Effects of Fasting and Handling Stress on Plasma Concentrations of Glucose, Stress-associated Enzymes and Carcass Quality of Market Pigs (절식과 출하취급이 출하돈의 스트레스 관련 혈액성상과 도체품질에 미치는 영향)

  • One hundred and forty-four cross-bred market pigs weighing approximately 110 kg were randomly divided into six groups in a 3 (duration of fasting prior to loading; 0, 12 and 24 h) ${\times}$ 2 (handling stress; minimal vs stimulated handling stress) factorial arrangement of treatments. The stimulated handling stress group received overally rough handling including electric prod stimulation during loading, transport and lairage at least once at each step. All the animals received 3-h lairage prior to slaughter. Blood and longissimus dorsi muscle (LM) samples were taken at slaughter and after overnight chilling of the carcass, respectively. Mean plasma glucose concentration, as expected, was less in the 12 h- or 24 h-fasting group than in the 0 h-fasting, whereas cortisol concentration was greater (P<0.05) in the 24 h- vs 0 h-fasting group. Plasma concentrations of stress indicators glucose, cortisol, creatine kinase and lactate dehydrogenase were greater in the stimulated vs minimal handling stress group. There were no interactions between the duration of fasting and handling stress in their effects on these blood variables. The incidence of pale, soft and exudative (PSE) carcass and drip loss of LM were reduced in the 12 h- or 24 h- vs 0 h-fasting group, whereas the 24-h postmortem LM pH and color including the lightness and redness were not affected by the duration of fasting. The incidence of PSE carcass and physicochemical characteristics of LM, however, were not changed by the stimulated vs minimal handling stress. In conclusion, results suggest that fasting the market pig overnight prior to transport is desirable in terms of reducing the incidence of PSE carcass. Rough handling of market pigs may not affect the carcass quality of the animals when an enough lairage time is provided. However, rough handling inflicts a stimulated stress on the animal, which is manifested by increased blood concentrations of stress indicators, and therefore should be avoided fer animal welfare.

Effects of ${\alpha}_1-Adrenergic$ Stimulation on Membrane Potential, Twitch Force, Intracellular $Na^+,\;and\;H^+$ Activity in Hyperthyroid Guinea Pig Ventricular Muscle (갑상선 기능 항진 기니픽 심근에서 ${\alpha}_1-Adrenergic$ 수용체 자극이 막전위, 수축력 및 세포내 $Na^+$$H^+$ 활성도에 미치는 영향)

  • The roles of ${\beta}-adrenoceptor$ were well known in hyperthyroidal heart, but not with ${\alpha}-adrenoceptor$. So we studied the effects of phenylephrine on membrane potential, intracellular sodium activity ($a^{i}_{Na}$), twitch force, and intracellular pH ($pH_i$) by continuous intracellular recordings with ion-selective and conventional microelectrodes in the papillary muscles of hyperthyroid guinea pig heart. ${\alpha}_1-adrenoceptor$ stimulation by phenylephrine (10^{-5}\;or\;3{\times}10^{-5}M$) produced the following changes: variable changes in action potential duration, a hyperpolarization ($1.5{\pm}0.1mM$) of the diastolic membrane potential, an increase in $a^{i}_{Na}\;(0.4{\pm}0.15mM)$, a stronger positive inotropic effect ($220{\pm}15%$), an increase in $pH_i\;(0.06{\pm}0.002\;unit)$. These changes were flocked by prazosin and atenolol. This indicated that the changes in membrane potential, $a^{i}_{Na}$ twitch force, and $pH_i$ are mediated by a stimulation of the ${\alpha}_1-adrenoceptor$. Ethylisopropylamiloride ($10^{-5}$) also blocked the increase in $a^{i}_{Na}$ and twitch force. On the other hand, strophanthidin, tetrodotoxin, $Cs^+$ or verapamil did not block the increase in $a^{i}_{Na}$ and twitch force. Thus, it was suggested that ${\alpha}_1-adrenoceptor$ stimulation increased $a^{i}_{Na}\;and\;pH_i$ by stimulation of $Na^{+}-H^{+}$ exchange, thereby allowing intracellular alkalinization and $a^{i}_{Na}$ increase. These results were very different from euthyroidal heart which showed ${\alpha}_1-adrenoceptor$-induced decrease in $a^{i}_{Na}$ and initial negative inotropic effect. From the above results, it was concluded that ${\alpha}_1-adrenoceptor$ had a important role in hyperthy-roidal heart.

Effects of Electrolytes and Drugs on the Inhibitory Junction Potentials Recorded from the Antrum of Guinea-pig Stomach (기니피그 유문동에서 기록되는 억제성 접합부 전압에 미치는 전해질과 약물의 효과)

  • The effects of electrolytes, adenosine, ATP, 5-hydroxytryptamine (5-HT, serotonin) and ketanserin on the inhibitory junction potentials (IJPs) were investigated to clarify the interactions of these drugs with the neurotransmitters released from non-adrenergic, non-cholinergic nerves in the antrum of guinea-pig stomach. Electrical responses of antral circular muscle cells were recorded intracellularly using glass capillary microelectrode filled with 3 M KCI. All experiments were performed in Tris-buffered Tyrode soluition which was aerated with 100% $O_{2}$ and kept at $35^{\circ}C$. The results obtained were as follows: 1) Inhibitory junction potential (IJP) was recorded in antral strip, while excitatory junction potential (EJP) was recorded in fundic strip. 2) IJP recorded in antral strip was not influenced by atropine $(10^{-6}\;M)$ and guanethidine $(5{\times}10^{-6})$. 3) The amplitude of IJP increased in high $Ca^{2+}$ solution, while that of IJP decreased in high $Mg^{2+}$ solution or by $Ca^{2+}$ antagonist (verapamil). Apamin, $Ca^{2+}$-activated $K^{+}$ channel blocker blocked IJP completely. 4) ATP and adenosine decreased the amplitude of IJP. 5) 5-HT decreased the amplitude of IJP with no change of the amplitude of slow waves, while ketanserin (5-HT type 2 blocker) decreased the amplitude of slow waves markedly with no change in that of IJP. From the above results, the following conclusions could be made. 1) IJP recorded in antral strip is resulted from neurotransmitters released from non-adrenergic, non-cholinergic nerves. 2) An increase in the concentration of external $Ca^{2+}$ enhances the release of neurotransmitters from non-adrenergic, non-cholinergic nerves which activate the $Ca^{2+}$-dependent $K^{+}$ channel.

Effects of Dietary Ginseng By-product on Growth Performance and Pork Quality Parameters in Finishing Pigs (인삼부산물 급여에 따른 비육돈의 생산성 및 육질 특성에 미치는 영향)

  • The objective of the present study was to investigate the effect of dietary supplementation of the ginseng by-product on growth performance and meat quality in finishing pigs. The animals used in the experiment were a total of 24 Landrace×Yorkshire and weighted 65.81±2.02kg. The experimental diets were basal diet(CON) and 2.5% ginseng by-product(GBP), which replaced lupin in basal diet. The pigs were allotted at 4 pigs per pen with three replicate pens per treatment by completely randomized design. In growth performance, ADFI was significantly lower(P<0.0001) in GBP than in CON. In plasma biochemical composition, total protein(P<0.01), blood urea nitrogen(P<0.03), glucose(P<0.01), albumin(P<0.02), calcium(P<0.01) and inorganic phosphate(P<0.01) were significantly higher in GBP than in CON. Carcass and meat quality were not significantly different between treatments. Total ginsenoside content on meat was significantly higher(P<0.0001) in GBP than in CON. TBARs was significantly lower in GBP than in CON for 6 days(P<0.03) and 12 days (P<0.06), respectively. Our research indicates that plasma biochemical composition, total ginsenoside content and TBARs were affected when replaced with ginseng by-product. Ginseng by-product in the pig diet increased pig muscle ginsenoside, indicating that ginseng by-product can be used as a feed additive.

  • Stress-susceptible pigs have been known as the porcine stress syndrome (PSS), swine PSS, also known as malignant hyperthermia (MH), is characterized as sudden death and production of poor meat quality such as PSE (pale, soft and exudative) meat after slaughtering. PSS and PSE meat cause major economic losses in the pig industry. A point mutation in the gene coding for the ryanodine receptor (RYR1) in porcine skeletal muscle, also known calcium (Ca$^{2+}$) release channel, has been associated with swine PSS and halothane sensitivity. We used the PCR-RFLP(restriction fragment length polymorphism) and PCR-SSCP (single strand conformation polymorphism) methods to detect the PSS gene mutation (C1843T) in the RYR1 gene and to estimate genotype frequencies of PSS gene in Korean pig breed populations. In PCR-RFLP and SSCP analyses, three genotypes of homozygous normal (N/M), heterozygous carrier (N/n) and homozygous recessive mutant (n/n) were detected using agarose or polyacrylamide gel electrophoresis, respectively. The proportions of normal, carrier and PSS pigs were 57.1, 35.7 and 7.1% for Landrace, 82.5, 15.8 and 1.7% far L. Yorkshire, 95.2, 4.8 and 0.0% for Duroc and 72.0, 22.7 and 5.3% for Crossbreed. Consequently, DNA-based diagnosis for the identification of stress-susceptible pigs of PSS and pigs producing PSE meat is a powerful technique. Especially, PCR-SSCP method may be useful as a rapid, sensitive and inexpensive test for the large-scale screening of PSS genotypes and pigs with PSE meat in the pork industry.y.

'OFF' Response and Its Characteristics of Guinea Pig Ureter (기니픽 요관(尿管)에 있어서 OFF Response 발생과 그 특징)

  • The in vitro guinea pig ureter responded to 5 sec trains of electrical stimuli with two contractions; the first an 'on response' (ON) occurred with $0.1{\sim}0.3$ sec after the onset o the stimulus train, the second an 'off response'(OFF) occurred $0.2{\sim}1.0$ sec after the termination of the stimulus train. Relaxation occurred between the two responses during a time when the stimulus was still being delivered. Longer duration and/or higher frequencies of stimuli within the train were required to elicit the OFF than the ON. Decreasing temperature from $37^{\circ}$ to $22^{\circ}$ decreased ON amplitude and increased OFF amplitude. $Ca^{++}$-free solution, 2 mM EDTA, 1 mM $Mn^{++}$ or $1{\mu}M$ verapamil rapidly abolished ON. OFF persisted when ON had disappeared by repeated stimulation at 0.12 train per sec. Conversely, caffeine, $50{\mu}M$ and theophylline, $10{\mu}M$ abolished OFF with only slight reduction of ON, and sodium nitroprusside decreased preferentially ON amplitude rather than OFF. Relaxation between ON and OFF was incomplete in low $Na^+$ solution. ON and OFF were not affected by the neural blockers tetrodotoxin, atropine or phentolamine, also pyrilamine and methysergide, and relaxation between ON and OFF was $Na^+$ dependent. Furthermore, ON depends on free $Ca^{++}$ and OFF is more dependent on bound or stored $Ca^{++}$.

Characterization of Phosphoinositide-3-kinase, Class 3 (PIK3C3) Gene and Association Tests with Quantitative Traits in Pigs

  • This study deals with the characterization of porcine PIK3C3 and association tests with quantitative traits. PIK3C3 belongs to the class 3 PI3Ks that participate in the regulation of hepatic glucose output, glycogen synthase, and antilipolysis in typical insulin target cells such as those in the such as liver, muscle system, and fat. On the analysis of full-length mRNA sequence, the length of the PIK3C3 CDS was recorded as 2,664 bps. As well, nucleotide and amino acid identities between human and pig subjects were 92% and 99%, respectively. Five SNPs were detected over 5 exons. We performed genotyping by using a SNP C2604T on exon24 for 145 F$_2$ animals (from a cross between Korean native boars and Landrace sows) by PCR-RFLP analysis with Hpy8I used to investigate the relationship between growth and fat depot traits. In the total association analysis, which doesn' consider transmission disequilibrium, the SNP showed a significant effect (p<0.05) on body weight and carcass fat at 30 weeks of age as well as a highly significant effect (p<0.01) on back fat. In an additional sib-pair analysis, C allele still showed positive and significant effects (p<0.05) on back fat thickness and carcass fat. Moreover, the effects of C allele on the means of within-family components for carcass fat and back fat were estimated as 2.76 kg and 5.07 mm, respectively. As a result, the SNP of porcine PIK3C3 discovered in this study could be utilized as a possible genetic marker for the selection of pigs that possess low levels of back fat and carcass fat at the slaughter weight.

Effects of wheat supplementation levels on growth performance, blood profiles, nutrient digestibility, and pork quality in growing-finishing pigs

  • Objective: This study was conducted to evaluate various wheat supplementation levels on growth performance, blood profiles, nutrient digestibility, and pork quality in growing-finishing pigs. Methods: A total of 120 growing pigs ($[Yorkshire{\times}Landrace]{\times}Duroc$), with an average $27.75{\pm}1.319kg$ body weight, were used in growth trial. Pigs were allotted into each treatment by body weight and sex in 4 replicates with 6 pigs per pen in a randomized complete block design. Fourphase feeding programs were used in this experiment. The treatments included the following: i) corn-soybean meal (SBM) - based diet (CON), ii) corn-SBM - based diet+15% of wheat (W15), iii) corn-SBM - based diet+30% of wheat (W30), iv) corn-SBM - based diet+45% of wheat (W45), and 5) corn-SBM-based diet+60% of wheat (W60). Results: There was no significant difference in growth performance among the dietary treatments. However, the gain-to-feed (G:F) ratio tended to increase (quadratic, p<0.08) when the pigs were fed a higher wheat diet during the finishing period. The digestibility of crude ash and fat tended to decrease as the wheat supplementation level increased (p<0.08). The proximate analysis of the longissimus muscle was not affected by the dietary level of wheat. The crude ash content in pork was decreased linearly as the wheat supplementation level increased (p = 0.05). There was no significant difference in the pH level, shear force, water holding capacity, and cooking loss of the pork. In pork and fat, $L^{\star}$, $a^{\star}$, and $b^{\star}$ values were not significantly different among dietary treatments. Conclusion: Wheat can be supplemented up to 60% in a growing-finishing pig without detrimental effects on growth and pork quality. The G:F ratio tended to improve in the finishing period by wheat inclusion.

Comparative Feeding Values of Soybean Hulls and Wheat Bran for Growing and Finishing Swine

  • Feeding values of soybean hulls (SH) were compared to those of wheat bran (WB) for swine diets by chemical compositions, a digestion trial, a preference test by self-selection, and two feeding trials. The SH and the WB appeared to have, on airdry basis, 11.1 vs. 15.4% CP, 32.5 vs. 8.7% crude fiber (CF), 36.8 vs. 10.7% ADF, 0.6 vs. 0.1% Ca, and 492 vs. 92 ppm Fe, respectively. Lysine and total sulfur-containing amino acids in the SH were 0.66 vs. 0.37%, respectively. Apparent digestibility values of the SH were 71% for dry matter, 50% for CP, and 74% for CF. Apparent digestible energy and MEn values of the SH were 2,420 and 2,370 kcal kg$^-1$, respectively, which were comparable to those of the WB, 2,420 and 2,275 kcal kg$^-1$ (NRC, 1998), respectively. The first feeding trial was conducted with 72 crossbred growing pigs with an average weight of 29.6 kg. The pigs when fed the diets containing 0, 6 and 10% SH by replacing the WB on a weight basis for 42 days did not show significant differences in body weight gain and feed/gain ratio among the treatments. The same trends were observed in the second trial with 60 crossbred finishing pigs with an average weight of 64.5 kg when fed the diets containing 12% SH or WB for 41 days. Back-fat thickness and adjusted loin eye muscle area of the finisher pigs were also not significantly different between the two groups. When allowed to self-select from two different feed troughs containing 10% SH or WB for two weeks, two groups of 80 pigs with 10 pigs per pen consumed the two diets exactly in equal proportion. In conclusion, the soybean hulls can be included up to 10 and 12% for growing or finishing pig diets, respectively, replacing the wheat bran on a weight basis without any adverse effects on palatability of diets and animal performances.

Quality characteristics, fatty acid profiles, flavor compounds and eating quality of cull sow meat in comparison with commercial pork

  • Objective: Although the slaughter of cull sows (CS) for human consumption and meat products processing appears quite common throughout the world, relatively limited scientific information regarding the meat quality parameters of this pork type is available. The present study aimed at providing the technological quality characteristics and eating quality of CS meat, and comparing with those of commercial pork. Methods: Longissimus thoracis et lumborum muscle samples of CS and finisher pigs (FP) at 24 h postmortem were collected and used for investigation of the meat quality traits (pH, color, shear force, cooking loss, water holding capacity), fatty acids, flavor compounds and sensory characteristics. Results: The CS meat had significantly higher moisture content (p = 0.0312) and water holding capacity (p = 0.0213) together with lower cooking loss (p = 0.0366) compared to the FP meat. The CS meat also exhibited higher (p = 0.0409) contents of unsaturated fatty acids, especially polyunsaturated fatty acids (PUFA, p = 0.0213) and more desirable PUFA/total saturated fatty acids ratio (p = 0.0438) compared to the FP meat. A total of 56 flavor compounds were identified, amongst the amount of 16 compounds differed significantly between the two pork groups. Most of the PUFA-derived flavor compounds (e.g., hexanal, benzaldehyde, and hydrocarbons) showed higher amounts in the CS meat. While, 3-(methylthio)-propanal and 4-methylthiazole associated with pleasant aromas (meaty and roast odor notes) were only found in the FP meat. Furthermore, no differences were reported by panelists for flavor, juiciness, tenderness, and acceptability scores between the two pork groups studied. Conclusion: The sow meat exhibited better technological quality and its eating quality could be comparable to the commercial pork. This study provides meat processors and traders with valuably scientific information which may help to improve the utilization and consumption level of sow meat.
