사람 및 가축 유래 분변 미생물 군집과 항생제 내성 유전자 간 상관 관계에 대한 연구 (Co-occurrence Analyses of Antibiotic Resistance Genes and Microbial Community in Human and Livestock Animal Feces)

  • BACKGROUND: Antibiotics used in animal husbandry for disease prevention and treatment have resulted in the rapid progression of antibiotic resistant bacteria which can be introduced into the environment through livestock feces/manure, disseminating antibiotic resistant genes (ARGs). In this study, fecal samples were collected from the livestock farms located in Jeju Island to investigate the relationship between microbial communities and ARGs. METHODS AND RESULTS: Illumina MiSeq sequencing was applied to characterize microbial communities within each fecal sample. Using quantitative PCR (qPCR), ten ARGs encoding tetracycline resistance (tetB, tetM), sulfonamide resistance (sul1, sul2), fluoroquinolone resistance (qnrD, qnrS), fluoroquinolone and aminoglycoside resistance (aac(6')-Ib), beta-lactam resistance (blaTEM, blaCTX-M), macrolide resistance (ermC), a class 1 integronsintegrase gene (intI1), and a class 2 integrons-integrase gene (intI2) were quantified. The results showed that Firmicutes and Bacteroidetes were dominant in human, cow, horse, and pig groups, while Firmicutes and Actinobacteria were dominant in chicken group. Among ARGs, tetM was detected with the highest number of copies, followed by sul1 and sul2. Most of the genera belonging to Firmicutes showed positive correlations with ARGs and integron genes. There were 97, 34, 31, 25, and 22 genera in chicken, cow, pig, human, and horse respectively which showed positive correlations with ARGs and integron genes. In network analysis, we identified diversity of microbial communities which correlated with ARGs and integron genes. CONCLUSION(S): In this study, antibiotic resistance patterns in human and livestock fecal samples were identified. The abundance of ARGs and integron genes detected in the samples were associated with the amount of antibiotics commonly used for human and livestocks. We found diverse microbial communities associated with antibiotics resistance genes in different hosts, suggesting that antibiotics resistance can disseminate across environments through various routes. Identifying the routes of ARG dissemination in the environment would be the first step to overcome the challenge of antibiotic resistance in the future.

Digestibility of nitrogen and dry matter of oilseed meals and distillers dried grains supplemented in swine diets

  • This study was conducted to investigate the digestibility of dry matter (DM) and nitrogen (N) in oilseed meals and distillers dried grains (DDG) fed to growing-finishing pigs. As experimental animals, eleven barrows (initial body weight, $71.7{\pm}17.0kg$) were housed in individual metabolism cages. The experimental design consisted of an $11{\times}8$ incomplete Latin square with 11 dietary treatments and 8 replication periods. The diets were individually formulated with dehulled soybean meal produced in Korea (SBM-KD), soybean meal produced in India (SBM-I), soybean meal produced in Korea (SBM-K), corn high-protein distiller dried grains (HPDDG), tapioca distillers dried grains (TDDG), canola meal (CAM), corn germ meal (CGM), copra meal (COM), palm kernel meal (PKM), sesame meal (SM), and perilla meal (PM). Pigs with SBM-KD and SBM-K showed greater (p < 0.05) intake of N than SBM-I, HPDDG, and PKM. Total feces output was decreased (p < 0.05) in SBMs (SBM-KD, -I, and -K), HPDDG, and CGM compared with TDDG, SM, and PM. The DM in excreted feces was decreased (p < 0.05) in SBMs and CGM compared to TDDG, SM, and PM. Similarly, the SM and PM fed to pigs resulted in greater (p < 0.05) fecal excretion of N than the others. Apparent total tract digestibility (ATTD) of DM in SBMs and CGM was greater (p < 0.05) than TDDG, SM, and PM. The SBMs fed to pigs showed higher (p < 0.05) ATTD of N than TDDG, COM, SM, and PM. In conclusion, our results provided nutritional information about various ingredients and would be useful to contain more precise amounts of nutrients included in feed ingredients of pig diet.

Genomic Approaches for Understanding the Characteristics of Salmonella enterica subsp. enterica Serovar Typhimurium ST1120, Isolated from Swine Feces in Korea

  • Salmonella enterica subsp. enterica serovar Typhimurium, one of the most common foodborne pathogens, is transmitted mainly through contaminated food derived from infected animals. In this study, S. Typhimurium ST1120, an isolate from pig feces in Korea, was subjected to whole-genome analysis to understand its genomic features associated with virulence. The genome of ST1120 was found to have a circular chromosome of 4,855,001 bp (GC content 52.2%) and a plasmid of 6,863 bp (GC content 46.0%). This chromosome was predicted to have 4,558 open reading frames (ORFs), 17 pseudogenes, 22 rRNA genes, and 86 tRNA genes. Its plasmid was predicted to have three ORFs. Comparative genome analysis revealed that ST1120 was phylogenetically close to S. Typhimurium U288, a critical isolate in piggery farms and food chains in Europe. In silico functional analysis predicted that the ST1120 genome harbored multiple genes associated with virulence and stress resistance, including Salmonella pathogenicity islands (SPIs containing SPI-1 to SPI-5, SPI-13, and SPI-14), C63PI locus, ST104 prophage locus, and various antibiotic resistance genes. In accordance with these analysis results, ST1120 showed competence in invasion and survival abilities when it was added to host cells. It also exhibited robust resistance against antibiotics in comparison with other S. Typhimurium strains. This is the first report of the complete genome sequence of S. Typhimurium isolated from swine in Korea. Comparative genome analysis between ST1120 and other Salmonella strains would provide fruitful information toward understanding Salmonella host specificity and developing control measures against S. Typhimurium infection.

  • The purpose of this study was to investigate to potective effects against porcine epidemic diarrhea virus (PEDV) infection in piglets by administration of the PEDV antiserum orally at 2 hrs, 24hrs and 36hrs after birth. six piglets administered the antiserum were experimentally infected with PEDV at five-day-old. Control group were four piglets infected with PEDV only. Serum antibody titers against PEDV were examined by serum neutralization (SN) test, dectection for PEDV or PEDV antigen from feces and small intestines was tested by reverse transcription-polymerase chain reaction (RT-PCR) and indirect immunoflurescence (IFA). The results obtained were as follows; 1. The piglets administered the PEDV antiserum showed higher antibody titers than those of control group and sustained during the experimental period. 2. The detection rate of PEDV in feces and small intestines by RT-PCR were 26.2% and 16.7% in PEDV antiserum treated group and 48.1 % and 75.0% in control group, respectively. 3. The detection rate of PEDV antigen in the small intestine by IFA were 0% in PEDV antiserum treated group and 50.0% in control group, respectively. It was concluded that oral administration of antiserum against PEDV to piglets was effective in preventing PEDV infection.

  • 산란계의 분변으로부터 생균제 특성을 가진 유산균을 분리하기 위하여, 무작위 선발법과 agar well diffusion assay법을 사용하여 다수의 유산균을 1차 선발하였으며 이 중에서 대장균 억제능이 가장 우수한 CPM-7 균주를 분리하였다. 최종 선발된 CPM-7 균주는 형태학적 특징, 당 이용성 및 16S rRNA 서별 분석을 통하여 Lactobacillus salivarius CPM-7으로 동정되었다. L. salivarius CPM-7은 내산성 및 내담즙성이 우수한 것으로 나타났는데, pH 2에서 30분 동안 및 pH 3에서 6시간 동안 생균수가 거의 유지되었으며, bile salt 0.2%가 첨가된 MRS 배지에서 증식할 수 있었다. L. salivarius CPM-7은 a-galactosidase를 포함한 다수의 효소를 생산하는 것으로 확인되었으며, 돼지의 장 상피세포에 흡착할 수 있었다. L. salivarius CPM-7의 배양액 및 중화액은 자돈 설사를 유발하는 것으로 알려진 E. coli K88에 대한 강력한 억제능을 나타내었다.

Analysis of Outer Membrane Proteins of Yersinia enterocolitica Isolated from Mountainspring Water and Pig

  • Yersinia enterocolitica causes various diseases in humans, including enteritis. The onset of such diseases is closely related with the expression of important virulence factors, particularly outer membrane proteins (OMPs). The expression of OMPs depends on several factors, including temperature, and origin, biotype and serotype of the bacteria. Recently, concerns over food safety have increased along with the demand for the development of sensitive, rapid, and pathogen-specific detection methods. To develop a suitable detection method for Y. enterocolitica isolated from Korean moutainspring water and pig feces, the OMP expression patterns were analyzed phenotypically and immunologically using 12 representative strains from 51 Y. enterocolitica Korean isolates. A 38-kDa OMP was commonly observed in all strains. However, additional OMPs were also observed in different biotypes and serotypes as well as bacterial origins, by incubating Y. enterocolitica at a low temperature. The specificity of the 38-kDa OMP was confirmed by a Western blot analysis with antisera against Y. enterocolitica and Brucella abortus. The results, therefore, indicate that the 38-kDa OMP could be used as a marker for detecting Y. enterocolitica in the environment or for seromonitoring.

국내(國內) 돼지의 Sarcocystis 감염과 Sarcocystis종의 동정에 관한 연구 (Sarcocystis infection and identification of Sarcocystis species in pigs in Korea)

  • Five hundred and forty-eight samples of pig heart muscle were collected from the abattoirs of many regions in Korea to reveal the frequency of Sarcocystis infections and to identify the species from June 1988 to April 1989. Heart muscle of the pigs was inspected for sarcocysts by the direct detection technique and for bradyzoites by the trypsin digestion technique. For examination of development of the parasites in the final host, 5 cross bred mature dogs, 5 puppies and 5 kittens were fed 100g, 50g and 50g of the infected meat respectively, four times in 2 days. Of 402 fattened and 146 older culled breeding pigs, 3 fattened pigs and 39 culled pigs were positive for Sarcocystis. Sarcocystis cysts from heart muscle measured an average of $425{\times}169{\mu}m$ and bradyzoites an average of $15.6{\times}3.5{\mu}m$. Of 15 animals, only 2 puppies were infected with Sarcocystis. The prepatent period was 11 to 12 days and patent period was not examined since the puppies were infected with some another infections and one died on day 11 and another died on day 12 after ingestion of the meat. The sporulated oocysts were detected 11 days after ingestion of the meat and sporocysts 12 days from the puppy feces. The sporulated oocysts measured an average of $16.5{\times}11.5{\mu}m$ and sporocysts an average of $12.6{\times}7.9{\mu}m$. On scraping examination of the intestinal mucosa, fully sporulated oocysts were detected in the tip of the intestinal villi. Considering above all descriptions, Sarcocystis in pig heart muscle in Korea was identified with Sarcocystis suicanis.

Exposure to low concentrations of mycotoxins triggers unique responses from the pig gut microbiome

  • The aim of this study is to investigate how the gut microbiome shifts when pigs were exposed with low concentrations of mycotoxins, deoxynivalenol (DON) and zearalenone (ZEN) in feed. Fifteen of pigs, 15 kg in weight which were negative for PRRSV and PCV2 were purchased, acclimatized until 20 kg in weight, and randomly divided into 3 groups; the DON group (DON treated), the ZEN group (ZEN treated) and the CTL (untreated negative control). DON and ZEN administered to each group for 30 days at 0.8 mg/kg (800 ppb) and 0.20 mg/kg (200 ppb) in feed, respectively. After extraction of microbial DNA from intestine and fecal samples, sequencing procedures were performed in the Ion PGM using an Ion 316 V2 chip and Ion PGM sequencing 400 kit. The results suggested that the bacterial communities in duodenum, jejunum and ileum of the DON and ZEN groups presented low-abundant OTUs compared with the CTL group. OTUs in cecum, colon and feces were determined more than in small intestine of all three groups. However, the CTL group yielded more OTUs than other two groups in inter-group comparison. It is not fully clarified how the richness and abundance in microbiome functions in the health condition of animals, however, the exposure to DON and ZEN has caused microbial population shifts representing microbial succession and changes following the diversity and abundance of porcine gut microbiome. The metabolomic analysis correlate with microbiome analysis is needed for further study.

Isolation and Identification of a Lactic Acid Bacterial Strain KJ-108 and Its Capability for Deodorizing Malodorous Gases Under Anaerobic Culture Conditions

  • A number of different sources, such as composts, leachates, and pig feces samples, were collected from different pig farms in Korea, and several microorganisms were screened for their ability to deodorize the malodorous gases. Consequently, a novel malodorous gases-deodorizing bacterial strain, KJ-108. was isolated, because it was highly abundant in nitrate-supplemented minimal medium ($MM-NO_3^-$) under anaerobic culture conditions. Airtight crimp-sealed serum bottles containing $MM-NO_3^-$ , medium were inoculated with KJ-108. Nitrate concentration was decreased rapidly after 20 h of incubation, and incubation was carried out until nitrite production reached almost zero. Taxonomic identification, including 16S rDNA base sequencing and phylogenetic analysis, indicated that the isolate had $100\%$ homology in its 165 rDNA base sequence with Lactobacillus pentosus. Among the volatile fatty acids, acetic acid contained in large amounts in fresh piggery slurry was decreased by about $40\%$ after 50 h incubation with strain KJ-108. n-Butyric acid, n-valeric acid, and isovaleric acid were gradually decreased, and isobutyric acid and capronic acid were dramatically eliminated at theinitial period with the treatment. Moreover, NH, removal efficiency reached a maximum of $98.5\%$ after 50 h of incubation, but the concentration of $H_2S$ was not changed.
