Effects of plant-derived natural products on inflammatory bone destructive disease

  • Rheumatoid arthritis, osteoarthritis, and periodontal disease are bone destructive diseases mainly caused by inflammation. Various studies are being conducted to develop treatments for inflammatory bone destructive diseases. Many of these studies involve plant-derived natural compounds. In these studies, cell differentiation, signal transduction pathways, and bone resorption were measured at the cellular level. In disease-induced animal models, the amount of inflammatory mediators or matrix destructive enzymes and serum metabolic markers were measured. This study examined the effects of plant-derived natural compounds, such as flavonoids, on inflammatory bone destructive diseases. In addition, we structurally classified various substances used to maintain bone health and summarized the biological effects and related mechanisms of the components.

Enhancing Resistance of Red Pepper to Phytophthora Blight Diseases by Seed Treatment with Plant Growth Promoting Rhizobacteria

  • Plant growth promoting rhizobacteria (PGPR) have been shown to suppress phytopthora blight. This suppression has been related to both microbial antagonism and induced resistance. The PGPR isolates were screened by dual culture plate method and most of the isolates were showed varyinglevels of antagonism. Among the PGPR isolates pyoverdin, pyochelin and salicylic acid producing strains showed the maximum inhibition of mycelial growth of Phytophthora capsici and increased plant growth promotion in red pepper. PGPR isolatesfurther analysed for its ability to induce production of defence related enzymes and chemicals. The activities such as Phenyle alanin ammonia lyase (PAL), Peroxidase (PO), Polyphenol oxidase (PPO) and accumulation of phenolics were observed in PGPR pretreated red pepper plants challenged with Phytophthora capsici. The present study shows that an addition of direct antagonism and plant growth promotion, induction of defense related enzymes involved to enhance resistance against invasion of P. capsici in red pepper.

Development of an Analytical Approach for the Utilization of Edible Tree Sprouts

  • This study evaluated the general nutritional ingredients such as crude fats, crude ashes, crude proteins, total polyphenols, and total flavonoids in 18 kinds of edible tree sprouts. The tree sprouts of Philadelphus schrenckii, Lycium chinense, and Morus alba had the highest crude fat, crude ash, and crude protein content, respectively. The tree sprouts of Cedrela sinensis (CSS) with high ABTS+ radical scavenging activities had a high content of total polyphenols (175.65 mg/g ext.) and total flavonoids (75.18 mg/g ext.). The simultaneous determination of flavonoids such as rutin, isoquercitrin, quercitrin, afzelin, and quercetin in CSS was conducted using high-performance liquid chromatography with a wavelength of 270 nm. Among the flavonoids, the content of quercitrin in CSS was the highest at 59.28 mg/g ext. This study also aids the quality control of many edible tree sprouts by analyzing the general components, total polyphenols, and total flavonoids.

Anti-inflammatory Effect of Natural Plant Extracts on in vitro Rumen Fermentation and Methane Emission (천연 식물 추출물의 항염 효과가 in vitro 반추위 발효성상과 메탄 생성에 미치는 영향)

  • This study was conducted to investigate the effects of anti-inflammatory plant extracts on the in vitro rumen fermentation characteristics and methane emission. Anti-inflammatory plant extracts from Morus bombycis Koidz, Mallotus japonicus L., Morus alba L., Paulownia coreana Uyeki, Isodon japonicus Hara and Ginkgo biloba L. were used in the study. The ruminal fluid(5 mL), McDougall buffer(10 mL), timothy as a substrate(0.3 g) and each anti-inflammatory plant extract(5% of substrate) were dispensed anaerobically into 50mL serum bottle. The mixtures were incubated for 3, 9, 12, 24, 48 and 72h at $39^{\circ}C$ without shaking. Supplementation of the anti-inflammatory plant extracts did not effects characteristics(pH, digestibility of dry matter, glucose concentration, ammonia concentration, protein concentration, VFA) on rumen fermentation. Total gas was showed a different pattern depending on treatments. Carbon dioxide was significantly(p<0.05) higher in Morus alba and Isodon japonicus than in control at 48h. Methane was significantly(p<0.05) lower in treatment than in control at initial fermentation. However the more incubation time was increased, the more methane emission was higher in treatment than in control. The concentrations of polyphenol and flavonoid were higher in Ginkgo biloba. In conclusion, supplementation of the anti-inflammatory plant extracts did not effect on rumen fermentation and methane emission was decreased in initial fermentation.

Antioxidative Activities of Water Extracts from Different Parts of Taraxacum officinale (민들레의 부위별 열수 추출물의 항산화 활성)

  • This study was conducted to examine antioxidative and physiological activities of water extracts from different parts (flower, leaf, root, and the whole plant) of Taraxacum officinale. The water extracts from different parts were measured to obtain total flavonoids content, total polyphenol content, electron donating ability, SOD-like activity, nitrite-scavenging ability and tyrosinase inhibition effects. Total flavonoids and total polyphenol contents in flower extract were 32.91 mg/g and 49.31 mg/g, respectively, which were much higher than those of any other parts. The electron donating abilities of flower, leaf, the whole plant, and root extracts were 87.07%, 87.66%, 81.06% and 66.20%, respectively at a concentration of 1 mg/mL. The activities increased in a dose-dependent manner. The SOD-like activity of water extracts from different parts showed 9.07~10.97% at a concentration of 1 mg/mL. The nitrite-scavenging abilities of flower and leaf extracts measured at pH 1.2 were 36.34% and 38.16%, respectively at a concentration of 1 mg/mL. Tyrosinase inhibition activity of the leaf extract at a concentration of 1 mg/mL was the highest (34.19%) and that of the whole plant and root extracts was shown to be more than 20%. These results suggest that water extracts from different parts of Taraxacum officinale could be used as an antioxidative functional food source.

Effects of Sclerophyllous Plant Leaves Addition on Fermentative and Sensory Characteristics of Kimchi (경엽식물 잎의 첨가가 김치의 발효 및 관능특성에 미치는 효과)

  • The effects of persimmon, mulberry and bamboo leaves addition on the fermentative and sensory characteristics of kimchi were investigated. Total polyphenol content, DPPH radical scavenging activity and lactic acid bacteria growth inhibition of EtOH extract from persimmon leaves were significantly higher than those from mulberry and bamboo leaves. From the sensory evaluation of kimchi added with three chopped plant leaves, kimchi added with chopped persimmon leaves gave the highest point in color, flavor, texture and overall preference significantly (p<0.05). When the chopped persimmon leaves were added at a ratio of 0.3% based on cabbage weight, the changes of pH and total acidity (TA) during the storage for 14 days were significantly lowered, compared to general kimchi (p<0.05). Total polyphenol content and DPPH radical scavenging activity of kimchi supplemented with chopped persimmon leaves at 0.3% also significantly increased after storage for 14 days (p<0.05), and this kimchi provided higher total polyphenol content and DPPH radical scavenging activity than general kimchi. These results suggest that the addition of perisimmon leaves have significant influences on the fermentation and sensory characteristics of kimchi.

Comparative Evaluation of Biological Activities and Active Compounds of Some Invasive Alien Plants (주요 생태계 교란 외래식물의 생리활성 비교 평가 및 유효성분)

  • To evaluate functional biomaterials of 5 invasive alien plants, total polyphenol and flavonoid contents, antioxidant activity, anti-inflammatory activity, and antibacterial effect were measured. The total polyphenol and flavonoid contents of the extracts were in the order of Rumex acetosella L. > Hypochaeris radicata L. ≥ Lactuca scariola L. > Humulus japonicus Siebold & Zucc. ≥ Solanum viarum Dunal. The DPPH and ABTS radical scavenging activities of the extract were the highest in R. acetosella and correlated well with the total polyphenol contents. In RAW 264.7 cells stimulated with lipopolysaccharide (LPS), nitric oxide (NO) and prostaglandin E2 (PGE2) production inhibitory effect of the extracts (100 ㎍ SE/mL) were 20~60% and 10~70%, respectively, showing the highest inhibitory effect in R. acetocella. The extracts of R. acetosella, H. japonicus and S. viarum showed antibacterial activity against food poisoning-causing microorganisms such as Bacillus subtilis, Escherichia coli, Vibrio parahaemolyticus and Vibrio vulnificus. Furthermore, the H. japonicus extract was found to have effective antibacterial activity against oral microorganisms such as Enterococcus faecalis, Lacticaseibacillus casei, Rothia dentocariosa, Staphylococcus epidermidis and Streptococcus mutans, and its major active ingredients were predicted to be pentadecylic acid, palmitic acid and clionasterol. These results suggest that alien plants have potential as biomaterials with antioxidant, anti-inflammation and antibacterial effects.

Antioxidant Polyphenol Glycosides from the Plant Draba nemorosa

  • Bioassay-directed fractionation of Draba nemorosa led to the isolation of two new phenolic glycosides, 1-O- (sinapoyl)-glucitol (2) and 1,3-disinapoylgentiobiose (5) along with five known phenolic glycosides (1, 3, 4, 6, and 7). Their structures were characterized based on spectroscopic methods (2D NMR, HRTOFMS, IR, and UV). The isolated compounds showed antioxidant activities (IC50) in the range of 14-98 mM which were estimated by DPPH radical-scavenging assay.

Antioxidant Effect of 80% Ethanol Extracts Obtained from Three Dendranthema Species (국화과 Dendranthema속 식물 3종 80% 에탄올 추출물의 항산화 효과)

  • Flowers and shoots of three species of Dendranthema boreale, Dendranthema indicum, Dendranthema zawdskii var. lucidum, were extracted with 80% ethanol by reflux, and polyphenol content, scavenging activity on DPPH and ABTS radicals, ferrous ion chelating effects and inhibition effects on lipid peroxidation were analyzed. Total polyphenol and flavonoid contents were highest in D. zawdskii var. lucidum, especially in the flower part. Scavenging activity on DPPH and ABTS was also highest in D. zawdskii var. lucidum flower with less activity in shoot. Ferrous ion chelating effects was highest with D. boreale flower and lowest in D. zawdskii var. lucidum flower. Inhibition activity on lipid peroxidation was highest in D. zawdskii var. lucidum shoot with 41.01% inhibition activity showing 32 days after reaction, which is higher than synthetic antioxidant BHT. Due to higher antioxidant level and activity of shoot and flower of D. zawdskii var. lucidum is promising material for natural plant antioxidant. It was also shown that antioxidant activity is different according to plant part ever in same plant, and proper plant species should be used for antioxidant after careful studies.

Mycelial growth and antioxidant effects of Pleurotus ostreatus in medium supplemented with Aronia berries (아로니아 첨가 배지 내 느타리(Pleurotus ostreatus) 균사체 생장 및 항산화 효과)

  • This study was conducted to investigate the growth and antioxidant activity of Pleurotus ostreatus mycelia grown in medium supplemented with Aronia. The diameter and dry weight of the mycelia were increased in the medium supplemented with Aronia compared with the untreated medium. The total polyphenol content of mycelia grown in medium supplemented with Aronia and untreated medium was 6.54 mg GAE/g and 5.77 mg GAE/g, respectively. The 2,2-diphenyl-1-picrylhydrazyl radical scavenging activity of mycelia grown in medium supplemented with Aronia and untreated medium was 61.81% and 49.65%, respectively. Moreover, the 2,2'-azinobis-(3-ethylbenzothiazoline-6-sulfonic acid) radical scavenging activity of mycelia grown in medium supplemented with Aronia and untreated medium was 59.83% and 52.66%, respectively. These results confirmed that P. ostreatus mycelial growth and antioxidant activity were increased when Aronia was added to the culture medium.
