Effect of relative humidity in swine house on pathogenesis of swine pleuropneumonia (돈사의 상대습도가 돼지흉막폐렴의 병인에 미치는 영향)

  • The effect of relative humidity in swine house on swin pleuropneumonia was examined in piglets experimentally infected with Actinobacillus pleuropneumoniae serotype 5. A total of 20 piglet were grown under 30~40%, 41~50%, 51~64% and 65~80% relative humidity chambers after intratracheal inoculation of A pleuropneumoniae. Characteristic fibrinous pleuropneumonia was observed in the pigs grown at the low relative humidity groups. The detailed results were as follows : 1. Growth performance and environment conditions were lower than high relative humidity groups. 2. Characteristic histopathological findings were fibrinous pleuritis and pneumonia accompanied congestion, hemorrhage, thrombosis and edematous change. 3. Antigenic distribution of inoculated bacterium was found mainly in alveolar macrophages or accumulated foci of macrophages adjacent to necrotic area. 4. Characteristic electron microscopic findings were proliferation of type II pneumocyte with increased lamella bodies and activated alveolar macrophages with pseudopods and widening of interstitium.

Silybum marianum seed extract supplementation positively affects the body weight of weaned piglets by improving voluntary feed intake

  • This study was conducted to evaluate the effects of dietary supplementation of Silybum marianum seed (SMS) extract on the growth performance, nutrient digestibility, fecal noxious gas emission, and hematology parameters in weaned piglets. A total of 120, 21-day-old weaned piglets ([Yorkshire × Landrace] × Duroc) were randomly assigned to 3 groups based on the average initial body weight (6.46 ± 0.45 kg). There were 8 replicate pens per treatment and 5 pigs (mixed sex) per pen. The experimental period was 42 days. Dietary groups included a basal diet, and a basal diet supplemented with 0.05% or 0.10% SMS extract. Feeding weaned piglets with SMS extract containing diet significantly increased average daily gain and average daily feed intake. Additionally, the supplementation of SMS extract had no significant effects on nutrient digestibility, serum hematology, and fecal noxious gas emission parameters. We considered that the supplementation of SMS extract had positive effects on the voluntary feed intake in weaned piglets, thus improving growth performance.

  • The present study was focussed mainly on the morphological changes of the glandular tubules in the large intestine according to age of piglets. Samples were taken from large intesine of 1-, 10-, 20-, 35- and 45-day-old piglets, 2 to 3 piglets in each age group. The intestinal samples were fixed in 10% neutral formalin solution, dehydrated, and then paraffin sections were stained with H-E. The results observed were summarized as follows: 1. The mucosal glands in the cecum and colon tend to be unbranched simple straight tubular glands, or often two or more branched simple stright tubular glands. 2. The number of the longitudinal folds and the number of the crypts per cross section of piglet colons, respectively, were 1-day-old piglets-$3.8{\pm}0.8$, $92.1{\pm}6.9$; 10-day-old piglets-$7.1{\pm}1.1$, $164.2{\pm}10.3$; 20-day-old piglets-$15.2{\pm}0.8$, $178.5{\pm}6.8$; 35-day-old piglets-$19.3{\pm}3.0$, $454.9{\pm}25.3$; 45-day-old piglets-$20.6{\pm}3.1$, $524.6{\pm}37.2$, and the regression equation between age and these two number were $\hat{Y}=0.40X+4.32$ and $\hat{Y}=10.4X+51.52$, respectively. 3. The length and cell number per single side wall of a glandular tubule in the colon section were 1-day-old piglets-$196.3{\pm}7.1{\mu}m$, $40.0{\pm}3.3$; 10-day-old piglets-$236.0{\pm}34.5{\mu}m$, $47.9{\pm}5.3$; 20-day-old piglets-$262.8{\pm}39.6{\mu}m$, $54.3{\pm}9.0$; 35-day-old piglets-$291.75{\pm}48.3{\mu}m$, $56.9{\pm}4.9$; 45-day-old piglets-$364.8{\pm}61.5{\mu}m$, $67.7{\pm}7.4$, respectively, and the regression equation between age and these two data were $\hat{Y}=3.45X+193.8$ and $\hat{Y}=0.56X+41.0$, respectively. 4. The overall percentages of the cell number and length of glandular tubules in piglet colons were the pit and isthmus-$75.3{\pm}11.1%$, $78.8{\pm}12.3%$; gland-$24.7{\pm}5.4%$, $21.2{\pm}5.3%$, respectively. 5. The length and cell number of single side wall of glandular tubules in cecal sections were 1-day-old piglets-$190.3{\pm}31.1{\mu}m$, $37.6{\pm}4.8$; 10-day-old piglets-$235.6{\pm}25.3{\mu}m$, $46.2{\pm}3.6$; 20-day-old piglets-$295.3{\pm}45.6{\mu}m$, $52.0{\pm}6.2$; 35-day-old piglets-$351.3{\pm}28.3{\mu}m$, $60.4{\pm}8.5$; 45-day-old piglets-$366.3{\pm}48.5{\mu}m$, $64.7{\pm}8.2$, respectively, and the regression equation between age and these two data were $\hat{Y}=4.11X+196.6$ and $\hat{Y}=0.60X+38.9$, respectively.

Toxicity of lectin extracted from Korean mistletoe (Viscum album coloratum) in piglets and its effects on the immunogenicity of Aujeszky's disease virus vaccines (한국산 겨우살이(Viscum album coloratum)로부터 추출된 lectin의 돼지에 대한 독성 및 오제스키병 백신의 면역원성에 미치는 영향)

  • In the present study toxicity and immunostimulating activity of the lectin(KML-C), which was extracted from Korean mistletoe(Viscum album coloratum) were investigated in swine. To determine the toxicity, lectin was injected into thigh or cervical muscles of 4-week-old piglets(Landrace) and observed clinically and pathologically. For determination of the immnunostimulating activity, lectin($0.7{\mu}g/kg$ of body weight)-adjuvanted vaccine of Aujeszky's disease virus(ADV)(NYJ1-87) which was inactivated by 0.2% formalin was injected into the cervical muscle of antibody-negative piglets in the same age group. Subpopulation of the immune cells and serum neutralizing(SN) antibodies in the piglets were examined after vaccination, and resistance of the piglets against challenge by virulent NYJ1-87 was further examined. The results were also compared with those from piglets injected with aluminum hydroxide [$Al(OH)_3$]-adjuvanted vaccine of inactivated NYJ1-87 and NYJ1-87 vaccine without adjuvant, and the results are as follows. By injection of lectin with $30{\mu}g/kg$ of body weight to the thigh muscle, all of 12 piglets died after signs such as dyspnea, fever, systemic erythema and subcutaneous hemorrhages, and lesions pertaining to poisonous hepatitis and dysfunction of kidney were observed. By injection of lectin with $7{\mu}g/kg$ of body weight to the thigh muscle, all of 12 piglets showed signs such as edema and cutaneous hemorrhage in the injected area, lameness and depression, and lesions pertaining to poisonous hepatitis and dysfunction of kidney were observed. By injection of lectin with 1, 3 and $5{\mu}g/kg$ of body weight to the thigh muscle of each one piglet, signs such as congestion, induration and grayish coloration in the injected area, depression and inappetence were observed in all piglets. Toxic changes were also observed in the liver and kidney of piglets by lectin of 3 and $5{\mu}g$. By injection of lectin with 0.5 and $0.7{\mu}g/kg$ of body weight to the cervical muscle of each 9 piglets, all piglets were clinically normal and there were no significant changes in blood counts and chemistry values. Whereas, epithelial swelling and vacuolation of convoluted tubules were observed from one piglet injected with lectin of $0.7{\mu}g$, and necrosis and fibrosis of muscular fiber were observed in the muscle of one piglet injected with lectin of $0.5{\mu}g$. Only population of sIgM+ B lymphocytes increased among immune cells in all of 15 piglets immunized with lectin($0.7{\mu}g/kg$ of body weight)-adjuvanted vaccine, while compared to those in $Al(OH)_3$-adjuvanted vaccine and vaccine without adjuvant. No additional stimulation to the immune cells was recognized when lectin was added to $Al(OH)_3$-adjuvanted vaccine. In piglets immunized with lectin-adjuvanted vaccine, SN titers in reciprocal values for loge were 1.3-4.0 at 1-4 weeks after vaccination, which was similar to those with 1.0-3.3 by vaccine without adjuvant but lower than those with 2.0-5.7 by $Al(OH)_3$-adjuvanted vaccine. Also, no additional increase in the SN titers was recognized when lectin was added to $Al(OH)_3$-adjuvanted vaccine. Piglets immunized with lectin-adjuvanted vaccine were resistant to challenge by the virulent NYJ1-87 at 4 weeks after vaccination, and the SN titers reached to 5.0 one week after challenge, which was higher than those with 4.0 by vaccine without adjuvant but somewhat lower than those with 7.7 by $Al(OH)_3$-adjuvanted vaccine.

The Effects of Gilts Housed Either in Group with the Electronic Sow Feeding System or Conventional Stall

  • This experiment was conducted to assess the welfare and productivity of gestating gilts in groups with the electronic sow feeding (ESF) system compared to conventional stalls. A total of 83 gilts ($Yorkshire{\times}Landrace$) were housed into individual stalls to be artificially inseminated. Gilts confirmed pregnant were introduced to their treatment, conventional stalls (ST) or groups with the ESF system. All gilts were taken to the farrowing crates one week prior to their expected farrowing date. In the gestation period, there were no significant differences between gilts allocated to ST and ESF on growth performance. However, backfat thickness gain (p = 0.08) and body condition score (BCS) at 110 days of gestation (p = 0.10) tended to be higher in ESF gilts than ST. Likewise, gilts housed in group showed significantly higher estimated body muscle contents at 110 days of gestation (p = 0.02) and body muscle change during gestation (p = 0.01). There was a trend for a shorter parturition time in ESF gilts (p = 0.07). In the lactation period, group housed gilts showed a tendency to increased BCS changes (p = 0.06). Reproductive performance did not differ with the exception of piglet mortality (ST = 0.2 no. of piglets vs ESF = 0.4 no. of piglets; p = 0.01). In blood profiles, ST gilts showed a higher cortisol level at 110 days of gestation (p = 0.01). Weaning to estrus interval was shorter in gilts housed in ESF than ST (p = 0.01). In locomotory behaviors, ESF gilts recorded a tendency to elevate locomotion score at 36, 70, and 110 days of gestation (p = 0.07, p = 0.06, and p = 0.06, respectively). Similarly, ESF gilts showed significantly higher incidence of scratches at 36, 70, and 110 days of gestation (p = 0.01). Moreover, farrowing rates were higher in stall treatment (97.6%) compare to group housing treatment (95.2%). In conclusion, while group housed gilts with ESF system positively affected welfare status in combination with less physiologically stressful environments and activity, it negatively effects piglet mortality, farrowing rates and injuries of gilts.

Effect of Dietary Lysine Supplement on the Performance of Mong Cai Sows and Their Piglets

  • The objective of this study was to determine optimal lysine requirement of lactating Mong Cai sows and their piglets. An experiment was conducted using 30 Mong Cai sows in a factorial randomized design with 5 dietary total lysine levels (0.60, 0.70, 0.85, 1.0 and 1.15%) for one-week pre-partum and 5 dietary total lysine levels (0.60, 0.75, 0.90, 1.05 and 1.2%) for lactation diets. Mong Cai sows were about 1 to 2 years old and had an initial body weight of 120 kg (sd = 2.5) after farrowing. Sows were restrictively fed 1.7 kg feed during gestation and were fed ad libitum during lactation. Diets of sows contained about 12% CP during pregnancy and about 14% CP for the lactation period. DE concentration of the diets ranged between 12.5-13.0 MJ of DE. Water was supplied at up to 8 liters per sow per day in a basin. Studied traits were related to both sows and their progeny. Sows were weighed at 107 days of gestation, after farrowing and at weaning. Sow back-fat depth was measured at 110 days of gestation, after farrowing, at 21 days of lactation and at weaning. Number of piglets born, at 24 h after birth, at 21 days of age and at weaning were recorded. Piglets were weighte at birth, at 21 days and at weaning. Supplying lysine one week pre-partum had no effect on the number of piglets born nor litter weight at birth (p = 0.776 and p = 0.224). A positive effect of increasing dietary lysine level during lactation from 0.60 to 1.20% was observed with regard to less sow weight loss, and increased piglet weight at 21 days and at weaning. The level of lysine that resulted in the lowest sow backfat loss and the highest weaned piglet weight was 1.05%; this may be the optimum level of lysine for the diet of lactating Mong Cai sows. At this lysine level, the number of weaned piglets was also highest.

Effects of HACCP System Implementation on Medicine Use and Productivity of Medium Scale Swine Farms in Korea (HACCP시스템 적용이 중규모 양돈농장의 동물용의약품 사용 및 생산성에 미치는 영향)

  • The aim of this study was to examine the effect of HACCP system implementation on the budget for medicine use, antibiotic utilization, and productivity of swine farms in Korea. Data were collected form thirty swine farms before and after implementation of HACCP system. Parturition rate, total number of piglets, number of initial weaning piglets, number of weaning piglets, survival rate of piglets after weaning, date of weaning piglets, piglet per sow per year, and market piglet per sow per year tended to be higher after HACCP system implementation. In contrast, price for medicine used per month, number of antibiotic used, the farm number of feeds containing antibiotics in each, growing stages added at the feed company, and the farm number of feeds containing antibiotics in each growing stages added at the swine farm were lower after HACCP system implementation. Due to increase in the feed cost recently, production cost for shipment in HACCP system implemented swine farms had increased in this study. In conclusion, our results indicated that the implementation of HACCP system might be effective in reducing medical expenses and improving the productivity.

Effect of increasing levels of rice distillers' by-product on growth performance, nutrient digestibility, blood profile and colonic microbiota of weaned piglets

  • Objective: This study was conducted to evaluate the effects of diets containing different wet rice distillers' by-product (RDP) levels on growth performance, nutrient digestibility, blood profiles and gut microbiome of weaned piglets. Methods: A total of 48 weaned castrated male crossbred pigs, initial body weight 7.54±0.97 kg, and age about 4 wks, were used in this experiment. The piglets were randomly allocated into three iso-nitrogenous diet groups that were fed either a control diet, a diet with 15% RDP, or a diet with 30% RDP for a total of 35 days. Chromium oxide was used for apparent digestibility measurements. On d 14 and d 35, half of the piglets were randomly selected for hemato-biochemical and gut microbiota evaluations. Results: Increasing inclusion levels of RDP tended to linearly increase (p≤0.07) average daily gain on d 14 and d 35, and decreased (p = 0.08) feed conversion ratio on d 35. Empty stomach weight increased (p = 0.03) on d 35 while digestibility of diet components decreased. Serum globulin concentration decreased on d 14 (p = 0.003) and red blood cell count tended to decrease (p = 0.06) on d 35, parallel to increase RDP levels. Gene amplicon profiling of 16S rRNA revealed that the colonic microbiota composition of weaned pigs changed by inclusion of RDP over the period. On d 14, decreased proportions of Lachnospiraceae_ge, Ruminococcaceae_ge, Ruminococcaceae_UCG-005, and Bacteroidales_ge, and increased proportions of Prevotellaceae_ge, Prevotella_2, and Prevotella_9 were found with inclusion of RDP, whereas opposite effect was found on d 35. Additionally, the proportion of Lachnospiraceae_ge, Ruminococcaceae_ge, Ruminococcaceae_UCG-005, and Bacteroidales_ge in RDP diets decreased over periods in control diet but increased largely in diet with 30% RDP. Conclusion: These results indicate that RDP in a favorable way modulate gastrointestinal microbiota composition and improve piglet performance despite a negative impact on digestibility of lipids and gross energy.

The Effect of Acupuncture on the Intestinal Smooth Muscle Dysfunction Caused by Enteric Bacterial Infection in Weaning Piglet (돼지의 세균성 위장관 질환에 따른 평활근 기능 이상에 대한 침술의 효과)

  • It is now generally accepted that acupuncture is effective in diarrhea caused by bacterial infection. However, its effect on the intestinal smooth muscle dysfunction is not clear. Therefore, we investigated the effect of acupuncture therapy at Jiao-chao (GV-1) on the intestinal muscle dysfunction in weaning piglets orally infected by Escherichia coli. The animals are divided into four groups; 1) E. coli + no-treatment, 2) E. coli + antibiotic, 3) E. coli + acupuncture, 4) normal group. In the three E. coli infected groups, low frequency electrical field stimulation (EFS, 1 Hz) provoked triphasic responses composed of initial relaxation followed by on-contraction and off-contraction. While in the normal group, EFS (1Hz) induced biphasic responses composed of relaxation during the stimulation and off-contraction. At the high frequency (16Hz) EFS, both on-contraction and off-contraction of the E. coli + antibiotic, E. coli + acupuncture and the normal group were larger than those of the E. coli + no-treatment group. In the non-adrenergic non-cholinergic (NANC) condition, only biphasic responses occurred to EFS in all experimental groups and the off-contraction of E. coli + antibiotic, E. coli + acupuncture and the normal group were larger than those of the E. coli + no-treatment group. The response to carbachol of those three groups was also significantly greater than that of the E. coli + no-treatment group. These results suggest that acupuncture is as effective as antibiotic in the dysfunction of colonic circular muscle caused by E. coli infection. The maintenance of contractile neuromuscular transmission seems to be involved in the mechanism of the acupuncture effects on diarrhea.

Dietary Conjugated Linoleic Acid (CLA) increases milk yield without losing body weight in lactating sows

  • This study was conducted to evaluate the effects of dietary conjugated linoleic acid (CLA) on the performance of lactating sows and piglets as well as the immunity of piglets suckling from sows fed CLA. Eighteen multiparous Duroc sows with an average body weight (BW) of $232.0{\pm}6.38kg$ were randomly selected and assigned to two dietary treatments (n = 9 for each treatment), control (no CLA addition) and 1% CLA supplementation. For the control diet, CLA was replaced with soybean oil. Experimental diets were fed to sows during a 28-day lactation period. Litter size for each sow was standardized to nine piglets by cross-fostering within 24 hours after birth. Sow milk and blood samples were taken from sows and piglets after 21 and 27 days of lactation, respectively. Loss of BW was significantly (p < 0.05) higher in sows fed control diet compared to sows fed CLA diet. Piglet weights at weaning and weight gain during suckling were significantly (p < 0.05) higher in sows fed CLA compared to sows fed control diet. Serum non-esterified fatty acid (NEFA) and urea nitrogen concentrations were significantly (p < 0.05) lower in sows fed CLA than in sows fed soybean oil. IgG concentrations of the groups supplemented with CLA increased by 49% in sow serum (p < 0.0001), 23% in milk (p < 0.05), and 35% in piglet serum (p < 0.05) compared with the control group. Sows fed CLA showed an increase of 10% in milk yield compared with sows fed soybean oil (p < 0.05), even though there was no difference in daily feed intake between the treatments. Milk fat content was significantly (p < 0.05) lower in sows fed CLA than in sows fed soybean oil. Solid-not-fat yield was significantly (p < 0.05) higher in sows supplemented with CLA than in sows fed control diet and also protein-to-fat ratio in milk was significantly (p < 0.05) higher in sows fed CLA compared with the control group. The results show that CLA supplementation to sows increased milk yield without losing BW during lactation, whereas soybean oil supplementation resulted in severe BW loss.
