Variation in Photosynthesis and Leaf Pigments of Susceptible Pinus densiflora and Resistant Pinus rigida Following Pine Gall Midge Attack (솔잎혹파리 침해(侵害)에 따른 감수성수종(感受性樹種)인 소나무와 저항성수종(抵抗性樹種)인 리기다소나무에 있어서 광합성(光合成) 및 엽(葉)의 색소변이(色素變異)에 관한 연구(硏究))

  • Susceptible trees of Pinus densiflora and resistant trees of Pinus rigida following pine gall midge (Tnecodiplosis japonensis Uchida et Inouye) attack were seasonally compared to examine the variation in needle growth and photosynthetic ability, respiration rate, chlorophyll contents, carotenoid and anthocyanin contents. Also, carotenoid and anthocyanin contents of larvae both from soil and from galled tissue were compared during March and September, respectively. The plantation damaged severely by this insect consisted mostly of 10-to 15-year old P. rigida and P. densiflora. The results obtained in this study were as follows: 1) The length of the infested needles of P. densiflora decreased by 48.1 percent compared with the normal needles, while that of P. rigida did 37.4 percent. 2) All of P. densiflora and P. rigida showed higher photosynthetic ability in normal needles than in infested needles. The maximum photosynthetic ability of P. densiflora was shown in mid-August, while that of P. rigida in mid-October. In contrast to that, respiration rate of infested needles was higher than that of normal needles in both species. The respiration rate of P. rigida was higher than that of P. densiflora. 3) P. rigida had higher total chlorophyll contents than P. densiflora. The total carotenoid contents tents in infested needles were higher than those in normal needles of both species. 4) Total carotenoid contents were generally higher in P. rigida than in P. densiflora during the growing season. The total carotenoid content (0.094mg/g) in larvae from soil was similar to that (0.092mg/g) in larvae from galled tissues. 5) Infested needles of both species showed higher anthocyanin contents than normal needles. Higher anthocyanin contents in galled needles were due primarily to its active formation stimulated by larval attack. Thus, reddish-brown coloration occurred only in galled needles of P. densiflora.

Seasonal Variations of Physico-Chemical Characteristics and Phytoplanktonic Pigments in Coastal Sea Water from Ullungdo and Dokdo Islands (울릉도와 독도 근해해역의 이ㆍ화학적 특성과 광합성색소의 계절적 변화)

  • The physico-chemical characteristics and the concentrations of chlorophylls of coastal seawater were investigated to know the seasonal variations of biological oceanographic environments in the Islands of Ullungdo(U) and Dokdo(D). The samplings of sea water according to different depths were performed four seasons (May, June, August and November) in five stations along the coast of Ullungdo Island and 3 times (June, August and November) in three stations around the coast of Dokdo Island. The seasonal variations of sea water temperature showed that the formation of thermocline in August was distinct in comparison to the other seasons. The sea water in the surface was influenced by low temperature-high salinity in May and with high temperature-low salinity in the investigated area. The amount of seston was high in May (5.3-15.0mg/l) and was low in August (1.4-4.9mg/l) in ullungdo island. for the nutrients or sea water in Ullungdo Island, the concentrations of nitrate and ammonium were higher than Dokdo Island (nitrate-max. of U in August : 0.10-11.50$\mu\textrm{g}$/1, max. of D in August : 2.92-8.10$\mu\textrm{g}$/l : ammonium-max. of U in November : 14.18-20.69$\mu\textrm{g}$/l, max. of D in June : 0-1.78 $\mu\textrm{g}$/l). The high concen-tration of chlorophylls showed on the deeper layer from 30 m to 50 m in August (U 30 m : 0.85$\mu\textrm{g}$/l ; D 50m : 1.02 $\mu\textrm{g}$/l), while the concentrations of chlorophylls were even in May, June and November in the deeper layer of surface layer. In conclusion, the establishment of thermocline in deeper area of the euphotic layer in August was a trigger far the development of phytoplankton, while the complex physico-chemical system by diverse currents and vertical mixing of sea water in the area induced the even distribution of phytoplankton in both epilimnion and hypolimnion in May, June and November.

A Comprehensive Study on the Forced Aging of Flue-cured Tobacco-Leaves (황색종 잎담배의 발효숙성 촉진에 관한 종합적 연구)

  • The process of the forced aging of flue-cured tobacco leaves were studied extensively from various scientific points of view. The Flue-cured tobacco leaves were inoculated and fermented with nicotine resistant Hansenula yeast, or the leaves were subjected under simple forced aging. The above two processes of forced aging were studied from the summarized points of microbiology, physics, chemistry, and biochemistry, and the resulted products ware compared in their physical, chemical and biochemical quality determining factors with that of raw material tobacco leaves (dried-tobacco leaves) and 2 years aged high quality tobacco leaves. The summary results were as follows. 1) The Korean flue-cured tobacco leaves, were forcedly aged under the basic optimum aging condition, temperature $40^{\circ}C$, moisture contents 18%, relative humidity 74%. It was found that this aging condition was the best in bringing the quality of forcedly aged tobacco leaves to the utmost state. 2) Under this optimum temperature and moisture condition of forced aging in about 20 days the forcedly aged tobacco leaves both with yeast inoculation and without yeast inoculation showed the equivalent tobacco qualities comparable with that of more than 2 years aged tobacco leaves. 3) The forcedly aged tobacco leaves both with and without yeast inoculation under $40^{\circ}C$ temperature and $74^{\circ}C$ relative humidity achieved the necessary quality determining physical and chemical changes in about 20 days. 4) The microbial changes during the forced aging were as follows. The population of yeasts and bacteria increased until to 15 days of aging, then decreased thereafter. Whereas the molds grew continously until the end of fermentation. 5) The tobacco quality determing physico-chemico-properties of yeast inoculated aged and simple forcedly aged tobacco leaves, progressed as the follows in time. As the forced aging progresses, swelling and combustibility properties were improved. The pH, total reducing materials, total sugars, alkaloids contents decreased. The contents of organic and ether extractable materials increased. The total nitrogen, protein, crude fiber, ash contents showed no changes. The color properties, excitation purity, luminance, main wave length, showed equivalent changes comparable with that of 2 years aged tobacco leaves. 6) The changes in chemical components in yeast treated and simple forcedly aged tobacco leaves during $15{\sim}20{\;}days$ of forced aging were as follows. The following chemical components decreased as the aging. Sugars-sucrose. rhamnose, glucose. Pigments-chlorophyll, carotenes, xanthophyll and violax anthine. Polyphenols-rutin, chlorogenic and, coffeic acid. Organic acids-iso-butylic, crotonic, caprylic, galacturonic, tartaric, succinic, citric acid. Alkaloids-nicotine, nornicotine. The following components increased as the forced aging progressed. Sugars-frutose, maltose, raffinose. Amino acids-proline, cystine. Organic acids-formic, acetic, propionic, n-butyric, iso-valeric, n-valeric, malic, oxalic, malonic, ${\alpha}-ketoglutaric$, fumaric, glutaric acid. 7) During the forced aging of tobacco Leaves the oxygen-uptake decreased gradually. The enzyme activities of polyphenol oxidase, ${\beta}-amylase$ ${\alpha}-amylase$ decreased gradually. The activities of the enzymes, catalase, and invertase increased once then decreased at the later stage.

Quality Changes of a Fully Ripe Korean Native Pumpkin, Yangsan, during Long-term Storage, and High Temperature and Pressure Treatment (장기저장 및 고온고압 처리에 따른 한국재래종 호박 '양산'의 품질변화)

  • We have studied quality changes of fully ripe fruit of Korean native pumpkin 'Yangsan' regarding the following parameters: pH, sugar content, weight, water content, contents of crude protein and amino acids during 60 days storage at room temperature. As the results, there was no changes in sugar contents according to the storage period, but pH was changing to a little acidic direction with slight decrease in weight and water content. Contents of total crude proteins and comprising amino acids were increased during the storage period. The main contents of amino acids of the Korean native pumpkin, Yangsan, were glutamic acid (15.5%), aspartic acid (10.1%), lysine (8.7%), valine (7.5%), leucine (7.1%) and alanine (6.6%), which were not highly influenced during storage period. Additionally we have investigated the content of free amino acids and color changes during processing of Yangsan under high temperature at $121^{\circ}C$ and high pressure at $1\;kg/cm^2$. In fully ripe fruits, a total of 29 kinds of free amino acids were detected including 8 kinds of essential amino acids (histidine, isoleucine, leucine, lysine, phenylalanine, methionine, threonine and valine). More than 35% of total free amino acids were aspartic acid (20.3%) and asparagine (15.4%); ornithine, citrullin, and arginine, which are related to Ornithine cycle, were also detected in fully ripe fruits. But when treated with high temperature and high pressure, glutamic acid and arginine were decreased rapidly whereas ammnonium chloride was relatively increased. Moreover "b" value as yellow color indicator was decreased from 17.45 to 9.14 while treated for 60 minutes with high temperature and pressure, caused by the degradation of ${\beta}-carotene$ and other yellowish pigments in Yangsan.

Description and Application of a Marine Microalga Auxenochlorella protothecoides Isolated from Ulleung-do (울릉도 거북바위 조수웅덩이에서 분리된 해양 미세조류 옥세노클로렐라 프로토테코이드 균주의 기술 및 응용)

  • A unicellular green alga was axenically isolated from a tidal pool on Ulleung-do, Korea. Morphological, molecular, and biochemical analyses revealed that the isolate belonged to Auxenochlorella protothecoides. The current study is the first record of this species in Korea. The microalgal strain was named as A. protothecoides MM0011 and its growth, lipid and pigment compositions, and biomass properties were investigated. The strain is able to thrive in a wide range of temperatures ($5{\sim}35^{\circ}C$) and to withstand up to 1.5 M NaCl. The results of GC/MS analysis showed that the isolate was rich in nutritionally important polyunsaturated fatty acids (PUFAs). Its major fatty acids were linoleic acid (27.6%) and ${\alpha}-linolenic$ acid (39.6%). Thus, this indigenous microalga has potential as an alternative source of ${\omega}3$ and ${\omega}6$ PUFAs, which currently come from fish and plant oils. Also, the HPLC analysis revealed that the value-added antioxidant, lutein, was biosynthesized as the accessory pigments by the microalga. A proximate analysis showed that the volatile matter content was 85.6% and an ultimate analysis indicated that the gross calorific value was $20.3MJ\;kg^{-1}$. Since 40.5% of total nitrogen and 27.9% of total phosphorus were removed from the medium, respectively, it also has potential as a feedstock for biofuel applications which could be coupled to wastewater treatment. In addition, the biomass may also serve as an excellent animal feed because of its high protein content (51.4%). Therefore, A. protothecoides MM0011 shows promise for application in production of microalgae-based biochemicals and as a biomass feedstock.

Comparison of the Antioxidant Activity of Absolute Ethanol Extracts and 90% Ethanol Extracts obtained at Successive Stages of a Maillard-type Browning Reaction Mixture (Maillard 형(型) 갈색화(褐色化) 반응액(反應液)에서 얻어진 Absolute Ethyl Alcohol 과 90% Ethyl Alcohol 추출물(抽出物)의 항산화효과(抗酸化效果)의 비교(比較))

  • The color intensity (Absorbance at 490nm) and the antioxidant effects of absolute and 90% ethanol extracts obtained from a Maillard-type browning reaction mixture (0. 5M glucose and 0. 5M glycine mixture, heated at $100^{\circ}C$) were determined. The color intensity of the absolute and 90% ethanol extracts were compared with the length of reaction time and the antioxidant effects of the extracts of both types were compared one another. The results obtained are as follows. 1. The color intensity of the absolute ethanol extracts remained almost unchanged as the browning reaction proceeded. The color intensity of the 90% ethanol extracts appeared to increase nearly in proportion to the length of reaction time. 2. The absolute and the 90% ethanol extracts seemed to possess significant antioxidant activity on the autoxidation of an edible soybean oil. which was kept at $45{\pm}0.5^{\circ}C$ for 21 days. It was noteworthy that the absolute ethanol extracts showed stronger antioxidant effects than those of the 90% ethanol extracts, which contained a far greater amount of brown-colored pigments. Since the PVs of the controls in both groups, after the end of the storage period, did not differ much from one another, the possibility of residual water playing some prooxidant role in the substrates containing the 90% ethanol extracts should be ruled out. Extracts of both types obtained at earlier stages of the brownig reaction demonstrated less but comparable antioxidant activity to that of extracts taken at later stages of the reaction. 3. The results of the present study appeared to suggest that the effective antioxidant compounds, produced in the Maillard-type browning reaction, were probably intermediate products such as reductones formed at fairly earlier stages of the browning reaction.

Effect of Shading Practices on the Chemical Compounds and Antioxidant in Aruncus dioicus (차광재배가 눈개승마(Aruncus dioicus var. kamtschaticus)의 성분 및 항산화 특성에 미치는 영향)

  • This study was conducted to determine chemical compounds and antioxidant in Aruncus dioicus. Further, effects of cultural practices such as the shading conditions on the general compounds, essential oils and antioxidants. The chemical compounds and antioxidant activity of edible extracts on the shading cultivation and harvesting time were measured by crude fiber, pigments, higher fatty acids, essential oils and DPPH free radical scavenging ability and activities of SOD and POD in edible shooting parts of Aruncus dioicus. The contents of crude fiber, total chlorophyll and total carotenoids in extracts of edible shooting by shading cultivation and later harvesting were higher than those of non-shading cultivation and early harvesting. Phenolic compounds such as benzoic acid were identified as the aromatic compounds in the edible extracts of Aruncus dioicus. The contents of azulene, benzaldehyde and linalool among those compounds on the cultivation conditions increased in shading cultivation. $IC_{50}$ values of DPPH free radical scavenging activities were high from 6.644 to $14.499{mu}g/m{\ell}$ in extracts of edible parts and those of edible shooting parts was high such as edible shooting cultivated by 60% shading, 30% shading and non-shading, respectively. The activity of POD and SOD in seeds was lower than that of edible shooting and that by shading cultivation was high in extracts cultivated by non-shading. POD activity of extracts on harvesting time was high in earlier harvesting but SOD activity was low. The numbers of isozyme pattern of POD and SOD in seedling showed 7 bands and 3 bands, respectively, especially, bands of POD and SOD in the first year-growing plant did not show and show a difference according to plant positions, respectively.

Effects of Elevated Atmospheric CO2 and Nitrogen Fertilization on Growth and Carbon Uptake of Yellow Poplar Seedlings (대기 이산화탄소 증가와 질소 시비가 백합나무 유묘의 생장과 탄소 흡수에 미치는 영향)

  • To investigate the responses of yellow poplar (Liriodendron tulipifera L.) seedlings to the interactive effects of the elevated atmospheric $CO_2$ level and nitrogen addition, we measured biomass, photosynthetic pigments, photosynthesis, and the contents of nitrogen (N) and carbon (C) from the seedlings after 16 weeks of the treatments. Yellow poplar seedlings were grown under the ambient ($400{\mu}mol\;mol^{-1}$) and the elevated (560 and $720{\mu}mol\;mol^{-1}$) CO2 concentratoins with three different N addition levels (1.2, 2.4, and $3.6g\;kg^{-1}$) in the Open Top Chambers (OTC). The dry weight of the seedlings enhanced with the increased N levels under the elevated $CO_2$ concentrations and the increment of the dry weight differed among the different N levels. Photosynthetic pigment content of the yellow poplar leaves also increased with the increase of the $CO_2$ concentration levels. The effects of the N levels on the photosynthetic pigment content, however, were significantly different among the $CO_2$ levels. Photosynthetic rates were affected by the levels of $CO_2$ and N concentrations. Stomatal conductance and transpiration rates increased with increasing $CO_2$ concentration. The carboxylation efficiency of the seedlings without N addition increased under the higher $CO_2$ concentrations whereas that with N addition decreased under the elevated $CO_2$ concentrations. Nitrogen and carbon uptake in leaf, stem, and root increased with the elevated $CO_2$ concentration level and N addition. In conclusion, under the elevated $CO_2$ concentrations, physiological characteristics and carbon uptake of the yellow poplar seedling were improved and increased with N addition.

  • Properties of enzymatic hydrolysates from ascidian tunic were assessed on supernatant ratio, solid yields and solid concentration. The concentartion of solid and yields in the extracts were increased as the enzyme concentration raised from $100\;{\mu}l\;to\;1000{\mu}l$ during the extraction period. The optima concentration and reaction time of each enzyme on digestion were $400\;{\mu}l$ 60 minutes, through treated with Duncan's multiple test. The percent of yields of solid, protein and carotenoids for 60 minutes extraction at $400\;{\mu}l$ were $32.32\%,\;1.34\%\;and\;74.60\;mg\%$, respectively, in Viscozyme systems. The extracts were composed with many kinds of carbohydrates such as arabinose, ribose, xylose, galactose, glucose, N-acetyl-D-galactosamine, and N-acetyl-D-glucosamine. Aspartic and glutamic arid were noted as predominant amino acids in all parts. Amino acid profiles of various ascidian tunic part were similiar to each other, but most of essential amino acids content of inter coat was higher than that of root and tunic (body). About sixty six fatty acids components were observed, and their distribution among neutral and polar lipids was compared. The main fatty acids were found to be 14:0, 16:0, 16:1n7, 18:0, 18:1n9, 18:1n7, 18:2n6, 20:5n3, and 22:6n3.

  • To utilize the ascidian tunic as a natural pigment and dietary sources for rainbow trout (Oncorhynchus mykiss), juvenile were fed on experimental diets containing enzymatic hydrolysates of ascidian tunic treated with three commercial mined enzymes (ultrazyme, cellulase, viscozyme) for 12 weeks. From the results of feeding experiment, similar growth rate was checked in the enzymatic hydrolysis group compared with control, and those were a higher than of ascidian tunic powder group. The total acetone extractable pigment in muscle of the enzymatic hydrolysates group was lower than that of the ascidian extracts group and carophyll pink group until 8 weeks, but the level of those pigment of the enzymatic hydrolysates was similar to the ascidian extracts and carophyll pink group after 12 weeks. The lipid content was increased with the pigment concentration in the all experimental group. But the ascidian tunic pigment did not influence on the composition of the fatty acids in the muscle and liver. From the consideration of results for pigmentation, the enzymatic hydrolysates of ascidian tunic were suitable for both a natural pigment and dietary protein and carbohydrates sources as a substitute synthetic pigment for aquaculture use.
