_{5}{\cdot}H_2O$ (polyphenol) were isolated from seeds of both plant.

  • Bioassay-guided fractionation has led to the isolation of triterpenoid saponins such as Acutoside A (3-O-[O-${\beta}$-D-glucopyrano$yl-(1${\to}$2)-O-${\beta}$-D-glucopyranosyl] oleanolic acid) from the whole plants of Viola hondoensis. Among them, Saponin 1 exhibited potent inhibitory activity against matrix metalloproteinase (MMP)-1, and prevented the UV-induced changes in the MMP-1 expression. In addition, compound was isolated from this plant for the first time.

  • This study was conducted to provide basic information about the drying methods (daylight, hot-air, and freeze drying) used for Platycodon grandiflorum radix. We investigated the mineral, free sugar, and saponin contents of dried P. grandiflorum. The potassium and calcium contents of hot-air-dried samples were the highest (22.6 and 9.2 mg%, respectively), when compared to those of daylight- or freeze-dried samples. Glucose and sucrose contents were the highest in freeze-dried samples (1,552 and 145.0 mg%, respectively), while fructose content was the highest in hot-air-dried samples (611.9 mg%). Platycodin D content was the highest in hot-air-dried samples (622.0 mg%); however platycodin D3, polygalacin D, and deapioplatycodin D contents were the highest in daylight-dried plant (113.5, 756.6, and 109.2 mg%, respectively). Glucose content was highly negatively correlated (p<0.01) with platycodin D, platycodin D3, and deapioplatycodin D (-0.924, -0.957, -0.861, p<0.01, respectively). These results suggest that the drying method affects the saponin content of P. grandiflorum and daylight and hot-air drying methods are more suitable and beneficial than freeze-drying.

  • This study was carried out to compare growth characteristics, and extract and crude saponin contents of 4-year-old ginseng cultured by direct seeding and transplanting cultivation at several farms of the main producing district, Geumsan and Eumsung. Though root weight per plant of direct seeding cultivation was lower than that of transplanting cultivation, but yield of the former was higher than that of the latter owing to high rate of survival plant and Leaf Area Index. Dry matter partitioning ratio of direct seeding cultivation was high in primary root and low in secondary root because direct seeding cultivation elongated the length of primary root, while it suppressed the growth of secondary root. Ratio of rusty root was decreased in condition of direct seeding cultivation, while the contents of extract and crude saponin were lower than that of transplanting cultivation.
