The volcanic aspect on determining Site of nuclear power plant in Indonesia: Gap analysis between standard and regulations

  • The development of nuclear power plants is in three phases. The first phase is a consideration before the decision on the NPP construction program is approved, the second phase is the preparatory work for making contracts and preparing for the construction of NPP after the NPP construction policy is approved, and the third phase is contracting, licensing and building the first NPP. As a volcanically active country, Indonesia contains over 130 active volcanoes that are part of the Pacific Ring of Fire. The volcanic aspect is one of the safety factors considered while deciding the location of an NPP. Research on the potential of natural external risks to the determination of nuclear power plants in Indonesia, including the volcanic aspect, has been conducted based on the safety reference or safety guide of the IAEA and the Nuclear Energy Regulatory Body (BAPETEN) Regulation. Due to technological advancements, safety needs have evolved so the existing Indonesia National Standard (SNI) must be updated to comply with BAPETEN regulations. The substance in SNI 18-2034-1990 relating to volcanic features seems less relevant in actual conditions, given that more complete and exact criteria for determining a site guarantee the safety and health of residents and surrounding the environment site. The study intends to conduct a gap analysis of volcanic issues in SNI and volcanic regulations. The method used is identification requirements for volcanic aspects in SNI 18-2034-1990 about Determining Site of Nuclear Reactor Guidance with BAPETEN Chairman Regulation (BCR) number 4 of 2018 about Nuclear Installation Site Evaluation Safety Provisions and BCR number 5 of 2015 about Evaluation of Nuclear Installation Sites for Volcanic Aspects, and analysis uses a qualitative method of inductive techniques. The outcome of this research applies to suggesting a revision of SNI number 18-2034-1990, especially the volcanic aspect.

  • Recently, wind power has been gaining attention as a highly efficient renewable energy source, leading to various technological developments worldwide. Typically, wind power is operated in the form of large wind farms with many wind turbines installed in areas rich in wind resources. However, in developing countries or regions isolated from the power grid, off-grid small wind power systems are emerging as an efficient solution. To efficiently operate and expand off-grid small-scale power systems, the development of real-time monitoring systems is required. For the efficient operation of small wind power systems, it is essential to develop real-time monitoring systems that can actively respond to excessive wind speeds and various environmental factors, as well as ensure the stable supply of produced power to small areas or facilities through an Energy Storage System (ESS). The implemented system monitors turbine RPM, power generation, brake operation, and more to create an optimal operating environment. The developed small wind power system can be utilized in remote road lighting, marine leisure facilities, mobile communication base stations, and other applications, contributing to the development of the RE100 industry ecosystem.

Structural analysis, anti-inflammatory activity of the main water-soluble acidic polysaccharides (AGBP-A3) from Panax quinquefolius L berry

  • Background: Panax quinquefolius L, widely recognized for its valuable contributions to medicine, has aroused considerable attention globally. Different from the extensive research has been dedicated to the root of P. quinquefolius, its berry has received relatively scant focus. Given its promising medicinal properties, this study was focused on the structural characterizations and anti-inflammatory potential of acidic polysaccharides from the P. quinquefolius berry. Materials and methods: P. quinquefolius berry was extracted with hot water, precipitated by alcohol, separated by DEAE-52-cellulose column to give a series of fractions. One of these fractions was further purified via Sephadex G-200 column to give three fractions. Then, the main fraction named as AGBP-A3 was characterized by methylation analysis, NMR spectroscopy, etc. Its anti-inflammatory activity was assessed by RAW 264.7 cell model, zebrafish model and molecular docking. Results: The main chain comprised of α-L-Rhap, α-D-GalAp and β-D-Galp, while the branch consisted mainly of α-L-Araf, β-D-Glcp, α-D-GalAp, β-D-Galp. The RAW264.7 cell assay results showed that the inhibition rates against IL-6 and IL-1β secretion at the concentration of 625 ng/mL were 24.83 %, 11.84 %, while the inhibition rate against IL-10 secretion was 70.17 % at the concentration of 312 ng/mL. In the zebrafish assay, the migrating neutrophils were significantly reduced in number, and their migration to inflammatory tissues was inhibited. Molecular docking predictions correlated well with the results of the anti-inflammatory assay. Conclusion: The present study demonstrated the structure of acidic polysaccharides of P. quinquefolius berry and their effect on inflammation, providing a reference for screening anti-inflammatory drugs.

The Growth Characteristics and Germanium Uptake by Water Celery in Soil Treated with Germanium (게르마늄 처리 토양에서 미나리 생육 특성과 게르마늄 흡수)

  • In order to obtain the basic information for agricultural utilization of Germanium(Ge), the growth characteristics and the germanium uptake by water celery were investigated at different concentration of germanium in soil. This experiment was carried out in the Wagner pot(1 $5,000^{-1}a$). Germanium concentrations in soil for water celery cultivation were maintained at 0.26, 25.0, 62.5, and 125.0 mg $kg^{-1}$, respectively. The treatment of over Ge 25.0 mg $kg^{-1}$ in the soil led to germanium phytotoxicity such as reduction of plant height and fresh weight. The contents of germanium in water celery were increased with the increase of germanium concentration in the soil. When water celery was cultivated from soil maintained with Ge 25.0 and 62.5 mg $kg^{-1}$, its germanium contents in plant were 89.9 and 371.6 mg $kg^{-1}$, respectively. Then, the efficiency of germanium uptake of water celery in Ge 25.0 and 62.5 mg $kg^{-1}$ maintained plots was 1.7 and 2.4%, respectively. When water celery was cultivated from soil maintained with Ge 25.0, 62.5 and 125.0 mg $kg^{-1}$, its content of amino acid was found to be 89.8, 198.4, and 318.2 mg $g^{-1}$, respectively. To investigate the effect of N fertilizer application in uptake of germanium by water celery, these were treated with nontreatment, 1.0, 1.5 and 2.0 times of N application based on soil testing for cultivation of water celery. However, the amount of the N fertilizer application did not affect the contents of germanium in the water celery. When water celery was cultivated from soil maintained with two kinds of inorganic and organic germanium 50 mg $kg^{-1}$, respectively, the content of germanium were 24.2 mg $kg^{-1}$ in the Ge-132 treatment and 11.8 mg $kg^{-1}$ in the $GeO_2$ treatment.

The Status of Endangered Plants Distributed in the Middle Eastern Area of Korea and Evaluation of the Risk Factors (우리나라 중동부지역에 분포하는 멸종위기야생식물 현황과 위험요인 평가)

  • Evaluation of the extinction risk of endangered plants at international, national as well as at regional levels is essential to the implementation of plans for direct conservation activities. Reports indicate that 34 endangered plants are distributed in the middle eastern area of Korea. For each endangered plant, we investigated the sites, area of extents, population size, and factors that affect population extinction. We assessed risk factors based on 10 evaluation criteria including the results from the investigation and the life traits each endangered plant has. As a result of evaluating the risk factors, these 34 endangered plants are classified into 3 groups: the first category comprises 12 endangered plants that require active and urgent conservation of habitats due to multiple risk factors; the second group has 16 endangered plants that should be able to persist with the removal of a few direct risk factors; the third category has 6 endangered plants that can persist with minimal management due to comparatively large distributed area and numerous individuals. It was found that most major risk factors in the population of endangered plants are caused by disruption of habitats and population extinction due to the increase of human habitation in the concerned areas, development and illegal harvesting. Futhermore, ecological collapse from decreasing habitats and malfunctioning mechanism of extinction and regeneration due to the changes of vegetational environment can be the other causes. From the area of the present investigations, we selected 5 regions according to the number of species and the frequency of appearance and importance of conservation measures. Also, we suggested a conservation strategy according to the regional characteristics. We suggest that the method for evaluating extinction risk of endangered plants includes distributional data and life traits of species. In addition, we underscore the necessity for understanding population dynamics and ecological niche of the each target species.

Reconsideration of Acer pictum complex in Korea (한국산(韓國産) 고로쇠분류군(分類群)에 대한 재고(再考))

  • Acer pictum complex (A. pictum Thunb. ex Murray with varieties, A. okamotoanum Nakai, A. truncatum Bunge) in eastern Asia causes frequent difficulty in identification. One hundred twenty five specimens from A. pictum complex of China, Korea and Japan and A. cappadocicum var. sinicum of China were compared to investigate patterns of intra- and interspecific variation and to evaluate a recognition of several species as well as many varieties using 22 characters for morphometric analysis. The first three PCA accounted for 59% of the total variance. No strong discontinuities existed among taxa with respect to fruit and leaf characters. Much overlap among all taxa occurred the central region of the scatter diagram. Many characters appeared to show some clinal variation with changes from east of China to Japan through Korea. This was true not only when all species as considered as a single taxon, but when characters of individual taxa were compared with geography. As one considers a path from the western part of the ranges to areas to the east, the leaves become larger in most respects and become increasingly many lobed (five to seven or nine). In general, there was a tendency toward larger nutlet with smaller wing in the area toward northeast of China (=A. truncatum), while in the east of ranges (Island Ullung-do), plants were larger with respect to characters of fruit and leaves (=A. okamotoanum). The morphological differentiation between A. okamotoanum and Japanese and Korean individuals of A. pictum was not considered sufficient to warrant recognition of either specific or varietal status and should be treated as con specific under A. pictum var. mono. Since the lectotype of Acer pictum had minute hairs uniformly on the under surface of leaves(A. pictum var. pictum), the glabrous type of A. pictum was called A. pictum var. mono as Ohahsi suggested. The univaraite analysis (the mean and maximum/minium of nutlet size and wing/nutlet length ratio) indicated geographical differentiation of northeastern populations, A. truncatum, was distinctive, but Korean individuals of A. truncatum showed an affinity between Chinese individuals of A. truncatum and Korean individuals of A. Pictum var. mono. The current results, together with qualitative character, trunk features, justify subspecific status for this taxon. The previous varieties of A. mono in Korea were indistinguishable from typical form of A. Pictum var. mono on the basis of the wing angle and nutlet size, rejecting continued recognition of these taxa as distinctive varieties. Therefore, it is recommended that only one polymorphic species of A. pictum be recognized in addition to three varieties.

Antioxidant Activities of Phenolic Compounds from Medicinal Plants (Hibiscus esculentus, Cirsium japonicum, Zizania latifolia and Kalopanax pictus) (약용식물(오크라, 엉컹퀴, 엄나무, 줄풀) 유래 페놀성 물질의 항산화 활성)

  • In this study, the antioxidant activity of water and ethanol extracts from Hibiscus esculentus, Cirsium japonicum, Zizania latifolia and Kalopanax pictus for functional food source were examined. The optimal conditions for phenolic compounds extraction from medicinal plants were at 50% ethanol with Hibiscus esculentus and Cirsium japonicum var. ussuriense, at 40% ethanol with Kalopanax pictus and at 60% ethanol with Zizania latifolia. The total phenolic contents from the extracts of medical plants were determined to be 2.72~34.15 mg/g in the water extracts and 2.83~34.23 mg/g in the ethanol extracts. The electron-donating abilities (EDA) of the water and ethanol extracts were both above 74% at the low concentration of $50{\mu}g/mL$. The ABTS radical-cation decolorization was above 88% at $100{\mu}g/mL$ concentration in all the extracts of various medicinal plants. The antioxidant protection factor (PF) in the water and ethanol extracts of the Cirsium japonicum var. ussuriense extracts was $1.73{\pm}0.02PF$ and $1.76{\pm}0.01PF$ at $50{\mu}g/mL$ concentration respectively, and was higher than those of the other medicinal-plant extracts. The TBARs inhibition rates of all the medicinal-plant extracts, were above 80% at the $50{\mu}g/mL$ concentration except Hibiscus esculentus. These results confirmed that the various oriental medicinal plants (Hibiscus esculentus, Cirsium japonicum var. ussuriense, Kalopanax pictus and Zizania latifolia) that were included in this study are useful anti-oxidant and functional-food resources.

Consideration on coexistence strategy of GM with non-GM, environmentally friend crops in South Korea (GM과 non-GM, 친환경작물의 공존을 위한 제도 보완의 필요성)

  • The current legislation in South Korea clearly states that the tolerance threshold on the adventitious presence of GMO in environment-friendly agricultural products is 3.0% and no GM seed should be detected in their planting seed batches. To date, in Korea, there is no approved GM crop for commercial cultivation in field. However, several GM crops including rice, Chinese cabbage, potato and wild turf grass are currently under risk assessment for their environmental release. Also Korean government (Rural Development Administration, RDA) announced that 11 institutes including universities have been currently certified to carry out a risk assessment of GM crops. Meanwhile, the cultivated area and certified quantities of environment-friendly crops (organic, pesticide-free and low-pesticide) are sharply increasing every year according to the report of National Agricultural Products Quality Management Service (NAQS). In detail, in 2007, the certified quantities of environment-friendly agricultural products were elevated up to 100-fold for organic, 171-fold for pesticide-free and 2,324-fold for low-pesticide crops when compared with those in 1999. The total certified quantity of environment-friendly cereal crops in 2007 was equivalent to 6.4% of total production of cereal crops. Moreover, 24% of total production of root and tuber crops such as potato and sweet potato were certified for environment-friendly agricultural products. In these circumstances, I strongly suggest that current legislations on GM crop's safety management should be revised to include strategies for the coexistence of GM with non-GM crops, especially environment-friendly crops before GM crop is approved to be cultivated for commercialization. Since all types of crops are grown in an open environment, the adventitious presence of GM crops among non-GM crops is inevitable if appropriate measures for coexistence are not established for species by species such as isolation distance, workable management measures to minimize admixture.

Effect of Application Level of Swine Slurry on Agronomic Characteristics and Yield of Corn and $NO_3-N$ Content of Corn Field (돈분액비 시용수준이 사료용 옥수수의 생육특성과 수량 및 토양 중 $NO_3-N$ 함량에 미치는 영향)

  • Livestock manure has been utilized as fertilizer, and trying to make resources natural circulation of organic material. This experiment was conducted to investigate the effect of application level of swine slurry on agronomic characteristics and yield of silage corn and $NO_3-N$ content of soil for three years in the experimental field of Grassland and Forage Crops Division, National Livestock Research Institute. Summary of the results were as follows. The experiment was conducted according to a randomized complete block design. Six treatments were non fertilizer(NF), chemical fertilizer(CF), $100\%$ swine slurry(SS100), $150\%$ swine slurry(SS150), $200\%$ swine slurry(SS200) and mixture fertilizer(MF, $100\%$ swine slurry + $50\%$ chemical fertilizer) with three replications. The application level of swine slurry(SS) $100\%$ was highest in Brix as $8.6^{\circ}$ and stay green was as swine slurry application increased. Deficiency of plant, plant height, ear height and lodging were increased in above $100\%$ swine slurry application level. Dry matter yield was decreased in SS 100 but increased in SS 150 and SS 200. Significant effect was observed for all treatments. The content of $NO_3-N$ in infiltration water was high in above swine slurry nitrogen $150\%$ but that content in run off water was very small. Conclusively, application to swine slurry below $150\%$ would be recommended to produce higher yield and to conserve environment in corn field.

Genomic Analysis of Satellite RNA of Cucumber mosaic virus-Paf Related with Mild Symptoms (Cucumber mosaic virus Paf 계통의 약독 병징과 관련된 satellite RNA의 유전자 해석)

  • Recently, we reported a satellite RNA (Paf-satRNA) which is encapsidated in a pepper isolate of Cucumber mosaic virus (CMV-Paf) regulated symptom attenuation of the helper virus. To characterize mild symptom domain of Paf-satRNA, a series of chimeric cDNAs of satRNAs were created by using full-length cDNA clones of Paf-satRNA and a Pep-satRNA, chlorosis-inducing satRNA in pepper plants, and analyzed for determinants of symptom attenuation. When compared the nucleotide sequences, the 3' and 5' terminal sequences of the two wild-type (wt) satRNAs contained relatively conserved sequences which are the typical to CMV satRNA. Ten bases insertions were found in PepY-satRNA, and two variable regions, 81st to 113th and 183rd to 265th from the 5'-end, were located in the middle parts of the satRNAs. To delineate the attenuated symptom-related domain for the Paf-satRNA, in vitro transcripts RNAs transcribed from the wt cDNAs and constructed chimeric cDNAs were combined with genomic RNAs, RNA1, RNA2 and RNA3, of CMV-Fny and inoculated onto Nicotiana benthamiana plants. These transcripts were fully infectious onto the N. benthamiana and infectivity was confirmed by the RT-PCR. Chimeric Paf(H/N)-satRNA and PepY(N/A)-satRNA as well as Paf-satRNA induced very mild mosaic or symptomless infection on N. benthamiana. By contrast, typical mosaic symptom and stunting of infected plants were induced when PepY-satRNA, PepY(H/N)-satRNA and Paf(N/A)-satRNA were infected to N. benthamiana. Paf-satRNA coinfected with CMV-Fny RNAs induced very mild to sympomless on pepper plants whereas PepY-satRNA-infected pepper expressed typical chlorosis mosaic symptom. Two kinds of chimeric mutants, Paf(H/N)-satRNA and PepY(N/A)-satRNA, induced mild mosaic or symptomless infection onto pepper plants, while PepY(H/N)-satRNA and Paf(N/A)-satRNA showed typical chlorosis and mosaic symptom with stunting. This results suggest that mild symptom-related domain for the Paf-satRNA was located on HpaI-NarI region.
