자색 고구마 색소의 추출에 미치는 고전압 펄스 전기장의 영향 (Effect of High Voltage Pulsed Electric Fields on Extraction of Purple Sweet Potato Pigment)

  • 천연 식용 색소원으로서 최근 주목받고 있는 자색 고구마로부터의 색소 추출에 대한고전압펄스전기장의 영향을 조사하였다. 자색 고구마 색소는 일정한 주파수와 처리시간 하에서 전기장의 세기를 변화시켜 추출하였을 경우 전기장의 크기가 증가함에 따라 색소의 추출량도 점차 많아지고 있음을 나타내었으며 최적 전압은 35kV/cm임을 알 수 있었다. 동일한 전기장의 세기와 처리시간을 주고 주파수만을 변화시키면서 추출을 했을 경우 낮은 주파수에서 500Hz까지는 추출량이 큰 폭의 증가를 보였으나, 500 Hz 이상에서는 spark발생에 의한 색소의 파괴로 약간씩의 감소를 나타내었다. 전기장의 세기나 주파수가 추출에 큰 영향을 미치는 것으로 나타났지만 일정한 전기장과 주파수 하에서 처리 시간은 색소의 추출양에 큰 영향을 미치지 못하는 것으로 나타났다.

곰팡이 RF101이 생성하는 적색 색소의 안정성 및 추출물의 생리활성 검정

  • 적색 색소를 생성하는 곰팡이 RF10l 로부터 여러 용매를 이용하여 추출물을 얻어낸 후 각 추출물의 특성을 조사한 결과 chloroform 용액은 510nm 에서 흡수 peak를 나타냈으며, pH 5-9 영역에서 안정함을 보였고, 암상태의 저온과 실온에서 5 일이 경과 한 후90% 이상이 구조적으로 안정함을 보였다. 금속이온들 중 NaCl을 제외하고 KCl. $CaCl_2$, $MgCl_2$ 용액에서 5 일이 경과한 후에도 60% 이상의 안정함을 나타내었고, 균사체 powder 가 0.5% 첨가된 배지에서는 모든 균주의 생장이 억제되었으며 70% methanol 추출물 . $70^{\circ}C$ D.W 추출물에서도 생장억제 효과가 크게 나타났다. 이러한 결과로 볼 때, 더 많은 연구가 진행된다면 곰팡이 RFl0l로부터 산업적 이용 가치가 있는 물질을 생산할 수 있을 깃으로 생각되며 그에 대한 연구는 계속 수행중에 있다.

키토산과 천연색소를 이용한 Eco-Printing(제1보) (Eco-printing Using Chitosan and Natural Colorants(1))

  • The aim of this study is to develope eco-printing method using natural pigments and chitosan as a natural binder. Three chitosans with different molecular weights were employed to find appropriate conditions including chitosan concentration and pigment/binder ratio. Dye uptake, color and fastnesses of the printed fabrics were evaluated to find optimum conditions within the range of experiments carried out in this study. The effectiveness of chitosan as a printing binder was examined in comparison with color, dye uptake, and fastnesses of conventional synthetic binder and guar gum. It was found that chitosans with low or medium molecular weight were appropriate. Using low molecular weight chitosan, optimum concentrations were 1.7% for charcoal, madder and chlorophyll, whereas 2.2% for ocher, yellow soil, indigo and cochineal. Regardless of molecular weight and concentration of chitosan, the color fastnesess of fabrics printed with mineral pigments were superior to those of the fabrics printed with plant and animal pigments. As pigment/chitosan ratio became higher, rubbing fastness was decreased by 1-3 grade. The colorfastness of printed fabric with chitosan binder was similar to that with synthetic binder, which was higher than that with guar gum.

Biocide에 의한 담수조류의 생장에 관한 연구 I. Chlamydomonas reinhardii의 생장에 미치는 영향 (Studies on the Growth of Freshwater Algae by Biocides I. On the Growth of Chlamydomonas reinhardii)

  • The effects of 6 biocides on the growth of Chlamydomonas reinhardii in pure culture were studied. For the batch culture assay of biocides, the growth rate in control tubes should be compared with in the test tubes and the effective concentration calculated on the basis of the percent decrease in growth rate at the different toxicant concentration. The concentrations at which 50% decrease in growth rate was observed are $40{\mu}g$/l for butachlor and $350{\mu}g$/l for alachlor in herbicide, $900{\mu}g$/l for phenazine-5-oxide and $3,400{\mu}g$/l for isoprothiolane in fungicide, and $3,330{\mu}g$/l for fenthion and $332,500{\mu}g$/l for trichlorfon in insecticide. The inhibitory effect on the growth of Chlamydomonas reinhardii by the treatment of various biocide concentrations was decreased in order of herbicide>fungicide>insecticide. Chlorophyll and carotenoid content per cell were increased, whereas chlorophyll a/b ratio was hardly affected by biocides. The effects of biocides on pigment content were also decreased in order of herbicide>fungicide>insecticide, which suggested the relationship between the effect of biocides on the pigment content and inhibition of growth rate.

Changes of Anthocyanidin, Growth Characteristics and Brown Rice Yield of Red Colored Rice at Two Region of Eastern Coast in Gyeongsangbuk-do Province

  • The study was carried out to determine the changes of growth characteristics, anthocyanidin, and brown rice yield grown at coast and inland areas apart from the east coastline of Gyeongsangbuk-do province. Number of spikelets per panicle was much more increased in inland area. Regional difference in number of spikelets per panicle was observed in Jeongjinju cultivar. Among these red rice cultivars, the highest brown rice yield was Jeongjinju rice cultivar having 702 kg in inland area and 692 ㎏ in coast area, respectively. Anthocyanidin content ranged 524 to $610{\mu}g/g$ dry weight basis. Cyanidin content was 11.4 to 14.0 times higher than that of delphinidin under coast and inland area. Anthocyanidin content was higher in rice cultivar grown at coast area. Highest head brown rice rate was only observed in Geonganghongmi to 95.2 at coast area and 95.4 inland area. In considering brown rice yield and pigment content, Hongjinju rice cultivar was recommended in optimal pigment rice cultivar in eastern coast of Gyeongsangbuk-do Province.

액침진탕 배양에 의한 Monascus sp..가 생산하는 적색색소에 관한 연구 제1보 균주의 분리 및 색소생산 배양 조건 (Studies on the Red Pigment Produced by Monascus sp. in Submerged Culture. Part I Isolation of Strain and Cultural Conditions of Pigment Produced)

  • 적색색소의 생산을 위하여 태국산 tapioca chips와 곡자, 공기, 식물의 잎으로부터 우수한 균주를 선별하였고 이의 형태적 특성을 조사 동정하였다. 배양조건으로 온도, pH, 탄소원, 질소원, 통기효과, amino산, vitamin, 무기염류 등이 색소생산에 미치는 영향을 조사하여 최적조건을 구하여 다음과 같은 결론을 얻었다. 1) 선별된 균주는 형태적 특성을 근거로 균사에 격벽과 균사체끝에 피자기를 가지고 자낭포자와 분생포자를 만들며 특수구조인 후피포자가 있는 Monascus sp. 로 동정되었다. 2) 최적 배양조건은 온도 32~33$^{\circ}C$, pH 6.5, 탄소원으로 Tapioca chips 분말 3.5%, 질소원으로 NaNO$_3$ 0.2%, 금속이온은 MnO$_2$ 0.001% 첨가가 가장 좋았고 통기효과는 100/500mι(v/v) 진탕배양 (180rpm)에서 4일 배양이 가장 효과적이었다.

Transgenic plants with cyanobacterial genes

  • Over the years, cyanobacteria have been regarded as ideal model systems for studying fundamental biochemical processes like oxygenic photosynthesis and carbon and nitrogen assimilation. Additionally, they have been used as human foods, sources for vitamins, proteins, fine chemicals, and bioactive compounds. Aiming to increase plant productivity as well as nutritional values, cyanobacterial genes involved in carbon metabolism, fatty acid biosynthesis, and pigment biosynthesis have been intensively exploited as alternatives to homologous gene sources. In this short review, transgenic plants with cyanobacterial genes generated over the last two decades are examined, and the future prospects for transgenic crops using cyanobacterial genes obtained from functional genomics studies of numerous cyanobacterial genomes information are discussed.

천연색소 함유 화장료 조성물의 항균효과 및 항산화활성 (Antimicrobial Effects and Antioxidative Activities of the Cosmetic Composition Having Natural Plant Pigments)

  • 천연색소를 이용한 천연 색조화장품 개발을 위한 기초자료로 활용하고자, 15종의 천연색소를 사용하여 제조한 화장료 조성물에 대하여 항균활성 및 항산화능을 조사하였다. 실험에 사용한 6종의 균주 중 Bacillus subtilis, Staphylococcus aureus, Listeria monocytogenes, Vibrio parahaemolyticus, Aspergillus flavus에 대해 대부분의 색소혼합조성물에서 항균활성이 나타나는 것으로 확인되었으며, Proteus vulgaris에서는 다른 균주에 비해 항균활성이 상대적으로 낮았다. 색소종류별로는 자색고구마, 여주, 파프리카, 뽕잎, 지치 등의 색소 조성물에서 다른 색소 조성물에서보다 더 높은 항균활성을 보였다. 또한 DPPH 라디컬 소거능은 양파껍질, 파프리카, 적양배추, 뽕잎, 포도과피, 오디, 레드비트 색소 함유 조성물에서 상대적으로 높은 활성을 보였으며, 아질산염 소거능에 있어서는 특히 pH 1.2에서 적양배추, 포도과피, 오디, 백년초를 제외한 대부분의 색소에서 70% 이상의 높은 활성을 나타냈다. 다양한 천연색소를 이용하여 수행한 본 실험을 통해 천연색소의 높은 항균효과와 항산화능을 확인할 수 있었으며, 향후 이러한 기능성을 활용한 안전한 천연 색조화장품 개발에 유용한 자료가 될 것으로 기대한다.

Production of Pigments and $\alpha$-Tocopherol by Cell Cultures in Safflower (Carthamus tinctorius L.)

  • Safflower is an important medicinal plant that has been used in China, Korea and Japan for thousands of years. The red and yellow pigments obtained from the petals of safflower can invigorate blood, release stagnation and promote menstruation. In addition, these pigments are used safely in processed foods and soft drinks as naturally harmless rotor additives. On the other hand, the seed of safflower contains 30-40% oil with higher level of mono- and poly-unsaturated fatfy acid profiles and elevated levels of $\alpha$-tocopherol. In this paper, we describe advances in the production of pigments and $\alpha$-tocopherol by cell culture in safflower.

식물의 뿌리와 근권으로부터 Salicylic acid를 생성하는 근권세균의 검색 (Screening of Salicylic acid Producing Rhizobacteria Isolated from Plant Roots and Rhizosphere)

  • Twenty two rhizobacteria were isolated from the roots and rhizosphere of radish, carnation, potato and tomato. There isolates produced a fluorescent pigment in King's B medium and identified as Pseudomonas spp. These isolates colonized roots and rhizosphere of the host plants. In the study of cultural characteristics of the bacteria, the pH of the culture broth was changed from neutral (7.0) to alkali (8.8∼9.41) and the numbers of cells were increased from 106 to 108 after 40 hr of incubation in basal standard succinate medium. The salicylic acid production identified by pink color reaction were observed in 7 bacteria. Out of these 7 salicylic acid producing bacteria, only 2 strains of bacteria such as Pseudomonas fluorescens RS006, and Pseudomonas sp. EN401 were confirmed as salicylic acid producers by optical density measurement. Therefore, for screening of salicylic acid producing bacteria from the roots and rhizosphere, color reaction of the culture medium should be done in the first step, and then optical density measurement of culture extract should be made for the confirmation of salicylic acid production.
