Inhibition Activity of Plants on IgE-mediated Degranulation of RBL-2H3 Cells

  • In this study, the inhibitory activities of fifty plant extracts on IgE-mediated degranulation in the rat basophilic leukemia cell line (RBL-2H3 cells) were measured; the release of interleukin (IL)-4 and β-hexosaminidase from IgE-sensitized cells treated with the plant extracts was measured; and the effects of the plant extracts on cell viability were tested. The results of the analysis of plant extracts at 20 μg/ml, including the aerial part of Magnolia sieboldii K. Koch, exhibited suppressive activities upon the release of IL-4. Furthermore, several plant extracts including methanol extracted from Lindera erythrocarpa Makino (aerial part) at the same concentration significantly inhibited the release of β-hexosaminidase. Twenty-six of the plant extracts, including methanol extract of Weigela subsessilis (Nakai) L. H. Bailey (branch), showed a cell proliferation effect of over 80% at 100 μg/ml. In conclusion, the results suggest that the leaf/stem of Geum japonicum Thunb. and the stamen/ovary of Nelumbo nucifera Gaertn., which exhibited effective inhibition on β-hexosaminidase release and IL-4 release from mast cells and showed high cell viability, could be useful candidates as anti-allergy materials.

Plant Regeneration from Zygotic Embryo-Derived Callus in Citrus junos SIEB. (유자(Citrus junos SIEB.)의 접합배로부터 캘러스 유도 및 식물체 재분화)

  • Calli were successfully induced from immature embryos of Citrus junos SIEB. cultured on 1/2 MS medium supplemented with 40.4 BA. Plant were regenerated from immature embryo derived callus on MS medium with 5 $\mu$M BA. The calli were morphologically characterized by two types: one was whitish and the other was yellowish. After 16 weeks of culture, shoots and root were formed on calli. Plantlets were transplanted to soil and successfully grown to a whole plant Also, the arrangement of the cells showed many differences according to developmental stages of callus and organogenesis. The small cells were compact in callus cultured for 6 weeks and the extended cells which divided actively appeared in it after 8 weeks of culture. The globular protrusion of compacted cells occurred in callus after 10 weeks of culture, and the neighboring cells were liquefied. Oil sac surrounded by the liquefied cell was observed in the leaf and was formed by rupture of liquefied cells.

Roles of Plant Extracts and Constituents in Cervical Cancer Therapy

  • Cervical cancer is a major health problem worldwide and is the most frequent cause of cancer in women in India. Early detection and affordable drugs with clinical efficacy have to go hand-in-hand in order to comprehensibly address this serious health challenge. Plant-based drugs with potent anticancer effects should add to the efforts to find a cheap drug with limited clinical side effects. Keeping this very purpose in mind, an attempt has been made in this review to explore the potential of plant extracts or constituents known to exhibit antitumorigenic activity or exert cytotoxic effect in human cervical carcinoma cells. Alkaloids such as those isolated from C. vincetoxicum and T. Tanakae, naucleaorals A and B, isolated from the roots of N. orientalis, (6aR)-normecambroline, isolated from the bark of N. dealbata appear promising in different human cervical carcinoma cells with the $IC_{50}$ of 4.0-8 ${\mu}g/mL$. However, other compounds such as rhinacanthone and neolignans isolated from different plants are not far behind and kill cervical cancer cells at a very low concentrations. Among plant extracts or its constituents that enhance the effect of known anticancer drugs, noni, derived from the plant M. citrifolia perhaps is the best candidate. The cytotoxic potency and apoptotic index of cisplatin was found to significantly enhanced in combination with noni in different human cervical carcinoma cells and it therefore holds significance as promising herbal-based anticancer agent. However, efficacy needs to be further investigated in various cervical cell lines and more importantly, in in vivo cervical cancer models for possible use as an alternative and safe anticancer drug.

Direct somatic embryogenesis, plant regeneration and genetic transformation of Panax ginseng

  • Somatic embryogendesis is one of good examples of the basic research for plant embryo development as well as an important technique for plant biotechnology. This paper describes the direct somatic embryogenesis from zygotic embryos of Panax ginseng is reversely related to normal axis growth of zygotic embryos by the experiment of various chemical treatments. Under the normal growth condition, the apical tips of embryo axis produced an agar-diffusible substance, which suppressed somatic embryo development from cotyledons. Although the cells of zygotic embryos were released from the restraint of embryo axis, various factors were still involved for somatic embryo development. Electron microscopic observation revealed that the ultrastructure of cells of cotyledon epidermis markedly changed before initiation of embryonic cell division, probably indicating reprogramming events into the cells embryogenically determined state. Polar accumulation of endogenous auxin or cell-cell isolation by plasmolysis pre-treatment is the strong inducer for the somatic embryo development. The cells for the process of somatic embryogenesis might be determined by the physiological conditions fo explants and medium compositions. Direct somatic embryos from cotyledons fo ginseng were originated eithrer from single or multiple cells. The different cellular origin of somatic embryos was originated either from single or multiple cell. The different cellular origin of somatic embryos was depended on various developmental stages of cotyledons. Immature meristematic cotyledons produced multiple cell-derived somatic embryos, which developed into multiple embryos. While fully mature cotyledons produced single cell-derived single embryos with independent state. Plasmolysis pretreatment of cotyledons strongly enhanced single cell-derived somatic embryogenesis. Single embryos were converted into normal plantlets with shoot and roots, while multiple embryos were converted into only multiple shoots. GA3 or a chilling treatment was prerequisite for germination and plant conversion. Low concentration of ammonium ion in medium was necessary for balanced growth of root and shoot of plantlets. Therefore, using above procedures, successful plant regeneration of ginseng was accomplished through direct single embryogenesis, which makes it possible to produce genetically transformed ginseng efficently.

Relationship between ganglioside expression and anti-cancer effects of a plant-derived antibody in breast cancer cells

  • Production of therapeutic monoclonal antibodies (mAbs) using a plant platform has been considered an alternative to the mammalian cell-based production system. A plant-derived mAb CO17-1AK ($mAb^P$ COK) can specifically bind to various types of cancer cell lines. The target protein of $mAb^P$ COK is the epithelial cell adhesion molecule (EpCAM) highly expressed in human epithelial cancer cells, including breast and colorectal cancer cells. It has been hypothesized that its overexpression supports tumor growth and metastasis. A ganglioside is extended well beyond the surfaces of the various cell membranes and has roles in cell growth, inflammation, differentiation, and carcinogenesis. However, the regulation of EpCAM gene expression in breast cancers and the role of gangliosides in oncogenesis are unclear. Here, the purpose of this study was to determine the effects of $mAb^P$ COK on human breast cancer cell proliferation, apoptosis, and ganglioside expression patterns. Our results show that treatment with $mAb^P$ COK suppressed the growth of breast cancer cells and induced apoptotic cell death. It also upregulated the expression of metastasis-related gangliosides in breast cancer cells. Thus, treatment with $mAb^P$ COK may have chemo-preventive therapeutic effects against human breast cancer.

Cytotoxic Activities of Panax ginseng and Euphorbia humifusa in Human Brain Tumor Cells (인삼 비당부와 땅빈대의 뇌암세포 독성작용)

  • The effects of acid hydrolysis product of Panax ginseng and MeOH extract of Euphorbia humifusa on the growth of human brain tumor cells were evaluated using U-373 MG human astrocytoma and SK-N-MC human neuroblastoma cells as model cellular systems. These plant extracts induced cytotoxicity in both cells in a dose-dependent manner. These cytotoxic effects were significantly inhibited by GSH, an antioxidant, in both cells. BAPTA/AM, an intracellular $Ca^{2+}$ chelator, significantly blocked the cytotoxic effects of these extracts in U-373 cells, but enhanced these effects in SK-N-MC cells. These results suggest that the plant extracts may be a valuable choice for the studies on the treatment of human brain tumors.

  • The role of S-adenosylmethionine (SAM) as an intermediate in interrelation between polyamine and ethylene biosynthesis was studied in suspension cultures of Nicotiana tabacum L. Exogenous SAM stimulated the polyamine and ethylene biosynthesis in 4 day-cultured cells, which were in active cell divisions, and 10 day cultured cells, which went on with active cell elongation and senescence. SAM-induced ethylene production was more effective in 10 day-cultured cells than in 4 day-cultured cells, but SAM-induced polyamine biosynthesis was more effective in 4 day-cultured cells than in 10 day-cultured cells. Polyamine contents were increased by the blockage of ethylene biosynthetic pathway in the conversion of SAM to ethylene via 1-aminocyclopropane-1-carboxylinc acid (ACC) with aminooxyacetic acid (AOA). Also, ethylene production was increased by the inhibitors of polyamine biosynthesis such as methylglyoxal bis-(guanylhydrazone) (MGBG), dicyclohexylamine (DCHA), $\alpha$-difluoromethylarginine (DFMA) and $\alpha$-difluoromethylorinithine (DFMO). These results suggest that there may be interrelations between polyamine and ethylene biosynthesis for the competition of SAM and the inherent mechanism of switch on-off in polyamine and ethylene biosynthetic activity with the progress of cell growth and senescence.

  • The origin of cambial initials from procambium was studied in the seedling root of Acer saccharinum. In transverse view, the first periclinal divisions of procambial cells occurred just outside of each early metaxylem and resulted in meristematic strips. As root development progressed, the division activities appeared subsequently outside of each late metaxylem and then in pericycle cells opposite the four protoxylem poles. Eventually, such meristematic strips were connected completely each other. Thus, a nearly rectangular shaped meristematic layer in outline was formed outside the xylem in a whole root transectioned. In tangential section, early procambium showed a homogeneous structure consisted of uniform short cells with transverse end walls. However, some of the procambial cells did elongate, whereas others divided transversely. The former become more elongate, tapered, and vacuolated. Finally, they differentiated into fusiform initials. Short cells consisting axial strands divided continuousely in transverse plane and became ray initials, while some short cells elongated and transformed into long cells. The early ontogeny of vascular cambium in Acer saccharinum root was interpreted to be established by a gradual process.

The Fine Structure of Callus Derived from Panax ginseng Leaves (인삼의 잎에서 유기된 Callus의 미세구조)

  • The fine structure of the callus induced from epidermis of Panax ginseng leaves cultured on Murashige & Skoog medium plus kinetin 0.1mg/l, NAA 0.2mg/l and 2.4-D 0.5mg/l was observed. The cells composing callus tissue are mononucleus. Three types of cells were identified; cells with abundant cytoplasm, cells with relatively differentiated vacuoles and with numerous starch grains in the plastids and ones with highly differentiated vacuoles and with unsaturated lipid granules. Prolamellar body, plastid lamellae, plastid globules, stromacenter, fine tubules, crystal-containing body and DNA-like structures were observed in the stroma of the plastids. The chromoplasts were identified in some cells believed as the mother cells of secretory cells in secretory ducts. Curved or straight micro-fibrils of 100~150A in diameter were observed in the cytoplasm. And the characteristics of cell organelles and cell inclusions and the vacuole formation in callus tissues were discussed.

Origin and Development of Fascicular Cambium in Hypocotyl of Eucommia ulmoides Seedlings (두충나무 유식물의 하배축에서의 유관속내 형성층의 기원 및 발생)

  • The early ontogeny of the vascular cambium in the hypocotyl of Eucommia ulmoides seedling was studied anatomically. In transverse view, the procambial cells repeated periclinal divisions and then gradually differentiated into vascular cambium; however, in the transectional plane, it was difficult to reveal the origin of cambial initials. In tangential view, the early procambial cells repeated periclinal divisions and then the procambium consisted of homogeneous short cells. Some cells in late procambium did not show transverse divisions, but elongated continuously. Thus, the procambium showed a heterogeneous structure: The long cells became fusiform initials with tapering end walls and the short cells differentiated into ray initials. Conclusively, the two types of cambial initials were gradually originated from the homogeneous cells at the early procambial stage, and they consisted vascular cambium.
