Characterization of a new variety 'Chung' by intra-specific hyphal anastomosis in Pleurotus ostreatus (계통간 교잡에 의한 느타리 품종 '청'의 육성 및 그 특성)

  • The Oyster mushroom is saprophytic fungus. The pileus is stemmed at the side and later depressed. It grows to 5-15 cm, and is of grey, grey-lilac, blackish-grey, steel grey, grey-brown, and blue-blackish. Various kinds of Oyster mushrooms such as golden, pink, brown, grey, white, and blue make marketing an interesting challenge depending upon the market niche. A new commercial strain "Chung" of oyster mushroom was developed by hyphal anastomosis. It was improved with hybridization between monokaryotic strain derived from Pleurotus ostreatus ASI 2194 and ASI 2487. The pileus of parental strain ASI 2194 and ASI 2487 was grey and light blue-grey, respectively. Most of intra-specific hybrids between strain ASI 2194 and ASI 2487 were showed similar pileus color of parents. By the way, the pileus color of variety 'Chung' was blue to bluish grey. The optimum temperatures of mycelial growth and fruiting body development was $25{\sim}30^{\circ}C$ and $12{\sim}16^{\circ}C$, respectively.

Characterization and breeding of a new cultivar Pleurotus ostreatus 'Heuksol' (느타리버섯 신품종 '흑솔'의 육성 및 특성)

  • Oyster mushroom is a type of mushroom that is commonly cultivated and consumed in Korea. P. ostreatus 'Suhan' is a preferred cultivar for many mushroom farmers because it has a dark pileus and thick stipe. However, as it is very sensitive to environmental conditions, farmers consistently demand an alternative cultivar. To develop a new cultivar, the parental strains KMCC01680 ('Suhan') and KMCC00478 ('Gosol') were selected from various collected P. ostreatus strains by cultivating genetic resources. P. ostreatus 'Heuksol' was developed by the method of Mon-Mon crossing between monokaryotic strains derived from 'Suhan' and 'Gosol'. Thirty strains of 174 crossed strains were initially selected by cultivation experiments. After bulk cultivation tests, 'Heuksol' was selected. The nuclear DNA profile of 'Heuksol' was similar to those of the parental strains, 'Suhan' and 'Gosol', when RAPD (random amplified polymorphic DNA) primers and UPF (Universal PCR Fingerprinting) 2, 3, and 4 were used. The optimum temperature for mycelial growth was $30^{\circ}C$ for 'Heuksol', but medium-high temperatures were also appropriate, especially $13-20^{\circ}C$. The fruiting body production per bottle (1,100 mL) was approximately 140.8 g. When compared to the control strain 'Suhan', the thickness of the stipe of 'Heuksol' was greater than that of 'Suhan' (13.5 mm vs 9.4 mm). The pileus diameter of 'Heuksol' was similar to that of 'Suhan' and the pileus thickness of 'Heuksol' and 'Suhan' was 19.7 mm and 21.8 mm, respectively. 'Heuksol' had more a productive stipe number than 'Suhan' and the pileus of 'Heuksol' was dark gray, even at high temperatures. Therefore, it was suggested that this new cultivar, 'Heuksol', could provide an alternative to 'Suhan' and contribute to the profit of oyster mushroom farms.

Growth Characteristics of Oyster Mushroom Upon Replacement of Cotton Seed Meal with Rapeseed Meal (면실박 대체 채종박 첨가에 따른 느타리 생육 특성)

  • This study was conducted to investigate the optimum supplementation ratio when replacing cotton seed meal with rapeseed meal for cultivating oyster mushrooms. The chemical properties among the treatments were pH 4.5~4.7, total carbon content was 46.3~46.5%, total nitrogen content was 1.6~1.7%, and carbon-nitrogen ratio was 27.0~27.8. These did not differ significantly from the control. Mycelial growth was 85.1 mm (Heuktari), and 72.8 mm (Suhan-1Ho) in medium containing 5% rapeseed meal, with no significant difference in mycelial density between cultivars. Fruiting body characteristics are as follows: In 'Heuktari', the color of the pileus was darker than control with an L value of 32.5, and pileus thickness and diameter were 3.2 mm and 27.6 mm, respectively. Stipe length and diameter were 86.0 mm and 9.1 mm, respectively. Valid stipe number per bottle (1,100 cc) was 25.9, and a yield of 189.7 g/1,100 cc was recorded in mixed medium containing 5% rapeseed meal (higher than control). In mixed medium containing 10% rapeseed meal, the yield was 184.5 g/1,100 cc (comparable to control). In 'Suhan-1Ho', supplemented with 5% rapeseed meal, pileus color was darker than control color, with an L value of 28.8. Pileus thickness was 4.5 mm. Stipe diameter was 12.3 mm (higher than control), and yield was 145.5 g/1,100 cc (comparable to control).

  • Drying of Shiitake mushroom was investigated to see the effect of temperature, relative humidity of drying air and diameter of the pileus on its rates. The drying rate was increased with the increase of the air temperature and the decrease of the relative humidity. The external color was dark brown at higher drying temperature and higher relative humidity. Exponential and Thompson nodel were found to describe well the raying process of the Shiitake mushroom.

  • Differential display of reverse transcription(DDRT)-PCR was conducted to have a profile of the differentially expressed genes during the formation of fruiting body of Lentinus edodes. The lines of L. edodes(ImHyup-1) employed were cultivated in the artificial blocks of sawdust, and the fruiting body was induced from the mycelia or the mass protruded from the brown surface of the sawdust blocks. RNAs were prepared from the four different developmental stages; mycelial, primordial, and stipes and pileus of fruiting body. The fragments of cDNA were synthesized from the combinations of the arbitrary primers and 3' one anchored Oligo-dT primer. Twelve combinations using the primers have been tested, and among them nineteen bands were identified as differentially expressed. Those genes were further analyzed by DNA sequencing and followed by homology search. Characterization of one clone was conducted as a preliminary data and more are under investigation.

Cultural characteristics and consumer acceptance of Pleurotus pulmonarius (산느타리버섯 재배특성 및 기호도 조사)

  • Pleurotus pulmonarius usually grows on the dead wood of deciduous trees in mid summer to early fall. The pileus color is white to tan and fruiting body is generally smaller than P. ostreatus. This study was carried out to investigate the characteristics of cultivation and consumer acceptance of P. pulmonarius. The period of incubation and primordial occurrence were required about 33days, 5days in bottle cultivation repectively, and the yield was 170g/850cc bottle. In bed cultivation, the period of primordial occurrence was similar to that of bottle cultivation and the yields was $35.7{\sim}46.2kg/3.3m^2$. Taste and favor of P. pulmonarius were superior to P. oostreatus., but the appearance, color and shape, was slightly inferior to P. ostreatus.

  • Two new species of Fulvifomes are described from specimens collected in rainforests of Nonggang Nature Reserve of southern China, based on morphological characteristics and molecular phylogenetic analysis of the internal transcribed spacer (ITS) and nuclear large subunit ribosomal DNA (nLSU) sequences. Fulvifomes nonggangensis sp. nov. is characterized by perennial, sessile and solitary basidiocarps, applanate pileus, small cystidioles of 9.9-15.4 × 2.9-3.5 ㎛, large pores of 5-6 per mm, a dimitic hyphal system, and broadly ellipsoid basidiospores of 4.3-5.3 × 3.3-4.2 ㎛. F. tubogeneratus sp. nov. is characterized by perennial, sessile, and imbricate basidiocarps, a duplex context, small pores of 7-8 per mm, a dimitic hyphal system, and ovoid to subglobose basidiospores of 5.72 × 5.00 ㎛.

  • Amylosporus sulcatus sp. nov. is described from Nonggang Nature Reserve, southern China, on the basis of morphological and molecular data. The morphological description and illustrations for the new species are provided. The species is characterized by pileate and stipitate basidiocarps. The pileus surface is obviously concentrically and radiately sulcate and tomentum, and the pore surface is snow white. Phylogenetic analyses based on sequences of the internal transcribed spacer and nuclear large subunit ribosomal DNA confirmed it to be a new species.

Changes in the Levels of Ergosterol and Methionine as Indicators of Lentinula edodes Quality According to the Relative Humidity During the Storage Period

  • Lentinula edodes mushrooms cultivated under different relative humidities were wrapped at 4℃ and the results of storage characteristics were investigated. Changes in water content of fruiting bodies during the storage period showed the highest water content in fruit bodies harvested from the treatment with the highest relative humidity. The luminosity of the fresh fruiting bodies showed no significant change during the storage period, and the redness was higher in the relative humidity 95% treatment than in the other treatments. According to this study, the relative humidity of the pileus was 65%, and the content of Ergosterol was 0.67 ± 15 g / L at relative humidity of 65%, 80% and 95%. In addition, amino acid analysis and Principal Component Analysis (PCA) confirmed that methionine was the main cause of changes in storage time and relative humidity.

Effect of "Green-one" organic nutrient on mycelial growth and fruiting of Pleurotus ostreatus (유기농 자재 『그린원』이 느타리버섯(Pleurotus ostreatus)의 균사생장 및 자실체 생육에 미치는 영향)

  • The objective of this study was to determine the effects of "Green-one" organic nutrient on mycelial growth and fruiting of P. ostreatus. The dilution concentrations of "Green-one" was treated as follows. There was control, 100, 200, 400 concentrations. That treatments were treated with step of each mycelial growth step. The best of growth steps was mycelial scratching step. At that time, DPI(Day required for primordial formation after inoculation) was shortened for 1 day. Valid germination stipe are 15 pieces, 3 pieces more than control. Stipe length and stipe diameter was long each 4mm, 3mm more than control. Pileus size is shortened than control. Yields per one bottle(g/850cc) was 146g increased 6.5% than control 137g/850cc.
