Estimation of Nitrogen Storage Potential and Aboveground Biomass of Tree Species Treated with Liquid Pig Manure (양돈분뇨 처리에 따른 수목의 질소저장 및 지상부 바이오매스 추정)

  • This study was conducted to estimate aboveground biomass and nitrogen storage potential of tree species-eight clones of a poplar and hybrids, one clone of Salix alba L., dawn redwood (Metasequoia glyptostroboides Hu and W.C. Cheng), yellow poplar (Liriodendron tulipifera L.), Okamoto maple (Acer okamotoanum Nakai), and pin oak (Quercus palustris $M{\ddot{u}}nchh.$)- after treating with liquid pig manure. Stems showed the highest percentage of aboveground biomass, and followed by branches and leaves. Nitrogen content in aboveground biomass components was the highest in leaves, and followed by branches and stems. Average aboveground biomass production was higher in the clones and species treated with manure than those of not treated, 30 ton/ha and 16 ton/ha, respectively. In the manure-treated site, clone 'Dorskamp' of Populus deltoides${\times}$Populus nigra showed the highest aboveground biomass (48.3 ton/ha). Average nitrogen storage potential was superior in the clones and species treated with manure than those of not treated, 159 kg/ha and 90 kg/ha, respectively. Clone 'Dorskamp' also showed the greatest nitrogen storage potential (286.5 kg/ha) among tested tree species. Therefore, 'Dorskamp' is the most suitable clone for treating liquid pig manure, but additional studies are needed to determine any damages or tolerance from the treatment.

Detection of Meat Origin (Species) Using Polymerase Chain Reaction

  • A quick and reliable method for identifying meat origin is developed to ensure species origin of livestock products for consumers. The present study examined the identification of meat sources (duck, chicken, goat, deer, pig, cattle, sheep, and horse) using PCR by exploiting the mitochondrial 12S rRNA and mitochondrial cytochrome b genes. Species-specific primers were designed for some or all mitochondrial 12S rRNA nucleotide sequences to identify meat samples from duck, chicken, goat, and deer. Mitochondrial cytochrome b genes from pig, cattle, sheep, and horse were used to construct species-specific primers, which were used to amplify DNA from different meat samples. Primer sets developed in this study were found to be superior for detecting meat origin when compared to other available methods, for which the discrimination of meat origin was not equally applicable in some cases. Our new development of species-specific primer sets could be multiplexed in a single PCR reaction to significantly reduce the time and labor required for determining meat samples of unknown origin from the 8 species. Therefore, the technique developed in this study can be used efficiently to trace the meat origin in a commercial venture and help consumers to preserve their rights knowing origin of meat products for social, religious or health consciousness.

  • A total of 113 Staphylococcus hyicus subsp. hyicus which had been isolated from healthy and diseased pigs were examined for their ability to produce staphylococcins. For selection of staphylococcin indicator strains, 80 strains were randomly selected from 113 strains, and were tested as indicator and as donator. The 8 indicators were chosen and used to type all of the strains. The ability to produce staphylococcin could be demonstrated in 43 strains (38.1%) of 113 Staph hyicus subsp. hyicus. The frequency of activity was higher in diseased pig strains (44.4%) than in healthy pig strains(36.8%). 43 staphylococcin producer strains were differentiated into 10 reaction types. The inhibitory effects of the staphylococcin (N27 and R 3) on other staphylococci differed by the staphylococcal species tested. All the gram-negative species were completely resistant to the three producing strains.

  • The purpose of this study was undertaken to evaluate of cryopreservation efficiency in ${\alpha}$ 1,3-galactosyltransferase knock-out(GalT KO) cloned miniature pig sperm. To compare ability of frozen-thawed sperm characteristics, three different pig strains (GalT KO) cloned miniature pig, PWG miniature pig and Duroc were used. The ejaculated semen from the three pig species was diluted with same volume extender and added to LEY solution for freezing. The diluted semen was placed in 0.5 ml straws, and freezing was initiated by exposing the straws to liquid nitrogen ($LN_2$) vapours for 10 min before placing them into $LN_2$ for cryopreservation. After thawing, the sperm ability were assessed for viability (SYBR-14/PI staining), abnormality (Rose Bengal staining), and acrosome status (intactness, intensity and capacitation) (chlorotetracycline, CTC staining). The viability of frozen-thawed GalT KO pig sperm had no significant difference as compared with Duroc and PWG miniature pig sperm. However, The CTC pattern of frozen-thawed GalT KO cloned miniature pig spermatozoa showed significantly lower rates in F pattern and AR pattern (p<0.05) and significantly higher rates in B pattern than Duroc and PWG miniature pig (p<0.05). The abnormality of GalT KO cloned miniature pig sperm was significantly lower as compared to Duroc and PWG miniature pig sperm (p<0.05). In conclusion, GalT KO cloned miniature pig semen can be cryopreserved successfully and used for artificial insemination reasonably.

Botanical Composition, Herbage Production and Plant Mineral Contents as Affected by Application of Chemical Fertilizer and Fermented Sawdust Pig Manure on Cheju Brown Volcanic Ash Pasture Soil

  • This experiment was carried out during the period from September, 1997 to October, 1998 to determine the effect of fermented saw-dust pig manure (FSP) application on the herbage production on a mixed pasture in the Cheju brown volcanic ash soil. Split plot design (main plot: 3 nitrogen application levels of 0, 150 and 300 kgiha; sub plot: 4 pig sawdust manure levels of 0, 3, 6 and 12 tonha) was used. Plant height and dry matter yield increased significantly with an increase of nitrogen and FSP level. There was no difference in the botanical composition of grasses as affected by FSP application level, but herbage yields of grass species were increased by nitrogen application compared to that without nitrogen application. Botanical composition of white clover decreased with an increase of nitrogen application, but increased with an increase of FSP application level. Percentages of weeds were not affected by application rates of chemical nitrogen fertilizer and swine manure in the mixed species pasture. Nitrogen, phosphorus, and potassium contents of species in the pasture significantly increased with increasing application rates of nitrogen fertilizer. In conclusion, it would be an optimum to apply 150 kgha of inorganic chemical fertilizer and plus either 3 or 6 toniha of fermented swine manure with sawdust for optimum production of mixed pasture on Cheju Island.   • Although successful pronuclear formation and apposition were seen in porcine oocytes following mouse sperm injection, little is known on the morphology of male and female pronuclei following sperm injection. The objective of this study is to describe the ultrastructure of porcine zygote following murine sperm injection in relation to the chronology of pronuclear S phase. At 40h ~ 44h following in vitro maturation, Cumulus cells were removed in TCM-HEPES with 0.1% hyaluronidase. Then, spermatozoa was injected into the cytoplasm of oocytes. After. injection, all oocytes were transferred to NCSU23 medium and cultured at 39$^{\circ}C$ under 5% $CO_2$ in air. Oocytes were fixed in 2% glutaraldehyde in Dulbeccos phosphate-buffered saline and observed by Transmission Electron Microscopy. Nuclear precursor bodies were observed in each pronucleus. A cluster of large and small granules was attached in the nucleolus precursor body. After the apposition of male and female chromatin, chromatin condensation was observed throughout the nucleoplasm and nucleolus precursor bodies and condensed chromatin in contact with clusters of small and large granules and the nuclear envelope were found in apposed pronuclear regions. These results suggest that non-species specific nuclear cytoplasmic interactions take place during pronuclear formation and apposition following sperm injection.

Onset of Pronuclear Formation and DNA Synthesis in Porcine Oocytes following Intracytoplasmic Injection of Porcine or Murine Spematozoa

  • The onset of pronucleus formation and DNA synthesis in porcine oocytes following the injection of porcine or murine sperm was determined in order to obtain insights into species-specific paternal factors that contribute to fertilization. After 44h in vitro maturation, spermatozoa was injected into the cytoplasm of oocytes. After injection, all oocytes were transferred to NCSU23 medium and cultured at 39'E under 5% CO2 in air. Similar frequencies of oocytes with female pronuclei were observed after injection with porcine sperm or with murine sperm. In contrast, male pronuclei formed 8 to 9 h following the injection of porcine sperm, and 6 to 8 h following the injection of murine sperm. After pronucleus formation maternally derived microtubules were assembled and appeared to move both male and female pronuclei to the oocyte center. A few porcine oocytes entered metaphase 22 h after the injection of murine sperm, but normal cell division was not observed. The mean time of onset of S-phase in male pronuclei was 9.7 h following porcine sperm injection and 7.4 h following mouse sperm injection. These results suggested that DNA synthesis was delayed in both pronuclei until the sperm chromatin fully decondensed, and the sperm nuclear decondensing activity and microtubule nucleation abilities of the male centrosome are cell cycle dependent.

Distribution Channel and Microbial Characteristics of Pig By-products in Korea

  • The distribution channel of meat by-products from the pig farm to the final consumer can include a meat processor, wholesale market, wholesaler, retailer, and butcher shop. Bacterial contamination at any of these steps remains to be a serious public health concern. The aim of this study was to evaluate the distribution channel and microbial characteristics of pig by-products in Korea. Upon evaluation of pig by-products in cold storage, we found that the small and large intestine were significantly (p<0.05) higher in pH value compared to the heart and liver. The total plate counts were not significantly different among offals until cold storage for 7 d. The coliform count after 1 d of cold storage was significantly (p<0.05) higher in small and large intestine than in the other organs. The coliform count of heart, liver, and stomach showed a higher coliform count than small and large intestine until 7 d of cold storage. As determined by 16S rRNA sequencing, contamination of major pig by-products with Escherichia coli, Shigella spp., and other bacterial species occurred. Therefore, our results suggest that a more careful washing process is needed to maintain quality and hygiene and to ensure the safety of pig by-products, especially for small and large intestine.

Identification of Korean Native Pork Using Breed-Specific DNA Marker of KIT Gene

  • Accurate methods for the identification of closely related species or breeds in raw and processed meats must be developed in order to protect both consumers and producers from mislabeling and fraud. This paper describes the development of DNA markers for the discrimination and improvement of Korean native pig (KNP) meat. The KIT gene is related to pig coat color and is often used as a candidate marker. A 538 bp fragment comprising intron 19 of the pig KIT gene was amplified by PCR using specific primers, after which the PCR amplicons of a number of meat samples from KNP and three major improved breeds (Landrace, Duroc and Yorkshire) were sequenced in order to find a nucleotide region suitable for PCR-RFLP analysis. Sequence data showed the presence of two nucleotide substitutions, g.276G>A and g.295A>C, between KNP and the improved pig breeds. Digestion of KIT amplicons with AccII enzyme generated characteristic PCR-RFLP profiles that allowed discrimination between meats from KNP and improved pig. KNP showed three visible DNA bands of 264/249, 199, and 75 bp, whereas DNA bands of 249, 199, and 90 bp were detected in the three improved pig breeds. Therefore, the 75 bp DNA fragment was specific only to KNP, whereas the 90 bp DNA fragment was specific to the improved breeds. The breed-specific DNA markers reported here that target the KIT gene could be useful for the identification of KNP meat from improved pig meats, thus contributing to the prevention of falsified breed labeling.
